Studies on the Preparation of Improved Soysauce Kojis (메주제조개선(製造改善)에 관(關)한 연구(硏究))

  • In order to determine proper ratio of wheat to soybean of soysauce koji material for Korean people's taste, sensory evaluation for the soysauces made from soysauce kojis with various above-mentioned ratio and analysis of chemical components through the brewing period of six months were carried out. The results obtained were summarized as follows: 1. The specific gravity of all the soysauces increased gradually as the time passed and had maximum values in two months with nearly constant values thereafter. Concerning soybean/wheat-ratio, generally, the higher soybean/wheat-ratio, the lower specific gravity was observed but when this ratio was higher than 10 : 6, the gravity was on the similar levels. 2. The solid material in all the soysauces increased in the earlier stage and there were no significant differences among the treatments but soysauces without wheat had a little low content. 3. The lesser amount of wheat as raw material was, the higher contents of total nitrogen were and the contents of total nitrogen in the every ratio of raw material increased parallely as the mash ages. 4. The amino-nitrogen levels were found to have the simllar tendency to total nitroghen levels. 5. The reducing sugar content was higher in the soysauces with more-wheat and increased rapidly up to the maximum level in two months and then the higher wheat/soybean-ratio was, the slower decrease of the sugar content was observed. 6. The maximum total acidity was observed in the soysauces with 10; 4-6 (soybean/wheat-ratio) and increased similary in all the soysauces throughout the period. 7. According to the sensory evaluation, the short (2-3 months) fermented soysauces showed best taste when soybean/wheat-ratio was 10 : 8 and 10 : 6 if diluted, but in long (4-6 months) fermented soysauces it was 10 : 6 and 10 : 4 if diluted.

A Review of the Characteristics of Early Apparatus and Methods for Hemoglobin Estimation (Hemoglobin 평가를 위한 초기 기구의 특성 및 측정법 고찰)

  • Since the late 19th century, scientific logic and techniques have been used extensively in the field of clinical pathology, including many laboratory tests utilizing various apparatuses and instruments. Among the techniques to measure hemoglobin, the visual color comparison method was most popular around this time; the specific gravity method and gasometric method were not widely adopted. Instruments that use the visual color comparison method include Gowers' hemoglobinometer, von Fleischl's hemoglobinometer, Dare's hemoglobinometer, Oliver's hemoglobinometer, Haden-Hausser hemoglobinometer, and Spencer Hb meter. Initially, the visual color comparison methods were used to diluate and hemolyze blood with distilled water and then to measure its color. Later, these methods were further developed to measure hemoglobin without dilution, and improved with the formation of acid or alkaline hematin ensuring the stability of color development. Hammerschlag's method as well as the Schmaltz and Peiper's methods were based on specific gravity measurement, but they were not widely used. The gasometric method used the Van Slyke gasometer, indirectly measuring the hemoglobin concentration. This method provides the most accurate results. This survey examined the characteristics and limitations of hemoglobinometers and methods used to measure hemoglobin from the late 19th century to the early-and mid-20th century. Moreover, this study aims to improve the understanding and applicability of the current methods and emerging technologies used in measuring hemoglobin. It is also expected that this investigation is the starting point to promote awareness of the need to organize historical data for a variety of historical relics of the diagnostic laboratory tests.

Survival and Physiological Responses of the Tunicate Halocynthia roretzi to Salinity Changes (염분변화에 따른 멍게 Halocynthia roretzi의 생존과 생리적 반응)

  • We investigated survival and osmolarity, oxygen consumption, amonia extetion and filtration rates associated with physiological responses of the tunicate Halocynthia roretzi salinity changes. Acclimation times for osmolatity in different salinities took $20{\sim}26$ hours in 60% SW (19.8 psu) and $20{\sim}25$ hours in 80% SW (26.4 psu), while their times took $7{\sim}8$ hours in 110% SW (36.3 psu). Accordingly, acclimation times for high salinities were faster than those for low salinities. Survival (%) was more than 80% at salinity over 26.4 psu, and 6 $days-LS_{50}$ was 25.4 psu. physiological responses such as oxygen consumption, amonia excretion and filtration rates of H. roretzi showed more clear reactions in the longer exposure period (four days after exposure) than that in the beginning of the exposure. To sum up the results, the tunicate might be stressed from the beginning of the exposure in low salinity.

