Extended Adaptation Database Construction for Oriental Medicine Prescriptions Based on Academic Information (학술 정보 기반 한의학 처방을 위한 확장 적응증 데이터베이스 구축)

  • The quality of medical care can be defined as four types such as effectiveness, efficiency, adequacy, and scientific-technical quality. For the management of scientific-technical aspects, medical institutions annually disseminate the latest knowledge in the form of conservative education. However, there is an obvious limit to the fact that the latest knowledge is distributed quickly enough to the clinical site with only one-time conservative education. If intelligent information processing technologies such as big data and artificial intelligence are applied to the medical field, they can overcome the limitations of having to conduct research with only a small amount of information. In this paper, we construct databases on which the existing medicine prescription adaptations can be extended. To do this, we collect, store, manage, and analyze information related to oriental medicine at domestic and abroad Journals. We design a processing and analysis technique for oriental medicine evidence research data for the construction of a database of oriental medicine prescription extended adaption. Results can be used as a basic content of evidence-based medicine prescription information in the oriental medicine-related decision support services.

  • 한의학은 대체의학이나 예방의학으로서의 활용이 가능한 우리나라의 독창적인 의료 체계이지만 세계시장 진출은 고사하고 국내에서의 인지도마저 낮은 실정이다. 이를 위해 한의학의 진단 기술과 IT 기술을 융합하여 한의학의 진단의 객관성을 확립함으로서 한의학의 대중화 및 인지도 상승을 위해 많은 연구가 진행되고 있으나 망진에서 가장 중요한 색윤에 대한 연구가 이루어지지 않고 있는 실정에서 본 논문에서는 색윤을 특정 할 수 있는 방법을 제안하고 이를 실험을 통해 입증하고자 한다.

Effects of a Glycoprotein Isolated from Ulmus davidiana Nakai on Toluene-Induced Ecotoxicity and its Mechanism in Human Intestinal Epithelial Cells (소장상피세포에 있어서 느릅나무 당단백질이 톨루엔에 의해 유도된 환경독성 기작에 미치는 효과)

  • Ulmus davidiana Nakai (UDN) has been traditionally used as a herbal medicine to treat inflammatory diseases in Korea. In the present study, we investigated the anti-ecotoxic potential of a 116 kDa glycoprotein isolated from UDN (UDN glycoprotein) in human intestinal epithelial INT-407 cells. We demonstrated that UDN glycoprotein ($20{\mu}g/mL$) could inhibit the production of lactate dehydrogenase (LDH) induced by toluene, an ecotoxic substance. Additionally, we found that the toluene-induced intestinal cytotoxicity was mediated by the phosphorylation of p38 Mitogen-Activated Protein Kinase (MAPK) via the production of intracellular Reactive Oxygen Species (ROS). The UDN glycoprotein significantly decreased the levels of ROS production and p38 MAPK activation in toluene-stimulated INT-407 cells. Moreover, the UDN glycoprotein inhibits the phosphorylation of nuclear factor-kappa B ($NF-{\kappa}B$), which is responsible for the production of LDH, in toluene-stimulated INT-407 cells. Collectively, our data indicate that UDN glycoprotein is a natural antioxidant and a modulator of ecotoxicity signaling pathways in human intestinal epithelial cells.

Protective effect of Citri Unshius Pericarpium against cadmium-induced liver damage in mice (카드뮴으로 인한 마우스 간 손상에 대한 진피의 보호효과)

  • Objective : Citri Unshius Pericarpium (Citrus unshiu peel) has been used in Korean medicine to treat indigestion, vomiting, coughing and phlegm. This study investigated the hepatoprotective effect of ethanol extract of Citrus unshiu peel (CEE) in cadmium (CdCl2)-treated mouse model. Methods : CEE was dissolved in water and administered orally to mice once a day for 7 consecutive days. The mice were then exposed to a single intraperitoneal (i.p.) injection of cadmium (4 mg/kg body weight) to induce acute hepatotoxicity. At the end of the experiment, blood and liver tissue samples were collected, analyzed for alanine aminotransferase (ALT), aspartate aminotransferase (AST), and histopathological evaluation. Liver damage was assessed as the percentage of degenerative areas of the hepatic parenchyma, the number of degenerative hepatocytes, and the number of infiltrated inflammatory cells. Results : In cadmium-treated rats, pretreatment with CEE significantly reduced the serum ALT and AST levels associated with liver damage. Histopathologically, CEE prevented degenerative changes on the hepatic tissues including confluent necrosis, congestions and infiltration of inflammatory cells. CEE also reduced the elevation of oxidative stress markers (nitrotyrosine and 4-hydroxynonenal) and apoptosis markers (cleaved caspase-3 and cleaved PARP) positive cells. PARP protein expression in liver tissue was also restored by CEE. Conclusion : This study showed that CEE exerted antioxidant and anti-apoptotic effects against cadmium-induced liver injury. Thus, it can be concluded that CEE can be used to prevent liver damage caused by cadmium.

