Effects of a Blend of Prunus Mume Extract as an Alternative to Antibiotics on Growth Performance, Activity of Digestive Enzymes and Microflora Population in Broiler Chickens (항생제 대체 사료첨가제로서 매실추출 혼합물이 육계의 성장, 소화 효소 활성도 및 장내 미생물 균총에 미치는 영향)

  • The current study was designed to define whether a blend of prunus mume extract(25%) containing lactic acid(75%) and grape seed extract(10ppm) could affect in vitro antimicrobial activity and growth performance, intestinal microflora, plasma biochemical profiles and digestive enzymes activities in broiler chickens. In paper disc agar diffusion test, we clearly observed antimicrobial activity against E. coli in response to prunus mume extract or a blend of prunus mume extract. For in vivo test, a total of ninety six 3-d-old male broiler chicks were assigned to basal diet(CON), basal diet supplemented with antibiotics (ANTI) and 0.5% a blend of prunus mume extract(PRNUS) until 35 days of age. Throughout the entire experimental period(3-35 days), there were no differences in BW and FCR between the birds fed the basal diet with antibiotics and the diet supplemented with a blend of prunus mume. However, ANTI group showed a significant increase in BW and total gain compared to CON group. The weights of digestive organs such as the pancreas and mucosal tissues were not affected by dietary treatments. There was no difference in plasma levels of glucose, cholesterol, AST and ALT activity. However, triglyceride in plasma increased(P<0.05) in the birds fed the diet supplemented with 0.5% a blend of prunus mume extract compared to those fed antibiotics supplemented diet. The activities of pancreatic trypsin and amylase, and intestinal hydrolase including disaccharidase were not affected by dietary treatment. The colony forming units(CFU) of lactobacillus in the lower ileal-cecum of the birds fed the diet supplemented with a blend of prunus mume extract was significantly(P<0.05) higher than that of birds fed antibiotic supplemented diet without affecting the CFU of E. coli. In conclusion, the birds fed the diet supplemented a blend of prunus mume as an alternative to antibiotics showed a similar growth performance and an significant increase in lactobacillus population compared with the birds fed basal and antibiotics supplemented diets.

Effect of the Use of Antibiotics on Meat Quality Grade of Broiler Carcass (항생제 사용 유무가 계육의 품질 등급에 미치는 영향)

  • The objective of this study was to investigate the effect of the supplementation of antibiotics on meat quality grade of broiler carcasses under the Korean meat grading system. A total of 34,000 broiler carcasses, which were randomly selected 100 broilers from each farm in 340 broiler farms (305 farms with antibiotics; 35 farms without antibiotics), were used in this study. Average daily weight gain of broilers was not significantly affected by the use of antibiotics. The ratio of quality grade A was higher in the broiler carcasses from the farms with antibiotics supplementation (78.87%) than those without antibiotics (73.46%, P<0.05). The numbers of carcass defects, which are down-grade factors of meat grading system, were much higher in the broiler carcasses produced from the farms without antibiotics supplementation when compared with those with antibiotics. In particular, the incidence of the carcass defect on conformation was approximately 1.8 times higher in the carcasses from the farms without antibiotics supplementation than those with antibiotics, which showed the highest negative correlation with quality grade A. Therefore, the improvement of farm management system, the use of antibiotics alternatives from natural products, and a proper incentive should be considered in various ways to encourage farmers to reduce antibiotics usage in their farm and earn consumers' trust for antibiotics-free products.

  • 지난 4월 19일 생산지수 405를 기록하여 체리부로 계열농가 중 가장 높은 수치를 기록한 태성농장을 찾아가 보았다. 태성농장은 3만수 규모로 계사 $2,046m^2$ (619평), 부지 $4,298m^2$(1,300평)의 육계농장으로 부지 $3,306m^2$(1,000평)규모의 증축을 계획하고 진행 중에 있다. 2008년 무항생제축산물 인증을 받았으며 질병예방과 천연 항생제대체물질 사용으로 면역력 증가에 중점을 두고 있다. 태성농장 김응수(54) 사장은 본회 홍성육계지부 이사와 체리부로지역(홍성 부여)농가협의회 회장으로 활동 중이다. 충남 홍성군 광천리에 위치한 태성농장 김응수 사장을 만나 농장운영 방식과 국내 육계산업 발전 방향에 대한 생각을 들어 보았다.

