Analytical Method Development of (-)-Epicatechin gallate in Penthorum chinense Pursh Extract using HPLC (HPLC를 이용한 낙지다리 추출물의 (-)-­Epicatechin gallate 분석법 개발)

  • This study attempted to eatablish a High Performance Liquid Chromatography (HPLC) analysis method for the determination of (-)-epicatechin gallate as a part of the quality control for the development of functional cosmetic materials from Penthorum chinense Pursh. HPLC was performed on a Unison $US-C_{18}$ column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and methyl alcohol at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 280 nm. The HPLC method was performed in accordance with the International Conference on Harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation were 0.11 and 0.33 mg/mL, respectively. Calibration curves showed good linearity ($r^2$ > 0.9999), and the precision of analysis was satisfied (less than 0.6%). Recoveries of quantified compounds ranged from 99.51 to 101.92%. This result indicates that the established HPLC method is very useful for the determination of marker compound in P. chinense Pursh extracts.

Development of Individual Residue Analysis Method for Cyanazine in Agricultural Commodities as an Unregistered Herbicide in Korea (국내 미등록 제초제 cyanazine의 농산물 중 개별 잔류분석법 개발)

  • Cyanazine is a member of the triazine family of herbicides. Cyanazine is used as a pre- and post-emergence herbicide for the control of annual grasses and broadleaf weeds. This experiment was conducted to establish a determination method for cyanazine, as domestic unregistered pesticide, residue in major agricultural commodities using HPLC-DAD/MS. Cyanazine was extracted with acetone from representative samples of five raw products which comprised apple, green pepper, Kimchi cabbage, hulled rice and soybean. The extract was diluted with saline water and partitioned to dichloromethane for remove polar extractive in the aqueous phase. For the hulled rice and soybean samples, n-hexane/acetonitrile partition was additionally employed to remove non-polar lipids. The extract was finally purified by optimized florisil column chromatography. On a $C_{18}$ column in HPLC, cyanazine was successfully separated from co-extractives of sample, and sensitively quantitated by diode array detection at 220 nm. Accuracy and precision of the proposed method was validated by the recovery experiment on every major agricultural commodity samples fortified with cyanazine at 3 concentration levels per agricultural commodity in each triplication. Mean recoveries were ranged from 83.6 to 93.3% in five major representative agricultural commodities. The coefficients of variation were all less than 10%, irrespective of sample types and fortification levels. Limit of quantitation(LOQ) of cyanazine was 0.02 mg/kg as verified by the recovery experiment. A confirmatory method using LC/MS with selected-ion monitoring(SIM) technique was also provided to clearly identify the suspected residue.

Determination of 11-nor-9-carboxy-Δ9-tetrahydrocannabinol (THCCOOH) in human urine by solid-phase extraction and GC/MS (고체상 추출과 GC/MS를 이용한 소변 중 대마 대사체 (THCCOOH) 분석)

  • 11-nor-9-carboxy-${\Delta}^9$-tetrahydrocannabinol (THCCOOH) is the major metabolite of tetrahydrocannabinol (THC) which is the primary psychoactive component of marijuana. It is also the target analyte for the discrimination marijuana use. A method using solid-phase extraction (SPE) and gas chromatography/mass spectrometry (GC/MS) was developed for the determination of THCCOOH in human urine. Urine samples (3 mL) were extracted by SPE column with a cation exchange cartridge after basic hydrolysis. The eluents were then evaporated, derivatized, and injected into the GC/MS. The limits of detection (LOD) and quantitation (LOQ) were 0.4 and 1.2 ng/mL, respectively. The response was linear with a correlation coefficient of 0.999 within the concentration range of 1.2 (LLE 1.3)~50.0 ng/mL. The precision and accuracy were stable within 1.20% and the recovery was 83.6~90.7%. The recovery of SPE method was lower than that of liquid-liquid extraction (LLE), but there were no apparent differences in LOD, LOQ, precision and accuracy between the two methods. While SPE method is used as a very effective and rapid procedure for sample pretreatment, and clean extracts, LLE method was not suitable for the extraction procedure of THCCOOH in urine. The applicability of the method was proven by analyzing a urine samples from a marijuana abusers.

Development of Ceramide NP Analysis Method in Cosmetic Formulations Using Liquid Chromatography (액체크로마토그래피를 이용한 화장품 제형 내 세라마이드엔피 분석법 확립)

  • In this study, a quantitative analysis method was developed using high-performance liquid chromatography (HPLC) to analyze the content of ceramide NP in lotion, cream, and cleanser formulations in cosmetics. The analysis was performed using a C18 column, and the mobile phase was set at a ratio of 70 : 30 for acetonitrile and methanol, the flow rate was set to 0.8 mL/min, and the column temperature was set to 20 ℃. The method was verified by analyzing specificity, linearity, limit of detection, limit of quantitation, accuracy, and precision in accordance with the ICH guidelines. As a result of validating the method, the linearity of the calibration curve was excellent (R2 = 0.99984). The accuracy of the lotion, cream, and cleanser formulations was confirmed with a recovery rate ranging from 95.11% to 100.48%. The precision analysis showed a low relative standard deviation (RSD) of less than 0.26%. The limit of detection was 0.902 ㎍/mL, and the limit of quantitation was 2.733 ㎍/mL. Through this quantitative analysis method of ceramide NP applied in cosmetics, it is expected to assist in the quality control of products by enabling measurement even when it is difficult to separate the main peak due to the influence of interfering substances.

