Inhibitory Effects of Dunaliella salina Extracts on Thermally-Induced Skin Aging (두날리엘라 살리나 추출물의 피부 열노화 억제 효과)

  • Just like UV radiation, heat increases collagen degradation and accumulation of abnormal elastin fiber and this is termed thermal skin aging. Dunaliella salina (DS), a green alga, is known for its beta-carotene accumulation, having various applications in the health and nutritional products. However, the effects of DS on heat-induced skin aging remain unexplored. In this study, we performed anti-thermal aging tests of the ethanol extract of DS (DSE). We measured the cellular levels of type I procollagen and MMP-1 using ELISA in human dermal fibroblast cells after heat shock. DSE reduced the expression of MMP-1 protein and increased the expression of type I procollagen. In addition, DSE upregulated the mRNA expression of HSP47 reduced by heat shock, which is involved in collagen synthesis. Also, DSE reduced the expression of inflammation mediator (TGF-${\beta}$, IL-12, etc). We demonstrate that DSE regulates the heat-induced solar elastosis through the regulation of tropoelastin and fibrillin-1, two major proteins of elastic fibers, and MMP-12 expression. These results suggest that DSE may be effective for preventing thermally induced skin aging.

  • Shaving dust is a collagen fiber that is the leather waste occurred for thickness adjustment during the natural leather manufacturing process, and causes problems such as an environmental contamination because of a chromium (Cr) contained when it comes to reclaiming process. Various studies applying the shaving dust are currently being conducted in many countries across the world with an initiative by the EU. Of those applications, the bonded leather is being highlighted as a substitute for natural leather. Since the bonded leather, however, uses latex as a binder, accordingly it entails a high weight and a poor ventilation, which are deemed as disadvantages due to its dense internal tissues compared to other synthetic leathers. To address such disadvantages, this study employed the thermally expandable micro sphere to improve its air permeability and light weight by alleviating the internal structure. This is a study on the manufacturing of light bonded leather using the shaving dusts. In the study, the shaving dusts were forced to foam under 100~120℃ considering the heat resistance of collagen fiber after applying the thermally expandable micro sphere, and then the tendency was analyzed. In the analysis results, the most excellent foaming rate was exhibited when the shaving dusts were treated under 120℃ for 8 minutes and the variation of internal structure according to a foaming was observed through SEM analysis for the cross-section of the bonded leather.

Anti-wrinkle effects of extracts and solvent fractions from Nymphoides peltata on CCD-986sk (CCD-986sk 세포내에서의 노랑어리연꽃 추출물 및 분획물의 주름개선 효과)

  • The purpose of this study was to investigate the antiaging effect in the fibroblasts by using the extracts and fractions of Nymphoides peltata for the development of new functional cosmetic materials. The anti-aging effects of the extracts and fractions of N. peltata were measured using CCD-986sk cells. Ethyl aceate (EA) fraction inhibited UVB-induced reactive oxygen species expression by more than 50%, and n-butyl alcohol (BuOH) fraction increased expression of pro-collagen mRNA by more than 50% at $25{\mu}g/mL$. In addition, EA and BuOH fractions inhibited MMP-1 protein and mRNA expression in a concentration-dependent manner, BuOH fraction inhibited MMP-1 mRNA expression by more than 40% at $10{\mu}g/mL$. Especially, the high efficacy of EA and BuOH of the N. peltata was confirmed. Therefore, the EA and BuOH of N. peltata are expected to be useful as anti-aging materials in cosmetics field.

Effect of Green Tea Catechins on the Expression and Activity of MMPs and Type I Procollagen Synthesis in Human Dermal Fibroblasts (사람 섬유아세포에서 녹차 카테킨이 노화 인자인 MMP와 type 1 Procollagen 발현에 미치는 영향)

  • Although many studies have been performed to elucidate the molecular consequence of factors that regulate skin aging, little is known about the effect of green tea catechins except EGCG. The matrix metalloproteinase (MMP), can degrade matrix proteins and results in a collagen deficiency in photodamaged skin, are known to play an important role in photoaging. This study, investigated the effects of green tea catechins on the UVA-induced MMP-1 expression, activity of MMP-2 and synthesis of type I procollagen in human dermal fibroblasts. We examined eight catechins that naturally exist in green tea leaves and compared their efficacies among them. Most of catechins inhibited the expression of MMP-1 in dose dependent manner, and the levels were reduced, especially, 57.4 and 68.2% by treatment with $1{\mu}M$ of epigallocatechin-3-gallate (EGCG) and gallocatechin-3-gallate (GCG), respectively. Also, catechins significantly suppressed the activities of MMP-2. Catechins also induced the expression of type I procollagen, however, they acted only at the concentration below $1{\mu}M$ interestingly. Furthermore, when EGCG:GCG:ECG had the ratio of 0.5:1.5:.1.3, they presented the most effective on procollagen synthesis. Therefore, we concluded that catechins significantly inhibited MMPs and induced collagen synthesis. Taken together, all these results suggested that green tea catechins might be good natural materials act as an anti-photoaging and a skin-aging improving agent.

