The objective of the present study was to investigate the inhibitory effect of physiologically pulsatile pattern of testosterone(T) on luteinizing hormone(LH) in wethers. To do this, 3 separate experiments were conducted. Infusion rates and patterns needed to produce normal T secretory profiles found in intact rams were established in Experiment 1, the time-course of the suppressive effect of T on circulating LH concentrations was determined in Experiment 2, and the effectiveness of a pulsatile versus a constant pattern of T to suppress LH secretion in wethers was compared in Experiment 3. In Experiment 1, three different doses(25, 50 or $100{\mu}g$) of T were injected intravenously to animals to do pharmacokinetic analysis of T. Elimination rate constant, volume of distribution, and total body clearance of T averaged $0.18min^{-1}$, 0.531/kg BW, and 0.091/min/ kg BW, respectively. In Experiment 2, three different doses(192,384, or $768{\mu}g/kg/24h$) of T were infused at 4h intervals for 3 days into animals to evaluate the time course of the inhibitory effect of T on mean LH concentration. As duration of T infusion increased, mean LH concentrations gradually reduced. Mean LH concentrations were significantly lower at day 2 or day 3 than at day 0. However, mean LH concentrations did not differ between day 0 and day 1 or between day 2 and day 3. In Experiment 3, animals were subjected to two different intravenous infusion regimens for 3 days: constant T($768{\mu}g/kg/24h$) and pulsatile(one pulse every 4h) T($768{\mu}g/kg24h$). Blood samples were collected at 10-min intervals for 4h both prior to infusion and during the last 4h of the infusion. Mean LH was more suppressed(p=0.045) by constant T than by pulsatile T. LH pulse amplitude was not affected by constant T or pulsatile T. LH interpulse interval was increased more(p=0.034) by constant T than pulsatile T.
Although there is a growing interest in male menopause, a phenomenon associated with male hormone depletion, current kits using antibodies to quantify male hormones are expensive. In this study, we constructed an in vitro system for verifying the activity or concentration of male steroid hormones using a transcriptional activity test. A reporter plasmid, pGL2-Neo-ARE-AdE1BTATA, which reacts to testosterone, was constructed. In this plasmid, the ARE-AdE1bTATA sequences can be bounded by the testosterone - androgen receptor complex to express luciferase as a reporter. Then, a stable transfection was performed on the human prostate cancer cell line, LNcap-LN3. The constructed LNcap-LN3/pGL2-Neo-ARE-AdE1BTATA testosterone compound detection (TCD) system showed quantitatively proportional luciferase activities to concentrations of $10^{-13}$ to $10^{-8}M$ of standard testosterone. The established in vitro TCD system will contribute to the development of materials for health/functional foods and drugs as it will be possible to search en masse for testosterone-like or testosterone-inhibiting substances derived from natural materials.
The purpose of this study was to screen the testosterone-like activities of eight oriental medicines with an in vitro testosterone compound detection (TCD) system. Distilled water and 80% ethanol, both at 80℃, were used to obtain extracts from Cervus nippon temminck (Nokgaksang), Cynanchum wilfordii (Baeksuo), Lespedeza cuneata (Yagwanmun), Panax ginseng (Hongsam), Smilax china (Toryeong), Taraxacum platycarpum (Mindlre), Tribulus terresteis (Namgase), and Trigonella foenum-graecum (Horopa), and extracts at concentrations of 5, 50, and 500 ㎍/ml were assessed using the TCD system. The testosterone-like activities of the 80% ethanol extracts were seen to increase with concentration and ranged from 0.9 to 5.0 times higher than those of the negative control; Horopa and Yagwanmun exhibited superior testosterone-like activities, followed by Mindlre and Hongsam at the same concentration. Toryeong and Namgase, on the other hand, showed low activity with Nokgaksang and Baeksuo exhibiting very low levels. Compared with the activities of the positive control (5α-DHT), the 80% ethanol extracts of Horopa and Yagwanmun at a concentration of 500 ㎍/ml showed higher testosterone-like activities than 10-8 M 5α-DHT. The water extracts showed lower activity levels than the ethanol samples, and the change of activity with concentration was also lower. It was confirmed that the 80% ethanol extracts of Horopa, Yagwanmun, and Mindlre etc. enhance the transcription efficiency of testosterone-related genes and may therefore be useful in developing health food or medicinal treatments for the improvement of male menopausal symptoms.

