Diabetes mellitus (DM) is one of the main global health problems. Chronic exposure to hyperglycemia can lead to cellular dysfunction that may become irreversible over time, a process that is termed glucose toxicity. Our perspective about glucose toxicity as it pertains to the pancreatic β-cell is that the characteristic decreases in insulin secretion are caused by regulated apoptotic gene expression. In this study, we examined whether ferulic acid protects INS-1 pancreatic cells against high glucose-induced apoptosis. High glucose concentration (30 mM) induced glucotoxicity and death of INS-1 pancreatic β cells. However, treatment with 1, 5, 10, or 20 μM ferulic acid increased the cell viability in a concentration-dependent manner. Treatment with ferulic acid dose-dependently decreased the intracellular levels of reactive oxygen species, thiobarbituric acid reactive substances, and nitric oxide in INS-1 pancreatic β cells pretreated with high glucose. These effects influence the apoptotic pathway, increasing the expression of the anti-apoptotic protein Bcl-2 and reducing the levels of pro-apoptotic proteins, including Bax, cytochrome C, and caspase 9. Annexin V/propidium iodide staining indicated that ferulic acid significantly reduced high glucose-induced apoptosis. These results demonstrate that ferulic acid is a potential therapeutic agent to protect INS-1 pancreatic β cells against high glucose-induced apoptosis.
Cultured pancreatic beta cells and nerve cells, it is given normal condition of 10% FBS (fetal bovine serum), 11.1 mM glucose and hyperglycemia codition of 1% FBS, 30 mM glucose. For low LET X-ray irradiated with 0.5 Gy/hr dose-rate(total dose: 0.5 to 5 Gy). Survival rates were measured by MTT assay. When non irradiated, differentiated in the pancreatic beta cells experiment is hyperglycemia conditions survival rate compared to normal conditions survival rate seemed a small reduction. However increasing the total dose of X-ray, the survival rate of normal conditions decreased slightly compared to the survival rate of hyperglycemia conditions, the synergistic effect was drastically reduced. When non irradiated, undifferentiated in the nerve cells experiment is hyperglycemia conditions survival rate compared to normal conditions survival rate seemed a large reduction. As the cumulative dose of X-ray normal conditions and hyperglycemia were all relatively rapid cell death. But the rate of decreased survivals by almost parallel to the reduction proceed and it didn't show synergistic effect.
제2형 당뇨병은 인슐린 저항성과 인슐린 분비능의 장애에 의해서 복합적으로 나타나는 대사성 증후군 중 하나이다. 인슐린 저항성은 용어에서도 알 수 있듯이 인슐린 신호전달의 저하로 인한 인슐린 작용 감소가 나타나고 이는 인슐린 분비가 증가하더라도 포도당 이용은 감소된 상태이다. 인슐린 분비능은 직접적으로 인슐린 작용과는 무관한 것처럼 보이지만 자세히 살펴보면 인슐린 분비능의 중요한 역할을 하는 췌장 베타세포의 성장과 생존은 인슐린/IGF-1 신호 전달과 밀접한 관계가 있으므로 인슐린 분비능도 인슐린/IGF-1 신호전달과 관련이 있다. 그러므로 인슐린/IGF기 신호전달의 장애는 제2형 당뇨병의 병인에 매우 중요한 요인이다. 그러므로 제2형 당뇨병을 효과적으로 예방 또는 치료하기 위해서는 근육, 지방세포와 간등에서 인슐린 신호전달을 향상시켜 포도당 이용을 증가시키고, 시상하부에서 인슐린 신호전달을 향상시켜 식욕을 적절하게 조절할 수 있도록 하여 과체중이나 비만의 발생을 방지하고 췌장의 베타세포에서 인슐린/IGF-1 신호전달을 향상시켜 베타세포의 성장과 생존을 향상시켜 포도당에 대한 인슐린 분비 능을 촉진시키는 것이다. 최근에 FDA에서 승인받은 Exenatide는 glucagons like peptide-1 receptor agonist로 시상하부에 작용하여 식욕조절도 하고 췌장의 베타세포에서 IRS2의 발현을 증가시켜 인슐린/IGF-1 신호전달을 향상시켜 insulinotropic 작용을 가지는 것으로 알려졌다. 이 Exenatide는 도마뱀의 독침에서 분리한 peptide로 천연물로부터 유래된 약물이므로 천연물로부터 제2형 당뇨병을 치료할 수 있는 약물을 발견할 가능성은 매우 높으므로 이에 대한 연구는 체계적으로 이루어져야 하겠다.
