  • Pine nut oil (PNO) is well known to impart beneficial effects in overweight individuals, but the mechanisms underlying PNO-mediated weight loss remain unclear. To investigate how PNO promotes weight loss, its composition was determined by gas chromatography coupled with mass spectrometry (GC-MS). In addition, the effects of PNO on cytotoxicity, lipid accumulation, expression of lipid metabolism-related biomarkers, and leptin secretion were assessed in 3T3-L1 cells. GC-MS analyses revealed that PNO contains several components, including linoleic acid, oleic acid, palmitic acid, and stearic acid. Moreover, PNO did not have a cytotoxic effect on 3T3-L1 cells. However, it inhibited the expression of peroxisome proliferator-activated receptor (PPAR) and adipocyte protein 2 (aP2). Finally, PNO significantly increased leptin secretion in a dose-dependent manner. Taken together, these results support the notion that PNO is useful for weight management in overweight individuals.

  • This study investigated the anti-obesity effects of Eriobotrya japonica leaves extract (EJE) in a high-fat diet-induced obese mice model. For the analysis, 40 mice were randomly divided into five groups: normal diet (ND), high-fat diet group (HD), high-fat diet containing EJE 100 mg/kg b.w. (EJE-L), 500 mg/kg b.w. (EJE-H), and Garcinia cambogia extract 100 mg/kg b.w. in each AIN 93G supplement for 8 weeks as positive control. During the study period, food intake and body weight were measured separately. Body weight, serum triglyceride level, and total cholesterol levels of the EJE group were significantly lower than those of the HD and ND. Moreover, mice fed an EJE-containing high-fat diet showed decreased leptin levels compared with high-fat diet-induced obese mice. These results suggest that EJE has potential as an anti-obesity agent by reducing body weight and improving levels of triglycerides, total cholesterol, low density lipoprotein-cholesterol, and leptin in serum.

  • We investigated the effects of feeding diet containing medicinal plant water extracts (MPWEs) on body weight, epididymal adipose tissue weight, adipocyte size of epididymal adipose tissue and plasma lipid levels in high fat (HF) diet-induced obese mice. To test antiobese effects of diet containing the MPWEs, C57BL/6J mice were fed with HF diet for 11 weeks. In the last 6 weeks, the HF diet was supplemented with 0 (HFD) or MPWEs (5 g/kg, HFD+MPWEs) or orlistat [0.5 g/kg, HFD+orlistat (antiobesity drug)]. The HF-free diet group was fed normal chow for 11 weeks. Eleven-weeks feeding with HFD resulted in significant increase in lipid levels, body weight, liver and epididymal adipose tissue weights, compared with the HF-free group. Diet containing MPWEs significantly reduced plasma total cholesterol, LDL-cholesterol, triglyceride and glucose concentrations as well as body weight, liver weight and epididymal adipose tissue weight. Plasma triglyceride levels were significantly lower in the HFD+Forlistat group after 6 weeks and a similar effect was found with HFD+MPWEs group. The adipocyte size of epididymal adipose tissue in HFD group was significantly larger than those of HF-free group. MPWEs and orlistat (positive control) significantly decreased the size of epididymal adipocytes but orlistat was slightly more effective than MPWEs. These results suggest that oral feeding of the MPWEs may have antiobesity effects by suppressing body weight gain, adipose tissue formation and adipocyte size increase.

  • There is a rising trend in obesity due to various factors, including changes in eating habits, lack of exercise, and genetic and psychological factors. Citrus peel has been reported to prevent obesity via antioxidative, antihypertensive, and LDL cholesterol-lowering effects. This study investigated the effects of citrus peel extract fermented with or without Aspergillus oryzae in a mouse model of diet-induced obesity. The animals were divided into four groups: a high-fat diet group (HFD), a normal fat diet (NFD) group, a citrus peel extract (CP) group, and a citrus peel extract fermented with A. oryzae (CPA) group. The citrus peel extract improved lipid metabolism and weight loss in the high-fat diet-induced obese mouse model. As expected, the body weight was higher in the HFD group compared with the NFD, CP, and CPA groups. However, the concentrations of total cholesterol (TG) and triglyceride (TC) in the serum and liver of the CP and CPA groups were lower than in the HFD group. There were no significant differences in the HDL cholesterol concentration among the groups. Taken together, our results suggest that extract of citrus peel biotransformed with A. oryzae had more antiobesity activity than citrus peel not transformed by A. oryzae through the fermentation of metabolites.

