Schiff base ligand columns and glow discharge detector have been developed for the trace analysis of metal ions desolved in water. Various types of hydrazide Schiff base ligands have been used and, additionally, they were examined as a filling material of a HPLC column. The hydrazide Schiff base ligands used were N, N'-oxalybis(salicylaldehydrazone) (OBSH), N, N'-malonylbis(salicylaldehydrazone) (MBSH), and N, N'-succinylbis(salicylaldehydrazone) (SBSH). A mixture of Schiff base ligand and poly(styrene divinylbenzene) was examined and it showed a smooth flow of solution. The OBSH-polymer column demonstrated different effluent factors for different metal ions. Metal ions in eluates were detected by Hollow Cathode Glow Discharge-Atomic Emission Spectrometry(HCGD-AES). HCGD-AES showed good sensitivity and selectivity. This is only the preliminary results of new OBSH-polymer column and glow discharge detector.
In this paper, we propose a forward vehicle detection algorithm using column detection and bird's-eye view mapping based on stereo vision. The algorithm can detect forward vehicles robustly in real complex traffic situations. The algorithm consists of the three steps, namely road feature-based column detection, bird's-eye view mapping-based obstacle segmentation, obstacle area remerging and vehicle verification. First, we extract a road feature using maximum frequent values in v-disparity map. And we perform a column detection using the road feature as a new criterion. The road feature is more appropriate criterion than the median value because it is not affected by a road traffic situation, for example the changing of obstacle size or the number of obstacles. But there are still multiple obstacles in the obstacle areas. Thus, we perform a bird's-eye view mapping-based obstacle segmentation to divide obstacle accurately. We can segment obstacle easily because a bird's-eye view mapping can represent the position of obstacle on planar plane using depth map and camera information. Additionally, we perform obstacle area remerging processing because a segmented obstacle area may be same obstacle. Finally, we verify the obstacles whether those are vehicles or not using a depth map and gray image. We conduct experiments to prove the vehicle detection performance by applying our algorithm to real complex traffic situations.

The portable gas chromatography system was developed which was consisted of ambient vapor sampler(AVS), short capillary column(3 m long, 0.32 mm i.d. GC(SCCGC), photoionization detector (PID) and vacuum pump which was operated at subambient pressure. The seletion of capillary column was based on the theoretical calculation from Golay equation. The pressure ratio of column inlet and outlet appropriated between 1.03 and 1.2 in the system. The available column flow were 0.87∼4.63 ml/min at the pressure ratios. The AVS consisted of three concentric tubes and enables rapid, repetitive introduction of vapor samples directly into capillary column and showed good reproducibility. The subambient column outlet pressure with PID resulted in a significant increase in the optimum column flow, permitting rapid analysis. The baseline separation of m-xylene and o-xylene was able to within 40 second with the system. Parameters affecting the column resolving power were sampling duration, column length and diameter, and the pressure ratio. Effects of these parameters were investigated using bezene derivative compounds.
Simple post-wash method by ion chromatography (IC) was established for the rapid and precise determination of fluoride ion in wastewater from mine in fluorite mineralized area. High sulfate in sample was retained in a pre-column and less strongly held fluoride ion was transferred to the principal separation system using modified conventional IC with switching technique. An analytical column with high capacity (AS 9 HC) was used as a pre-column to retain the amount of high sulfate. A guard column (AG 14) as a separation column was used to increase the response of fluoride and reduce the system pressure. According to the recovery of fluoride ion with one detector and the observation of sulfate peak with another conductivity detector, the optimum switching time of 10-port chromatographic injector was 4.3 min. The limit of detection (S/N = 3) of fluoride in synthetic solution containing $500mg\;L^{-1}$ sulfate was $2.4{\mu}g/L$, with $25{\mu}L$ sample volume.
Black soybean has been widely utilized as foods and oriental medicinal materials. The pigmentation in the seed coat of black soybean is due to accumulate anthocyanins in the epidermis palisade layer. The anthocyanins of black soybean seed coat are considered as a parameter of quality evaluation of black soybean. Therefore, the purpose of this study was to investigate the most suitable HPLC condition for simultaneous determination of anthocyanins in black soybean seed coats extracts. The efficient HPLC analytical condition of D3G, C3G, and Pt3G contained extracts of black soybean seed coats was developed. The gradient elution employed a $250\;mm\;{\times}\;4.6\;mm$ i.d. YMC-pak ODS-AM 303 column. The gradient system was used two mobile phases. A gradient elution was performed with mobile phase A, consisting of 5% aqueous formic acid, and mobile phase B, comprising 5% formic acid - acetonitrile, and delivered at a flow rate of 0.7 mL/min as follows: $0{\sim}35\;min$, $90%\;A{\sim}60%\;A$; 36 min, 90% A; 46 min, 90% A. The UV-VIS. detection wavelength was set at 520 nm. The limit of detection (LOD) for D3G, C3G, and Pt3G were under 10 ng/mL.

