Laboratory Study for Phosphate Coating on Pyrite Surface for Reduction of Acid Rock Drainage (건설현장의 산성암반배수 발생저감을 위한 철인산염 피막형성 최적조건 도출 실내연구)

  • Acid drainage occurs when sulfide minerals are exposed to an oxidizing environment. The objective of this study was to examine the optimum condition for creating a phosphate coating on standard pyrite surfaces for reduction of pyrite oxidation. The solution of $10^{-2}M\;KH_2PO_4\;10^{-2}M\;H_2O_2$ pH 6 was identified as the best phosphate coating agent for the reduction of pyrite oxidation. The formation of an iron phosphate coating on pyrite surfaces was confirmed with ore microscope and scanning electron microscope equipped with energy dispersive spectroscopy. The temperature did not significantly affect on the formation of phosphate coating on the surface of pyrite. However, the phosphate coating was less stable at higher temperature than at lower temperature. The phosphate coating was quitely stable at wide range of pH and $H_2O_2$ concentration. The less than 3.4% of phosphate was dissolved at pH 2.79 and 10.64 and less than 1.0% of phosphate was dissolved at 0.1M $H_2O_2$. On the basis of these results, the phosphate coating can effectively reduce the negative environmental of acid rock drainage.

Production of Levulinic Acid from Chitosan by Acidic-Hydrothermal Reaction (산성 수열반응을 통한 키토산으로부터 레불린산의 생산)

  • Recently, many chemicals produced from renewable resources such as lignocellulosics, micro-algae and marine macro-algae, were introduced to chemical industry. Chitin/chitosan is secondly abundant feedstock on Earth. It is easily obtained from crusraceans' shells such as crab, shrimp and insects. In this work, we performed the acidic-hydrothermal hydrolysis to produce levulinic acid from chitosan using statistical approach. By design of response surface methodology, the effect of reaction temperature, catalyst amount, and reaction time and their reciprocal interactions were investigated. As a result, higher reaction temperature and catalyst amount increased the higher concentration of levulinic acid. However, reaction time did not caused large increase of levulinic acid after some reaction period. Levulinic acid of 2.7 g/L produced from chitosan in the optimized condition of reaction temperature of $175^{\circ}C$, sulfuric acid of 2.4% and reaction time of 40.7 min.

  • Four species of Trichoderma causing green mildew of Agaricus bisporus were isolated from 38spots of mushroom growing areas in Korea. These are T. koningi, T. lignorum, T. glaucum and an unidentified species, and their frequencies of occurrence are $50\%,\; 32\%,\; 13\%; and\; 5\%$ respectively. All of these species grew well in potato dextrose, Waksman's and Richard's solution, and preferred acid (pH. 4) to neutral. The temperature in mushroom trouse should be kept at $15^{\circ}C$ during cropping period, not only for the high yield of mushroom but also for the prevention of green mildew of mushroom caused by Trichoderma spp . T. lignorum was killed in soil on an exposure of 60 minutes at $70^{\circ}C$ and when exposed for 30minutes at $80^{\circ}C$. Peak heat procedure of compost eliminated T. lignorum and T koningi.

Conditioning of the Extraction of Acidic Polysaccharide from Red Ginseng Marc (홍삼박으로부터 산성다당체의 최적 추출 조건 분석)

  • This study was carried out to investigate the optimum conditions for extraction of acidic polysaccharides from red ginseng marc produced by manufacturing alcoholic extract from red ginseng. Method of carbazole-sulfuric acid was applied to determine the amount of acidic polysaccharides in red ginseng marc. The amounts of acidic polysaccharides in water extract of red ginseng marc were increased with increasing extraction temperature. The contents of acidic polysaccharides were not significantly different despite of the extraction time increasing from 6 hours to 48 hours. The contents of starch in water-extract of red ginseng marc were increased with increasing extraction temperature. The starch amounts in water extract of red ginseng marc extracted for 48 hours were increased. The yields of polysaccharide precipitated from water-extract of red ginseng marc were increased with increasing extraction temperature. The hydration rate of acidic polysaccharides and starch from water-extract of red ginseng marc were decreased with increasing extraction temperature. The contents of starch were not significantly different despite of the extraction time increasing from 6 hours to 48 hours at $8^{\circ}C$. However, the rehydration rate of acidic polysaccharide for 48 hours were decreased at $8^{\circ}C$. The rehydration rate of acidic polysaccharide and starch extracted from 6 hours to 24 hours at $25^{\circ}C$ were not significantly different, but those extracted for 48 hours were increased. From the above results, we suggest that by altering the extraction conditions in red ginseng marc it is possible to develop optimum conditions for extraction that modulate the proportions of acidic polysaccharide and starch.

Effect of Soybean Protein Isolate on the Properties of Noodle (분리대두단백질의 첨가가 제면적성에 미치는 영향)

  • This study was conducted to investigate the effect of soybean protein isolate (SPI) on the properties of noodle which was made of composite flour blended with SPI extracted at acidic (pH 2.0, 3.0), neutral (pH 7.0) and alkaline (pH 10.0, 12.0) conditions. L-value of dry and cooked-moodle which were made of composite flour was lower than that of 100% wheat flour, but a and b-value wete higher than those of 100% wheat flour, Optimal cooking time of dry-noodle which was made of composite flour was longer than that of 100% wheat flour, but the weight, volume and water absorption of the cooked-noodle were lower than those of cooked-noodle of 100% wheat flour. Breaking force of dry-noodle which was made of composite flour blended with $SPI-2,\;SPI_3,\;SPI_{7}$, and $SPI-{10}$ was lower than that of 100% wheat flour, but the breaking force of dry-noodle which was made of composite flour blended with $SPI-{12}$ at level of 5% and 10% was same as that of 100% wheat flour. Springiness and cohesiveness of the cooked-noodle which was made of composite flour were same as those of 100% wheat flour, but chewiness and hardness were higher than those of 100% wheat flour.

