Antioxidative Constituents from Fruit of Sorbaria sorbifolia var. stellipila MAX. (쉬땅나무(Sorbaria sorbifolia var. stellipila MAX.) 열매의 항산화 활성 성분)

  • The purpose of this study was to evaluated the antioxidative constituents and their activities of the 80% methanolic extracts from fruit of Sorbaria sorbifolia var. stellipila MAX. The isolation of active compound was performed in three steps: solvent partition, open column chromatography, and high-performance liquid chromatography (HPLC). The solvent fractions were tested for their antioxidant activities by oxygen radical absorbance capacity (ORAC). The antioxidant activity of 80% methanolic extracts by various solvent partitions was in the order of 80% MeOH (1.68 ${\pm}$ 0.027), n-hexane (1.02 ${\pm}$ 0.036), $CH_2Cl_2$ (0.95 ${\pm}$ 0.025), EtOAc (1.98 ${\pm}$ 0.065), n-BuOH (1.94 ${\pm}$ 0.054) and Water (1.28 ${\pm}$ 0.032). Therefore, the results indicated that the potential antioxidant activities and functional values were observed significantly at EtOAc fraction from fruit of S. sorbifolia, flavonoid compound isolated.

  • The objective of this study was carried out in order to evaluate the antioxidative activities of n-hexane fractions from medicinal herbs. Certain parts from 6 species of edible and medicinal herbs were extracted with methanol. Radical scavenging activity of the n-hexane fractions against DPPH and ABTS radicals were observed to be high in Psoralea corylifolia, and Scutellsria barbata; it also significantly increased (p<0.05). For a comparison of the effect of various concentration of fractions, as the concentration of treatment increased, the antioxidative effects on radical scavenging activity also oncreased; yet it was not significant. The antioxidative effect of medicinal plants was dependent on the concentration but was not significant. Therefore, these results indicate that edible wild plants, such as Psoralea corylifolia, and Scutellsria barbata may be useful as potential antioxidant sources for improving the human antioxidant defense system against disease.

Chromaticity Analysis of Natural Dyes Extracted from Sappan Wood, Gardenia, and Mugwort (소목, 치자, 쑥으로부터 추출된 천연염료의 색도분석)

  • Extracting natural dyes have been widely studied since the needs of eco-friendly and non-toxic natural dyes increased. In this paper, the natural dyes were extracted from sappan wood, gardenia, and mugwort containing brazilein, crocin, and chlorophyll process. After the extraction with variables of pH of solvent and reaction time, the chromaticity of extracted natural dyes was analyzed using quantitative values from brightness and color coordinate (L, a, b) based on the target colors of red, yellow and green. For the case of brazilein and crocin, the cadmium red and cadmium yellow, respectively were extracted. In the case of sappan wood and gardenia, the red pigment under pH 12 (${\Delta}$ = 18.2) and the yellow pigment at pH 9 (${\Delta}$ = 18.4) were extracted respectively. However, the color of extracted chlorophyll from mugwort was different from the target chrome green.

가공반찬류의 저장성 향상을 위한 천연 항균복합제재의 처리효과

  • 식품조리 및 가공은 만들어진 제품의 저장성을 전제로 해야 하기 때문에 식품의 변화 혹은 변질에 깊은 이해를 바탕으로 모든 처리와 가공 방법이 검토되어야 한다. 식품의 변질이나 부패는 미생물 작용에 의한 변질이 그대부분을 차지하고 있으며 미생물학적 변질을 방지하기 위해서 화학적 합성보존 제가 상업적으로 사용되고 있으나 최근 소비자의 건강 지향적 욕구가 증대됨에 따라 인공합성품의 기피현상이 두드러지고 있다. 따라서 천연 항균물질의 개발과 이용은 인공 합성보존제의 대처라는 의미와 소비자기피현상을 유발시키지 않으면서 각종 가공식품의 저장성 향상 및 저온 유통식품의 안정성확보라는 견지에서 그 중요성이 있다. 본 실험에서는 식품의 미생물작용에 의한 변질의 저해 효과가 뛰어난 식물성천연항균제품(Botanical antimicrobial agents-Citrus product : 이하 BAAC라 칭함)에 천연보조제(Ginseng extract, Aloe, 매실추출물)를 첨가하여 Paper disk법에 의한 항균성 검사를 실시한 결과 Ginseng extract를 첨가한 경우 생육저해환이 가장 크고 뚜렷하게 나타났다. 따라서 BAAC에 보조제 (Ginseng extract)를 첨가하여 제조 된 천연 항균복합제재(이하 BAAG라 칭함)를 가공 반찬류인 마늘종 무침과 돈육장조림에 첨가하여 처리한 후 꺼내어 상온에서 저장하면서 주사전자현미경 관찰, 대장균수, 총균수의 변화, 외관상의 변화에 미치는 영향을 무처리 대조구와 비교하면서 조사하였다. 주사전자현미경 촬영사진의 결과 분석을 통하여 BAAG를 처리한 균체 세포는 세포막 및 세포벽 기능이 파괴되어 세포내용물이 균체외부로 유출되어 균체의 생육이 억제되며 성장을 저해 또는 사멸시키는 것으로 나타났다. 대장균수는 모든 처리구에서 BAAG의 첨가에 의해 성장이 억제되었으며 농도가 증가시킬수록 균증식 억제 효과가 뚜렷하였으며, 총균수도 압도적으로 낮은 값을 보여주었다. 외관상의 변화와 상품 가치를 측정한 결과는 무늘 종 무침과 돈육장조림의 대조구에서는 4일 경과후 강한 부패취와 아울러, 표피의 갈변정도가 심하되어 관능적으로 부패상태를 인지할 수 있었으며 7일경과 후부터 점질성 갈변물질이 생성되어 상품가치가 크게 떨어진 상태였다. 이와 같은 결과로 볼 때, BAAG의 처리는 BAAC의 경우보다 가격은 저렴하면서도 항균력은 우수한 천연 항균복합제재로써 가공 반찬류의 저장성을 최대한 연장 할 수 있음을 확인 하였고, 반찬류뿐만 아니라 여러 식품군에 다양하게 적용가능 할 것으로 기대된다.