  • 인체의 여러 요소들의 상호균형을 중요시하는 한의학 이론은 의학적 가치가 높이 평가되고 있지만 이를 뒷받침할 만한 체계적이고 과학적인 제시가 부족한 것이 현실이다. 이에 본 논문에서는 한의학의 사진(四診)중 망진(望診) 이론을 영상공학적 기술과 융합하여 손바닥 영상 분석을 통해 간단하게 질환을 진단할 수 있는 방법에 대해 제안하고자 한다. 특히 현대인의 스트레스로 인해 늘어가고 있는 간질환자의 증가에 대비하여 정상인과 간 질환이 있는 환자간의 손바닥 색상을 비교, 분석을 하고자 한다. 이를 위해 본 논문에서는 한의학적으로 간 질환자는 정상인보다 RGB값 및 K값이 상대적으로 낮게 나타난다는 것을 규명하기 위해 정상인과 간 질환자의 손바닥 영상 분석을 통해 RGB값 및 K값의 검출로 두 개체 집단간의 유의성을 실험으로 입증함으로써 간 질환이 의심스러울 경우 조기진단에 도움을 주고자 한다.

Protective effect of Citrus unshiu peel on the cadmium-induced apoptosis in HepG2 cells (카드뮴으로 유발한 간세포 자멸사에서 진피의 보호효과)

  • Objective : Citrus unshiu peel (Citri Unshius Pericarpium) has been prescribed to suppress coughing and phlegm in Korean medicine. In this study, the effect of ethanol extract of Citrus unshiu peel (CEE) on apoptosis was investigated using cadmium chloride (CdCl2) treated HepG2 cells. Methods : CEE was prepared by extracting 300 g of Citri Unshius Pericarpium in 3 L of ethanol for 72 h. Apoptosis was determined by the TUNEL assay. The mitochondrial membrane potential (MMP) was monitored using the membrane-permeable fluorescent dye Rh123. The expression level of each protein was monitored by Western blot analysis. Results : CEE protected HepG2 cells from apoptosis as determined by the TUNEL assay. A decrease in MMP was observed in cells exposed to cadmium, indicating that mitochondria are involved in the induction of apoptosis. However, CEE recovered the reduction in MMP caused by cadmium. In addition, decreased expression of B-cell lymphoma 2 (Bcl-2), procaspase, and poly(ADP-ribose) polymerase (PARP) by cadmium was increased by CEE. The anti-apoptotic effect of CEE was found to be associated with inhibition of JNK and p38 phosphorylation when examining the expression of phosphorylated MAPK by Western blot. Conclusion : This study showed that CEE exerted anti-apoptotic effects in cadmium-induced HepG2 cells by inhibiting the reduction of MMP and changes in the expression level of apoptotic proteins. These results suggest the potential for CEE to be used for heavy metal-induced liver damage.

Neuroprotective Effect of Gardeniae Fructus against Oxidative Damage Induced by tert-Butyl Hydroperoxide in PC12 Cells (PC12 cell에서 tert-butyl hydroperoxide로 유도된 산화적 손상에 대한 치자의 신경보호효과)

  • Objective : Gardeniae Fructus (GF) is the ripe fruit of Gardenia jasminoides Ellisa with a bitter taste and cold properties. Ingredient compounds including geniposide are known to have anti-inflammatory, antioxidant, and neuroprotective effects. The purpose of this study was to investigate the neuroprotective effect of GF on tBHP-induced PC12 cells. Methods : Cell viability was measured by the MTT assay, and apoptosis was determined by the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expression level of each protein was monitored by Western blot analysis, and reactive oxygen species (ROS) were analyzed using DCFH-DA. Results : In PC12 cells, tBHP induced cell death through apoptosis with caspase activation and PARP inactivation. Cells treated with tBHP showed an increase in intracellular ROS and depletion of GSH. Pretreatment with GF prevented tBHP-induced apoptosis, reduced ROS, and increased GSH. GF also maintained increased Nrf2 expression in the presence of tBHP. Phosphorylation of JNK and p38 MAPK was increased by tBHP, whereas phosphorylation of ERK was decreased. GF restored changes in ERK and p38 phosphorylation, but not JNK phosphorylation. Conclusion : These results indicate that GF has neuroprotective effects through anti-apoptotic and antioxidant effects mediated by regulation of Nrf2 expression and phosphorylation of ERK and p38. It also demonstrates the potential use of GF as a source of antioxidant and neuroprotective substances.
