Tyrosinase Inhibition Effect of extracts from Mistletoe Cultivated on Prunus mume (매실나무(Prunus mume) 겨우살이 추출물의 tyrosinase 저해활성)

  • 인공재배 매실나무겨우살이의 의약 화장품의 소재 및 산업화 가능성을 검토하기 위해 80% 에탄올 및 증류수 초음파추출물의 Tyrosinase 저해활성을 측정하였다. 인공재배한 매실나무겨우살이(PM, 동결건조) 및 자연산 굴참나무겨우살이(QM, 열풍건조, $30^{\circ}C$, 7day)의 80% 에탄올, 증류수 초음파추출물에 대한 tyrosinase 저해활성을 측정한 결과, 물 추출물의 경우 100 ppm의 농도에서 PM과 QM 모두 5분이 경과 하였을 때 PM은 88.37%, QM은 87.69%로 최대치를 나타내었고, 농도와 시간이 증가함에 따라 tyrosinase 저해활성이 감소하는 경향을 보여주었다. PM 80% 에탄올 추출물은 5분이 경과하였을 때 2,500ppm이 92.08%로 최대값을 보여주었고 이후 시간이 지남에 따라 저해활성이 감소되는 경향을 나타내었다. 반면 5,000ppm은 5분이 경과하였을 때 66.38%로 최소값을 나타낸 반면, 20분까지 저해활성이 증가하였고 10분이 경과한 이후로 거의 일정하게 유지되어 지속적으로 감소되는 1,000ppm보다 높은 저해효과를 보여 주었다. QM 80% 에탄올 추출물은 5분이 경과하였을 때 1,000ppm이 88.14%로 가장 높았고 5,000ppm은 63.49%로 가장 낮은 저해활성을 보여주었다. 15분까지 1,000ppm이 가장 높은 저해활성을 보여주었으나 꾸준히 감소한 반면, 2,500ppm의 경우 시간이 경과함에 따라 비교적 일정한 저해활성을 보여주어 15분이 경과한 이후로 2,500ppm의 저해활성이 가장 높게 나타났다. 25분이 경과하였을 때 2,500ppm은 70.27%로 가장 높은 저해활성을 나타내었고, 5,000ppm은 30.09%로 가장 낮은 저해활성을 보여주었다. 본 연구 결과에 따르면, 물 추출물의 경우 겨우살이의 농도가 낮을수록 더 높은 tyrosinase 저해활성을 나타내었고, 80% 에탄올 추출물의 경우 2,500ppm에서 가장 높은 저해활성을 나타내어 의약 화장품의 소재 및 산업화 가능성이 높음을 확인할 수 있었다.

Inhibitory Effect of Gardenia Fruit Extracts on Tyrosinase Activity and Melanogenesis (치자 열매 추출물의 Tyrosinase 효소활성 저해 및 Melanogenesis 억제 효과)

  • To determine the inhibition of tyrosinase activity and melanogenesis, gardenia fruit was extracted initially with $95\%$ ethanol (w/v), The ethanol extract was fractionated subsequently with hexane, chloroform and ethyl acetate in that order. The inhibitory effect of the ethanol extract on tyrosinase activity was higher than water extract. When 10 mg/mL of ethyl acetate fraction was applied, the inhibition ratio of tyrosinase activity was much higher ($99.7{\pm}0.1\%$) than that of arbutin. The inhibition of melanogenesis using B16F10 melanoma cell, the ethanol extract also showed higher inhibitory effect than water extract. The highest inhibitory activity of melanogenesis was also shown in ethyl acetate fraction ($89.1{\pm}1.4\%$ at the cone. of $100\;{\mu}g/mL$). These results suggests that gardenia extracts might be used to be a potential agents for whitening.

