생약재의 식품부패 세균에 대한 항균활성

  • 본 연구에서는 식품의 부패에 관여하는 미생물인 Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Staphylococcus aureu, Salmonella typhimurium Lactobacillus plantarum에 항균 활성을 나타내는 생약제의 효능을 살펴보았다. 금산 약초시장에서 구입한 생약재로부터 물 추출물과 70%에탄을 추출물을 제조하여 6종의 세균에 대해 항균활성을 탐색한 결과, 생약재 물 추출물의 경우에 가자 추출물은 E. coli, P. aeruginosa B. subtilis, S. aureus, S. typhimurium, L. plantarum의 6가지 균종에 대해서 모두 높은 항균활성을 나타내었으며, 복분자와 지유의 물추출물에서는 L. plantarum의 균종을 제외한 5가지 균종에 대해 높은 항균 활성을 나타내었다. 또한 소목 과 황련의 추출물에서는 E. coli를 제외한 5가지 균종에서 항균활성을 나타내었으며, 정향 추출물에서는 E. coli균주에 대해서는 약한 항균활성을 나타내었으나, B. subtilis, P. aeruginosa, S. aureus, S. typhimurium의 4종의 균주에 대해서는 우수한 항균활성을 나타내었다. 이 외에 감초 추출물은 P. aeruginosa, 석곡 추출물은 S. aureus, S. typhimurium, 계피 추출물은 S. typhimurium, 산수유 추출물은 B. subtilis, S. aureus, 파고지는 S. aureus균종에 대해서 항균활성을 나타내었다. 70% ethanol 추출물은 대부분의 생약재에서 높은 항균 활성을 나타내었다. 특히 가자, 감초, 강황, 복분자, 석곡, 소목, 오미자, 작약 ,정향, 지유, 파고지, 호장근, 황련 의 13가지 생약재 추출물에서 높은 항균활성을 나타내었다. 가자의 70% ethanol추출물의 경우는 물 추출물의 결과에서 같이 6가지 균종에 대해 모두 높은 항균활성을 나타냈으며, 복분자, 작약, 정향, 지유의 70% ethanol 추출물은 L. plantarum균을 제외한 5가지 균종에 대해 높은 항균 활성을 나타내었다. 소목의 70% ethanol 추출물의 경우에는 E. coli 균을 제외한 5가지 균종에서 높은 항균 활성을 보였으며, 이외에 생약재는 강황, 석곡, 황련, 호장근의 70% ethanol 추출물은 4가지의 균주에 대해서 항균 활성을 나타내었다. 또한 파고지, 오미자의 70% ethanol 추출물은 3가지 균주에 대해서 항균활성을 나타냈으며, 천궁, 육두구, 오수유, 갈근, 계피, 가시오가피의 70% ethanol 추출물은 2가지 균주에 대해 항균 활성을 나타내었다. 항균활성이 우수한 생약재를 농도별로 활성을 조사한 결과, 물 추출물과 10% Ethanol 추출물 모두 낮은 농도에서도 우수한 항균활성을 나타내었다.

  • 79종의 식물재료의 Alternaria alternata에 대한 항균활성을 조사한 결과, 7가지 식물재료가 항균활성이 있었고, 11가지는 확실치 않았으며, 나머지 61가지는 항균활성이 나타나지 않았다. A. alternata에 항균활성이 나타난 7가지 식물재료 중 계피가 활성이 가장 높았으며, 오미지와 천궁이 중정도의 활성을 보였으며, 나머지는 약한 활성을 나타내었다. 항균 활성 성분은 계피와 천궁의 경우는 diethyl ether에 분리되었고, 오미자의 경우는 butanol에 분리되었다. 계피의 diethyl ether 추출물은 1,000 배액 이하에서 식별 가능한 항균활성이 나타났다. 계피의 성분으로 cinnam aldehyde가 가장 높은 활성을 보여, 이 물질이 계피의 주요 항균물질로 사료된다.

Antimicrobial Activity of Garlic Extracts according to Different Cooking Methods (조리방법을 달리한 마늘 추출물의 항균활성)

  • This study was conducted to find the antimicrobial activity of garlic extracts by various processing methods(boiled, pan fried, microwave heated, pickled). Ethanol and water extracts from garlic sample were prepared and antimicrobial activities were determined against 10 microoganisms ; food borne pathogens, food poisoning microoganisms, food-related bacteria and yeasts. The ethanol extracts from the fresh and pickled garlic showed antimicrobial activities for test microoganisms, except lactic acid bacteria and yeast. However, the antimicrobial activities were decreased by heat treatment. The minimum inhibitory concentration(MIC) of the fresh garlic extracts was determined to 0.1 mg/mL against an gram positive bacterium and 0.5 mg/mL against an gram negative bacterium. The antimicrobial activities of the ethanol extracts were affected by heating methods and not by pHs.

Ethanol extract exhibited antimicrobial activities for the microorganisms tested, but not on lactic acid bacteria and yeast Especially, minimum inhibitory concentration(MIC) for Escherichia coli were as low as 0.5 mg/mL. Antimicrobial activity of the ethanol extract was stable by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of Lentinus edodes exhibiting high antimicrobial activity. The highest antimicrobial activity adjust bacteria tested was found in the ethylacetate fraction.

