Acute Toxicity of Leuconostoc kimchii GJ2, An Exopolysaccharide-Producing Lactic Acid Bacteria Isolated from Kimchi, in Mice (김치로부터 분리한 Exopolysaccharide생성 유산균 Leuconostoc kimchii GJ2의 마우스에 대한 급성독성)

  • The present study was carried out to investigate the acute toxicity of Leuconostoc kimchii GJ2(Leu. kimchii GJ2), an exopolysaccharide-producing lactic acid bacteria isolated from kimchi, in ICR male and female mice. The test article was intraperitoneally or orally administered to both sexes of mice. Then followed a 14-day period of observation. In intraperitoneal acute toxicity test, mice were injected intraperitoneally with dosage of 0, 700, 900, 1,100, 1,300, 1,500, 1,700, 1,900, 2,100, 2,300 and 2,500 mg/kg. Intraperitoneal 50% lethal doses($LD_{50}$) of Leu. kimchii GJ2 was >2,500 mg/kg in male and female mice. In oral acute toxicity test, male and female mice were gavaged with Leu. kimchii GJ2 of five doses(0, 625, 1,500, 2,500 and 5,000 mg/kg). Oral $LD_{50}$ of Leu. kimchii GJ2 was consider to be higher than 5,000 mg/kg. Furthermore, no significant changes in general conditions, body weights, clinical signs and any gross lesions were observed in both sexes of mice administered intraperitoneally or orally with Leu. kimchii GJ2. In conclusion, our results suggest that no adverse effect level of Leu. kimchii GJ2 is estimated to be over 2,500 mg/kg in the intraperitoneal route and 5,000 mg/kg in the oral route, respectively.

Studies on the Antioxidative Effect of the Buckwheat (Fagopyrum esculentum Moench) Extract and its Protective Role against Cadmiun-mediated Stress (메밀의 항산화 및 카드뮴 방어 효능에 관한 연구)

  • In the present study, the Chuncheon buckwheat extracts prepared from its seed coats, seeds and stems were used to determine anti-oxidative effects, the content of rutin and phytic acid, and the protective role against cadmium at the cellular level. futhermore, it was evaluated whether the buckwheat, mainly known as a healthy food source, might be applicable to functional cosmetics. Up to $100 {\mu}g/mL$ of the extract was not toxic in HaCaT and B16F10 cell lines using MTT assay. The anti-oxidative capacity of superoxide radicals was shown in seed coats extracts > stem extracts=seed extracts. Although its content of rutin, known as one of effective anti-oxidants, mainly exists in the stem, any extract did not eliminate hydroxyl radicals. Phytic acid, known as a heavy metal-chelate agent, was highly concentrated in the stem. The Chuncheon buckwheat extract had $10\%$ protective effect against the treatment of $50{\mu}M$ cadmium at which $50\%$ of HaCaT cells survived. Confocal laser scanning microscope revealed the intracellular generation of reactive oxygen species (ROS) by cadmium treatment. Finally, we identified that the stem extract had the most protective effect on the elimination of ROS.

Toxicity Evaluation of 'Bt-Plus' on Parasitoid and Predatory Natural Enemies (기생성 및 포식성 천적에 대한 작물보호제 '비티플러스'의 독성 평가)

  • Effect of a new crop protectant 'Bt-Plus' on natural enemies was analyzed in this study. Tested natural enemies included two parasitic species of $Aphidius$ $colemani$ and $Eretmocerus$ $eremicus$, and four predatory species of $Harmonia$ $axyridis$, $Orius$ $laevigatus$, $Amblyseius$ $swirskii$, and $Phytoseiulus$ $persimilis$. 'Bt-Plus' was formulated by combination of three entomopathogenic bacteria ($Xenorhabdus$ $nematophila$ (Xn), $Photorhabdus$ $temperata$ subsp. $temperata$ (Ptt), $Bacillus$ $thuringiensis$ (Bt)) and bacterial metabolite (BM). All three types of 'Bt-Plus' showed significantly higher toxicities against fourth instar $Plutella$ $xylostella$ larvae than Bt single treatment. Two types of bacterial mixtures ('Xn+Bt' and 'Ptt+Bt') showed little toxicity to all natural enemies in both contact and oral feeding assays. However, 'BM+Bt' showed significant toxicities especially to two predatory mites of $A.$ $swirskii$ and $P.$ $persimilis$. The acaricidal effects of different bacterial metabolites were evaluated against two spotted spider mite, $Tetranychus$ $urticae$. All six BM chemicals showed significant acaricidal effects. The BM mixture used to prepare 'Bt-Plus' showed a high acaricidal activity with a median lethal concentration at 218.7 ppm (95% confidence interval: 163.2 - 262.3). These toxic effects of bacterial metabolites were also proved by cytotoxicity test against Sf9 cells. Especially, benzylideneacetone, which was used as a main ingredient of 'BM+Bt', showed high cytotoxicity at its low micromolar concentration.