Identification of Flavor Components in Tomato Fruit (토마토 향기성분의 동정)

  • The volatiles of whole and homogenated tomato fruits collected by the headspace trapping method using Tenax GC and the simultaneous steam distillation method were identified by GC and GC-MS. Among over 100 GC peaks, 10 alcohols, 6 aldehydes, 4 ketones, 3 esters, 1 phenol and 1 acid were identified from whole tomato fruits, whereas 12 alcohols, 6 aldehydes, 5 ketones, 5 esters, 2 phenols, 1 hydrocarbon and 1 acid were identified from homogenated tomato fruits. By simultaneous steam distillation-extraction, 19 alcohols, 13 hydrocarbons, 9 esters, 9 ketones, 8 aldehydes, 2 phenols, 2 lactones,2 furans, 1 acids and 2 others were identified among over 300 peaks.

Acaricidal and antibacterial toxicities of Valeriana officinalis oils obtained by steam distillation extraction (수증기 증류 추출법에 의해 얻어진 미국산 길초근 정유의 살비효과 및 항균효과)

  • The chemical compositions of the essential oil of Valeriana officinalis roots obtained by steam distillation method were analyzed by GC-MS. The 16 constituents were identified in the V. officinalis oil, and the most abundant compounds were patchouli alcohol (18.69%) and ${\beta}$-gurjunene (15.26%). Acaricidal effects of the V. officinalis oil were evaluated against Tyrophagus putrescentiae, Haemaphysalis longicornis larva and H. longicornis nymph by contact bioassay. The $LD_{50}$ values against T. putrescentiae, H. longicornis larva and H. longicornis nymph were 28.01, 178.26 and $207.98{\mu}g/cm^2$, respectively. Agar disc diffusion bioassay showed the antibacterial activity of the V. officinalis oil against foodborne pathogens, especially L. monocytogenes. These results showed that the essential oil of V. officinalis roots derived from USA has a potential for development as acaricide and antimicrobial.

Analysis of Anti-Allergic Activities by Chaff Vinegar Liquor (왕겨초액의 추출물에 의한 아토피 완화 효과 비교)

  • To evaluate whether chaff vinegar liquor (CVL) has potential in ameliorating allergic symptoms in mice, we tested the anti-allergic activities with a DNFB (dinitrofluorobenzene)-induced allergic mouse model. The DNFB-induced allergic symptoms were reduced by 20% compared to the control by chaff vinegar liquor supplemented with Perilla frutescens var. acuta. Allergic symptoms were examined by measuring the width of epidermis swelling. The hematoxyline & eosin (H&E) staining also revealed that the chaff vinegar liquor dramatically decreased the allergic symptoms in the epidermis of the ear. These results collectively suggest that the chaff vinegar liquor supplemented with P. frutescens var. acuta has potential in ameliorating allergic symptoms, showing that the liquor could be a useful biomaterial(s) for the nutraceutical or cosmetic industries.

State-of-the-Art Knowledge Distillation for Recommender Systems in Explicit Feedback Settings: Methods and Evaluation (익스플리싯 피드백 환경에서 추천 시스템을 위한 최신 지식증류기법들에 대한 성능 및 정확도 평가)

  • Recommender systems provide users with the most favorable items by analyzing explicit or implicit feedback of users on items. Recently, as the size of deep-learning-based models employed in recommender systems has increased, many studies have focused on reducing inference time while maintaining high recommendation accuracy. As one of them, a study on recommender systems with a knowledge distillation (KD) technique is actively conducted. By KD, a small-sized model (i.e., student) is trained through knowledge extracted from a large-sized model (i.e., teacher), and then the trained student is used as a recommendation model. Existing studies on KD for recommender systems have been mainly performed only for implicit feedback settings. Thus, in this paper, we try to investigate the performance and accuracy when applied to explicit feedback settings. To this end, we leveraged a total of five state-of-the-art KD methods and three real-world datasets for recommender systems.