  • Innate immunity is the ability to differentiate infectious agents from self. The innate immune system is comprised of a complicated network of recognition and effector molecules that act together to protect the host in the early stage of an infectious challenge. Mannose-binding lectin (MBL or mannose-binding protein, MBP) belongs to the family of $Ca^{2+}$-dependent lectins (C-type lectin with a collagen-like domain), which are considered an important component of innate immunity. While it is associated with increased risk and severity of infections and autoimmunity, the most frequent immuno-deficiency syndrome was reported to be low MBL level in blood. Deficiency of human MBL is caused by mutations in the coding region of the MBL gene. Rat homologue gene of human MBL gene was used to study functions of wild type and mutant MBL proteins. Although extensive studies have yielded the structural information of MBL, the functions of MBL, especially mutant MBL, still require investigation. We previously reported the cloning of rat wild-type MBL gene and the production of a truncated form of MBL protein and its antibody. Here, we present the cloning of mutant MBL cDNA in collagen-like domain (R40C, G42D, and G45E) using site-directed mutagenesis and differential behaviors of wild type and mutant MBL in cells. The major difference between wild type and mutant MBL was that while wild type MBL was secreted, mutant MBL was inhibited for secretion, retained in endoplasmic reticulum, and still functioned as a lectin.

A Study on Anti-oxidant and Anti-wrinkle Effect of Supercritical Fluid Extraction of Black Carrot as a Functional Cosmetic Material (기능성화장품소재로서 자색당근 초임계추출물의 항산화 및 항주름 효능 연구)

  • The aim of this study was to evaluate the anti-oxidant and anti-wrinkle effect of the black carrot(BC) extracted by supercritical dioxide(SC-CO2). DPPH/ABTS radical scavenging and total polyphenol contents were measured to investigate the anti-oxidant activity of the BC supercritical extract. Collagen production and MMP-1 expression were assessed in normal human dermal fibroblasts(NHDF) for anti-wrinkle activity, The black carrot extract showed the highest total phenolic content(9.037±1.123 mg GAE/g extract) and the best antioxidant properties as shown by DPPH and ABTS assay. The supercritical fluid extracts of black carrot exhibited low toxicity to NHDF cells. In addition, the supercritical fluid extracts showed MMP-1 inhibition and type I pro-collagen synthesis inducing activities in a dose-dependent manner, respectively. Therefore, these results suggest that the black carrot is a potential source of high anti-oxidative, solvent-free and anti-aging material with promising applications in cosmetic, food, and beauty-food industries.

Anti-aging Effect of Asterosaponin P1 Isolated from Asterina pectinifera (별불가사리로부터 분리된 Asterosaponin P1의 항노화 효능)

  • The starfish, Asterina pectinifera Muller and Troschel (Asterinidae) is an indigenous species commonly found in all coasts of Korea causes damages to shellfish farms. In order to exterminate A. pectinifera, they are dried and used as fertilizer. Although various studies have been conducted to create high added value from the retrieved A. pectinifera, their actual utilization is relatively low. Accordingly, this study aimed to find new practical uses of starfish by identifying useful ingredients for skin anti-aging. Two polyhydroxysteroids and one asterosaponin were isolated from the A. pectinifera. The structures of these compounds were identified as $5{\alpha}$-cholestane-$3{\beta},6{\alpha},7{\alpha},8,15{\alpha},16{\beta},26$-heptol, $5{\alpha}$-cholestane-$3{\beta},4{\beta},6{\alpha},7{\alpha},8,15{\beta},16{\beta},2$6-octol, and asterosaponin $P_1$ on the basis of chemical and spectroscopic analysis. Among these compounds, we have found that asterosaponin $P_1$ increased epidermal stem cell proliferation and the expression of hyaluronan synthase-2 and hyaluronan synthase-3 gene, which are enzymes that synthesize water-binding matrix hyaluronic acids in keratinocytes. In addition, asterosaponin $P_1$ increased synthesis of pro-collagen type I, a major dermal collagen in fibroblasts. As a result, asterosaponin $P_1$ isolated from A. pectinifera could be used as a useful cosmetic ingredient that improves skin symptoms accompanying skin aging.

