Nematode-Trapping Fungi Showed Different Predacity among Nematode Species (선충 종류별 4종 포식성곰팡이의 포식력 차이)

  • Nematode-trapping fungi develop trap and consume nematodes are an important part of the subsoil ecosystem and they share a special predator-prey relationship. Four nematode-trapping species, there with adhesive network, Arthrobotrys oligospora, A. sinensis, A. thaumasia and one with constricting ring, Drechslerella brochopaga were collected from soils in Korea and tested their predacity against 12 different nematode species. They were three feeding groups, plant-parasitic (Meloidogyne incognita and Pratylenchus penetrans), fungivorous (Aphelenchus avenae), bacteriovorous (Betlerius sp. and Diplogasteritus sp. in diplogasterid, Panagrolaimus labiatus, P. multidentatus in panagrolaimid, Mesorhabditis irregularis, Pelodera strongyloides and Rhabditis sp., in rhabditid, and Acrobeloides sp. in cephalobid). Results showed that nematode-trapping fungi successfully captured most of nematodes in Petri dish in the group of plant-parasitic nematodes and rhabditids, moderately and variably in other nematodes in 15 days. But it didn't captured A. avenae and Acrobeloides sp. both belongs to c-p group 2. Numbers of Acrobeloides sp. and A. avenae even increased during the test period. The results of this study indicated that nematode-trapping fungi may have specificity among nematode species.

Diversity Census of Fungi in the Ruminal Microbiome: A meta-analysis (반추위 곰팡이 다양성 조사 : 메타분석)

  • This study was designed to examine the diversity census of fungi in rumen microbiome via meta-analysis of fungal 28S rDNA sequences. Both terms, "rumen" and "ruminal," were searched to retrieve the sequences of rumen fungi. As of September 2016, these sequences (n=165) of ruminal origin were retrieved from the Ribosomal Database Project (RDP; http://rdp.cme.msu.edu), an archive of all 28S rDNA sequences and were assigned to the phyla Ascomycota, Neocallimastigomycota, and Basidiomycota, which accounted for 109, 48, and 8 of the 165 sequences, respectively. Ascomycota sequences were assigned to the genera Pseudonectria, Magnaporthe, Alternaria, Cochliobolus, Cladosporium, and Davidiella, including fungal plant pathogens or mycotoxigenic species. Moreover, Basidiomycota sequences were assigned to the genera Thanatephorus and Cryptococcus, including fungal plant pathogens. Furthermore, Neocallimastigomycota sequences were assigned to the genera Cyllamyces, Neocallimastix, Anaeromyces, Caecomyces, Orpinomyces, and Piromyces, which may degrade the major structural carbohydrates of the ingested plant material. This study provided a collective view of the rumen fungal diversity using a meta-analysis of 28S rDNA sequences. The present results will provide a direction for further studies on ruminal fungi and be applicable to the development of new analytic tools.

Enhanced antimicrobial effectiveness of Omija (Schizamdra chinesis Baillon) by ClO2 (chlorine dioxide) treatment (오미자 수확 후 이산화염소수를 이용한 표면 세척에 따른 미생물 저감 효과)

  • The purpose of this study was to investigate antimicrobial effectiveness of Omija (Schizandra chinensis Baillon) treated with $ClO_2$ (chlorine dioxide) concentration (10, 15, 20, 25 and 30 ppm), washing time (30, 60 and 90 sec) and multiple proportion (x1, x2, x3 and x4). The seven groups were divided into control (Omija without washing water treatment), W-T (Omija treated by tap water ($20{\pm}1^{\circ}C$) for 30 seconds), $ClO_2$-10 (Omija treated by 10 ppm $ClO_2$), $ClO_2$-15 (Omija treated by 15 ppm $ClO_2$), $ClO_2$-20 (Omija treated by 20 ppm $ClO_2$), $ClO_2$-25 (Omija treated by 25 ppm $ClO_2$), $ClO_2$-30 (Omija treated by 30 ppm $ClO_2$), and then they were detected number of total aerobic bacteria, yeast and mold. The rate of inactivation was found, for microorganisms of total aerobic bacteria, yeast and mold, to increase with a increase of $ClO_2$ treatment concentration and multiple proportion. No total aerobic bacteria, yeast and mold in $ClO_2$-30 sample treated for 30 sec, $ClO_2$-15 treated for 60 sec and $ClO_2$-10 treated for 90 sec were detected, and in $ClO_2$-30 Omija with multiple proportion ${\times}1$ (Omija : 30 ppm $ClO_2$ solution ratio was 1:1 (w/w)), $ClO_2$-20 with ${\times}2$ (Omija : 20 ppm $ClO_2$ solution ratio was 1:2 (w/w)) and $ClO_2$-15 with ${\times}4$ (Omija : 15 ppm $ClO_2$ solution ratio was 1:4 (w/w)) respectively.