Antioxidant and Acetylcholinesterase Inhibitory Activities of Subcritical Water Extracts from Houttuynia cordata Thunb (어성초의 아임계수 추출물의 항산화 활성 및 Acetylcholinesterase 저해 활성)

  • Houttuynia cordata Thunb powder (0.05 g) was placed in a stainless vessel containing 10 mL of water, and subcritical water (SCW) extraction was carried out at various temperatures (50, 100, 200, and $300^{\circ}C$) for 10, 30, and 60 min. SCW treatment significantly affected physiologically important compounds, antioxidant activity, and acetylcholinesterase inhibitory activity of H. cordata Thunb extracts. H. cordata Thunb extracted at $300^{\circ}C$ for 30 min had the highest quercetin content, DPPH radical scavenging activity, total phenolic content, and acetylcholinesterase inhibitory activity, whereas the extract at $100^{\circ}C$ for 10 min possessed the highest flavonoid content. The highest ABTS radical scavenging activity was found in H. cordata Thunb extracted at 200 and $300^{\circ}C$ for 60 min. These results indicate that SCW extraction might be a useful processing method for obtaining valuable materials from H. cordata Thunb.

Mutagenic Activity by Ames Test of Bracken Grown in Korea (고사리의 돌연변이(突然變異) 유발성(誘發性))

  • The ethanol-and water-extracts of curled tops and stalks of young bracken (Pteridium aquilinum) fronds in raw and cooked states were examined for their mutagenicity by Ames test using Salmonella typhymurium mutants. Even a small amount of water extract from raw bracken was mutagenic to TA 1538 strain without addition of $S_9$ mix whereas a strong toxicity appeared at higher levels of the extractive. With $S_9$ mix, the water-extract from raw bracken was not mutagenic at all and not toxic to all strains tested, The amount of water-extract from cooked bracken was one-tenth of the amount from raw bracken and the water-extract showed no mutagenic effect to all strains tested. Ethanol-extract of bracken showed no mutagenicity in any case.

Extraction of Intercellular Material from Rice Leaf Tissue (수도의 세포간극 물질 추출)

  • The intercellular material was extracted from rice leaf tissue. The quantitative tests of protein and soluble carbohydrate, and activity of peroxidase, showed differences between tissue extract and intercellular material. Also electrophoretic patterns of peroxidase and esterase isozymes were not similar between them. It indicated that the intercellular material which was extracted, was not the mixture of cellular materials.

Effects of Treatments on the Distribution of Volatiles in Artemisia princeps Pampan (쑥의 처리조건에 따른 휘발성 성분 변화)

  • Volatiles in Artemisia princeps Pampan. cv. sajabal (sajabalssuk) and A. princeps Pampan. (ssuk) treated with different processing were analyzed using headspace-solid phase microextraction (HS-SPME)/gas chromatography- a mass selective detector (GC-MS). Sajabalssuk and ssuk were treated with steam distillation (SD) and freeze-dried/steam distillation (FD/SD) while controls were raw sajabalssuk and raw ssuk. Sajabalssuk had significantly more total volatiles than ssuk in control and FD/SD treated samples (p<0.05). Major volatiles in raw sajabalssuk were 2-hexenal, 1,8-cineol, trans-caryophyllene, and hexanal while those in raw ssuk were 1-hexanol, ${\beta}$-myrcene, limonene, and 2-hexenal, which implies that substantial lipid oxidation occurred in raw samples. Sajabalssuk with SD and FD/SD treatment had higher peak areas of 1,8-cineole, 4-terpineol, 1-octen-3-ol, and ${\alpha}$-terpineol while ssuk with SD and FD/SD treatment possessed 1,8-cineol, camphor, borneol, artemisia ketone, ${\alpha}$-thujone, and 1-octen-3-ol, which showed that steam distillation produced more volatiles from terpenoids than raw samples. Based on the results of HS-SPME/GC-MS, relative amounts of volatiles from lipid oxidation including 2-hexenal, hexanal, and 1-hexanol were reduced in sajabalssuk and ssuk with freeze-drying and/or steam distillation treatment.