  • Recently, there has been a marked increase in the use of bioactive products resulting from the fermentation of natural substances by microorganisms. In this study, Opuntia humifusa (OH) was fermented using Lactobacillus plantarum (fermented Opuntia humifusa; fOH). We then examined the anti-obesity effect of fOH in mice fed a 45% Kcal high fat diet (HFD). In this study, mice were treated with fOH concentrations of 100, 200, and 400 mg/kg. The mice in the control group were treated with OH at a concentration of 400 mg/kg based on previous animal experiments. All of the mice given a continuous HFD showed an increase in their weight, the density of abdominal fat, and the accumulated periovaric and abdominal fat. All of these obesity-linked factors, however, were significantly decreased in the groups treated with fOH at concentrations of 200 and 400 mg/kg. Mice treated with fOH at 100 mg/kg did not show a significant decrease in these obesity-linked factors compared to the control group. It appears that fOH fermented by L. plantarum has a greater anti-obesity effect in HFD-supplied mice compared to unfermented OH. While further studies of fOH are needed to examine its effect on obesity, hyperlipidemia, hepatic steatosis, renal function, and type II diabetes with its relevant complications, fOH may have significant therapeutic potential in the treatment of metabolic syndrome.

  • Obesity, the leading metabolic disease, is a prevalent health problem in industrialized countries and is closely associated with coronary heart disease, hypertension, diabetes, and even cancer. In this study, we investigated the effects of dip-sauce for meat containing Garcinia cambogia extract (GC) on the lipid accumulation and body weight reduction in rats fed high-fat diet for three months. Eighteen Sprague Dawley male rats of five-week-old were randomly assigned to one of three groups; normal chew diet (NCD) group, high-fat diet plus GC-noncontaining dip-sauce (HFD) group, and HFD plus GC-containing dip-sauce (HFD+GC) group. Co-administration of GC-containing dip-sauce (5 g/kg body weight/day) with HFD significantly attenuated cumulative body weight gain, compared with NCD or HFD groups. Both epididymal and perirenal fat pad weights in the HFD plus GC group were significantly lower than those of HFD or NCD groups. Administration of GC-containing dip-sauce also resulted in significant reduction in the serum levels of total cholesterol, total lipid, and triglyceride, compared with NCD or HFD groups. Thus, GC-containing dip-sauce confers beneficial effects to pathological states associated with metabolic disorder via its anti-obesity and lipid lowering properties.

  • This study tested the anti-obesity and hypolipidemic effects of red garlic extract in obese rats induced by a high fat diet over a period of 4 weeks. Red garlic extract of 15 brix was added in 1, 3, 5 and 7% ratios in diets. The obesity index and body fat content significantly decreased in rats fed a diet with over 3% red garlic extract compared to the control group. There was no significant difference in weight of visceral and epididymal fat in rats fed red garlic extract. Total lipid and triglyceride levels in serum were significantly decreased in a dose-dependent manner, and AI and CRF also fell. ALT and AST activities in groups fed red garlic extract were decreased compared to the control group. Total lipid level in liver tissue of the groups fed 5-7% red garlic extract exhibited a significant decrease compared to the control group. Total cholesterol and triglyceride levels in feces were significantly increased in rats fed a diet with over 5% red garlic extract. Lipid peroxide levels were significantly decreased in the groups fed diets with 5-7% red garlic and antioxidant activity in serum was significantly increased in the group fed a diet with 7% red garlic extract compared to the control group. Our results suggest that red garlic extract could have anti-obesity and hypolipidemic effects for suppressing obesity index and decreasing lipid profiles.