Effects of Aucklandia lappa Decne. Extract on Hair Growth in Depilated CL57BL/6 Mice (CL57BL/6 마우스의 발모에 미치는 목향 추출물의 영향)

  • Aucklandia lappa Decne., a traditional herbal medicine, has been used to treat various diseases, including asthma, coughs, rheumatism, and diarrhea. In the present study, the effects of the oral administration of Aucklandia lappa Decne. extract on hair growth was investigated in hair-removed mice. A 70% ethanol extract of Aucklandia lappa Decne. (ALD) was prepared, and the extraction yield and total polyphenol content of ALD were measured as 27.30±0.01 and 28.39 mg gallic acid equivalent (GAE)/g, respectively. The oral administration of ALD to hair-removed CL57BL/6 mice for six weeks had no significant effects on food intake and body weight changes. Biochemical and histological examinations also showed that the oral administration of ALD for six weeks had no significant effect on the liver and kidney functions. On the other hand, hair growth was significantly higher in the ALD group than the control group and the Pancidil group (positive control). In addition, the number of hair follicles and the degree of collagen production in the dermis were significantly higher in the ALD group than in the control and pancidil groups. These results suggest that ALD is a potential source of nutricosmetics with hair growth-promoting effects.

Chitosan-alginate Gel Modified Poly (L-Lactic-co-ε-Caprolactone) (PLCL) as a Scaffold for Cartilage Tissue Engineering (변형된 키토산 알지네이트 겔 poly (L-Lactic-co-ε-Caprolactone) 지지체의 연골 조직 재생 평가)

  • This study was designed in the fabricated poly (L-Lactic-co-${\varepsilon}$-Caprolactone) (PLCL) scaffold using chitosan-alginate hydrogel, which would be more suitable to maintain the biological and physiological functions continuing three dimensional spatial organizations for chondrocytes. As a scaffold, hydrogels alone is weak at endure complex loading within the body. In this study, we made cell hybrid scaffold constructs with poly (L-Lactic-co-${\varepsilon}$-Caprolactone) (PLCL) scaffold and hydrogels to make a three-dimensional composition of cells and extracellular matrix, which would be a mimic of a native cartilage. Using a particle leaching technique with NaCl, we fabricated a highly-elastic scaffold from PLCL with 85% porosity and $300-500{\mu}m$ pore size. A mixture of bovine chondrocytes and chitosan-alginate gel was seeded and compared with alginate as a control on the PLCL scaffold. The cell maturation, proliferation, extracellular matrix synthesis, glycosaminoglycans (sGAG) production and collagen type-II expressions were better in chondrocytes seeded in chitosan-alginate hydrogel than in alginate only. These results indicate that chondrocytes with chitosan-alginate gel on PLCL scaffolds provide an appropriate biomimetic environment for cell proliferation and matrix synthesis, which could successfully be used for cartilage repair and regeneration.

Development of Anti-Wrinkle Materials using Galloyl-Peptide Derivatives (갈릭산 펩타이드 유도체를 이용한 주름개선 소재 개발)

  • Conjugating a phytochemical, a strong antioxidant, with a functional peptide not only compensates for its stability, but also improves its solubility and anti-wrinkle effects, thereby contributing to the possibility of becoming an excellent cosmetic ingredient. Thus, in this study we examined the potential cosmetic use of a phytochemical-peptide derivative using gallic acid, a phytochemical with antioxidant, anti-inflammatory, and anti-cancer effects. To evaluate the antioxidant and wrinkle-improving efficacy of 5 synthesized gallic acids conjugated with LVH, IVH, KTTKS, YGGFM, and YGGFLRKYP respectively, we observed the expression of genes related to wrinkle improvement using DPPH radical scavenging activity and real-time PCR. As a result, all 5 derivatives had excellent free radical scavenging effects. The expression level of genes involved collagen synthesis also increased, and the secreted peptides during collagen production contributed to their antioxidant and wrinkle improving effects. These results mark the potential use of gallic acid peptide derivatives as a cosmetic ingredient for anti-oxidation and wrinkle improvement.