  • Antimicrobial drug-resistance is natural response to antimicrobial stress based on selection, which weakens chemotherapy effect. Introduction of large numbers of chemotherapeutic agents to clinical practice has generated strains of microorganisms that survive and multiply in vivo with high-drug concentrations. Methicillin-resistant Staphylococcus aureus (MRSA), bacteria found in normal daily life, can be easily ingested through milk vegetables, and meats, etc. MRSA emerged in many port of the world, increasing complex clinical problems. Therefore, new agents are needed to treat MRSA. Glycyrrhiza uralensis was extracted using 80% MeOH to investigate its antimicrobial activity against MRSA stains KCCM 11812, 40510, and 40512 through bacterial measurement, disc diffusion, and O.D. methods, MIC values, MRSA gene expression investigation, and scanning electron microscope observation. Results revealed MecA, Mecl, MecRI, and FemA were the most highly manifested MRSA genes. Methanolic extract of G. uralensis significantly inhibited MRSA and thus could be used in development of antibacteria.

  • 식품산업에서 박테리오파아지를 첫째, 동물 보건에서 항생제 대체품으로서, 둘째, 식품에서 바이오보존제(biopreservative)로서, 셋째, 식품 체인에서 병원성 세균을 검출하기위한 도구로서 다양하게 이용하고자 최근에 널리 인지하고 있다. 박테리오파아지는 바이러스로서 세균을 감염시키고 용해하는 특성이 있으며, 식품 안전과 관련하여, 1) 포유 동물세포에 무해하여 박테리오파아지를 안전하게 이용할 가능성, 2) 발효식품에 적절한 스타터 역할을 하거나 천연 미생물균총을 그대로 보존하는 박테리오파아지의 높은 숙주 선택성이라는 2가지 특징을 지니고 있다. 최근에는 식품첨가물로서 박테리오파아지를 '먹을 수 있는 바이러스'로 인정할 수 있을지 논의되고 있다. 식품유래 병원성 세균을 제어하기 위한 파아지의 이용가능성과 생물학 분야에서 관련되는 기초연구를 하고자 할 때 생기는 제한점 등에 대해서 고찰하고자 한다.

Antioxidant, Antimicrobial Acitivities and Cytotoxicity of Hot Water Extracts of Major Herbs in Korea (국내산 주요허브 열수 추출물의 항산화, 항균활성 및 세포독성)

  • In this study, the DPPH free radical scavenging activity, antimicrobial effects, growth inhibition and cytotoxicity of herb extracts were determined to screen alternative antibiotics. Hot water extracts of 10 species herbs (Origanum vulgare, Monarda didyma, Echinacea purpurea, Ocimum basilicum, Mentha piperita, Melissa officinalis, Thymus vulgaris, Stevia rebaudiana, Rosmarinus officinalis, Matricaria recutica) were used. DPPH free radical scavenging activity of all herb extracts was ranged from 31.4-49.9%, and significantly great activties were found at Rosmarinus officinalis, Origanum vulgare and Matricaria recutica (P<0.05). Hot water extracts of monarda didyma, origanum vulgare, thymus vulgaris and rosmarinus officinalis showed greater antimicrobial activities compared to others. Additionally, those four extracts represented relatively low cytotoxicity compared to others. As a result, it was found that Origanum vulgare and Rosmarinus officinalis which possessed great antioxidant and antimicrobial activity with less cytotoxicity. So these two herb extracts can be used as an alternative of antibiotics for organic farming.

