Allium victorialis Leaf Extract Prevents High Fat Diet Induced Obesity in Mice (고지방식이로 유발된 비만마우스에서 산마늘 잎추출물의 항비만효과)

  • The antiobese effects of Allium victorialis (AV) leaf methanol extract were evaluated in a high fat diet (HFD) supplied mice. The changes on the body weight, food consumption, leptin and adiponectin levels as well as the periovarian fat weights and histopathology of adipocytes were examined. The effects were compared with those of a group given 250 mg/kg of metformin. After 91 days of a continuous HFD supply, the mice were significantly showed obesity. However, the obesity induced by the HFD was inhibited by the AV extract treatment at the three different doses (62.5, 125 and 250 mg/kg) respectively. The results suggest that the AV methanol extract is beneficial for improving the diet-induced obesity in humans.

  • 최근 식생활의 변화로 인한 동물성 포화지방, 고칼로리 식품의 섭취 증가와 운동 부족, 스트레스 등의 여러 요인으로 비만인구 비율이 증가 추세를 보이고 있다. 유자, 석류등 천연소재 추출액에는 플라보노이드 성분이 풍부하여 항암, 항산화, 항염증, 항미생물 활성 등 생리활성이 있는 것으로 보고된 바 있다. 발효유로 대표되는 유산균 제품은 장 건강, 항산화 등 다양한 장점을 지니고 있으나, 짧은 유통기간 및 냉장유통 등 보존성에 있어 취약점을 가지고 있다. 따라서 천연 추출물을 주원료로 Lactobacillus acidophilus로 발효한 천연소재발효물을 활용하여 항비만 효과에 관한 연구를 수행하였다. 유산균 발효유의 풍미 유지하면서 유통기간을 늘릴 수 있는 가공법을 개발하고 건강식품 소재 개발 및 산업화를 통하여 국민의 건강 증진을 도모하고자 본 연구를 수행하였다. 천연소재 프로바이오틱스 발효물을 제조하여 세포독성, NO생성 억제능, 항비만 활성, 관능검사를 수행한 결과는 다음과 같다. 천연소재 추출물 및 천연소재추출발효혼합물을 각각 10, 50, 100, $500{\mu}g/mL$의 농도로 처리한 시험구들의 세포생존율을 측정한 결과 가장 고농도인 $500{\mu}g/mL$의 농도에서 각각 96.2, 95.1, 97.3, 96.2, 98.1, 97.0 %로 확인되어, 높은 수준의 안전성이 확인되었다. 천연소재 추출물 및 천연소재추출발효혼합물의 항염증효과를 분석하고자 수행한 Nitrite 생성저해 효과를 분석한 결과, NO 생성율은 $500{\mu}g/mL$의 농도에서 각각 98.1, 98.2, 96.2, 97.3, 91.2, 95.3 %로 확인되어 시험구 D의 항염증 활성이 가장 높은 것으로 나타났다. 동일한 농도로 처리하였을 때 lipid accumulation은 각각 100.0, 99.1, 98.2, 98.5, 87.6, 91.8 %로 나타나 D시험구와 E시험구에서 지방세포 분화억제효과가 확인되었다. 관능검사 시행 결과, 농축비율에 따른 관능적 기호도는 5 brix > 3 brix > 1 brix > 10 brix 순으로 나타났다.

The Anti-obesity Effect of Aureobasidium pullulans SM-2001 Extract (Polycan®) on 3T3-L1 Preadipocytes and Adipocytes (3T3-L1세포에서 흑효모 SM-2001 추출물(Polycan®)의 항비만 효과)

  • Obesity, the world's leading metabolic disease, is a serious health problem in both industrialized and developing countries. Natural substances are of great interest in preventative medicine, especially in the field of metabolic syndromes-from insulin resistance to obesity and diabetes. In the present study, we investigated the effect of A. pullulans SM-2001 Extract (Polycan®) on the adipocyte differentiation of 3T3-L1 preadipocytes and the anti-obesity effect of 3T3-L1 adipocytes. Although β-glucan has been found to have health benefits in the regulation of the immune system and blood cholesterol levels, its role in obesity has not been fully investigated. Polycan® suppressed lipid accumulation and glycerol-3-phosphate dehydrogenase (GPDH) activity without affecting cell viability in 3T3-L1 preadipocytes and adipocytes. Polycan® also inhibited cellular lipid accumulation through down-regulation of transcription factors, such as PPARγ and C/EBPα, and induced dose-dependent phosphorylation of AMP-activated protein kinase (AMPK)-a cellular energy sensor-while the total AMPK protein content remained unchanged. Taken together, this shows that the activation of AMPK by Polycan® in adipocytes plays a critical role in Polycan®-induced inhibition of adipocyte differentiation. Our results show that Polycan® has an anti-obesity action in vitro, suggesting a potential novel preventative agent for obesity and other metabolic diseases.

