Studies on the Estimation of the Genetic Parameters on All Traits in Korean Native Ogol Fowl II. Estimations of the Heritabilities and Genetic Correlations on Egg Components (한국재래오골계의 제형질에 대한 유전모수추정에 관한 연구 II. 난구성분에 대한 유전력 유전상관추정)

  • This study was conducted to estimate heritabilities and genetic correlations on egg compositions in Korean Native Ogol Fowl. The date analysis were a total of 58,320 eggs in 450 pullets producted from 150 dams and 20 sires of Korean Native Ogol Fowl raised at Chungnam National University from June 18. 1987 to April 6. 1989. The results obtained are summarized as follows: 1. On the egg compositions, the albumen weight at first egg, 300 and 500 days were $24.003\pm$2.499, $28.354\pm$2.755 and $31.863\pm$3.341g; the yolk weight were $10.272\pm$1.536, $16.346\pm$1.321 and $19.212\pm$1.661g: the shell weight were $3.906\pm$0.414, $4.336\pm$0.506 and $4.822\pm$0.515g, respectively. 2. The heritability estimates of egg compositions based on the valiance of sires, dams and combined components were 0.620-0.723, 0.206-0.300 and 0.413-0.511 for albumin weight;0.439-0.737, 0.484-0.544 and 0.492-0.615 for yolk weight:0.172-0.187, 0.412-0.642 and 0.390-0.503 for shell weight, respectively. 1. The genetic and phenotypic correlation coefficients of egg compositions were as follows; The coefficients between albumen weight and yolk weight were 0.089-0.654 and 0.084-0.255;between albumen weight and shell weight were 0.396-0.925 and 0.225-0.544; between yolk weight and shell weight were 0.227-0.357 and 0.098-0.358 respectively.

Comparison of principle components and quality of eggs by laying hen breeds (산란계 품종별에 따른 계란의 주요 성분과 품질 비교)

  • This study was to execute the random arrangement of total 120 heads (10 heads per treatment plot ${\times}$ 3 repeats ${\times}$ 4 treatment plots) for four breeds [Blue arakana commercial Fowl (BC), Yeonsan silky fowl (YS), Korean native fowl (KN), White leghorn fowl (WL)] with the initiation of laying hens at 32 weeks old in order to comparatively analyze egg quality and composition by sorts of laying hens. In case of evaluation of the quality of eggs, this study has shown that BC had higher degree of Haugh unit coefficient (82.82) and in case cholesterol content of eggs, BC had significantly low content as 317.55mg/100g, but YS had rather higher content as 381.30mg/100g. Furthermore, this study has found out that BC had significantly low Oleic acid (C18:1n9) and WL had higher Amino acid content by breeds than those of other breeds (p<0.05).

  • This study was conducted to estimate heritabilities and genetic correlations on egg compositions and egg qualities in Korean Native Ogol fowl. The data were gathered from a total of 58,320 eggs in 450 pullets produced from 150 dams and 20 sires of Korean Native Ogol fowl raised at Chungnam National University from June 18, 1987 to April 6, 1989. The heritability estimates of egg compositions based on the variance of sires, dams and combined components were 0.620 - 0.723, 0206 - 0.300 and 0.413 - 0.511 for albumen weight: 0.439 - 0.737, 0.484 - 0.544 and 0.492 - 0.615 for yolk weight: 0.172 - 0.187, 0.412 - 0.642 and 0.309 - 0.503 for shell weight, respectively. 2. The heritability estimates of egg qualities based on the variance of sires, dams and combined components were 0.202 - 0279, 0.557 - 0.819 and 0.428 - 0.508 for shell thickness : 0.202 - 0.394, 0.119 - 0.678 and 0.256 - 0.440 for albumen height : 0.108 - 0.443, 0237 - 0.698 and 0244 - 0.399 for Haught units, respectively. 3. The genetic and phenotypic correlations of egg compositions were as follows; The coefficients between albumen weight and yolk weight were 0.089 - 0.564 and 0.084 - 0.235; between albumen weight and shell weight were 0.396 - 0.925 and 0225 - 0.544; between yolk weight and shell weight were 0.220 - 0.357 and 0.098 - 0.358, respectively. 4. The genetic and phenotypic correlations of egg qualities as follows; between shell thickness and albumen height were 0.082 - 0.356 and - 0.163 - 0.060; between shell thickness and Haught units were - 0.076 - 0.167 and - 0.185 - 0.010; between albumen height and Haught units were 0.338 - 0.604 and 0.154 - 0285, respectively. 5. The genetic correlations of egg compositions and egg qualities were as follows: between albumen weight and shell thickness, albumen height. Haught units were - 0.380 - - 0.002, 0239 - 0.387, and - 0279 - - 0.127; between yolk weight and shell thickness, albumen height, Haught units were - 0.294 - - 0.133, - 0.049 - 0.133 and - 0.196 - - 0.136; between shell weight and shell thickness, albumen height, Haught units were 0.127 - 0.476, 0.140 - 0273 and 0.038 - 0223, respectively.