Chitosan for the Removal of Mercury, Hg (수중에서 카이토산에 의한 수은 제거)

  • Experiments were conducted with goldfish exposed to various levels of mercuric chloride(control group) and mercuric chloride with chitosan (experimental group). Dilutions of these two solutions were made in the concentration ranges 0.6 to 1.0 $mg/{\ell}$ and 1.2 to 2.0 $mg/{\ell}$, respectively. Fifty percent lethal concentration of mercuric chloride($LC_{50}$) for 48 hours with the species was between 0.6 and 0.7 $mg/{\ell}$. Exposure of goldfish to mercury produced a marked, dose-dependent mortality with elevation of concentration (P<0.05). However, at each concentration of mercuric chloride treated with chitosan, mortality decreased significantly compared to control group (P<0.05).

  • The nephrotoxicity of dichromate was reduced upon the administration of red ginseng $S_I-fraction$. Rabbits treated with dichromate showed acute renal failure and the majority of animals died within ten days. Mortality and physioligical status were significantly improved by the administration of ginseng. The effect of red ginseng $S_I-fraction$ was studied by hematological analysis. urinalysis. and histophathological examination. It was found that red ginseng $S_I-fraction$ has a potent effect on glomerulotubular imbalance induced by dichromate. In addition, $S_I-fraction$ normalized the rate of glycogenolysis. glycolysis. and the rate of lactate production. which had been accelerated by intravenous dichromate injection. Red ginseng $S_I-fraction$ was superior to ascorbic acid. CaEDT A and furocemide in the protection and recorvery from dichromate-induced acute renal failure.

  • This study suggests that ozonated olive oil can kill a wide variety of microorganisms while being less toxic but in the case of administration or abdominal administration, the human body is harmless. In Korea, there is no animal stability test yet. In this study, ozonated olive oil was used to test the stability of the animal through the eye. Specific experiments were performed to evaluate animal Ocular irritation safe test of ozonized oil (high concentration) in rabbits. and Observation of the treatment point on the test substance, ozonated olive oil showed no Ocular irritation in both the non-washing group and the washing group.

  • Since the usage of tributyltin(TBT) has been banned, many chemicals including Sea-Nine 211, Cu-pyrithione, and Zn-pyrithione were developed to use as antifouling agents for ships and coastal structures. However, the toxicity of these antifouling chemicals have not been systematically evaluated in ecotoxicological and biological studies. In this study, we investigated the effect of four antifouling substances on survival of estuarine rockfish, Sebastes schlegeli and amphipod, Monocorophium acherusicum. Survival of S. schlegeli and M. acherusicum during the 96-h exposure period were used to estimate the median lethal concentrations(LC50s) of test chemicals for each test species. Among antifouling agents, Cu-pyrithione($56{\mu}g{\cdot}1^{-1}$;96-h LC50) was most toxic to S. schlegeli, followed by $TBT(73{\mu}g{\cdot}1^{-1}),\;Sea-Nine(184{\mu}g{\cdot}1^{-1})\;and\;Zn-pyrithione(l707{\mu}g{\cdot}1^{-1})$, while TBT($26{\mu}g{\cdot}1^{-1}$) was most toxic to M. acherusicum followed by Sea-Nine($49{\mu}g{\cdot}1^{-1}$), Cu-pyrithione($119{\mu}g{\cdot}1^{-1}$) and Zn-pyrithione($334{\mu}g{\cdot}1^{-1}$). Effect concentrations of the antifouling chemicals estimated in this study can be used when assessing the potential risks of these substances, of which usage is increasing in the coastal environment.

