Anti-Obesity Effect of Red Garlic Composites in Rats Fed a High Fat-Cholesterol Diet (고지방-콜레스테롤 식이성 흰쥐에서 홍마늘 복합물의 항비만 효과)

  • Three kinds of dietary composites-R+T, R+F, and R+TF-were combined in green tea (T), dietary fiber (F), and green tea dietary fiber mixture (TF) to red garlic extract (RG), respectively. The effects of their diets on anti-obesity were investigated $in$ $vitro$ and $in$ $vitro$ in obese rats induced high fat-cholesterol. In $in$ $vivo$ rats, the total phenolic content of the R+T and R+TF was 1.9~2.0 times higher, and their total cholesterol adsorption was 9.5~11.5 times higher than that of RG. $In$ $vivo$, male Sprague-Dawley rats were divided into 6 groups (Normal, HFC, HRG, HR+T, HR+F and HR+TF). Afterwards, the diets of the HRG, HR+T, HR+F, and HR+TF groups were supplemented with 1% of RG and its dietary composites (R+T, R+F, and R+TF) for 4 weeks, respectively. The final body weight of the HRG, HR+T, HR+F, and HR+TF groups decreased significantly compared to the group fed high fat-cholesterol (HFC), but the food efficiency ratio was not significantly different from the HFC group. The liver weight of the HFC group doubled compared to the normal group, whereas that of HR+T and HR+TF groups decreased significantly. The weight of visceral and epididymal fat decreased significantly in the groups fed the composites compared to the HFC group. The obesity index of HR+TF group decreased significantly only when compared to the HFC group. The serum lipid profile such as total lipids, cholesterol, triglyceride, LDL- and VLDL-cholesterol, as well as the atherogenic index and cardiac risk factors decreased drastically in all experimental groups compared to the HFC group, and the levels of HR+T, HR+F and HR+TF groups were a similar trend. GPT activity was not significantly different among the groups fed the composites, and it decreased significantly in the HRG group. The content of the lipid peroxide level decreased significantly in the HRG group and in the groups fed the composites, compared to the HFC group. Serum antioxidant activity was the highest in the HR+T group. We suggest that the hypolipidemic and anti-obesity effect of the RG composites, achieved by mixing green tea extract and/or dietary fiber, was due to their total phenolic content and total cholesterol adsorption effect.

Effects of Vitexin from Mung Bean on 3T3-L1 Adipocyte Differentiation and Regulation According to Adipocytokine Secretion (녹두의 Vitexin이 비만전구세포에서 세포분화 및 아디포사이토카인 분비능에 미치는 영향)

  • Obesity is an important issue worldwide as it may associated with increased prevalence of metabolic diseases. Mung bean is known as a functional food for decreasing the glycemic index and lipid profile of plasma. The purpose of this study was to investigate the anti-obesity effects of vitexin from mung bean on the regulation of adipocyte differentiation and adipocytokine secretion. When 3T3-L1 adipocytes were treated with vitexin from days 0 to 14 at various levels of 25, 50, 100, and $200{\mu}M$, there was no change in cell viability. Vitexin treatment at 50, 100, and $200{\mu}M$ decreased triacylglycerol levels in cells, but only $100{\mu}M$ vitexin induced lipolysis. At $200{\mu}M$ of vitexin, phosphorylation of p38 and ERK, which causes secretion of inflammatory adipocytokines, was depressed, whereas there was an increase in expression of $PPAR{\gamma}$, the key regulator of adipocyte differentiation. Phosphorylation of AMPK increased at $100{\mu}M$ vitexin. TNF-${\alpha}$ and aP2 mRNA expression increased at $25{\mu}M$ vitexin, whereas only TNF-${\alpha}$ mRNA expression increased at $200{\mu}M$ vitexin. Further, the mRNA levels of TNF-${\alpha}$ and aP2 decreased at other concentrations in a dose-dependent manner. Since we observed that mRNA expression of C/EBP, SREBP1, and $PPAR{\gamma}$ did not change upon vitexin treatment, our future studies will investigate other genes such as mTOR, which is related with apoptosis signaling, or SIRT1, which is associated with inhibition of adipogenesis. Our results indicate that vitexin at concentrations between 100 and $200{\mu}M$ is suitable in vivo for the development of mung bean as an anti-obesity therapy or functional food.