The Study on Production and Performance of Crossbred Korean Native Chickens (KNC) (토종 순종계를 이용한 토종닭 생산 및 생산성 연구)

  • The current work was carried out to investigate the effect of crossbred Korean native chickens (KNC) on performance and carcass ratio. Seven hundred twenty 1-d-old chicks were divided into groups by strain (A, B, C and D) and sex (male and female). Strains were A) (KNC egg-meat type C strains $\times$ KNC meat type S strains) $\times$ KNC meat type H strains, B) (KNC egg-meat type C strains $\times$ KNC meat type H strains) $\times$ KNC meat type S strains, C) (KNC native R strains $\times$ KNC meat type S strains) $\times$ KNC meat type H strains and D) (KNC native L strains $\times$ KNC meat type H strains $\times$ Ross broiler. Experimental diets consisted of 3 phases such as starter(0~5 weeks; CP 20.0%, ME 3,050 kcal/kg), earlier (5~8 weeks; CP 18.0%, ME 3,100 kcal/kg) and finisher (8~12 weeks; CP 16.0%, ME 3,150 kcal/kg). Body weight (BW) and feed intake (FI) was measured every week and carcass ratio(CR) was calculated at 5 and 10 week after starting experiment. There was no difference in BW among strains until 5 weeks (P>0.05), however D strain resulted in a higher BW after 5 weeks (P<0.05). Body weight gain (BWG) and FI in D strain were also significant higher compared to the other strains for all periods. However, D strains showed the lowest (P<0.05) fee conversion ratio (FCR). The other strains except D showed a similar BW, BWG, FI and FCR among strains. In addition, there were no differences in carcass weight (CW) and carcass ratio (CR) among strains at 5 weeks, however D strain showed higher CW and CR at 10 weeks. These results suggested the basic data that needed to develope the new strains.

Physicochemical Characteristics of Silky Fowl(Gallus domesticus var. silkies) (백봉오골계육의 이화학적 특성)

  • In this study, the anatomic characteristics and the contents of nutritional ingredients and minerals of silky fowl and yeonsan ogolgye was investigated. Silky fowl is covered with white silky feather. There is a pinch of vertical white tassel on peak of the head, especially a silky fowl cock. The crown of a silky fowl has a nickname of phoenix crown, the crown of a cock is mostly like a rose, while that of a hen like strawberry or mulberry. The ear of silky fowl is mostly peacock green(Light blue turquoise), bronze for a small part. Peacock green is most common for Silky Fowl with a age of $60{\sim}150$ days, over the age of 150 days, the peacock green will be gradually replaced by purplish red. Beaks are leaden blue, short and stout while the face is smooth and fine. The lower jaw of silky fowl has comparatively long tiny hair, similar to beard. The two legs of silky fowl are covered with a handful of feather, or known as 'Putting on trousers'. Each leg of silky fowl has five talons. The whole skin, eyes, mouth, talons of silky fowl are grey black The bone and marrow of silky fowl are light black the periosteum is black. The whole meat, internal organs and abdominal fat of sillry fowl aye black the heart and leg meat are light black. Silky fowl meat had lower moisture and lipids content, but higher Ash and protein content than meats of yeosan ogolgye and general chickens. For mineral contents of leg muscle and breast muscle, silky fowl had higher contents of phosphorus (P), iron (Fe), potassium (K), zinc (Zn) than those of yeonsan ogolgye, while yeonsan ogolgye had higher contents of calcium (Ca) than that of silky fowl. The contents of iron (Fe) and zinc (Zn) is higher in leg muscle than in breast muscle for the silky fowl and yeonsan ogolgye. Leg muscle of silky fowl contains a lot of iron (Fe), about 4 times as much as that of leg muscle of yeonsan ogolgye. For the silky fowl and the yeonsan ogolgye, leg muscles contains a lot of zinc (Zn), about 5 times, 4 times respectively as much as that of breast muscle.

Studies on the Performance of Korean Native Chickens I. Effect of Various Feeding Systems on Performance of Korean Native Chickens (한국재래닭의 능력에 관한 연구 I. 서로 다른 사료 급여가 한국재래닭의 능력에 미치는 영향)

  • A feeding trial was carried out to evaluate optimum feeding system to Korean Native Chicken (KNC) with various feeding programs for 64 weeks. Eight hundreds and ten KNC'one day birds were assigned to three feeding systems, NRC(T1), Japanese Feeding Standard(T2), Korean Feeding Standard(T3). Each treatment had three replicates of 90 birds a replicate. Data were obtained growing performance to 20 week of age and laying performance from twenty one to sixty four week. Viability to 20 week of age was similar to treatments. Growth characteristics by feeding systems were not significantly difference on body weight, feed intake and 50% egg production days. However, body weight was less about 100g in T2. There were no different on egg production, egg weight, and egg mass during laying period. Feed intake of T1 was 104g daily and showed lower than other treatments, but was not different significantly. Feed conversion was significantly improved in T2(P <0.05) compared to other treatments(P<0.05). Egg production of all treatments was the highest between 27 to 30 week of age. ME intake of T1 was significantly maximized, whereas CP intake was the lowest of all treatments(P < 0.05). Nutrient requirements for egg mass tended to depend on nutrients intake. Interior egg and eggshell characteristics were not different among treatments except yolk color. Fertility and hatchability were similar to treatments, and the results at 39 week of age were higher than at 62 week in all treatments. In conclusion, treatments fed three or four phase feeding would be superior to other treatments during the growing period and suggest 2,800 ㎉/kg ME, 15% CP for Korean native laying hen.

