Isolation and Characterization of Bacillus Species Having Antifungal Activity Against Pathogens of Ginseng Damping Off (인삼모잘록병원균에 항균활성을 갖는 Bacillus 균의 분리 및 특성조사)

  • This study was performed to select potentially available biological control agent from soil bacteria for prevention of ginseng damping off. More than five hundred strains were isolated from ginseng rhizosphere soil. By testing antifungal activity, we have selected three soil bacteria strains and their ability to produce antibiotics and lytic enzymes such as cellulase, protease and pectate lyase was examined. Also, the presence of genes for biosynthesis of lipopeptide such as fengycin, bacillomycin D, surfactin, iturin A, and zwittermicin A was investigated in selected strains. All three strains produced cellulase, protease, and xylanase. Moreover, these strains had gene for biosynthesis of bacillomycin D, surfactin, and iturin A. ES1 and ES3 strains were identified Bacillus methylotrophucus and ES2 was confirmed Bacillus amyloliquefaciens using phylogenetic analysis on the basis of 16S rRNA gene sequences. In field test, control value of ES1, ES2 and ES3 treatment was 32.4%, 46.8% and 36.7%, respectively. This results indicate that antagonistic microbes with high ability of antifungal and lytic enzyme activity can be used as a useful biological control agent to control ginseng damping off.

Comparing of a Yearly Agronomic Characteristics and Yields of Festulolium Hybrids (Festulolium 교잡종의 연차간 생육특성 및 수량 비교)

  • This experiment was carried out to examine the winter hardiness, heading date, quality, and yields of Festulolium hybrids, Festuca pratensis and Lolium multiflorum, which were cultivated during September 2005 to July 2006 at Suwon and Namwon. The results are as follows; Perun, Boxer, and Fleurial showed stronger or same level of hardiness than Hwasan 101, which is one of the strongest hybrids in winter hardiness. Therefore, we did not detect any problem in winter hardiness when these hybrids were cultivated in Korea. These are turned out to be mid and late maturing hybrids. The average heading date of three hybrids were 24 to 27 May in Suwon and Namwon. These date are same or 3 days later than Hwasan 101. The average fresh yields of 3 hybrids were increased by $17{\sim}29%$ than that of Hwasan 101. And the dry matter yields of Perun, Boxer, and Fleurial were increased by 39%, 26%, and 29%, respectively, than that of Hwasan 101. We could not detect any difference in the average fresh and dry matter yields between region, however there were significant differences in hybrids(p<0.05). Crude protein contents of 3 hybrids were higher to $14.3{\sim}16.6%$ than that of Hwasan 101. The neutral detergent fiber and acid detergent fiber were decreased by 3.9 and 7.0%, respectively, comparing to Hwasan 101. Therefore these hybrids have turned out to be excellent feed quality hybrids.

Control of Phythophthora capsici and residual characteristics by drenching of pesticides on tomato in hydroponic culture system (약제 관주처리에 의한 양액재배 토마토의 역병 방제 및 농약잔류 특성)

  • To establish effective and safe control method against Phytophthora root rot caused by Phytophthora capsici on tomato in hydroponic culture, three pesticides, oxadixyl copper hydroxide 8% WP, metalaxyl copper oxychloride 15% WP, and dimethomorph. dithianon 38% WP at 4 concentration levels were tested on potato dextrose agar medium inoculated with Phytophthora capsici. All pesticides inhibited mycelial growth, but two pesticides of them, metalaxyl copper oxychloride WP and dimethomorph. dithianon WP, were selected as effective pesticides for the efficacy test in a hydroponic culture. Forty days after transplanting of tomato seedlings, 4 ml of sporangia of P. capsici (about 25 sporangi/ml) per plot was inoculated around tomato plant root, and then 5 days after inoculation, the pesticides diluted at 5,000 times were drenched 1, 2 or 3 times per plot on the culture cube at 15 days interval. Fifteen days after drenching, tomato fruits and hydroponic culture solution were sampled for the analysis of pesticide residues. Dimethomorph was detected 0.001 and 0.003 mg/kg in tomato of the plots sprayed 2 and 3 times with dimethomorph dithianon WP of which detection levels were far below compared with 1.0 mg/kg of the Korean MRL of dimethomorph on tomato. Incidences of Phytophthora root rot were $30.5{\sim}50%$ in the plots drenched at 1 or 2 times with metalaxyl.copper oxychloride WP, and $16.7{\sim}25%$ in the plots treated with dimethomorph dithianon WP. However, there was no incidence of Phytophthora root rot in the plots treated at 3 times with both of pesticides, showing no phytotoxic effect. Based on the results, the drenching of these pesticides on the culture cube could be recommended as a very safe and effective control method for Phytophthora root rot in tomato.

