Effect of Temperature and Dissolved Oxygen on the Survival Rate and Physiological Response of the Warty Sea Squirt Styela clava (수온과 용존산소 변화에 따른 미더덕 Styela clava의 생존율 및 생리적 반응)

  • Decrease in dissolved oxygen concentrations associated with temperature fluctuation is an important criteria to evaluate the mortality rate of the species. Based on this parameter, we investigated the survival rate, physiological response and histological change of warty sea squirt. It was found that the survival rate of the warty sea squirt species was 63.3% at $23^{\circ}C$ and 56.6% at $26^{\circ}C$ respectively. However, exposure of six days at $29^{\circ}C$ caused deaths among species, which indicated the 6day-$LT^{50}$ of the tested species to be $24.58^{\circ}C$ ($19.48{\sim}35.48^{\circ}C$). Further, after 11 day of exposure, the dissolved oxygen concentration has been found to decrease, with the survival rate of 20% at $4.0mg\;L^{-1}$ and deaths at $2.0mg\;L^{-1}$, thus 11day-$LC^{50}$ calculated to be $3.88mg\;L^{-1}$ ($3.29{\sim}4.57mg\;L^{-1}$). In addition, decrease in rate of oxygen consumption and excretion of ammonia was also noted at this critical water temperature and dissolve doxygen concentration. Moreover, there has been common histopathological changes were observed in warty sea squirt's gill pouch, digestive tract, and tunic as follows such as: proliferation of epithelial cells, condensation and necrosis, permeation of phagocyte and blood cell, loss of cilium and muscular fiber degeneration. Based on our study results, we suggest that these parameters can also be useful to evaluate the survival rate and physiological response in other species.

Effects of Electrical Stimulation on the Biochemical Properties of Plaice, Paralichthys olivaceus, Sarcoplasmic Reticulum and Myofibrils (넙치 근소포체 및 근원섬유의 생화학적 특성에 미치는 전기자극의 영향)

  • This study was undertaken to clarify the effects of electrical stimulation on the biochemical properties of plaice sarcoplasmic reticulum and myofibrils at early period of death. The plaices were electrically stimulated (110V/60 Hz) In sea water bath for 15, 35, and 60 seconds, and killed instantly by spiking at the head. Killed samples were investigated for the changes in $Ca^{2+}$-ATPase activity of FSR (fragmented sarcoplasmic reticulum), LSR (light SR), HSR (heavy SR), and SDS-PAGE pattern of FSR. $Ca^{2+}$-ATPase activity of FSR increased until $45^{\circ}C$ and inactivated over $50^{\circ}C$. $Ca^{2+}$-ATPase activity of FSR remarkably decreased according to the duration of electrical stimulation. Myofibrillar $Mg^{2+}$-ATPase activity of electrically stimulated plaices in the presence of $Ca^{2+}$ was higher than that of sample instantly killed by spiking. $Mg^{2+}$-ATPase activity of myofibrils increased by electrical stimulation and the activity decreased during storage at $5^{\circ}C$. Myofibrillar $Mg^{2+}$-ATPase activity in sample killed by spiking was not affected by $Ca^{2+}$ ion. Myofibrillar $Mg^{2+}$-ATPase activity of electrically stimulated sample in the absen-re of $Ca^{2+}$ decreased during storage at $5^{\circ}C$, whit $Mg^{2+}$-ATPase activity in unstimulated sample did not show any change. $Ca^{2+}$-sensitivity of myofibrils showed no differences between electrically stimulated sample and sample killed by spiking, and the was no change during at $5^{\circ}C$.