Estimation of Breeding Values for Economic Traits in Korean Native Chicken (한국재래닭의 경제 형질에 대한 육종가 추정)

  • This study was carried out to evaluate the breeding values for economic traits in Korean native chicken on the basis of 11,583 birds of record along 7 generations collected from 1995 to 2001 at National Livestock Research Institute, Korea. The results obtained are summarized as follows : The breeding value estimates of the body weights at 150 and 270 days in Dark Brown, Light Brown, Gray Brown, Black and White strains were $-16.563\~0.474\;and\;-18.836\~21.700,\;-14.714\~9.272\;and\;-23.025\~11.297,\; -25.186\~12.953\;and\;-28.368\~18.156,\;-15.549\~6.329\;and\;-18.264\~4.963,\;-10.708\~20.284\; and\;-25.112\~10.132g$, respectively. The breeding value estimates of the ages at first egg in Dark Brown, Light Brown, Gray Brown, Black and White strains were $-1.236\~0.047,\;-1.509\~0.298,\;-1.362\~0.114,\;-1.436\~0.199\;and\;-1.385\~0.718$, respectively. The breeding value estimates of the egg weight at first egg and at 270 days were $-0.105\~0.075\;and\;-0.120\~0.398,\;-0.109\~0.042\;and\;-0.205\~0.173,\;-0.166\~0.084\;and\;-0.256\~0.091,\;-0.119\~0.066\;and\;-0.051\~0.132,\;-0.112\~0.027\;and\;-0.070\~0.101g$, respectively. The breeding value estimates of the egg Production to 270 days in Dark Brown, Light Brown, Gray Brown, Black and White strains were $0.298\~l.807,\;-0.049\~l.146,\;-0.396\~l.125,\;-1.206\~l.173\;and\;-0.474\~2.641$, respectively.

Gene Expression Profiling by RNA Sequencing in Mature/Immature Oocytes of Chicken (닭의 성숙/미성숙란에서 RNA Sequencing을 이용한 유전자 발현 양상 고찰)

  • Chicken eggs undergo various physiological changes during egg maturation. To study genes associated with the egg maturation in pre-ovulation (immature) and post-ovulation (mature), we compared gene expression patterns between in the immature egg and mature egg using RNA sequencing data. Mature and immature eggs were obtained from a Heuksaek Jaerae-jong of Korean native chicken. Total RNAs obtained from the eggs were sequenced by Illumina HiSeq 2000 platform, and the generated sequence reads were mapped to Galgal4 reference sequence assembly using Tuxedo Protocol. From the comparison of the RNA sequencing data, 315 genes were differentially expressed between mature and immature eggs, and 46 genes were only detected in immature egg. Further gene ontology (GO) analysis was performed for the differentially expressed genes using DAVID, showing that 29 and 28 GO terms were independently clustered from mature and immature, respectively. From those clustered GO terms, genes related to germ cell development, sex differentiation and defense response to bacterium were mainly expressed in the immature egg, while genes related to regulation of apoptosis, steroid metabolic process and lipid homeostasis were mainly detected in the mature egg. Our results could contribute to understand egg maturation before and after ovulation, and develop genetic markers for improving egg quality and productivity.

Effects of N-Methylacetamide on the Viability, Fertility and Hatchability of Cryopreserved Ogye (Korean Native Black Fowl) Semen (N-Methylacetamide 동결보호제가 오계 동결정액의 생존성, 수정 및 부화율에 미치는 영향)

  • The use of methylacetamide (MA) as a cryoprotective agent for freezing Korean Native Black rooster Ogye semen was examined with artificial insemination. The diluted Ogye semen with HS-1 was subjected for 2 step dilution method of cryopreservation in which the final concentration of MA was adjusted to 7.5%. The sperm viability after thawing was reduced from $95.17{\pm}0.93%$ to $55.93{\pm}1.38%$ which was confirmed by live-death analysis based on Fluorescence-Activated Cell Sorting (FACS). The rates of fertilized eggs with fresh or frozen-thawed semen were reduced from $94.98{\pm}3.93%$ to $66.36{\pm}8.43%$ at day 7 with significant difference. However, the hatching rates of experiments at day 21 did not shown difference between $92.64{\pm}2.33%$ and $90.45{\pm}8.05%$ (P<0.05). With these results, the utilization of MA for freezing of Ogye spermatozoa could affect on viability of frozen-thawed semen but not on the fertility of lain eggs and hatchability of fertilized eggs and also provide possible tools of freezing for poultry genetic resource conservation.
