Development of Measurement System for Quantitative Measurement of Cantilever in Atomic Force Microscopy (원자간격 현미경의 캔틸레버의 정량적 특성평가를 위한 계측 시스템 개발)

  • In this study, the two methods of stiffness measurement(Spring constant) of cantilever were proposed for quantitative measurement in Atomic Force Microscopy(AFM). As the 1st method for the measurement of stiffness, the probe method, which is used in the measurement of the semiconductor mechanical and electrical properties, was applied to the measurement of the cantilever. Experiments by the probe method were performed finding the resistance value of cantilever. As the results, the resistance was measured differently along with the dimension and the thickness of cantilever that determined the stiffness(spring constant) of the lever. As the 2nd method, the vibration characteristics(Dunkerley expression) is used to obtain the stiffness of the complex structure which is combined by AFM cantilever and the standard cantilever. We measured the resonant frequency from the complex structure using the micro stages and stereo microscope. As the results, we confirmed that the vibration characteristics(Dunkerley expression) is effected the micro complex structure of AFM cantilever.

Proper Orthogonal Mode Analysis of AFM Microcantilevers in Dynamic Mode (동적모드 AFM 마이크로캔틸레버의 적합직교모드 해석)

  • Proper orthogonal decomposition (POD) is a method for extracting bases for modal decomposition from the ensemble of dynamic signals. Using the POD method, we analyzed the proper orthogonal modes (POMs) of AFM microcantilevers in dynamic mode operations such as Tapping Mode. The POMs and POVs (proper orthogonal values) were computed through MATLAB simulation for the 5-mode model of the microcantilever. We found that the POV portion of the higher POMs of the tapping microcanilever slightly increased in comparison with no tapping. This implies that the modal energy in the fundamental mode can be transferred to the higher modes during tapping.

  • Proper orthogonal decomposition(POD) is a method for extracting bases for modal decomposition from the ensemble of signals. We verified the connection of the proper orthogonal modes(POMs) and the linear normal modes(LNMs) through MATLAB simulation for the simple cantilever and AFM microcantilever models. Using the POMs, we can analyze and model effectively the dynamic mode of AFM microcantievers.

  • This paper deals with the free vibrations of cantilever arches with constant volume. Its cross-sectional shape is the regular polygon whose depth is varied with the linear functional fashion. The non-dimensional differential equations governing the free vibration of such arch are derived and solved numerically for calculating the natural frequencies. As the numerical results, the effects of arch parameters such as side number of cross section, section ratio and aspect ratio on the natural frequencies are reported in figures.

  • A method is developed for solving the elastica of cantilever beam subjected to a tip point load and uniform load. The Bernoulli-Euler differential equation of deflected beam is used. The Runge-Kutta method and the Regula Falsi method are used to perform the integration of the differential eqution and to determine the horizontal deflection, respectively. The horizontal and vertical deflections of the free end, and the free-end rotations are calculated for a range of parameters representing variations in tip point load and uniform load. All results are presented in nondimensional forms. And some typical elastic are also presented.

  • In this paper, we present details on fabrication of single-cell electroporation microdevice, practical experiments of single-cell electroporation with our fabricated microdevice. Also, the continuous electroporation for the continuous flow of cells is used for high-throughput electroporation. The delivery efficiency and cell viability tests are provided and the successful GFP transfection into cells is also evaluated with a fluorescent microscope after electroporation. This device enables to reduce the size of samples and thus the use of small amount of reagents. Also, it makes it possible to permit to avoid cell discrimination (transfected cells versus non-transfected cells) encountered when traditional bulk electroporation is held.