  • This study was aimed at expanding the tea plant culture in Korea to further north, and the leaf-form and the cold resistance of the tea plants selected form 7 districts(around Korea) were investigated. 1. The length of tea leaf is 5.1cm to 8.4cm, the width of the leaf is 2.3cm to 3.6cm, but the area of tea leaf in Mt. Mudung is $26.88cm^2$ and that is the widest of all. 2. The tooth number of the tea leaf in 43 to 73 but the number of Mt. Borim is remarkably number of all. The tooth number is increased or decreased in proportion to the leaf width and to the leaf length. 3. The lateral vein number is generally 13 to 19. The vein number of Mt. Borim is especially number of all. That is also increased or decreased in proportion to the leaf width and to the leaf length. 4. In general, the number of leaf formation index is 2 to 3, and ablong. 5. In general, those which come upper lands are remarkably small in length and width of the tea leaf and those which come from level lands are large. 6. All kinds of tea plant which is growing in Korea, area, are the same as those imported from China : Thea sinensis Linne var. Bohea. 7. I supposed that the external form of tea plant has a little changed by geography or climate for many a long day, since the tea plant had been transplanted in Mt. Samsin. 8. In the treatment of low temperature and duration of vernalization of their plants, those selected from Mt. Mudung and Mt. Hwaun were the coldest resistance, those from Mt. Samsin and Mt. Borim were medium and those from Mt. Joge and Nursery were cold sensitive. 9. The critical temperature of the tea plant from Mt. Mudung, Mt, Hwaum, Mt, Samsin and Mt. Borim was about $-10^{\circ}C$, and that from Mt. Joga and Nursery was about $-5^{\circ}C$. The critical temperature of frost injury of all tea plants in this experiments was $-15^{\circ}C$. 10. In spite of increasing the vernalization time, the critical temperature was not effected, but the treatments over critical temperature were increased their frost injuries. Based on these results, the coldest resistance, Mt. Mudung tea plant, was considered expanding their culture to further north improvement yields of the tea plants in Korea.

  • The purpose of this study was to monitor physicochemical changes of olive flounder (Paralichthys olivaceus) muscle by iced water pre-treatment. Moisture content, crude fat content, nucleotide content, texture (hardness and toughness), and rigor mortis were assessed. The sensory evaluation was performed with a nine-point hedonic test. K-values, a parameter of fish flesh freshness, were also calculated from the content of nucleotides and their corresponding decomposition products. Pre-treatment of flounder flesh with iced water was found to be fresher compared to the control, as determined by a difference in the K-values. Iced water pre-treatment hastened postmortem stiffness, as judged from the rigor index, and increased inosine monophosphate (IMP), which is known to be a savory taste compound, more quickly as adenosine triphosphate (ATP) degradation proceeded.

  • In the present study, in succession to the previous reports, the sterilizing values ($F_0$) of the thermal processes for the canned minced hen-clams in brine and the canned smoked baby-clams in oil were determined ana discussed. The heat penetration tests were carried out three times with three cans at a time for each canned product. The thermocouple was setted on the can so as the tip of the applicaotor fixed on the position a little below the geometrical center of the can. The test cans were placed in the middle layer of the crate in which the same canned products were loaded with, and test cans were arranged to the front, the middle and the rear in the retort. The heat penetration curve obtained for the canned minced hen-clams in brine showed a broken logarithmic heating curve, while that of the canned smoked baby-clams in oil showed a simple logarithmic heating curve. The calculated $F_0$ values for the canned minced hen-clams in brine were 47.79 for No. 2 can, 52.99 for Ne. 7 can. and 45.21 for No. 2 tuna can, respectively. And the $F_0$ value for th canned baby-clams in oil packed into No. 3B square can was 14.12. Additionally, the nomographs represent the relationship between $F_0$ vlaues and B values (process time including $42\%$ of come-up time) for the each canned product were constructed.

Cold Tolerance Assessment of Lagerstroemia indica and Pyracantha angustifolia with Dormant Branches (배롱나무와 피라칸사 겨울 휴면지의 내한성 평가)