Effects of fermented blueberry liquid in high-fat diet-induced obese C57BL/6J mice (블루베리발효액이 식이유도 비만 Mice의 비만에 미치는 효과)

  • Purpose: The objective of the present study was to determine whether fermentation can increase the protective effects of blueberry liquid in a high-fat diet-induced obese mice model. Methods: Male C57BL/6J mice were fed a high-fat diet (HD, 60% fat, w/w,), HD supplemented with 10 ml/kg BW/day of blueberry liquid (BHD, blueberry high-fat diet), or HD supplemented with 10 ml/kg BW/day of fermented blueberry liquid (FBHD, fermented blueberry high-fat diet) for 10 weeks. Results: There were significant decreases in the body, epididymal adipose tissue, and liver weights of blueberry-fed groups compared to HD, whereas there were no significant differences in food intake among the groups. Furthermore, blueberry liquid groups, especially fermented blueberry liquid, significantly attenuated the contents of hepatic triglycerides and total cholesterol induced by HD. Serum LDL-cholesterol was significantly lower in the BHD and FBHD-fed groups, whereas FBHD significantly increased the serum HDL-cholesterol level compared to the control. Concentrations of aspartate transaminase, alanine transaminase, and leptins in serum were also reduced by blueberry liquid supplementation. The mRNA expression of hepatic acetyl CoA carboxylase was significantly reduced in both the BHD and FBHD groups compared to HD. Furthermore, FBHD altered the mRNA expression level of hepatic lipolysis genes. Conclusion: In conclusion, these results suggest that blueberry, especially fermented blueberry liquid, may improve obesity-related abnormalities.

Anti-Obesitic Effects of Cathepsin S Inhibitory Fraction Derived from Paecilomyces tenuipes in Mice Fed a High-Fat Diet (고지방식이를 섭취한 쥐에서 눈꽃동충하초 유래 cathepsin S 저해 분획의 비만 억제 효과)

  • In this study, cathepsin S (CTSS) inhibitory fraction was isolated from Paecilomyces tenuipes and anti-obesitic effects of the fraction were evaluated in mice, fed a high-fat diet. Hot water extract of P. tenuipes (DHW) was divided into 2 fractions, water eluate fraction (DHP1) and methanol eluate fraction (DHP2) using Diaion HP-20. $IC_{50}$ values for DHW, DHP1 and DHP2 against CTSS were 108.7, 890.3 and 2.3 ${\mu}g$/mL, respectively. To evaluate anti-obesitic effects of the fractions, each fraction was administrated orally to C57BL/6 mice for 4 weeks with a high-fat diet. DHP2 had the highest inhibitory effect on CTSS activity, causing serious reduction in weight gain, a reduction in the amount of adipose tissue and in serum lipids levels. These results suggest that the inhibition of CTSS by compounds derived from P. tenuipes may be effective in preventing and in ameliorating obesity.

Hangbisan, Sulfur-based Oriental Medicine, Lowers the Blood Cholesterol Level of ob/ob Obese Mice (유전성 비만 마우스에 대한 항비산의 혈중 콜레스테롤 저하 효과)

  • Among oriental medicine, sulfur is known to generate heat in the human body. Since body warming reaction results in the consumption of energy source, the medicines containing sulfur could help in the weight loss of laboratory rats. This study was designed to determine the possible weight loss effects of Hangbisan, sulfur based oriental medicine, on ob/ob mice. The obese mice were fed with standard diet containing 10% (w/w) Hangbisan or 10% (w/w) cellulose during 12 weeks. Hangbisan affected the weight loss of obese mice as cellulose did during experimental periods, while also reducing the level of plasma total cholesterol. These results suggest that dietary Hangbisan improved the composition of blood profiles in obese mice, and therefore has potential as an anti-obesity ingredient in the application of oriental medicine compounds.

Combination of berberine and silibinin improves lipid metabolism and anti-obesity efficacy in high-fat diet-fed obese mice (고지방식이로 유도한 비만마우스에서 berberine과 silibinin 복합투여를 통한 지질대사 개선과 항비만 효능 증진)

