생물촉매를 이용한 고효율 바이오디젤 생산

  • 차세대 재생산성 에너지로 각광을 받고 있는 바이오디젤은 현재 주로 알칼리촉매를 이용하는 화학공정으로 생산하고 있으나 고에너지 요구성이며 대규모 생산시 폐수발생 등 환경오염 유발요인이 있기 때문에 친환경 생물공정의 필요성이 대두되고 있다. 생물촉매 리파제(lipase)를 이용하는 친환경 생물공정은 화학공정에 비해 다양한 장점을 제공하고 있으나 고가의 효소생산 비용문제로 실용화에 어려움이 있다. 따라서 본 연구에서는 저비용의 생물학적 바이오디젤 생산 시스템 구축을 위해 고활성의 효소 개발, 경제적 재조합 대량생산, 반복 재사용을 위한 효소고정화 등을 통해 고효율의 생산반응계를 개발하였다. 우선 바이오디젤 생산공정에 적합한 리파제로서 CalB(Lipase B of Candida antarctica)를 선택하고 분자 진화기술을 이용하여 효소활성을 17배 향상시킨 CalB14를 개발하였다. CalB14를 효모 발현시스템을 이용하여 경제적 대량생산하기 위해 단백질분비를 획기적으로 개선할 수 있는 맞춤형 분비융합합인자기술(TFP technology)을 이용하여 재조합 CalB를 2 grams/liter 수준으로 분비생산하였다. 생산된 효소를 반복 재사용이 가능하도록 다양한 레진에 고정화하였고 최적의 바이오디젤 전환반응용 고정화효소를 개발하였다. 고정화효소를 효율적으로 재사용하기 위해 바이오디젤 생산용 고정상반응기(packed-bed reactor)를 제작하였으며 기질을 12시간내에 95% 이상 바이오디젤로 수십회 이상 반복전환할 수 있는 경제적인 생물학적 바이오디젤 전환 시스템을 구축하였다.

  • A study on the cementation for the recovery of tin with aluminium in the hydrochloric acid solution was carried out. Parameters, such as aluminium metal equivalent, pH, reaction time, reaction temperature and the concentration of chloride ions were investigated. The experimental results showed that the cementation rate of Sn(II) ions increased with increase of the addition amount of aluminium powders, temperature, pH and the concentration of chloride ions in hydrochloric acid solution. From the results, the optinum conditions for recovery of metallic tin by cementation with aluminium metal powders were proposed.

  • RP-HPLC (Reverse phase-high performance liquid chromatography) assay which was introduced to measure quantitatively the amount of 4-methylthio-3-butenyl isothiocyanate in the radish root, proved to be convenient, accurate and reproducible, showing a good linearity between 10 n moles/ml and 120 n moles/ml (r=0.9997). The results of this assay showed that while the isohtiocyanate was hydrolyzed slowly in the basic medium, it decomposed rapidly in the acidic aqueous medium. On the other hand, the isothiocyanate was relatively stable in the organic solvent (65% acetonitrile). Also, it was found that intact root of Korean radish contain 210-420 ${\mu}moles/100g$, based on the measurement with radish homogenate (pH 8.5, 1 min).

  • An alkali-tolerant bacterium producing the xylanase was isolated from soil and identified as Bacillus alcaiophilus. This strain, named B. alcalophilus AX2000, was able to grow and produce xylanase optimally at pH 10.5 and $37^{\circ}C$. The maximum xylanase production was obtained when 0.5%(w/v) birchwood xylan and 0.5%(w/v) polypeptone and yeast extract were used as carbon source and nitrogen source, respectively. The biosynthesis of xylanase was under the catabolite repression by glucose in the culture medium, and inhibited in the presence of high concentration of xylose. The maximum activity of xylanase was observed at pH 10.0 and $50^{\circ}C$ and the enzyme activity remained was over 80% at $60^{\circ}C$ and from pH 5.0 to 11.0.

  • A continuous-automated method for the determination of nitrite ion using gas-sensing membrane electrode was developed. The pH electrode of tubular PVC membrane type was used as a detector of this system. The slope of linear response of the electrode measured at optimum conditions for the continuous-automated determination of nitrite ion was 63.5 mV/decade. The concentration range of linear response and detection limit were 2.5 ${\times}10^{-4}{\sim}\;7.5{\times}10^{-2}$M and $8.0{\times}10^{-5}$M, respectively. This detection system was not only less interfering to acidic gas species than other methods but also less time consumable for determination.