Type 2 diabetes is a serious chronic metabolic disease, and the goal of diabetes treatment is to keep blood glucose at a normal level and prevent complications from diabetes. Hyperglycemia is a key pathologic feature of type 2 diabetes that mainly results from insulin resistance and pancreatic β-cell dysfunction. Chronic exposure of β-cells to elevated glucose concentrations induces glucotoxicity. In this study, we examined whether an 80% ethanol extract of Oxya chinensis sinuosa Mishchenko (OEE) protected INS-1 pancreatic β-cells against glucotoxicity-induced apoptosis and oxidative stress. Pretreatment with a high concentration of glucose (high glucose = 30 mM) induced glucotoxicity and apoptosis of INS-1 pancreatic β cells. Treatment with OEE significantly increased cell viability. Treatment with 0.01-0.20 mg/ml OEE dose dependently decreased intracellular reactive oxygen species, lipid peroxidation, and nitric oxide levels and increased insulin secretion in high glucose-pretreated INS-1 β cells. OEE also significantly increased the activities of antioxidant enzymes in response to high-glucose-induced oxidative stress. Moreover, OEE treatment significantly reduced the expressions of pro-apoptotic proteins, including Bax, cytochrome C, caspase-3, and caspase-9, and increased anti-apoptotic Bcl-2 expression. Apoptotic cells were identified using Annexin-V/propidium iodide staining, which revealed that treatment with OEE significantly reduced high-glucose-induced apoptosis. These findings implicate OEE as a valuable functional food in protecting pancreatic β-cells against glucotoxicity-induced apoptosis and oxidative stress.
The purpose of this study was to investigate the relationship between VAI, insulin resistance, and pancreatic beta cell function according to the prevalence of metabolic syndrome in obese adults. From 2017 to 2019, 1,797 obese adults who received medical checkups at a general hospital in Bundang. Diagnosis of metabolic syndrome is NCEP-ATP III. HOMA index was used for insulin resistance and pancreatic beta cell function. VAI was higher in the metabolic syndrome than in the control(p<.001). As the number of risk factors for metabolic syndrome increased, the VAI value was higher(p<.001). The prevalence of metabolic syndrome increased as the VAI quartile increased(p<.001). VAI was also shown to be related to HOMA-IR and HOMA-β in the control, but not in the metabolic syndrome.

Perilla frutescens (L.) Britton var. sprouts (PFS) is a plant of the labiatae family. The purpose of this work was to assess the preventive effects of PFS ethanolic extracts (PFSEs) on cytokine-induced ${\beta}$-cell damage. Cytokines, which are released by the infiltration of inflammatory cells around the pancreatic islets, are involved in the pathogenesis of type 1 diabetes mellitus. The combination of interleukin-$1{\beta}$ (IL-1), interferon-${\gamma}$ (IFN-${\gamma}$), and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) induced formation of reactive oxygen species (ROS). Accumulation of intracellular ROS led to ${\beta}$-cell dysfunction and apoptosis. PFSEs possess antioxidant activity and thus lead to downregulation of ROS generation. Cytokines decrease cell viability, stimulate the expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), and induce the production of nitric oxide (NO). PFSEs prevented cytokine-induced cell viability in a dose-dependent manner. Incubation with PFSE resulted in significant reduction in cytokine-induced NO production that correlated with reduced levels of the iNOS and COX-2 protein expression. Furthermore, PFSE significantly decreased the activation of nuclear factor ${\kappa}B$ (NF-${\kappa}B$) by inhibition of $I{\kappa}B{\alpha}$ phosphorylation in RINm5F cells. In summary, our results suggest that the protective effects of PFSE might serve to counteract cytokine-induced ${\beta}$-cell destruction. Findings indicate that consumption of Perilla frutescens (L.) Britton var. sprouts alleviates hyperglycemia-mediated oxidative stress and pro-inflammatory cytokine-induced ${\beta}$-cell damage and thus has beneficial anti-diabetic effects.