Short Term Runoff Characteristics Change of Stream Water Quality with Different Rainfall Events in Planted Coniferous Forest (침엽수 인공림에서 강우사상별 계류수 수질의 유출특성 변화)

  • This study was carried out to investigate solutes concentration change with respect to discharge change in coniferous forest experiment watershed in Gyeonggi-do, Korea. From 2005 to 2008. Precipitation, discharge, solutes has been analyzed from 23 precipitation events. The results showed that low API induced low discharge. $NH_4{^+}$, $K^+$, and $Ca^{2+}$ were indicated by clockwise and $Cl^-$ and $NO_3{^-}$ were represented by counterclockwise hysteresis loop. ${SO_4}^{2-}$, $Na^+$, $Mg^{2+}$ showed no hysteresis loop pattern. $Cl^-$, $Na^+$, $NH_4{^+}$ was relatively constant due to groundwater during precipitation, $NO_3{^-}$ was increased due to soil water compared to early precipitation. $Cl^-$, ${SO_4}^{2-}$, $Na^+$, $Mg^{2+}$, $Ca^{2+}$ was diluted with respect to increased discharge and $NO_3{^-}$ was diluted in early precipitation and then increased in the end. $NO_3{^-}$ and $Ca^{2+}$ eluviated in early precipitation. This characteristics was presumed by the effect of API, discharge and ground water.

Synthesis and Properties of N-Acyl Glycine Used Triglyceride (트리글리세라이드를 이용한 N-아실 글리신의 합성 및 물성)

  • Amino acid based surfactants as protein-surfactants is eco-friendly compound. So, amino acid based surfactants is expected as next generation surfactants. Amino acid based surfactants has high biodegradability, low toxicity and surface active properties. In this experiment, amino acid based surfactants, cocoyl glycine, was synthesized by glycine and triglyceride such as coconut oil and palm oil. And it was tested the surface tension, emulsifying properties, foam stability and HLB value. The synthesized surfactants was confirmed by FT-IR. Surface tension of surfactants synthesized by coconut oil on diluted aqueous solutions of surfactants was 31.2 dyne/cm at $1.0{\times}10^{-4}mol/L$. Surface tension of surfactants synthesized by palm oil on the diluted aqueous solutions of surfactants was 42.1 dyne/cm at $3.2{\times}10^{-5}mol/L$. Foam stability measured the foam height as time passed. Initial foam height of surfactants synthesized by coconut oil is 14.5 cm, and 10.7 cm after five minutes. Initial foam height of surfactants synthesized by palm oil is 3.0 cm, and 2.8 cm after five minutes. Foam height of surfactants synthesized by coconut oil was higher than surfactants synthesized by palm oil. But foam stability of surfactants synthesized by palm oil was better than surfactants synthesized by coconut oil. The emulsifying properties of synthesized surfactants are observed in benzene and soybean oil and emulsifying properties in organic solvent is better than in soybean oil.

Effect of Oriental Melon Vinegar Treatment on Growth and Disease Control of Oriental Melon (참외식초 처리가 참외 생육 및 병충해 방제에 미치는 영향)

  • Oriental melon vinegar was prepared by two stage fermentations of alcohol and acetic acid. In the alcohol fermentation using oriental melon residual products, alcohol content showed 7.43% in 17$^{\circ}$ brix of initial sugar concentration and 80 h of fermentation time. In the acetic acid fermentation using oriental melon alcohol, acidity showed 5.25% in 250rpm of agitation rate and 200 h of fermentation time. The cultivation esults of oriental melons using its vinegar are as follows. Quantity and quality of samples treated with 500, 1,000 and 2,000 times of oriental melon vinegar were higher than that of control : weight, quality, sugar content and goods production rate were higher to degree of 33∼42 g/piece, 370 ∼460kg/10a, 0.6∼.9$^{\circ}$ Brix, 2∼5%, respectively. Coods production yield of samples treated with 500, 1,000 and 2,000 times of oriental melon vinegar was higher (400∼610 kg/10a) than that of control. The results of control of powdery mildew on oriental melons using oriental melon vinegar as the diluted solution with 500 and 1000 times were identical for control value that used by agrochemical. Powdery mildew were exterminated by 2nd treatment of the diluted solution. In case of aphids, the diluted solution with 500, 1,000 and 2,000 times of oriental melon vinegar exterminated thoroughly by 2nd treatment.