This study investigated the effects of a mixture of fenugreek seeds and Lespedeza cuneata extracts on testosterone synthesis in TM3 cells that were oxidatively stressed with $H_2O_2$. In order to oxidatively stress TM3 cells, the cells were treated with $50{\mu}M$ hydrogen peroxide for 4 hr in serum-free media. Yagwanmun-horopa mixture (YHM) showed neither cytotoxicity nor increment of cell proliferation in the oxidatively stressed TM3 cells in any concentration. When the cells were treated with hydrogen peroxide, testosterone levels decreased, but the testosterone level was returned to that of the control level in the presence of YHM. In order to find out the reasons for the increase of testosterone, the expression of the genes involved in the synthesis or disintegration of testosterone. On the other hand, the levels of $3{\beta}$-HSD4 and 17, 20-desmorase, which are involved in testosterone synthesis, were decreased through the use of hydrogen peroxide and were recovered through YHM treatment. Aromatase and $5{\alpha}$-reductase2, which convert testosterone to estradiol and dihydrotestosterone, respectively, were increased through the use of hydrogen peroxide, and were returned to control level through YHM treatment. These results suggest that YHM does not affect TM3 cell proliferation. However, YHM increases the expression of testosterone-synthesizing enzyme, which was decreased through oxidative stress, and decreases the expression of testosterone- converting enzyme, which was increased through oxidative stress. Therefore, it is reasonable that YHM has strong recovery activity on testosterone to normal level, even in the oxidatively stressed TM3 cells which mimics the andropause state.
The current study was undertaken to determine the effects of sex steroid hormones(estrogen, testosterone and 19-nortestosterone) on differentiation and proliferation of pig preadipocytes. The preadipocytes were isolated from the backfat of new-born female pigs by collagenase digestion. 10-8M and 10-7M sex steroid hormones were treated to the cultured preadipocytes. Sex steroid hormones treated during the early stage of cell growth did not affect differentiation and proliferation of preadipocytes. However, testosterone and 19- nortestosterone treated during the late stage of cell growth stimulated differentiation of pig preadipocytes.

Testosterone acts to protect the heart as a male hormone. If it is secreted suitably, health, but too less it will have an bad influence in organs of the body. In particular, when normal levels of testosterone secretion is less, arteriosclerosis will be increased risk of heart diseases. According to the amount of testosterone is influenced difference in the ring finger length of male. Therefore, we organize groups of subjects in heart diseases and normal who measure these groups' length of the ring finger and index finger. With this in mind, we carried out analysis of interrelationship in length of the ring finger and heart diseases through cross-comparison analysis in this paper.
Men's climactic syndrome, andropause, or testosterone deficit syndrome, is one of the new problems with the health of older men in the age of aging. This phenomenon is a natural phenomenon occurring in men as they age, clinically characterized by a decrease in blood testosterone levels and a marked decrease in physical and mental activity. The purpose of this study was to investigate the effect of hydrothermal extract of Taraxacum coreanum by comparing the levels of nitric oxide (NO) in the cavernosum and the levels of male hormone in the blood. Taraxacum coreanum extract increased NO production in vitro and in vivo in a dose-dependent manner. Levels of dihydrotestosterone and 17-hydroxyysteroid dehydrogenases, as well as levels of neurogenic nitric oxide synthase and cGMP, increased significantly in elderly rats (22 weeks) after 4 weeks of daily intake of Taraxacum coreanum extract. However, prostaglandin $E_2$, testosterone, and sexually-hormone-binding globulin levels were not different among all groups. Furthermore, total sperm and motile sperm counts were also no significant difference. Overall, these results suggest the possibility of Taraxacum coreanum extract as a safe and effective natural substance for enhancing NO, cGMP and free testosterone.

Results of this study were as follows: 1. There were no significant differences in the semen volume, pH and sperm concentration of Duroc boars between spring and summer. 2. Sperm motility and normal acrosome of raw semen in Duroc boars did not differ significantly between spring and summer. However, motility and normal acrosome of frozen-thawed sperm were higher in spring season than in summer season(P<0.05). 3. Serum testosterone concentrations in Duroc boars were 2.15 ng/$m\ell$ in spring and 0.65 ng/$m\ell$ in summer. Serum testosterone concentrations in spring were higher thin those in summer (P<0.05). 4. In conclusion, when serum testosterone concentrations in Duroc boars were higher, frozen-thawed sperm viability were higher.

In conclusion, when serum testosterone concentrations in Yorkshire boars were higher, frozen-thawed sperm viability was higher.
Cordyceps militaris(CM) has been used as a traditional folk medicine to strengthen the immune system and regain energy for hundreds of years in Far East Asian countries. In the present studies, we determined that the stimulatory effect of Cordyceps militaris on testosterone production in male mouse rats. Results illustrated that changes of the body weight, food and water intake of the rats were not observed in this study but the concentration of testosterone in the serum of the rats was significantly increased by CM(p < 0.05). Therefore fruiting bodies of CM grown on the drone bee medium may be an integrative medicine for the treatment of reproductive problems caused by insufficient testosterone levels in human males.