Inhibitory Effects of Ginseng Extracts on Histamine-release from Rat's Mast Cell (인삼추출물의 랫트 비만세포 히스타민 유리 억제 효과)

  • We investigated inhibitory effects of ginseng extracts against compound 48/80-induced responses in rat peritoneal mast cells. Initially, we optimized extraction condition with various temperature and time for recovery of high saponin contents in extracts. Using a primary rat peritoneal mast cells, we examined whether ginseng extracts inhibit compound 48/80-induced histamine release form rat mast cells. High red ginseng-spercific saponin containing extracts were recovered at $85^{\circ}C$ for 48 hr, and had no cytotoxicity with relatively high dose of extracts on rat peritoneal mast cells(<0.5 mg/ml). For examine of ameliorate effects of mast cells responses by ginseng extract, we pre-treated the extracts or saline to mast cells and treated compound 48/80. In results, compound 48/80 treatment was increased histamine release (approximately 30%) from mast cells than normal group, whereas ginseng treatment was completely inhibited histamine release. These results suggested that ginseng extracts inhibits the compound 48/80-induced mast cell activation, and ginseng extracts is a candidate for effective therapeutic tools of allergic diseases.

Anti-Allergic Effect of 1,2,3,4,6-Penta-O-Galloyl-β-D-Glucose on RBL-2H3 Cells (RBL-2H3 세포에서 1,2,3,4,6-Penta-O-Galloyl-β-D-Glucose의 항알레르기 효과)

  • 1,2,3,4,6-Penta-O-galloyl-${\beta}$-D-glucose (PGG) is a gallotannin isolated from various plants such as Galla Rhois. In a previous study, it was reported that PGG has anti-allergic effects by inhibiting interleukin (IL)-4 signaling in B cells. However, the effect of PGG on basophilic cells remains unclear. Therefore, the aim of this study was to investigate the inhibitory effect of PGG on mitogen and calcium ionophore-induced allergic responses. PGG had no effect on proliferation and cytotoxicity of RBL-2H3 cells. PGG significantly suppressed cell degranulation (histamine and ${\beta}-hexosaminidase$) as well as inflammatory cytokine production such as IL-4 and tumor necrosis factor-${\alpha}$. The underlying mechanism of PGG on these anti-allergic actions was correlated with inhibition on translocation of nuclear factor-${\kappa}B$ from the cytosol to nucleus. These data suggest that PGG is a potentially effective functional compound for prevention of allergic diseases.

Effects of the Sarcodon aspratus on the High Level of Blood Lipid and Obesity Induced by High Fat-diet in Rat (능이 버섯이 고지방식이에 의한 고지혈증 비만 쥐에 미치는 영향)

  • The mushroom, Sarcodon aspratus was tested for its possibility of being a health food against corpulence. The mushroom powder was fed to rats with high-fat diet-induced corpulence for 6 weeks. To measure its effects, various factors such as obesity index, serum lipid, and organ weight were analyzed and compared. The group fed a high-fat diet showed a significant increase in the levels of triglyceride, total cholesterol and LDL-cholesterol in serum. However, this increase was not shown in the group fed mushroom powder. The R$\ddot{o}$hrer index (one of many obesity indexes) was higher in the group fed a high-fat diet compared to that of control group. However, there was a significant decrease in the R$\ddot{o}$hrer index in the group fed with mushroom powder. Especially, the increased ratio of liver to body weight was observed in the group fed a high-fat diet compared to the control group. These results suggest that the significant changes in obesity index and organ weight in the group fed with mushroom powder are due to the decreased level of serum triglyceride. Thus, there is a possibility for Sarcodan aspratus to become a heath food that can prevent diseases related to corpulence.

The Anti-inflammatory and Antiallergic Effects of Allomyrina dichotoma Larva Hot-water Extract (장수풍뎅이 유충 열수 추출물에 의한 항알레르기와 항염증 효과)

  • Traditionally, the larvae of Allomyrina dichotoma (AD), a species of the rhinoceros beetle, have been widely used for their antidiabetic, antihepatofibrotic, antineoplastic, and antiobesity effects. The United Nations' Food and Agriculture Organization has reported on the possibility of using edible insects in human dietary supplements in the future. However, despite the growing interest in insect-based bio-active products, the biological activities of these products have rarely been studied. Previously, we reported that AD larvae inhibit the in vitro differentiation of adipocytes via transcription factor downregulation. In this study, our objective was to evaluate the effects of a hot-water extract of AD larvae on allergy and inflammation. To investigate the inhibitory effect of the extract on allergic reactions, we measured the levels of ${\beta}-hexosaminidase$, tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-4 (IL-4), and cyclooxygenase-2 (COX-2) after activation of RBL-2H3 cells using Compound 48/80. In addition, the inhibitory effect of the extract on inflammation was determined using Raw 264.7 cells after lipopolysaccharide (LPS) stimulation. The extract significantly inhibited the ${\beta}-hexosaminidase$, $TNF-{\alpha}$, IL-4, and COX-2 levels in RBL-2H3 cells. Furthermore, it effectively inhibited the inflammatory cytokine IL-6, nitric oxide, and inducible nitric oxide synthase expression in LPS-stimulated Raw 264.7 cells. These results suggest that AD larval extract can be potentially developed as an antiallergic and anti-inflammatory therapeutic agent.