  • This study was conducted to check danger of the cold injury by reviewing the lowest temperature by regions and the cold tolerance of Lagerstroemia indica and Pyracantha angustifolia. The cold tolerance of the samples treated at low temperature was evaluated by measuring electrical conductivity and observing browning of the cambium. It was proved that the lethal temperature of L. indica is below $-17.1^{\circ}C$, and that of P. angustifolia is below $-18.9^{\circ}C$. The frequency of the lowest temperature went down below $-17.1^{\circ}C$, was 7 times in Daejeon, 55 times in Cheongju and 72 times in Suwon for the last 45 years. In Daejeon, it has happened only once since 1975. The temperature dropped below $-18.9^{\circ}C$ 45 times in Cheongju and 32 times in Suwon during the same period, but it has not happened at all since 1991. It appears that the rising coefficient of the lowest temperature increases, as the latitude gets higher. As a result, L. indica and P. angustifolia which were planted in the central region of Korea, are considered in danger from freezing injury because the temperature can get very low intermittently in winter. But, the risk of freezing has reduced gradually as the lowest temperature raises each year.

국내 연근해 및 환자로부터 분리된 vibrio vulnificus의 세균학적 특징

  • Vibrio vuln$cus has been recognized as a pathogen of septicemia and wound infection, when the organism attacks high-risk persons with a history of hepatic disease. alcohol abuse. diabetes or any debilitative disease. Forty six strains of K vulnzjicus. isolated from 1025 marine specimens from May to Novemver for three years. from 1985 to 1987. were studied for their biochemical properties. growth requirements, serotype and drug susceptibilities. The isolates were different in their various biochemical reactions. Ninety-five percent of isolated strains were able to ferment lactose, while most strains didn't utilize sucrose in their biochemical test, for example ornithine, gelatin and mannitol were quite dit'ferent composition than those described in other reports. It was found that the biochemical test wasn't useful for identifying strain. The type of somatic 0 antiserum was determined in isolates from marine sources and in patients with Vibrio septicemia. In patient isolates. 1-2 group were 24% and 1-4 group were 42%. However. 02 group(33%) were more abundant in isolates from marine sources. Minimal inhibitory concentrations(M1Cs) of chloramphenicol, tetracycline. erythromycin and ampicillin were determinef for V vuln~ficus by broth dilution method. MIC90 was I , 0.25, :! and 4,ug/ml in patient isolates. 1, 0.25, 2 and 2 ,ug/ml in marine isolates. The divalent chelating agent, IDTA. inhibited the growth of V. vuln!'ficus at 6.25 mMlml of MIC90.

Studies on the Embryonic Development of the Oriental Tobacco Budworm, Heliothis assulta Guenee(Lepidoptera: Noctuidae) (담배나방의 배자발생(胚子發生)에 관한 연구)

  • Embryonic developmental rates of the Oriental tobacco budworm, Heliothis assulta Guenee, were compared at various constant temperatures and 16 hours of light, and detailed embryogenesis was also studied at $25^{\circ}C$. The egg was nearly globular in form and had an average equatorial diameter of 0.53mm. A single micropyle was in the center of the circular area at the anterior pole of the egg. Durations of embryogenesis at 20, 25, 30, and $35^{\circ}C$ were 147, 81, 61, and 67hrs, respectively. Embryonic death was especially higher at $35^{\circ}C$ than any other temperatures investigated. Embryogenesis progressed with changes in color and pattern, which are quite characteristic at each developmental stage of the embryo. At $25^{\circ}C$, organogenesis began in 14hrs after oviposition, formation of gut completed in 44hrs and eclosion occurred in 80hrs. The embryo formed along the long axis at early developmental stage, moved towards the equatorial plane in 24hrs, and made a half-turn on the plane in 36hrs. In 40hrs, head was oriented to the anterior pole of the egg until eclosion.

Establishment of Optimal Rearing Conditions for the Production of Tenebrio molitor Larvae (갈색거저리 유충 생산을 위한 효율적인 사육조건 확립)