Effect of Mulching with Easily-Decomposable Organic Materials on the Rice Growth and Earthworm Ecology Characteristics in Paddy Fields (분해성 피복자재의 멀칭처리가 벼 생육 및 지렁이 생태에 미치는 영향)

  • This study was conducted to investigate the effects of mulching of easily-decomposable organic materials including rice powder and corn starch on rice paddy fields. The treatments were including seven mulching materials each consisted of rice powder [Rice], starch with black color [Starch-B] and transparent [Starch-T] and stone powder [Stone], and conventional vinyl [Conventional], no-mulching without herbicide [Control-O] and no-mulching with herbicide [Control-C]. Rice growth and yield were different among the treatments; the highest yields were observed for [Rice] ($6.51ton\;ha^{-1}$) and [Conventional] ($6.47ton\;ha^{-1}$) followed by [Starch-T] ($6.25ton\;ha^{-1}$) and [Stone] ($6.20ton\;ha^{-1}$) which is comparable to the [Control-C] ($6.15ton\;ha^{-1}$). However, [Starch-B] showed lower rice yield ($4.93ton\;ha^{-1}$) than the [Conventional] or [Control-C]. Mean soil temperature ranged form 23.6 to $24.1^{\circ}C$ with the highest temperature for [Rice] treatment, and cumulative soil temperature was also significantly higher in the [Rice] treatment. Among the soil parameters, electrical conductivity, organic matter content, and phosphorus concentration were also different among the treatments. The amount of earthworm casting was higher for [Rice] treatment ($17.7kg\;m^{-2}$) compared to the others ($5.5{\sim}9.8kg\;m^{-2}$). It was suggested that mulching of rice fields with organic materials containing rice powder is better than others in respected to rice yield and earthworm activity.

Control of Phythophthora capsici and Residual Characteristics by the Pesticides Tank-Mixed in Tomato Hydroponic Culture System (농약의 양액 탱크내 혼합처리에 의한 토마토 역병 방제 효과 및 잔류 특성)

  • To control effectively and safely Phytophthora root rot caused by Phytophthora capsici on tomato in hydroponic culture, tank-mixing method was considered with two pesticides, metalaxyl copper oxychloride 50% WP and dimethomorph dithianon 38% WP. Forty days after transplanting of tomato seedlings, 4 mL of sporangia of P. capsici (about 25 sporangi/mL) per plot was inoculated around tomato plant roots, and at 5 days after inoculation, the pesticides tank-mixed at three dilution levels, 12,500, 25,000 and 50,000, were drenched 1, 2 or 3 times per plot on the culture cube every 15 days for metalaxyl copper oxychloride 50% WP and every 10 days for dimethomorph dithianon 38% WP. During the drenching period, the residue levels of metalaxyl and dimethomorph in hydroponic culture solution were similar to the initial levels but the level of dithianon was drastically decreased from one day after tank-mixing. In tomato drenched with metalaxyl copper oxychloride 50% WP, metalaxyl was detected $0.02\sim0.04$ mg/kg in all diluted plots. Dimethomorph was detected $0.012\sim0.021$, $0.001\sim0.006$ and $0.001\sim0.003$ mg/kg in 12,500, 25,000 and 50,000 times diluted plots, respectively, while dithianon was detected 0.005, 0.003 mg/kg in 12,500 and 50,000 times diluted plots, respectively. The detection levels of three pesticides were far below compared with the levels of Korean MRLs. Incidences of Phytophthora root rot were not found in all the plots, but phytotoxic responses were recognized in the 12,500 times diluted plots of both pesticides. Based on the above results, the drenching of the culture solution tank-mixed with these pesticides could be recommended as a very safe and effective method to control Phytophthora root rot in tomato in hydroponic culture.