Anti-Obesity Effects of Fermented Samjung-hwan in Hign Fat Diet Rats (고지방 식이를 섭취한 흰쥐에서 발효 삼정환의 항비만 효과)

  • Objectives: This study was performed to evaluate the effects of fermented Samjung-hwan (SJH) extracts on weight, serum lipids and blood glucose. Methods: SJH was fermented using three different probiotic bacterial strains (Lactobacillus plantarum [LP], Leuconostoc mesenteroides [LM], Bifidobacterium longum [BL]) separately. Thirty-six rats were divided into normal, control (high fat diet), SJH-UF (high fat diet+unfermented SJH 200 mg/kg), SJH-LP (high fat diet+LP fermented SJH 200 mg/kg), SJH-LM (high fat diet+LM fermented SJH 200 mg/kg) and SJH-BL (high fat diet+BL fermented SJH 200 mg/kg). For 8 weeks later, we examined body weight, total cholesterol, high-density lipoprotein (HDL)-cholesterol and blood glucose. Results: The control group showed significantly increased weight gain compared with normal group and SJH-LP and BL groups had less weight gain than control group, significantly. In the lipid serum tests, control group showed significantly increased total cholesterol levels compared with normal group and only SJH-LP represented decreased total cholesterol levels compared with control group. However there was no significant change in the HDL-cholesteol levels. In the blood glucose tests, that of control group significantly incereased more than that of normal group, SJH-BL showed significantly decreased blood glucose levels compared with control group. Conclusions: SJH-LP, SJH-BL showed weight control effect, SJH-LP decreased TC and SJH-BL reduced blood glucose.

In Vitro Anti-Obesity Effects of Raw Garlic and Pickled Garlic (생 마늘과 절인 마늘의 In Vitro 항비만 효과)

  • Objectives: This study aimed to compare the anti-obesity effects of raw and pickled garlic in vitro in 3T3-L1 preadipocytes. Methods: The pickled garlic samples comprised the following: garlic aged in vinegar (VG), garlic aged in soy sauce, and vinegar (1:1, v/v) (PG) and raw garlic (RG) as control. Hexane, butanol, and distilled water were used to prepare the fractions. The pancreatic lipase inhibitory activity was used as a measure of anti-obesity effects of the extracts. The lipid droplet accumulation and triglyceride content in the 3T3-L1 cells were measured using Oil red O staining and triglyceride assay kits, respectively. The adipogenesis related protein expression levels were analyzed using the kits and the western blot method. Results: The pancreatic lipase inhibitory activity of the garlic extracts (VG, PG, RG) was the highest in the butanol fraction, and the inhibitory effect was the highest in RG, followed by PG and VG. All garlic butanol extracts suppressed triglyceride accumulation in differentiated adipocytes (P<0.05) through the activation of cyclic adenosine monophosphate (AMP), AMP-activated protein kinase, carnitine palmitoyl transferase-1, and the inhibition of fatty acid synthase. Raw garlic extracts significantly inhibited the expression of proteins involved in adipogenesis as compared to pickled garlic. Conclusions: Raw garlic has the potential to be an effective natural material for reducing obesity compared to pickled garlic with vinegar or soy sauce.

Effects of Mixture Lactic Acid Bacteria and Sea Tangle on Anti-obesity and Gut Microflora in Rats (흰 쥐에서 복합 유산균과 다시마가 항비만 및 장내 미생물에 미치는 영향)

  • The present study was undertaken to investigate the effects of dietary provision of lactic acid bacteria (LB) and sea tangle (ST) on the obesity-associated intestinal microbiota in rats with obesity induced by a high-fat diet. Forty-eight 8-wk-old Sprague-Dawley rats were fed a basal diet (CON), a high fat diet (HFD; CON supplemented with 10% lard), HF supplemented with LB [HFL; $5{\times}10^8cfu$ of each of Lactobacillus rhamnosus, Lactobacillus johnsonii, Bifidobacterium longum and Bifidobacterium lactis], or HFL containing 10% ST (HFLS), with 4 replicates (cages) of 3 rats per dietary treatment, for 6 wk, and the intestinal microbiota were determined by pyrosequencing. The HFL and HFLS groups exhibited reduced rates of weight gain than the HF group, and the former groups had smaller ratios of Firmicutes and greater ratios of Bacteriodetes, with decreased Firmicutes/Bacteroidetes ratios, than the latter at the level of the phylum. Compared with the results for the HF group, HFL and HFLS had reduced ratios of the families of Roseburia, Mollicute, Erysipelotrichi, and Oscillibacter within Firmicutes associated with obesity and increased ratios of the families of Prevotella, Alistipes and Bacteroides within the Bacterioidetes phylum known to have an anti-obesity effect. The content of butyric acid in feces was greater in the HFLS group vs. HF and HFL. In conclusion, the present results suggest that dietary provision of LB plus ST has an anti-obesity effect and induced changes in intestinal microorganisms, and enhanced the content of butyric acid, which is an intestinal metabolite.