Evaluation of Offshore Water Quality by Bioassay Using the Gametes and Embryos of Sea Urchins (성게 알을 이용한 생물검정에 의한 연안해수 수질평가에 관한 연구)

  • The water quality of offshore waters around the Korean coast was evaluated by bioassay using gametes, embryoss and early development systems of a sea urchin species Hemicentrotus pulcherrimus. The results show that despite the inflows of several river systems, the west coast maintain the grade II water. This is thought to be due to the decrease in pollutant input resulting from the purification of inflow rivers (e.g., the control of waste water discharge and the construction of sewage treatment facilities), and the dilution of pollutants by a strong tidal mixing with a large difference between the ebb and flood tides. However, Asan, Hampyong and Chonsu bays, where circulations of water are relatively poor, and the Kunsan and Mokpo harbors which are influenced by pollutants from neighboring cities have shown the grade III water. The south coast has maintain the grade II or III because developments of sea urchins were moderately or strongly inhibited. Also, Kangjin, Duekryang, Kwangyang, Masan, and Jinhae bays where water circulations are relatively poor, show the grade III water, with strong inhibitions of the early development of sea urchins. The east coast has maintained the grade I and II due to monotonous coastlines and smooth circulation of sea water. However, Chongcho Lake shows the worst water quality due to the breakwater which is constructed to maintain port functions.

Effect of Presowing Treatment with Growth Regulators on Different Growth and Yield Contributing Parameters in Soybean [Glycine max (L.) Merril] (식물생장과 조절제의 종자처리가 콩의 생장과 수량구성요소에 미치는 영향)

  • To know the effect of presowing seed soaking with some growth substances (kinetin, IAA, ethrel and salicylic acid) on vegetative growth and yield contributing parameters in soybean [Glycine max (L.) Merrill] the investigation was undertaken. The salient features from the study are;-All the treatments showed the enhancement effect on vegetative growth, viz. seediling emergence, germination percentage, plant height, number of leaves and branches per plant as compared with control, whereas salicylic acid delayed the seedling emergence process and lowered the germination percentage.-Nodule initiation, number of nodules and nodule weight were hastened and increased respectively under all the treatments whereas IAA showed an opposite effect on all the parameters at the early stages of nodule development.-Kinetin, IAA and ethrel showed the significant enhancement effect on the levels of biochemical parameters throughout the time of seed development whereas salicylic acid and water showed the tendency similar to that of control.-Yield and its components, viz. number of flowers, pods and seeds per plant were significant increased under all the treatments but they didn't show any significant enhancement effect on number of seeds per pod and pod setting rate. One hundred seed weight was lower under all the treatments, indicating the dilution effect resulting from incapability of increased number and size of source to provide the increased size of sink with assimilates.

A Study on the Determination of Slip-up Time for Slip-Form System using Surface Wave Velocity (표면파 속도를 이용한 슬립폼 시스템 상승 시기 결정에 관한 연구)

  • The early setting time of concrete is an important factor determining the slip up velocity of the slip-form system. Accordingly, need is for a technique evaluating the early setting time in order to secure the safety of the slip-form system and the construction quality of concrete. This paper intends to estimate the early setting time by evaluating the setting degree of concrete using surface wave velocity so as to determine the slip up time of the slip-form system. Penetration resistance test and compressive strength test are performed first to clarify the relationship between the early setting time of concrete and the compressive strength. Then, compressive strength test and ultrasonic wave test are conducted to examine the relation between the compressive strength and the surface wave velocity. Continuous wavelet transform is adopted to measure the surface wave velocity. Numerical analysis is carried out to demonstrate the appropriateness of the application of continuous wavelet transform. Based on these results, the propagation velocity of the surface wave required for the slip up of slip-form system is suggested. Finally, a reduced model test of the slip-form system is conducted to verify the feasibility of the proposed surface wave velocity for the determination of th slip up velocity.

Strength Characteristics of Concrete Subjected ta Vertical Continuous Vibration during Initial Curing Period (초기양생 중에 수직방향 연속진동을 받은 콘크리트의 강도특성)

  • In construction site, there are some occasions where concrete under initial curing is being affected by nitration from nearby vibration sources. To study these effects, in this paper, strength characteristics of concrete specimens subjected to continuous vibration up to 12 hours in vertical direction after concrete placement were observed. And through the vibration time control experiment where a number of time combinations consisted of times before and after applying vibration during initial curing period were used as experimental parameters, possibility of concrete strength improvement was investigated. From the experimental results, it could be seen that the concrete strengths were mostly decreased due to the increase of vibration velocity during initial curing period. But fluctuation ratio of concrete strength did not have any close correlations with the vibration times. And results of vibration time control experiment showed that if times before applying vibration sustains at least more than 3 hours, subsequent vibrations after that hours do not affect the concrete strength in any unfavorable ways.