In this study we extracted DNA from 100 tissues of breast cancer patients and 103 normals. Then we confirmed single-nucleotide polymorphism(SNP) using PCR-DHPLC(polymerase chain reaction-denaturing high performance liquid chromatogrphy).Also, we studied SNP of samples using several columns to identify relation between packing materials of column and resolution.As a result, we identified 4 C/A, C/G genotypes(4%) in exon 5 and 37 T del genotypes(37%) in exon 8 among 100 breast cancer tissues and 2 in exon 5, 9 in exon 8 among 103 normal samples.In resolution test, we confirmed that PS-DVB(poly styrene-divinylbenzen) column is more efficient than C18 column.
This study was investigated to elucidate the pesticide mobility in three different soil textures(Heongseong sandy loam; Chuncheon, loam; Taeback, silty clay loam) and four different gravel contents(0, 20, 40, 60%) of Taebaek soil using soil column. Carbofuran, which ranks the highest water solubility among 7 pesticides(carbendazim, carbofuran, chlorpyrifos, cypermethrin, dimethomorph, diniconazole and endosulfan) was defected over 87% in leachate samples within all soil types from early sampling time. Amount of 5 residual pesticides excluding carbendazim and carbofuran were ordered silty clay loam > loam > sandy loam, indicating pesticide residues are related to percentage of clay contents in soils. Comparing the amount of residual pesticides in soil column(upper, middle and lower layer), 6 pesticides apart from carbofuran were found in the range of $50{\sim}92%$ on the upper layer of silty clay loam and loam. Mobility of pesticides either in soil or leachate samples is dependant on water solubility of pesticide and clay content of soil. The results obtained from four different gravel contents of Tacback soil were similar to the results of three different soil textures. Also it was found that more the gravel contents, faster the flow velocity of leachate water. These results possibly provide an idea to select proper pesticides and to reduce soil and water contamination at alpine and sloped-land.

Platycodin D was isolated from n-butanol extract of Platycodi radix(Platycodon grandiflorum and identified by the spectroscopic analysis using $^1H$ and $^{13}C$ NMR techniques. A new method of analysis of platycodin D by high performance liquid chromatography(HPLC) was established using a reversed phase system with YMC-Pack ODS-AM( 250 X 4.6 mm) column and 30% acetonitrile as a mobile phase. Evaporative light scattering detector was used as detector. The kinds of extraction solvents and methods were examined to determine the efficient extraction condition and HPLC analysis was carried out to establish the optimum drying condition for the root of Platycodon grandiflorum. The contents of Platycodin D was highest as 0.083% when platycodon roots were dried at $60^{\circ}C$ using dry oven.
Rapid and simple method was developed for simultaneous determination of vitamins $D_3\;and\;K_1$ contents in infant formula. Contents of vitamins $D_3\;and\;K_1$, extracted by column-switching HPLC with reversed phase column using enzymatic hydrolysis and organic solvent, in CRM determined by developed method were within certified ranges of standard values.
The separation and determination method of racemic isomers of flurbiprofen in urine samples have been investigated using column switching- HPLC, which include the sample treatment and concentration column. The optical rotation of two isomers separated were measured to identify d-form and l-form. The calibration curves are liear in the ranges of $0.11-5.4{\mu}g/mL$ for both d-form and l-form. The detection limits were $0.031{\mu}g/mL$ for l-isomes and $0.027{\mu}g/mL$ for d-isomer. The coefficients of variation of intra-day and inter-day precision of this method were about 1.8%. The present method was apllied to determine the concentration of racemic isomers in urine sample from human eating the drug.