  • The effect of media composition and culture condition on mycelial growth and cordycepin (3'-deoxyadenosine) production was determined using Cordyceps spp. Among the strains of C. militaris and C. sinensis tested, C. militaris KCTC 6862, C. militaris DGUM 32003 and C. militaris KCTC 16932 were excellent for the production of cordycepin. The optimal temperature and pH for production of cordycepin were $24^{\circ}C$ and pH ranged from 6.0 to 10, respectively. Among various sources of carbon and nitrogen tested, glucose and tryptone were very excellent for the production of cordycepin, respectively. After 5days cultivation with 1% of tryptone with nitrogen source, 39mg/l of cordycepin was produced. However, addition of phosphorus sources reduced the production of cordycepin.

Characteristics of improved Kochujang (개량식 고추장의 특성)

  • In order to investigate systematically characteristics of improved Kochujang in comparision with traditional Kochujang, changes of components in curing of two Kochujangs, enzyme activities of koji and Meju, and effects of salt and red pepper were measured. And it was found that the large differences of amino nitrogen and reducing sugar contents were appeared in the initial stage of the curing period, but the changes were parallel thereafter. The protease activity of koji was maximum in weak acidic pH and that of Meju was maximum in neutral pH. The optimum condition of substrate digestion was three hours at $60^{\circ}C$, and that was more effective than curing at room temperature. The activities of protease and saccharogenic amylase were decreased remarkably by adding salt and red pepper, Therefore, to decompose starch and protein effectively, the addition of salt and red pepper after substrate digestion was more favorable.

Method of Using Acid Hydrolysis to Increase the Efficacy of Decreasing Alcohol Concentration from Hovenia dulcis Extract (헛개열매 추출물의 산 가수분해에 의한 알코올 분해 효능 증대)

  • This work was a method that used an acid hydrolysis for increasing the efficacy of decreasing alcohol concentration from Hovenia dulcis extract. For acid hydrolysis, the best pH was 2.0 to obtain a maximum alcohol dehydrogenase activity at fixed reaction temperature and time. At pH 2.0, reaction temperature $80^{\circ}C$ and reaction time 4 hr gave the highest activity which was $124\%$ of control. The bioactive compound, (+)-dihydromyricetin, content increased to $30\%$ after acid hydrolysis. This is very simple and efficient method to increase the efficacy of decreasing alcohol concentration from Hovenia dulcis extract.

  • 본 연구는 반도체소자 제조공정에 적용되는 CMP공정 중 LPP(Landing Plug Poly) Contact간의 소자 분리를 위해 진행되는 LPP CMP의 후 세정 과정에서 유발되는 방사형 Defect 제거에 관한 내용이다. 방사형 Defect은 LPP CMP 후에 노출되는 BPSG, Poly, Nitride Film과 연마재로 사용되는SiO2 입자, 후 세정과정에서 적용되는 SC-1, DHF, $NH_4OH$ Chemical과 Brush와의 상호작용에 의해 발생되며, Cleaning시의 산성 분위기 하에서 각 물질간의 pH와 Zeta Potential의 차이에서 기인한다. 이 Defect을 제거하기 위해 LPP CMP후에 Film 표면에 노출되는 각 물질의 표면 특성과 CMP 후 오염입자의 흡착과 재 흡착에 영향을 미치는 Electrostatic force와 Van der Waals force, PVA Brush에 의한 Mechanical force의 상호작용을 고려하여 최적 후 세정 조건을 제시 하였다.

Polyethylenimine Mediated Gene Delivery with Various Liposomal Formulations (폴리에틸렌이민 및 그들의 리포좀이 중재된 Plasmid DNA의 운반)

  • The transfection efficiency of plasmid DNA was inspected using multi-cationic polymer, 5, 10, 25 and 50KD polyethylenimine (PEI). The optimal neutralization ratio of PEI/DNA complexes by agarose assay was 1.5-2.0 (nmol/nmol) without much difference in molecular weight of PEI.In vitro transfection assay, most of PEI-mediated plasmid delivery was better compared to the naked DNA. Especially, 25KD PEI at optimal condition gave higher transfection rather than the standard assay of DEAE-dextran or Lipofectin. To enhance the cell targeting delivery, the liposome formulations were introduced using phospholipids. As a result, PC/PE liposomes increased 2-2.5 times of the transfection efficiency of PEI single or PC/PE single delivery, but not the case of 25KD PEI. Moreover, the DOTAP/PE-introduced PEI delivery reduced the transfection of DOTAP/PE single delivery. All these results proved that the PEI can be used not only good transfectants and but also good DNA condensing agents in neutral/anionic liposome for cell targeting delivery.