  • Nematophagous fungi can capture, kill, and digest nematodes using a specific capturing organ. Of the nematophagous fungi, while Arthrobotrys flagrans and A. superba have been described previously, certain characteristics have not been described. For a detailed description of the two nematophagous fungi, the fungi were isolated from soil samples and produced in a pure culture. Morphological characteristics, such as predatory ability (according to the nematode species), shape, and size of predatory organ, conidia, and chlamydospore were investigated and they were used for identification of the fungal isolates along with molecular phylogenetic analysis. Furthermore, this study provides the classification key for 21 nematophagous species.

Effects of Chlorine Water and Plasma Gas Treatments on the Quality and Microbial Control of Latuca indica L. Baby Leaf Vegetable during MA Storage (염소수와 플라즈마 가스 처리가 왕고들빼기 어린잎채소의 MA저장 중 품질과 미생물 제어에 미치는 영향)

  • This study was carried out to investigate the effect of chlorine water and plasma gas treatment on the quality and microbial control of Latuca indica L. baby Leaf during storage. Latuca indica L. baby leaves were harvested from a plant height of 10cm. They were sterilized with $100{\mu}L{\cdot}L^{-1}$ chlorine water and plasma-gas (1, 3, and 6hours), and packaged with $1,300cc{\cdot}m^{-2}{\cdot}day^{-1}{\cdot}atm^{-1}$ films and then stored at $8{\pm}1^{\circ}C$ and RH $85{\pm}5%$ for 25days. During storage, the fresh weight loss of all treatments were less than 1.0%, and the carbon dioxide and oxygen concentrations in packages were 6-8% and 16-17%, respectively for all treatments in the final storage day. The concentration of ethylene in the packages fluctuated between $1-3{\mu}L{\cdot}L^{-1}$ during the storage and the highest concentration of ethylene was observed at 6 hours plasma treatment in the final storage day. The off-odor of all treatments were almost odorless, the treatments of chlorine water and 1 hour plasma maintained the marketable visual quality until the end of storage. Chlorophyll content and Hue angle value measured at the final storage day were similar to those measured before storage in chlorine water and 1 hour of plasma treatments. E. coli was not detected immediately after sterilization in all sterilization treatments. After 6 hours of plasma treatment, the total bacteria fungus counts were lower than the domestic microbial standard for agricultural product in all sterilization treatments. The total aerobic counts in the end storage day increased compared to before storage, whereas E. coli was not detected in all sterilization treatments. The sterilization effect against bacteria and fungi was the best in chlorine water treatment. Plasma treatment showed sterilization effects, but within a prolonged period of time. In addition, the sterilization effect decreased gradually. These results suggest that chlorine water and plasma treatment were effective in maintaining Latuca indica L. baby Leaf commerciality and controlling microorganisms during postharvest storage.

Effect of kimchi on the Microbiological properties of fermented sausages during ripening period (김치 첨가가 발효소세지 숙성중 미생물 특성에 미치는 영향)

  • We studied the effects that the addition of Kimchi has on microbiological properties of fermented sausages as we observed pH and microbiological changes of Kimchi-added fermented sausage by aging period. The pH figure of sausages added with commercial starter culture was 5.69 at the beginning stage and declined gradually to 5.01. The more Kimchi is added, the lower pH figure goes. Sausages with 10% and 20% of fresh Kimchi, and 2% and 4% of powdered Kimchi added, showed about pH 5.4 at the early stage and down to pH 4.87 as they are stored longer. In terms of microbiological changes, sausages mixed with starter culture showed 10$\^$7/cfu/g level in the number of total bacteria, lactic acid bacteria, yeast and fungi, lower than Kimchi-added sausages. However, the figure soared sharply to 10$\^$10/cfu/g during the aging period. Kimchi-added sausages recorded 10$\^$9/cfu/g, slightly high during the eurly aging stage, in the number of total bacteria, lactic acid bacteria, yeast and fungi, while showing a gradual increase up to 10$\^$10/cfu/g towards the end of aging period and became microbiologically stabilized.

Effects of Supplemental Synbiotics Composed of Anaerobic Bacteria, Yeast and Mold on the Aerobic Stability of Total Mixed Ration for Cattle (혐기성 박테리아, 효모 및 곰팡이로 제조된 synbiotics의 첨가가 축우용 완전혼합사료의 호기적 안전성에 미치는 영향)

  • This study was conducted to investigate the effects of supplementation of synbiotics manufactured with anaerobic bacteria, yeast and mold on preservation of total mixed ration (TMR) by exposing days. Eight treatments were composed of untreated synbiotics(US), bacterial synbiotics (BS), yeasty synbiotics (YS), moldy synbiotics (MS), bacterial and mouldy synbiotics (BMS), yeasty and moldy synbiotics (YMS), bacterial and yeasty synbiotics (BYS), and bacterial, yeasty and moldy synbiotics (BYMS). After 7 days of anaerobic fermentation, fermented-TMRs were exposed to the air during 1, 3, 5, 7, 14 and 21 days. One hundred forty four (8 treatments${\times}$6 days${\times}$3 replications) fermented-TMRs were manufactured by vinyl bag ($43\;cm{\times}58\;cm$). Although no significant differences in the activities of carboxymethylcellulase, xylanase and amylase were observed among treatments, theirs acivities were seemed to increase by treatment of BYS or YMS containing yeast. Total bacterial and mold counts also decreased in the treatments containing yeast. Potential pathogenic bacteria were less detected in BYS and BMYS for E. coli, BMYS and YS for Salmonella, and BMS and BMYS for Shigella than those of the other treatments, MS was, however, contaminated easier than US by pathogenic bacteria. From above results, synbiotics containing facultative anaerobic yeast have effects for preservation of TMR fermented anaerobically. Particularly, BMYS treatment having good results in nutrient contents, dry matter loss and pathogenic bacteria amounts was a resonable synbiotics for preservation of the fermented-TMR.