Effect of Selenium Yeast on MPTP (1-methyl-4-phenyl-propion-oxypiperidine)-Induced Neurotoxicity in Mice (Selenium이 MPTP(1-methy-4-phenyl-1,2,3,6-tetrahydropyridine)에 의해 유도된 생쥐의 신경독성에 미치는 영향)

  • This study is investigated the effect of selenium against neurotoxicity induced by MPTP(1-methy-4-phenyl-propion-oxypiperidine) in mice. In order to demonstrate neuroprotective activity of selenium, mice were administrated orally with selenium(25, 50, 100 ${\mu}g/kg$, once/day) for 10 days, and MPTP(10 mg/kg) was injected subcutaneously into the mice for 6 days from the beginning 1hr before selenium treatment. Test of rota road activity was inhibited by treatment with selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. Monoamine oxidase(MAO)-B activity and cerebral lipid peroxide content were significantly decreased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice and MAO-A was not affected. Activities of cerebral superoxide dismutase, catalase and glutathione peroxidase were significantly increased in the treatment of selenium in MPTP-induced neurotoxicity group when compared to MPTP treatment group in normal mice. These results suggest that selenium might be estimated the result from the cooperative action of its inhibitory effect on monoamine oxidase-B with that of the enhancement of antioxidant(SOD, catalase, GSH-Px) defence ability.

Study on Short-term Toxicological Evaluation of Treated Landfill Leachate Using Early Stage of River Puffer Fish, Takifugu obscurus (황복 Takifugu obscurus 자치어를 이용한 침출수 단기독성 연구)

  • Toxic effect of landfill leachate on the larvae and juvenile stage of the river puffer fish, Takifugu obscurus, were investigated in order to assess severity of environmental impact of leachate effluent on the general population of estuarine fishes. A short-term toxic experiment was designed to test both laboratory hatched larvae and juveniles (5, 10 and 15 mm in length) and in-situ juveniles (30 and 45 mm in length) using the leachate concentrations between 0 and 16%. Lethal concentrations of 50% mortality ($LC_{50}$) were observed using Spearman-karber Method. 24hr-$LC_{50}$ appeared at the leachate concentrations ranging from 3.03 to 8.57%, 48hr-$LC_{50}$ at 2.73 to 6.21 %, 72hr-$LC_{50}$o at 2.45 to 5.53%, and 96hr-$LC_{50}$ at 2.38 to 4.93%, respectively. Leachate concentrations between 0.69 and 1.51% induced 96hr-$LC_1$. Respiratory frequency was significantly affected even at low leachate concentrations between 0.5 and 1.0% (P < 0.05). These results suggest that the leachate effluents may cause a harmful impact on the physiology of river puffer fish, especially for younger cohorts.

A Study on the Evaluation of Teratogenecity of Chemical by Korean Brown Frog Embryo, Rana dybowskii (산개구리 배아를 활용한 화학물질의 기형성 평가에 관한 연구)

  • The Frog Embryo Teratogenesis Assay-Xenopus(FETAX) protocol was recently adopted as a valuable tool fur evaluating chemical toxicity and the effect of environmental contaminants in frogs. In this study, the teratogenecity of NiCl$_2$, Carbofuran, Diazinon were determined in the Korean frog, Rana dybowskii using the FETAX protocol. The mortality rate and the percentage of malformed larvae were investigated by probit analysis, The teratogenic concentrations(EC$_{50}$) of NiCl$_2$, Carbofuran and Diazinon were 0.4 and 1.6 and 1.9 mg/1. The embryolethal concentrations(LC$_{50}$) of NiCl$_2$, Carbofuran and Diazinon were 17.6 and 41.5 and 20.2mg/1. The teratogenic indices (TI=LC$_{50}$/EC$_{50}$) were 43.8 for NiCl$_2$, 26.0 for Carbofuran and 10.6 for Diazinon. NiCl$_2$, Carbofuran and Diazinon were shown to be potent teratogens for Rana dybowskii embryo, causing concentration related increase of edema, tail and abdomen. The study results reveal that NiCl$_2$, Carbofuran and Diazinon suppress the development of embryos at relatively low concentrations. Therefore, the Rana dybowskii embryo teratogenesis assay system was proven to be a useful tool to evaluate the toxicity of environmental pollutants.lutants.