제2형 당뇨병은 대사성 질환으로 간, 근육 그리고 지방 조직 세포에서 인슐린 작용의 장애로 나타나는 인슐린 저항성으로 혈당의 이용이 감소하여 혈당이 높아짐에도 불구하고 췌장의 베타세포에서 인슐린 분비가 충분하지 못할 때 유발된다. 서구에서는 비만 등으로 인해 인슐린 저항성이 증가하면 인슐린 분비가 높은 고인슐린혈증을 나타내어 당뇨병으로의 진전은 늦다. 하지만 우리나라를 비롯한 아시아의 사람들은 인슐린 저항성이 증가할 때 인슐린 분비가 충분치 못해 혈청 인슐린 농도가 정상인과 비슷하거나 더 낮은 상태에서 당뇨병으로 진전된다. 이러한 차이는 우리나라를 비롯한 아시아 사람들에게서 제2형 당뇨병의 발생이 급격하게 증가할 것이라는 보고되었다. 결국 당뇨병은 간, 근육 및 지방조직에서의 인슐린 작용의 장애와 췌장의 베타세포에서 인슐린 분비의 부족의 복합적인 장애에 의해서 나타나고 이것은 공통적으로 각 조직에서의 인슐린/insulin growth factor (IGF)-1 신호전달의 장애와 관련이 있다. 베타세포에서의 인슐린분비 자체는 인슐린/IGF-1 신호전달과 관계가 없지만 간접적으로 관련이 있다. 인슐린 분비능은 베타세포의 증식과 생존에 의한 베타세포의 양과 밀접한 관련이 있는데 인슐린/IGF-1 신호전달은 베타세포의 증식과 생존을 조절한다. 그러므로 혈당 조절에 관여하는 기능성 식품은 인슐린 작용을 향상시키는 인슐린 민감성 특성을 가지거나, 혈당이 높아질 때 인슐린 분비를 촉진시키는 insulinotropic 작용을 하는 성질을 가지고 있어야 하겠다. 전자의 대표적인 약은 1999년에 미국 FDA에서 승인 받은 peroxisome proliferator-activated receptor $(PPAR)-{\gamma}$ agonist 인 thiazolidinedione 계통의 약물인 troglitazone, pioglitazone, rosiglitazone 등이 있고, 후자는 2007년에 승인 받은 Exenatide는 glucagon like peptide (GLP)-1 agonist이다. 이 두 가지 약은 모두 자연계에 존재하는 동식물에서 유래된 것으로 식품에도 많이 다양한 종류의 인슐린 민감성 물질이나 insulinotropic 작용을 하는 물질이 함유되어 있을 것이다. 이러한 기능 이외에 혈당조절 약이나 식품으로 사용되는 것은 탄수화물의 소화를 방해하는 것으로 탄수화물 소화효소인 a-amylase 또는 maltase의 활성을 억제하여 식후 혈당의 급격한 상승을 방지하는 것이 있다. 우리나라 사람들은 탄수화물의 섭취가 너무 많아서 실제로 이러한 식품이나 약의 효능이 높지 않을 것이다. 혈당을 조절하는 기능성 식품은 이 세 가지 효능 중 일부를 가지고 있는 것이 될 수 있다. 이러한 기능을 스크리닝하기 위해서 3가지 단계를 거쳐야 한다. 먼저 시험관에서 또는 세포 실험을 통해서 앞서 언급한 3가지 기능을 가지고 있는 지 여부를 각각 조사한다. 이중에서 효과가 있는 것은 당뇨 동물 모델을 사용하여 in vivo에서 혈당 강하기능과 혈당 강하기전을 조사하는 실험을 한다. 효과가 있는 식품이 우리가 전통적으로 식품으로 섭취해 왔다면 독성 검사를 거쳐야 할 필요가 없지만 한약재이거나 특수 식품의 경우에는 in vivo 실험 전에 GLP 기관에서 반드시 독성 실험을 거쳐 독성 유무를 확인할 필요가 있다. 동물 실험에서 효과적인 것은 인체 실험을 거쳐 혈당 조절 기능성 식품으로 식약청에서 허가를 받을 수 있겠다. 결론적으로 식품에는 항당뇨 특성을 가진 물질들이 함유되어 있는 것들이 상당히 많다. 혈당 조절기능이 있는 기능성 식품으로 개발할 때 고려해야 할 것은 1) 그 양이 혈당 강하 기능성 식품으로 지정받을 수 있을 정도로 충분히 함유되어 있느냐, 2) 혈당을 강하시키는 기전이 단순히 당의 배설을 촉진시켜서 혈당을 저하시키는 것이 아니라, 인슐린 작용을 촉진시키거나, 포도당 자극에 의한 인슐린 분비를 촉진시키거나 탄수화물의 소화 흡수를 억제시킴으로 혈당을 강하시키는 지 등을 파악하는 것이다. 이러한 조건을 만족시키는 식품은 지속적으로 섭취할 때 당뇨병을 예방하거나 진전을 지연시킬 수 있는 혈당조절기능이 있는 기능성 식품으로 개발 가능성이 있겠다.