Effect of Strip-cast Conditions on the Formation of Microstructures in Nd-Fe-B alloys (Strip-cast 조건이 Nd-Fe-B 합금의 미세조직 형성에 미치는 영향)

  • In order to improve the microstructure of the strip cast Nd-Fe-B alloys that are frequently used for production of high energy sintered magnets, influence of various strip casting conditions on the microstructure and phase formation and distribution were investigated. Nd-Fe-B strips consisting of microstructures suitable for preparation of high energy sintered magnets could be obtained when the wheel speed was below 5 m/s. The compositional limit that can avoid the crystallization of free iron in the as-cast state was estimated to be Nd$\_$14/Fe$\_$79/B$\_$7/. Regardless of the compositional variation, <001> preferred orientation of Nd$_2$Fe$\_$14/B normal to the strip surface was always occurred below 5 m/s, which would eventually enhance the grain alignment during pressing the powder under a magnetic field. While the coercivity of the strip cast alloys increased with the increase of the wheel speed, mainly due to the refinement of Nd$_2$Fe$\_$14/B grains, it decreased with the reduction of Nd content in the alloy composition as the formation of free iron increased.

  • In this paper, we propose a new method to debond between mixed finite elements for porous media in ABAQUS (2014). ABAQUS just provides debonding algorithm for the u-p model using cohesive elements in standard version. However, this approach has a drawback that it is hard to simulate complex debonding problems like element separation, rigid body motion, and contact between separated elements in standard version. ABAQUS-explicit can resolve these complex problems, but cohesive elements for the u-p model cannot be applied. We introduce a new algorithm for debonding for porous media instead of using cohesive elements. In this method, subroutines VUMAT to apply constitutive models and VDISP to separate elements in ABAQUS are used to simulate debonding problems. In addition, a simple 2-D example is demonstrated in the ABAQUS-explicit solver.

  • In this study, track dynamic interaction characteristics caused by the vehicle running through transitional section such as bridge abutments were studied using the finite element analysis program. The geometric condition of track was generated by trigonometric function and allowable maximum track irregularity is determined by KORAIL track maintenance criteria. The sub-infrastructure under rail fastener system was modelled by 3D solid elements. To reduce computational cost only half track line is numerically considered and the roller boundary condition was applied to each side of model. In this study, the vehicle-track dynamic interaction analysis was carried out for standard Korean transition section of concrete track and the dynamic behaviors were investigated. The dynamic characteristics considered are wheel load variation, vertical acceleration at body, and maximum Mises stress at each part of transitional section.

Optimization of Culture Conditions for the Production of Diphtheria Toxin (디프테리아 toxin 생산을 위한 발효조건 최적화)

  • Experimental studies were carried out to optimize the culture conditions of Corynebacterium diphtheriae for the production of diphtheria toxin. A new media which does not contain any meat digest products was selected. The main ingredient of new medium was enzymatic digests of casein known as NZ-Case. In fermenter experiments, the toxin production was increased with the increase of cell growth. The optimum initial pH of media, air flow rate and agitation speed were 7.0, 0.22, vvm and 400 rpm, respectively. The contents of iron and calcium-phosphate precipitate were important for maximal cell growth and toxin production. The optimum concentration of iron was 0.3 mg/L and calcium-phosphate precipitate could serve in gradual supply of iron to maintain the optimal culture condition which is required for enhanced yield of toxin production. In potency test, the potency of toxoid from fermentor culture was higher than that from static culture. When diphtheria toxin is produced by fermentor culture, it is possible to produce higher levels of toxin and better toxoid quality in terms of safety, yield, productivity and immunity.

Analysis of GBAS Availability and Requirement with respected to Protection Level at Jeju International Airport (제주 국제 공항의 Protection Level 관점에서 GBAS 가용성 및 요구 조건 분석)

  • This paper presents the simulation results of GBAS availability and requirement (with respected to Vertical Protection Level) using simulated data at CAT I, CAT II/III DH point (Decision Height), which are generated using Jeju international GNSS reference position, aircraft horizontal velocity and reference/aircraft GNSS antenna performance index and so on. Two kinds of protection levels are presented, one is from a hypothesis (H0) and other is from a alternative hypothesis (H1). These protection levels are compared with AL (Alert Limit), and we analyse the GBAS availability and requirement for CAT I and CAT II/III at the airport.

A Case Study on Reinforcement of Ground and Foundation against Subsidence in Abandoned Mining Area (폐광지역 침하방지를 위한 지반 및 구조물기초 보강)

  • As the mechanism and effect range of subsidence are altered according to the various conditions (the ground condition, the earth pressure, the geometric condition of underground cavity and the structure load), the analysis and prediction of subsidence in abandoned mining area are very difficult. Also, as the geological characteristics and the mining methods are differed in each mines, the application of the pre-existing reinforcements without improvement has a lot of difficulties and limits. In this study, the various underground investigation such as long-depth core drilling, seismic tomography and BIPS (borehole image processing system) were performed, the distribution of underground cavity and coal seam and rock relaxation condition were analyzed. And we predicted the type of subsidence and estimated the subsidence by theories of mining subsidence. With these results, we analyzed the mechanism of subsidence occurrence in the research object area. Finally, we improved existing methods which were applied to the abandoned mining area and also we established the rational reinforcement for the ground and structure foundation against each subsidence cause.