  • Natural antimicrobial activities in leaf mustard Dolsan(Brassica juncea) were evaluated for food preservation. Antimicrobial activities and changes in volatile allylisothiocyanate(AIT) concentration were examined during incubation of leaf mustard homogenate for 72 hours at $30^{\circ}C$. The concentration of volatile AIT was highest at 2 hours of incubation. Antimicrobial activities, which were insignificant in raw homogenate without incubation, were occurred at 21 hours and were showed highest at 48~72 hours of incubation. Water-pretreated extract(WPE) showed the antimicrobial activity by 1.2~1.4 times higher than the water extract. Ethylacetate fraction of leaf mustard showed high antimicrobial activities. The WPE had strong antimicrobial activities against Salmonella typhimurium and Staphylococcus aureus. Heat treatment of the WPE for 30 min at $121^{\circ}C$ did not lose the antimicrobial activities.

The ethanol, extract of chestnut exhibiting the high antimicrobial activities was fractionated in the other of diethylether and butanol fractions. The highest antimicrobial activity against bacteria was shown in the ethanol fraction.

Especially, minimum inhibitory concentrations (MIC) of the ethanol extracts were determined as 0.25 mg/mL against bacteria and 2 mg/mL against target lactic bacteria and yeasts. Antimicrobial activity of the ethanol extracts were not destroyed by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of cornus exhibiting high antimicrobial activities were fractionated in the other of diethylether and butanol fractions to test antimicrobial activity The antimicrobial activity adjust bacteria test was highest in the ethanol fraction.

Antimicrobial Activity of Allyl Derivate in Multi-purpose Solution as Natural Preservative (다목적용액에서 천연보존제로서의 Allyl 유도체의 항균활성)

  • Purpose: The purpose of this study is to investigate antimicrobial activity of ally derivative of garlic against eye diseases, and to get basic data for an application development of the derivative, as a natural preservative, to multi-purpose solution (MPS). Methods: Antimicrobial activity of allyl derivative of garlic, diallyl sulfide (DAS), against Pseudomonas aeruginosa (P. aeruginosa) and Staphyloccus aereus (S. aereus) was determined. After DAS treatment in MPS of no microbial activity, its growth inhibition effect to bacteria was also determined. Results: DAS concentrations of $IC_{50}$ against P. aeruginosa and S. aereus were found when both bacteria were treated with DAS concentration of 0.25% and 0.64%, respectively. At MPS test, a product from Company A did not show any effects against both bacteria, but one from Company B showed antimicrobial activity against both bacteria. Antimicrobial activity test of the product from Company B in MPS did not showed any significant difference on both gram-negative and gram-positive bacteria. For a trial application of antimicrobial activity of DAS as a natural preservative, DAS was added to MPS of Company A which did not show any antimicrobial activity at treated concentration. Addition of DAS into MPS of Company A resulted in antimicrobial activity of DAS, which suggested a possible application of DAS as a germicide for contact lenses. Conclusions: This study showed allyl derivative of garlic has antimicrobial activity against both P. aeruginosa and against S. aereus, but more effects on P. aeruginosa. The result suggests that allyl derivative derived from garlic can be developed to a preservative of MPS as a natural substance.

Antimicrobial Effect of Caesalpinia sappan L. Extract on Foodborne Bacteria (식중독 세균에 대한 소목 추출물의 항균 활성)

  • To develop a natural antimicrobial agent, we investigated the antimicrobial activities of 13 species of edible herbal plant extracts against major Gram-positive foodborne bacteria. Among the 13 screened edible herbal plants, Caesalpinia sappan L. showed the highest antimicrobial activity. In the paper disc agar diffusion assay, Caesalpinia sappan L. extracts had strong antibacterial activities against most Gram-positive bacteria but did not have antibacterial activities against most Gram-negative bacteria. Minimum inhibitory concentrations of the ethanol extract were 0.06 mg/mL against Clostridium difficile and Listeria monocytogenes and 0.03 mg/mL against Staphylococcus aureus. Their inhibitory activities were not reduced by heat treatment or pH adjustment against C. difficile, L. monocytogenes, and S. aureus. Antimicrobial activities were higher in ethanol extract than in distilled water extract. These results support the potential use of Caesalpinia sappan L. ethanol extract as an antimicrobial agent or functional food components against Gram-positive bacteria.

Antibacterial Activity of Elven (맥반석의 항균활성)

  • Analysis of components in elven(trade name, Holon killer sand) and its antibacterial activity were studied. Zinc was the highest composition by the element analysis in the elven, and heavy metals concentration was quite low on the basis of drinking water standard of this country. The antibacterial activity for 24hr culture was the most effective in Bacillus Subtilis ATCC 6633. In which, there was no growth in 24hr when 50% extract of elven was employed for the culture. And the next strong activity was shown in Stapylococus aaeruginosa ATCC 27853. But, Streptococcus mutans did not reveal any gowth inhibition by when elven extract of 10% was added in the culture broth.