Purification and Characterization of Carboxymethyl Cellulase IV from Penicillium verruculosum (Penicillium verruculosum 으로부터 Carboxymethyl Cellulase IV 의 정제(精製) 및 특성(特性))

  • An endo-type cellulase, carboxymethyl cellulase(CMCase) IV, was purified from culture filtrate of cellulolytic fungus Penicillium verruculosum. The CMCase IV was acidic glycoprotein having carbohydrate of 13% as glucose and pI value of 4.0. The CMCase IV was 52 KDa of molecular weight in SDS-polyacrylamide gel electrophoresis and have optimum temperature and pH of $50^{\circ}C$ and 5.0 for enzyme activity. The CMCase IV liberated glucose and cellobiose as major products of the enzyme against carboxymethyl cellulose(CMC) and seemed to has transglycosylation activity simultaneously. Cellulase activity staining(zymogram) showed that the cellulase components of P. verruculosum were not aggregated in the medium. P. vrttuculosum mRNA was translated in vitro and translation product by the mRNA coding for CMCase IV was identified by immunoprecipitation.

Optimization of Extraction Conditions of Antioxidants from Sporophyll of Undaria pinnatifida by Response Surface Methodology (반응표면분석법에 의한 미역 포자엽의 항산화물질 추출 조건 최적화)

  • We selected a solvent for producing antioxidants from sporophyll of Undaria pinnatOptimum extraction condition for antioxidant ability(Y) of sporophyll of U. pinnatifida was investigated using response surface methodology(RSM). A compound central design was used with variables ($X_1$)temperature($^{\circ}C$) (3.4; 20; 60; 100; 116.6); ($X_2$) pH(1.8; 3; 6; 9; 10.2) and ($X_3$) treatment time(min) (1.7; 10; 30; 50; 58.3). Antioxidant activities were increased with decreasing pH and increasing temperature. The optimal extraction conditions for antioxidant ability(Y) of sporophyll of U. pinnatifida were found to be temperature $51.55^{\circ}C$, pH 4.2 and treatment time 28.2 min. Among the variables tested, pH showed greater significant facin extraction of antioxidants from sporophyll of U. pinnatifida.

Isolation and Purification of Ficin from Fig Latex (무화과(無花果)에서 Ficin의 분리(分離) 및 정제(精製))

  • Ficin, a proteolytic enzyme in Fig latex, was extracted and purified with using ammonium sulfate and CM-cellulose column chromatography, respectively, and studied for its chemical properties. The disc gel electrophoresis showed one major and three minor bands for $(NH_4)_2SO_4$ extract and only one band showed after CM-cellulose chromatography. The optimum conditions for ficin activity was found to be pH 7.0 and $50^{\circ}C$. The amino acids composition of the purified ficin were 21.8% as acidic, 3.5% as basic and 74.7% as neutral amino acids. The amino acids analysis indicated that the ficin was composed of 174 amino acids residue having molecular weight of 19,500.

Physiological Characteristics and Immunomodulation Activity of Lactobacillus fermentum 450 isolated from Raw Milk (원유에서 분리한 Lactobacillus fermentum 450의 생리적 특성과 면역활성)

  • The aim of this study was to investigate the physiological characteristics and immunomodulatory activity of Lactobacillus fermentum 450 isolated from raw milk. L. fermentum 450 showed optimum growth at $40^{\circ}C$ and exhibited immunomodulatory effects on $interleukin-1{\alpha}$, tumor necrosis $factor-{\alpha}$, and nitrous oxide at concentrations of >2,500 pg/mL, >2,000 pg/mL, and $11.55{\pm}2.95{\mu}M$, respectively. Of the 16 antibiotics tested, L. fermentum 450 exhibited the highest sensitivity to rifampicin, followed by penicillin-G, and the highest resistance to kanamycin, followed by neomycin and polymyxin B. The strain showed high acid phosphatase activity and was comparatively tolerant to bile juice and acid. Moreover, the strain displayed high resistance to Salmonella Typhimurium (63.86%). These results demonstrate that L. fermentum 450 has potential for use as a probiotic with immunomodulatory activity.

Isolation of Sorbic Acid from the Mountain Ash Berries (I) (국산(國産) 건조(乾燥)마가목 열매로 부터 Sorbic Acid의 분리(分離)에 관한 연구(硏究) (I))

  • This investigation was primarily carried out to establish the optimum conditions for the separation of sorbic acid, as an enzyme inhibitor, from dry mountain ash berries. The berries were crushed to $35{\sim}40\;mesh$, and leached with water to produce a reddish juice, which was concentrated to syrup after evaporation. Parasorbic acid was separated from the syrup by steam distillation in the presence of acid. In this study, the optimum experimental results for separation of parasorbic acid were obtained as follows : (1) the most applicable leaching time of the dry ash berries was about 6 hours at room temperature and less than 4 hours at $40^{\circ}C$ and (2) in steam distillation, after removal of malic acid, addition of 30 ml of sulfuric acid per 50 ml of syrup extracted from 100 g of the ash berries was very adequate. The purity of sorbic acid obtained experimentally through the isomerization technique was qualitatively examined by spectrophotometeric, paper chromatographic, and melting point measurements.