Identification and Chemotype Profiling of Fusarium Head Blight Disease in Triticale (국내 재배 트리티케일에 발생한 붉은곰팡이병의 다양성 및 독소화학형 분석)

  • This study aimed to assess the disease incidence and distribution of toxigenic in Korean triticale. The pathogen of triticale that cause Fusarium head blight were isolated from five different triticale cultivars that cultivated in Suwon Korea at 2021 year. The 72 candidate were classified as a Fusarium asiaticum by morphology analysis and by ITS1, TEF-1α gene sequence analysis. And the results of pathogenicity with 72 isolates on seedling triticale, 71 isolates were showed disease symptom. Also, seven out of 71 Fusarium isolates were inoculated on the wheat, to test the pathogenicity on the different host. The results showed more low pathogenicity on the wheat than triticale. The results of analysis of toxin type with 72 isolates, 64.6% isolates were produced nivalenol type toxin and other 4.6% and 30.8% isolates were produce 3-acetyldeoxynivalenol and 15-acetyldeoxynivalenol, respectively. To select fungicide for control, the 72 Fusarium isolates were cultivated on the media that containing four kinds fungicide. The captan, hexaconazole, and difenoconazole·propiconazole treated Fusarium isolates were not showed resistance response against each fungicide. However, six isolates out of 72 isolates, showed resistance response to fludioxonil. This study is first report that F. asiaticum causes Fusarium head blight disease of triticale in Korea.

Effects of Gamma Irradiation on the Decontamination of Animal Feeds : Sterilization of Protein Sources (배합사료 원료에 대한 방사선 살균 효과 -단백질원의 살균-)

  • The effect of gamma irradiation of microbiological and physicochemical properties of raw ingredients (thirteen kinds of protein sources) for the mixed feed were investigated. The total aerobic bacteria counts in the samples was ranged from $10^2$ to $10^7$cells/g. After 5 to 7 kGy irradiation, the total count was reduced by 3 to 4 log cycles. Coliforms were detected in seven samples and the range was from $10^4$ to $10^6$cells/g. Enteric pathogens were found only in fish meal and poultry byproduct as $10^2$ to $10^6$cells/g. They were sensitive to radiation and completely sterilized by 3 to 5 kGy irradiation. Total fungi count was ranged from $10^1$ to $10^4$cells/g in all samples. They were osmophiles such as Aspergillus, Penicillium, Cladosporium, Aureobasidium and Rhizopus and were destroyed by 3 to 7 kGy irradiation. Five species of potential mycotoxin producers including Aspergillus flavus were also identified. Physicochemical properties, such as total amino acid content, TBA value, POV and color difference showed that irradiation with optimum does was less detrimental than autoclaving.

Combined Treatment of Aqueous Chlorine Dioxide, Organic Acid, and Blanching for Microbial Decontamination of Wild Vegetables after Harvest (수확 후 산채류의 미생물 제어를 위한 이산화염소수와 유기산 및 Blanching 병합 처리)

  • To improve the microbiological safety of wild vegetables after harvest, Aster scaber and Cirsium setidens Nakai were treated with combinations of 50 ppm aqueous chlorine dioxide ($ClO_2$)/0.5% citric acid or fumaric acid, and 50 ppm $ClO_2$/0.5% fumaric acid/blanching at $90^{\circ}C$ for 2 min. Combined treatment of 50 ppm $ClO_2$ and 0.5% citric acid reduced populations of total aerobic bacteria, yeast, and molds in Aster scaber and Cirsium setidens Nakai by 2.80~3.64 and 2.02~2.67 log CFU/g, respectively, compared to those of the control. Combined treatment of 50 ppm $ClO_2$ and 0.5% fumaric acid reduced total aerobic bacteria, yeast and molds populations by 3.62~3.82 and 2.47~3.02 log CFU/g, respectively. Based on the results, combined treatment of $ClO_2$ and fumaric acid was more effective in controlling microorganisms in the wild vegetables than either $ClO_2$ or citric acid. In addition, combined treatment of $ClO_2$/fumaric acid/blanching reduced the populations of total aerobic bacteria by 4.59~5.12 log CFU/g, and populations of yeast and molds were not detected by treatment. These results suggest that combined treatment of $ClO_2$/fumaric acid/blanching is the most effective method for improving microbiological safety of wild vegetables after harvest.