Assessment of Physiological Activity of Entomopathogenic Fungi with Insecticidal Activity Against Locusts (풀무치에 대하여 살충활성을 보유한 곤충병원성 진균의 생리활성 평가)

  • Locusts, Locusta migratoria (Orthoptera: Acrididae) are periodical unpredictable agricultural pests worldwide and cause serious damage to crop production; however, little consideration has been given to the management of this pest. Herein, we constructed a locust-pathogenic fungal library and confirmed that some fungi could be used as resources for locust management. First, the entomopathogenic fungi were collected from sampled soils using a Tenebrio molitor-based baiting system. For the locust assay, a locust colony was obtained from the National Institute of Agricultural Science and Technology. A total of 34 entomopathogenic fungal granules, which were produced by solid cultures, were placed in the plastic insect-rearing boxes (2 g/box) and nymphs of locust were contained in the box. In 3-7 days, mycosis was observed on the membranous cuticles of the head, abdomen, and legs of locusts. In particular, Metarhizium anisopliae, M. lepidiotae, and Clonostachys rogersoniana exhibited high virulence against the locust. Given that the 34 isolates could be used in field applications, their conidial production and stability (thermotolerance) were further characterized. In the thermotolerance assay, Paecilomyces and Purpureocillium isolates had higher thermotolerance than the other isolates. Most of the fungal isolates produced ca. >$1{\times}10^8conidia/g$ on millet grain medium. In a greenhouse trial, the granular application of M. anisopliae isolate on the soil surface resulted in 85.7% control efficacy. This work suggests that entomopathogenic fungi in a granular form can be effectively used to control the migratory locust.

An Evaluation of Polycross Progenies of the High-yielding Genotypes Derived from a Variety "Fawn" in Tall Fescue (Festuca arundinacea Schreb.) (톨페스큐 품종(品種) "Fawn"에서 선발(選拔)된 몇가지 다수성(多收性) 유전자형(遺傳子型)의 다계교배(多系交配) 후대검정(後代檢定)에 관(關)한 硏究)

  • This experiment was conducted to select genotypes that were high-yielding and adapted to the soil and weather conditions in Korea, from 13 Fawn deri ved progenies which represented in the previous study. Comparisons were made in various leaf and plant characteristics, and forage productivities, between the 13 polycross progenies of high-yielding Fawn and control varieties including Fawn, Alta, and Kentucky 31. The results obtained were summarized as follow; Genetic differences between the polycross progeny group and the control group were recognized in leaf fresh weight but those between two groups in leaf dry weight were not observed. Leaf area of all derived progenies and control varieties were appeared to have individual genetic differences and, moreover, the leaf area of the polycross progeny group was larger than that of the control. Differences between polycross progeny group and control group in leaf width were not significant. However, leaf length exhibited significant differences among all polycross progenies, and control varieties between two groups, and among polycross progenies but no differences were observed among control varieties. Tiller number, plant height, and plant width of the polycross progeny group were larger than those of the control varieties when measured before and after harvest. Panicle length among all polycross progenies and control varieties showed minor differences. However, significant differences were found between the polycross progeny group and control group, as well as among the polycross progenies and control varieties within each group in culm length. Above characteritics in the leaf fresh and dry weight, area and length, plant width and height, and tiller number showed positive correlations with the forage yields, respectively. Forage yields of the polycross progenies were higher than those of the control varieties and the variation of the yields among the genotypes were great.

Association between Dental Fear and Anxious Thoughts and Tendencies and Dental Experience of Dental Patients (치과내원 환자의 불안취약사고 경향 및 치과경험과 치과공포의 관련성)

