Physicochemical Properties of Salicornia herbacea Powder as Influenced by Drying Methods (건조방법에 따른 함초 분말의 이화학적 품질 특성)

  • The effects of drying methods on the physicochemical properties of Salicornia herbacea powder were investigated. Samples prepared by freeze drying showed a significantly higher L$^{*}$-value but lower a$^{*}$-value as compared with those prepared by hot-air drying and vacuum drying (p<0.05). Scanning electron micrograph revealed that freeze drying produced more orderly and denser samples than did vacuum and hot-air drying. Water solubility of the freeze-dried sample was significantly higher than those of the other methods while swelling ratio of the same sample was significantly lower than those of others (p<0.05). Freeze-dried Salicornia herbacea powder contained significantly higher amounts of total sugar and reducing sugar as compared to the other samples (p<0.05).

Antiadipogenic Effect of Korean Glasswort (Salicornia herbacea L.) Water Extract on 3T3-L1 Adipocytes (세포배양 모델을 통한 함초(Salicornia herbacea L.) 물 추출물의 항비만 효과 탐색)

  • As a natural plant ingredients, glasswort (Salicornia herbacea L.) contains various physiological activities, mainly anti-oxidative and anti-diabetic activities. However, only a few studies have been carried out on its anti-adipogenic effect. This study investigated the anti-obesity effects of Salicornia herbacea L. on 3T3-L1 adipocytes. As adipogenesis of preadipocytes to adipocytes involves proliferation and differentiation of cells, we treated three concentrations (125, 250, and $500{\mu}g/mL$) of Salicornia herbacea L. water extracts (SLW) in both pre-processing and post-processing stages. When 3T3-L1 adipocytes were differentiated and dyed with Oil Red O, adipocytes size and the value of relative Oil Red O staining were reduced by all concentrations of SLW in post-processing stage. Following adipogenic differentiation, the concentration of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in the cell supernatant significantly increased upon treatment with $125{\mu}g/mL$ of SLW and further rose at concentrations of 250 and $500{\mu}g/mL$ during post-processing stage. There was no significant difference in glycerol production upon SLW treatment. Leptin production significantly decreased at all SLW concentrations during post-processing stage, whereas peroxisome proliferator activated receptor-${\gamma}$ (PPAR-${\gamma}$) and adiponectin secretions were significantly enhanced. Overall results showed that SLW might have an anti-adipogenic effect via enhancement of TNF-${\alpha}$ production, which causes dedifferentiation and inhibits lipid accumulations in adipocyte. Furthermore, SLW might prevent diabetes and cardiovascular disease, as it reduces leptin secretion and enhances production of both PPAR-${\gamma}$ and adiponectin. However, further research is needed to elucidate the exact mechanism and bioactive compounds of glasswort.

Effect of Salicornia herbacea L. Supplementation on Tissue Triglyceride Concentrations and PGC-1α & PPAR-γ Expression of Skeletal Muscle of Rats Fed a High-fat Diet (함초의 보충식이가 고지방식이 흰쥐의 혈청 및 조직의 중성지방 농도와 골격근 내 PGC-1α 및 PPAR-γ 단백질 발현에 미치는 영향)

  • This study examined whether the supplementation of Salicornia herbacea L. (SH), a member of the Chenopodiaceae subfamily, affects tissue specific triglyceride (TG) accumulation and the peroxisome proliferator-activated $receptor-{\gamma}$ $coactivator-1{\alpha}$ ($PGC-1{\alpha}$) and peroxisome proliferator-activated $receptor-{\gamma}$ ($PPAR-{\gamma}$) protein expressions of skeletal muscle in rats with a high-fat diet. Sprague-Dawley male rats were randomly divided into three groups: control normal diet group (CD), high-fat diet group (HD), and 5.0% SH supplemented high-fat diet group (SD). The weights of fat tissue of the SD group were reduced by approximately 25%(p<0.01), while the skeletal muscle weight of the SD group increased approximately 5% compared to those in the HD group (p<0.01). The serum and hepatic TG of the SD group decreased approximately 20% compared to those of the HD group (p<0.05). In the protein expression levels in the skeletal muscle, the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ expressions of the SD group were 1.5-folds higher than those of the HD group (p<0.01). From these results, SH supplementation contributes to the improvement of the serum and hepatic TG concentrations, and the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ protein expression levels in the skeletal muscle of fed a high-fat diet. Thus, SH supplementation was effective in reducing fat mass and increasing muscle mass.

Immunomodulating Activity of Salicornia herbacea Extract (함초 추출물의 마우스 면역 증강 활성)

  • Immunomodulating effect of Salicornia herbacea extract on the mouse splenocytes was investigated. Crude S. herbacea polysaccharide extract (CSP) and other kinds of fine S. herbacea polysaccharides (SPI and SPII) were prepared from S. herbacea by hot water extraction and further ultrafiltration and gel filtration chromatography. In vitro experiment, the mouse splenocytes and separated T cells were treated with CSP, SPI or SPII (0.5, 1, 2, 4 mg/ml). In vivo experiment, three different S. herbacea extracts were orally administrated everyday for one week. For the basic data, body weight and physiological parameters such as organ weight and spleen index were observed. The proliferation of the cells was used as an index for immunemodulating activity and the effect of proliferation was evaluated using MTS assay. The CSP, SPI and SPII directly induced the proliferation of splenocytes and separated T cells in a dose-dependent manner. In results, the proliferation was more increased in the SPI and SPII treated cells than in the CSP treated cells. The best proliferation was shown in the splenocytes cultured with SPI at the concentration of 4 mg/ml for 24 hr. The proliferation of splenocytes and separated T-cells was higher (3.2 and 3.5 times, respectively) than the control. Moreover, when the mouse splenocytes were treated with mitogen, the efficient proliferation was shown in the splenocytes cultured with SPI. In conclusion, polysaccharides from S. herbacea showed a substantial immunomodulating activity in the mouse immune cells.