  • In this study, we investigated whether the combined administration of berberine (BBR) and silibinin (SBN) was effective in improving hyperlipidemia and anti-obesity efficacy using a high-fat diet (HFD)-fed obese mouse model. HFD-induced obese mice were supplemented with the BBR and SBN combination (BBR-SBN) along with the HFD administration for 8 weeks. During the experiment, body weight, food intake, and levels of total cholesterol, triglyceride and high-density lipoprotein (HDL)-cholesterol were analyzed. Consumption of HFD in the mice caused rapid increases in body weight and the levels of total cholesterol and triglycerides compared to the normal control (NC) group. However, supplementation of BBR-SBN in these obese mice significantly reduced body weight gain and suppressed the levels of total cholesterol and triglyceride with the increment of HDL cholesterol level. In the HFD-fed group, abdominal fat weight was significantly increased and the adipocytes within the epididymal adipose tissue were found to have expanded sizes compared to the NC group. However, in the BBR-SBN group, the sizes of the adipocytes were comparable to those of the NC group and abdominal fat weight was significantly reduced. Moreover, the deposition of giant vesicular fat cells in liver tissues seen in the HFD-fed group was considerably reduced in the BBR-SBN group. These results suggest that the BBR-SBN combination tends to have synergic potential as an anti-obesity agent by significantly reducing body weight gain as well as lowering serum lipid levels and thus improving anti-obesity efficacy in HFD-induced obese mice.

Anti-Oxidative and Anti-Obesity Activities of Tetrapanax papyriferus and Siegesbeckia pubescens Extracts and their Synergistic Anti-Obesity Effects (통초.희렴 추출물의 항산화.항비만 활성 및 혼합물의 항비만 시너지 효과)

  • In this study, the anti-oxidative and anti-obesity activities of two medicinal herb extracts, Tetrapanax papyriferus (TP) and Siegesbeckia pubescens (SP), were evaluated using DPPH radical scavenging activity assay, lipase enzyme inhibition assay, and the cell culture model system. Both methanol extracts of TP and SP showed DPPH radical scavenging activities dose-dependently, and the $IC_{50}$ of DPPH radical scavenging activities of the two medicinal herbs were 65.23 and 47.79 ${\mu}g/ml$, respectively. Furthermore, both extracts suppressed effectively lipase enzyme activity dose-dependently. Moreover, TP and SP extracts significantly suppressed adipocyte differentiation, lipid accumulation, triglyceride (TG) contents on 3T3-L1 preadipocytes in a dose-dependent manner without cytotoxicity. Their anti-obesity effect was modulated by cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$ and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) gene and protein expressions. Furthermore, TP and SP possessed a synergistic effect on anti-obesity activity. The identification of the active compounds that confer the anti-obesity activity of TP and SP might be needed.

Anti-obesity and Anti-diabetes Effects of the Fermented White Jelly Fungus (Tremella fuciformis Berk) Using Lactobacillus rhamnosus BHN-LAB 76 (유산균을 이용한 흰목이 버섯의 발효를 통한 항비만과 항당뇨 효과)

  • White jelly fungus (Tremella fuciformis Berk) is effective for hypertension, diabetes, obesity, lung disease, beauty, and has been known as an elixir of life in ancient Chinese dynasty. In this study, we investigated the anti-obesity and diabetic effects of the fermented white jelly fungus extract by the bioconversion process. We fed an obesity-inducing mouse with 5% non-fermented Tremella fuciformis (TF), 2.5% fermented Tremella fuciformis (FTF), and 5% FTF containing High Fats Diet (HFD) and HFD for 8 weeks, respectively. The oral glucose tolerance test (OGTT) was performed analysis after 7 weeks of feeding and the dietary intake, food efficiency ratio, body weight, liver, epididymal fat weight, and serum insulin level were measured after 8 weeks of feeding. Also, HOMA-IR was analyzed. The concentrations of serum total cholesterol, triglycerides test was analyzed. The FTF compare with 5% TF and HFD confirmed that 5% FTF reduced body weight, tissue weight, triglycerides concentration, HOMA-IR, respectively. As a result, we confirmed that the fermented white jelly fungus has the anti-obesity effect. Finally, this study can be used a basic data for obesity treatment using fermented white jelly fungus.

Anti-obesity Effects of Sparassis crispa on High-fat Diet-induced Obese Mice (고지방식이로 유도한 비만 흰쥐에 대한 꽃송이 버섯의 항비만 효과)

  • The present study investigated the anti-obesity effects of Sparassis crispa (SC) on mice fed a high-fat (HF, 45 kcal% fat) diet. Mice were fed either a normal control diet and an HF diet or an HF diet supplemented with SC (1%, 3%, and 5%) for 12 weeks. The consumption of an HF diet compared to the NC group resulted in increases in body weight, the food efficiency ratio (FER), retroperitoneal and subcutaneous fat weights, cholesterol and triglyceride levels, fecal fat, and liver lipids. However, the administration of SC significantly decreased body weight gain, food intake, FER, cholesterol and triglyceride levels, and liver lipids in a dose-dependent manner. In particular, treatment with 5% SC significantly reduced the occurrence of fatty liver deposits and steatosis, which are associated with the increased adipocyte size in mice fed an HF diet. Therefore, these results suggested that dietary supplementation with SC exerts anti-obesity effects and could be used as a functional food to control obesity.