The present study was designed to examine the potential antidiabetic and antioxidant effect of ethanol extract from $Prunus$$mume$ fruit (PME) against alloxan-induced oxidative stress in pancreatic ${\beta}$-cells, HIT-T15. To evaluate the antidiabetic effect of PME, 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliu bromide (MTT) cell proliferation assay, lactate dehydrogenase (LDH) release assay, $NAD^+$/NADH ratio and insulin secretion were assessed. We also measured its antioxidant effect against alloxan-induced oxidative stress in the cells by assessing the levels of the antioxidant enzymes including superoxide dismutase (SOD), glutathione S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx). The results of this analysis showed that alloxan significantly decreased cell viability, increased LDH leakage, and lowered $NAD^+$ /NADH ratio and insulin secretion in HIT-T15 cells. However, PME significantly increased the viability of alloxan-treated cells and lowered LDH leakage. The intracellular $NAD^+$ /NADH ratio and insulin secretion were also increased by 1.5~1.9-fold and 1.4-fold, respectively, after treatment with the PME. The HIT-T15 cells treated with alloxan showed significant decreases in the activities of antioxidant enzymes, while PME significantly elevated the levels of antioxidant enzymes. Based on these results, we suggest that PME could have a protective effect against the cytotoxicity and dysfunction of pancreatic ${\beta}$-cells in the presence of alloxan-induced oxidative stress.
This study was to investigate the effect of betaine on the hypoglycemia and hepatoprotection of streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing around 280 g were randomly assigned to the three experimental groups: a healthy normal group and two groups with STZ-induced diabetes and fed either control diet or betaine diet. Betaine given to the STZ-diabetic rats had significant effect in lowering the serum glucose concentrations compared to the STZ-diabetic rats. The alanine aminotransferase (AST) and aspartate aminotransferase (ALT) activities and triglyceride contents in serum were dramatically higher in the STZ-diabetic rats, but these increases in relation to diabetes also decreased in the STZ-diabetic rats fed betaine. However, the total-cholesterol concentration in the STZ-diabetic rats was even increased by betaine. The morphology of the pancreatic islets in the normal rats showed a typical round form, but most of the islets in the STZ-diabetic rats showed severe morphological alterations by being markedly destroyed. However, the islet morphology of STZ-diabetic rats given betaine mostly maintained a normal rounded appearance. The present study strongly suggests that the administration of betaine showed a moderate hypoglycemic effect by protecting the pancreatic beta-cells by morphological examination from STZ-induced destruction.
(-)-Epigallocatechin-3-gallate (EGCG), a green tea polyphenol, has been shown to have strong antibacterial, antiviral, antioxidant, anti-inflammatory, and chemopreventive effects. However it is unknown whether EGCG can recover alcohol-associated pancreatitis. The aim of this study was to investigate the effects of EGCG on pancreatic enzyme activities and the expressions of pancreatic regenerating related markers, such as adenosine monophosphate-activated protein kinase (AMPK), raf-1 kinase inhibitor protein (RKIP), and Regenerating gene 1 (Reg1), in mice pancreatic primary acinar cells. Our results revealed that activities of ${\alpha}$-amylase and chymotrypsin were significantly increased in the cells treated with ethanol compared to the untreated control cells; however, the increased activities of both enzymes were markedly reduced by pretreatment with EGCG. Phosphorylation of AMPK and total expression of RKIP were decreased in the ethanol-treated primary acinar cells; however, these were both significantly increased in the EGCG-pretreated cells. In addition, when EGCG was treated, expression of Reg1 was markedly increased compared with that of the control or the ethanol-treated primary acinar cells, demonstrating that EGCG can modulate pancreatic regenerating related genes. Therefore, our findings suggest that EGCG may have therapeutic utility in the prevention or treatment of alcohol-associated pancreatitis.

제 5 장 계약 해지 및 이용 제한

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