Comparative Study of Food Components and Sensory Properties of Common Porphyra yezoensis and Functional Porphyra yezoensis (일반 김과 기능성 김의 식품성분과 관능평가 비교 분석에 관한 연구)

  • This study is to compare food components and the sensory properties of common and functional Porphyra yezoensis, Pyroligneous liquor, Salicornia herbacea L. and Salicornia herbacea L. treated with oak charcoal. The samples used in this study were provided by a local manufacturer. We analyzed their proximate composition, mineral content, heavy metal content and amino acids. We conducted the sensory evaluation before and after grilling Porphyra yezoenis. Common components are more plentiful in functional Porphyra yezoenis than in common except for carbohydrate and crude fiber. In the case of mineral content, generally functional Porphyra yezoensis contains much more mineral than common except for magnesium and zinc. Especially Salicornia herbacea L. treated with oak charcoal contains abundant iron. Also, Salicornia herbacea L. contains a lot of calcium potassium, copper and sodium. There is a little more heavy metal except for chromium, cadmium and lead in Salicomia herbacea L. treated with oak charcoal than in common that contains a lot of chromium, cadmium and lead, although a wide difference was not discovered between them. In the case of composed amino acid, there are much glutamic acid, glycine, isoleucine and phenylalanine in both Porphyra yezoensis. Especially alanine and phenylalanine are trebled in functional Porphyra yezoensis. Also, there are far more serine and lysine in Salicornia herbacea L. and Salicornia herbacea L. treated with oak charcoal than in common Porphyra yezoensis and Pyroligneous liquor. In the case of the sensory properties, there isn't a big difference before grilling Porphyra yezoensis; however, after grilling, most functional Porphyra yezoenis had good sensory properties results, especially Salicornia herbacea L. were better than Pyroligneous liquor.

Effect of Insoluble Dietary Fiber Extracted from Salicornia herbacea L. on Large Intestinal Function in Rats (불용성 함초 식이섬유의 섭취가 흰쥐 장기능에 미치는 효과)

  • This study investigated the effect of insoluble dietary fibers extracted from Salicornia herbacea L. (S. herbacea) on the improvement of intestinal function in rats. Sprague-Dawley rats were fed on diet containing 5% and 10% S. herbacea dietary fiber (SHDF) for four weeks. Rats receiving the SHDF diet showed a significant decrease in their triglyceride levels and an increase in HDL-cholesterol levels. In addition, compared with the control group, the SHDF group showed a significant increase in the total quantity of the feces and its moisture content. The intestinal transit time of the feces was also shorter in this group. The pH of the feces decreased in all the other experimental groups. Particularly, the bile acid content of the feces and the thicknesses of the mucus layers showed significant recovery on SHDF intake. These results suggest that dietary fiber isolated from S. herbacea has a marked effect on the improvement of bowel function in rats with loperamide (2 mg/kg)-induced constipation.

Effect of Salicornia bigelovii Extract on the Activities of Whitening and Anti-wrinkle (추출용매에 따른 함초 추출물의 피부미용 효과)

  • This study investigates the effect of Salicornia bigelovii extract on the activities of antioxidant, anti-wrinkle and whitening. Among amino acids, the glutamic acid concentration was the highest with an order of proline > alanine > $\gamma$-amino-n-butyric acid > arginine. The total polyphenol concentration and antioxidant activity were an order of ethyl acetate extract > n-butanol extract > hexane extract > hot water extract > ethanol extract. Especially, when ethylacetate was used as extractant, the total polyphenol concentration and antioxidant activity were 31.6 mg/mL and 82.6%, respectively. The inhibition activity of tyrosinase was increased with the increase concentration of ethylacetate extract. Especially, when the extract concentration was increased from 20 to 100 mg/mL, it was raised from 15.2 to 98.2%. In the case of inhibition activities of elastase, and collagenase, they were also increased with the increase concentration of ethylacetate extract. When the extract concentrations were increased from 20 to 100 ug/mL, they were enhanced from 2.9 to 32.5% and from 3.8 to 29.7%, respectively. The results show that the ethyl acetate extract of Salicornia bigelovii can be applied as sources for functional cosmetics and pharmaceuticals.

Anti-cancer effects of Salicornia herbacea extract in OVCAR-3 cells by induction of apoptosis pathway (함초 추출물의 세포사멸 유도에 의한 난소암세포에서 항암 효과)

  • This study investigated the anti-cancer effects of Salicornia herbacea L. fractions in human ovarian cancer cells (OVCAR-3). S. herbacea powder was extracted with 95% EtOH and sequentially fractionated with hexane, dichloromethane (DCM), ethyl acetate, butanol, and H2O. Further, the growth inhibitory effects of the six fractions were determined using the MTS assay. The DCM fraction dramatically decreased cell viability. Similarly, the cell cycle was arrested at the subG1 phase in DCM-treated cells. To confirm apoptosis, the cells were stained with annexin V/FITC-PI solution. Total, early, and late apoptotic cells were significantly increased in the DCM fraction. The mRNA expression of Bcl-2 was reduced, whereas the pro-apoptotic factors Bax and Bak were increased in DCM fraction-treated cells. These results indicated that the DCM fraction in S. herbacea exhibited strong apoptotic effects through the p53-dependent signaling pathway.