Effects of Drinking Rhus Tree-Extract on Performance of Broiler (옻나무 추출액의 음수투여가 육계의 생산성에 미치는 영향)

  • The effect of drinking rhus tree-extract on performance of broiler was performed to investigate development of natural antibiotic in process of broilers production. A total of 320 broiler chicks at 1 day of age were fed the commercial diet and water, drinking water containing 0 ppm(control), 500 ppm (T1), 1,000 ppm (T2) and 2,000 ppm (T3) of rhus tree-extract fur seven weeks. The body weight gain and fred conversion ratio were not different by drinking rhus tree-extract until 35 days of age, but body weight gain and feed conversion ratio were tend to improve by drinking rhus tree-extract over 35 days of age, the improvement was higher in both T$_1$, and T$_3$ than control group(p<0.05) when 35 to 42 days of age. There was a decrease in the microflora population of both E. coli. end Salmonella in the cecum contents and feaces in broilers by drinking rhus tree-extract. The digestibility of dry matter, crude fat, and crude ash of feed were tend to increased in broiler drinking rhus tree-extract. Digestibility of crude protein of feed in broiler three treated groups was significantly improved (p<0.15) as compared with those in control group. These results indicated that the drinking rhus tree-extract were effective in the body weight gain, feed conversion ratio, decreasing of microflora population of both E. coli and Salmonella in the cecum contents and feaces, and it had also effective the digestibility of nutrients in broilers.

  • This article describes the concept and the trend of antimicrobial coating agents, which will help to establish the direction of the research and development on antimicrobial coating agent. Antimicrobial agents are compounds that inhibit or kill microorganisms. They are classified into inorganic, metallic, low molecular weight organic, natural organic, and polymeric compounds. Antimicrobial coatings are applied to the surface of daily necessities, medical devices, industrial products, electrical appliances, fabrics, and interior building materials, etc. Conventional antibiotics penetrate microbes without damaging bacterial cell walls, leading to drug resistance which polymeric antimicrobials can prevent by disrupting cell walls. Most polymeric antimicrobials are focused on cationic polymers. Improvement in the selectivity and durability of antimicrobials and reduction of their toxicity will come true by more reasonable design of molecular structures and their combination in coating system.

Comparison of the Fastness of Dyed Fabric using Natural Extracts and its Antibacterial Efficacy against Antibiotic-resistant Strains (천연 추출물을 이용한 염색포의 견뢰도와 항생제 내성균주에 대한 항균효능 비교)

  • This paper aims to compare and verify the antibacterial effects of cotton fabrics naturally dyed with extracts of Ulmus davidiana var. japonica Nakai, Caesalpinia sappan, Saururus chinensis, and Artemisia princeps against antibiotic-resistant strains of bacteria. After natural dyeing of the test fabric, Al, Cu, Fe mordants were used. The color fastness against washing, rubbing, and perspiration of the cotton fabrics dyed with the 4 types of extracts were mostly excellent. However, the color fastness against light showed poor results for all four types. As for the antibacterial test method, MRSA (ATCC 33591) was applied to the cotton fabrics dyed with the four kinds of extracts and cultured for 24 hours. After that, the bacteria that proliferated on the fabrics were collected and spread on a solid medium. The bacteria were measured to find out the bacteriostatic reduction rate for the antibiotic-resistant strains. As a result of the analysis, all four extracts showed a high bacteriostatic reduction rate of more than 99% when the copper mordant was used. Even with the lack of a mordant, the bacteriostatic reduction rate was high, at 99.9% for Caesalpinia sappan and 94.6% for Saururus chinensis.

Verification of Antimicrobial Activities of Various Pine Needle Extracts against Antibiotic Resistant Strains of Staphylococcus aureus (다양한 적송잎 추출물의 항생제 내성 황색포도상구균에 대한 항균활성 검증)

  • We investigated antimicrobial activities of various pine (Pinus densiflora) needle extracts against antibiotic resistant strains of Staphylococcus aureus. Hot water extract showed the highest antimicrobial activity against normal and methicillin resistant Staphylococcus aureus (MRSA), however, it exhibited no antimicrobial activity against penicillin resistant S. aureus (PRSA). Hot water-hexane (HWH), hot water-ethanol (HWE), hexane, and ethanol extracts showed antimicrobial activity against S. aureus, PRSA and MRSA. Minimum inhibitory concentrations (MIC) of HWH, HWE, hexane, and ethanol extracts were 0.05, 0.05, 0.5 and 5 mg/ml, respectively, and HWH and HWE extracts showed the strongest antimicrobial activity among these extracts. Antimicrobial activities of pine needle extracts were stable after heating at $121^{\circ}C$ for 20 min. These results suggested that pine needle extracts can be used as an effective natural antimicrobial agent for food and medical industries.