  • Tenebrio molitor larvae contain large amounts of proteins, lipids and other functional materials, enabling this insect to be used as an edible food source in animal feeds and for industrialization. Although many efforts have been made to set up mass rearing systems, few studies have been conducted to establish optimal rearing conditions for the production of high quality T. molitor larvae. Herein we investigated 1) the effects of additional diets on the survival and fecundity of the insect, 2) the relationship between oviposition period and the uniformity of larval size, 3) the effects of rearing density and temperature on insect development, and 4) the storage stability of eggs and pupae at low temperatures given possible temporary production discontinuation. The addition of carrot and zucchini to the traditional wheat bran diet significantly increased the survival and fecundity rate of adult T. molitor. Of the three different oviposition sampling periods (3, 7, and 14 days) used to investigate the uniformity of the hatched larvae in each treatment, the period of 3 and 7 days provided higher uniformity than the 14 days oviposition period. Larval development was faster at $30^{\circ}C$ than at 20, 25, and $35^{\circ}C$. Interestingly, oviposition rates were highest at $20^{\circ}C$ but showed much slower larval development and lower uniformity at $30^{\circ}C$. Regarding the effect of larval rearing densities (1, 10, 20, 30, 40, and 50 larvae per 90 mm diam. dish), larval weight was significantly reduced at higher rearing densities, but larval longevity and length were not influenced by rearing density. The 30 larvae/dish is suggested to be a reasonable density to be applied to mass production systems. When kept at $4^{\circ}C$, T. molitor eggs showed a significant reduction in hatching rate; however, when stored under the same conditions, pupae emergence rates remained high until 10 weeks, suggesting that storage at low temperatures is more suitable for the pupal stage than the egg stage. Our findings suggest that an increase in T. molitor adult survival and fecundity rates and a uniformity of hatched larval development can be achieved with the following recommendations: a combination diet (including wheat bran), a 7-day oviposition period; a larvae-rearing temperature of $30^{\circ}C$, a rearing density of 30 larvae/dish, and the storage of pupal stages at low temperatures in the case of rearing discontinuation. This study serves as a strong foundation for the successful mass production of high quality T. molitor larvae.

Comparative Analysis of Cold Tolerance and Overwintering Site of Two Flower Thrips, Frankliniella occidentalis and F. intonsa (꽃노랑총채벌레와 대만총채벌레의 내한성과 월동처 비교 연구)

  • Two dominant thrips in hot pepper (Capsicum annuum) cultivating in greenhouses are Frankliniella occidentalis and F. intonsa in Korea. This study investigated their overwintering physiology. These two thrips were freeze-susceptible and suppressed the body freezing temperature by lowering supercooling point (SCP) down to -15~-27℃. However, these SCPs varied among species and developmental stages. SCPs of F. occidentalis were -25.7±0.5℃ for adults, -17.2±0.3℃ for pupae, and -15.0±0.4℃ for larvae. SCPs of F. intonsa were -24.0±1.0℃ for adults, -27.0±0.5℃ for pupae, -17.2±0.8℃ for larvae. Cold injuries of both species occurred at low temperature treatments above SCPs. Thrips mortality increased as the treatment temperature decreased and its exposure period increased. F. occidentalis exhibited higher cold tolerance than F. intonsa. In both species, adults were more cold-tolerant than larvae. Two thrips species exhibited a rapid cold hardening because a pre-exposure to 0℃ for 2 h significantly enhanced the cold tolerance to a lethal cold temperature treatment at -10℃ for 2 h. In addition, a sequential exposure of the thrips to decreasing temperatures made them to be acclimated to low temperatures. To investigate the overwintering sites of the two species, winter monitoring of the thrips was performed at the greenhouses. During winter season (November~February), adults of the two species were not captured in outside of the greenhouses. However, F. occidentalis adults were captured to the traps and observed in weeds within the greenhouses. F. occidentalis adults were also emerged from soil samples obtained from the greenhouses during the winter season. F. intonsa adults did not come out from the soil samples at November and December, but emerged from the soil samples obtained after January. To determine the adult emergence due to diapause development, two thrips species were reared under different photoperiods. Adult development occurred in all photoperiod treatments in F. occidentalis, but did not in F. intonsa especially under short periods. Tomato spotted wilt virus, which is transmitted by these two species, was detected in the weeds infested by the thrips during the winter season. These results suggest that F. occidentalis develops on weeds in the greenhouses while F. intonsa undergoes a diapause in the soil during winter.