  • Introduction The aim of this study, personal trait and their dental care based on experience level of dental fear is to analyze the differences. Catastrophizing is an irrational belief that something is far worse than it actually is. Because it makes it possible to analyze patients' anxious thoughts and tendencies. Methods The Dental Fear Survey(DFS) and Anxious Thoughts and Tendencies(AT&T) were used as measuring tools, and the independent sample t-test and ANOVA were performed on the basis of the mean value of the summated scale scores and the standard deviation. On the factors to dental anxiety, the multiple regression analysis was performed. Results Demographically, the total DFS score was higher in women($25.73{\pm}8.27$) than in men. Also, AT&T was higher in women($31.01{\pm}7.05$) (p<0.05). Dental fear was intense in the group of patients who visited dental clinics only problems($25.29{\pm}8.57$) than in those regular visited($22.29{\pm}7.78$). In relation to dental experiences, the DFS score was significantly higher in children. In many cases, patients experienced therapeutic pains in children($26.40{\pm}9.54$) (p=0.004). Also in many patients, dental anxiety began to occur in adolescence(44.3%). The multiple regression analysis result, it was found that dental treatment pain and their AT&T wielded great influence upon dental fear.

Characterization of New Bacillus thuringiensis Isolated with Bioactivities to Tobacco Cutworm, Spodoptera litura (Lepidoptera: Noctuidae) (담배거세미나방에 살충효과를 나타내는 새로운 Bacillus thuringiensis 균주의 특성)

  • Bacillus thuringiensis with selected high toxicities against tobacco cutworm, Spodoptera litura were isolated from domestic soils. When being observed under a phase-contrast microscope, the insecticidal crystal proteins were showed a bipyramidal crystal types. New CAB 109 isolate was identified to B. thuringiensis subsp. aizawai in the H serotype. As a results of insecticidal activities between CAB 109 isolate and 3 existing ready-made products against 3rd larva of S. litura, CAB 109 isolate showed 100% mortality with spore concentration $(1.3{\times}10^7cfu/ml)$. It was a very high insecticidal activity compared with a existing ready-made B. t. products. $LD_{50}$ values of CAB 109 isolate was $9.78{\times}10^5,\;6.87{\times}10^6\;and\;1.83{\times}10^7cfu/ml$ spore concentration against 2nd, 3rd and 4th larva of S. litura, respectively. Unlike Plutella xylostella, S. litura was slowly died after application up to 7 days. The weight of S. litura larva applied with CAB 109 isolate were 6-7 times less than controlled group. Even though it didn't die, it did not grow into next larva. The result observed with scanning electron microscope was that CAB 109 isolate of B. t. aizawai formed a typical bipyramidal crystal protein type. Otherwise, when CAB 109 isolate was examined with SDS-PAGE and with trypsin, there was no difference between CAB 109 strain and ready-made products of B. thuringiensis.

The study of awareness and practice of infection control on dental practitioners during the prosthodontic treatment (치과 보철물 제작 과정에서 감염 관리에 관한 치과 종사자의 인지도 및 실천도에 대한 연구)

  • Purpose: The aim of this study was to assess the degree of awareness and actual practice of dental practitioners regarding infection control during prosthodontic treatment. Materials and methods: Study participants were composed of dentists, dental hygienists, dental technicians working at dental university hospital, general hospital, dental hospital and dental clinics in Daejeon, Korea (n=126). The questionnaires included the data on general characteristics, education, awareness and practice of infection control procedures during prosthodontic treatment through personal self-administration survey. Awareness and practice were measured by Likert scale ranging from 1 to 5. Statistical analysis was done by one-way ANOVA. Influence of awareness on practice was estimated by regression analysis. Results: 27.7% of the respondents answered that the infection control education regarding prosthodontic treatment is insufficient. The mean value of awareness and practice of infection control during the prosthodontic treatment were $2.72{\pm}0.80$, $1.58{\pm}0.88$, respectively. The mean value of practice who answered that had education curriculum at college was $1.62{\pm}0.9$, and $1.31{\pm}0.49$ who did not. The mean value of awareness who equipped guideline was $3.01{\pm}0.9$, and $2.56{\pm}0.70$ who did not. Conclusion: 1. The respondents who received the infection control education by college curriculum showed higher degree of practice than the respondents who did not. 2. The respondents who have appropriate infection control guidebook in their workplace showed higher degree of awareness than the respondents who did not. 3. There was a correlation between the awareness and practice of infection control during prosthodontic treatment; the degree of awareness becomes higher, so does the degree of practice. 4. During the prosthodontic treatment, the degree of practice was lower than the degree of awareness on infection control.