The Study of Preservative System in Cosmetics using Botanical Antimicrobial (식물유래 천연항균제를 이용한 화장품에서의 방부시스템에 관한 연구)

  • Anti-microbial activities of Scutellariae Radix Extracts and Sophora flavecens Extracts was tested by formulation, such as skinsoftener, emulsion for 4 weeks. Control contained no preservative, test group contained 1.00% Scutellariae Radix extracts and 1.00% Sophora flavecens extracts and positive control contained parabens and imidazolidinyl urea. To determine the anti-microbial activity of these extract, the 4 germs such as Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Canida albicans were used. The test groups showed significant anti-microbial activities against the 4 germs at 2 and 3 weeks as compared with control. Anti-microbial activities of these extracts were similar to positive control. Considering that the Scutellariae Radix Extracts and the Sophora flavecens Extracts have a significant anti-microbial activities against 4 germs, it is possible as natural preservative in cosmetics.

Antimicrobial activity of Medicinal plants extracts against Streptococcus sobrinus KCOM 1157 (Streptococcus sobrinus KCOM 1157에 대한 약용식물들의 항균 활성)

  • Natural extracts were isolated from 20 kinds of medicinal plants that have been traditionally widely used in Korea to develop treatments for dental caries. Medicinal plants showing antibacterial activity against a cariogenic bacterium, Streptococcus sobrinus KCOM 1157 were investigated. The natural extracts of Glycyrrhiza uralensis, Sophora flavescens, Coptis Chinensis, Mentha arvensis, Pinus densiflora, Spirodela polyrhiza showed antimicrobial activities against S. sobrinus KCOM 1157. The extracts of Pinus densiflora, Sophora flavescens, Coptis Chinensis among these medical herbs showed significant antimicrobial activity. These results suggested that the natural products of Pinus densiflora, Sophora flavescens, Coptis Chinensis could be the potential source of anticariogenic agent against dental pathogens including S. sobrinus.

Evaluation of Anti-Oxidant from Natural Products (천연물로부터 유래한 천연 항산화제 규명)

  • In this study, we analyzed 80%MeOH extract of fruits of sorbaria sorbifolia var. stellipila MAX. to measure the total antioxidant capacity by oxygen radical absorbance capacity (ORAC) assay, individual flavonoid content by high-performance liquid chromatography (HPLC). n-Hexane ($1.02{\pm}0.036$), $CH_2Cl_2$ ($0.95{\pm}0.025$), EtOAc ($1.94{\pm}0.065$), n-BuOH ($1.98{\pm}0.054$), D.W. ($1.2{\pm}0.032$) fractions were examined antioxidative activity by ORAC assay. It was revealed that EtOAc($1.94{\pm}0.065$), n-BuOH($1.98{\pm}0.054$) fractions had significant antioxidative activity. The isolation and separation were facilitated using open column chromatography, while separation, purification and identification were accomplished by using high-performance liquid chromatography (HPLC) and nuclear magnetic resonance (NMR).

Antimicrobial Activities of the Extracts of Vegetable Kimchi Stuff (식물성 김치재료추출물의 항미생물활성)

  • In order to investigate the possible use of vegetable kimchi stuff as natural preservatives for kimchi, the methanol extracts of 15 kinds of vegetable kimchi stuff were solvent-fractionated and fractions were tested for antimicrobial activities against Leuconostoc mesenteroids, Pediococcus cerevisiae, Lactobacillus plantarum and Saccharomyces cerevisiae. The neutral fractions of the extracts of garlic and leek showed strong antimicrobial activities. The extract of leek showed particularly strong antimicrobial activities against Ped. cerevisiae and L. plantarum that were known to be main microorganism of fermentation and acidification of kimchi. The results suggest the possible use of the leek extract as natural preservatives for kimchi.

Therapeutic Effects in the RIP-treated liver Fibrosis Rat Model (천연식물추출물(RIP)이 쥐의 간섬유화 치료에 미치는 영향)

  • Chronic liver diseases and hepatic cancer have been reported as 10% of cause of death in Koreans. Regardless of various causes, chronic liver disease accompanies commonly hepatic fibrosis. But still the mechanism of hepatic fibrosis remains poorly understood. Using the dimethylnitrosamine(DMN)-induced hepatic fibrosis rat model, We performed to evaluate the possible therapeutic effect of RIP(extracts of Phellodendron amurense and Patrinia scabiosaefolia) and to investigate the changes in referential connective tissue proteins($TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin, and vimentin) as a marker of fibrogenesis. For these purposes, liver tissues were stained with H & E, and Azan staining for estimation of developing fibrosis. In the DMN-treated rat liver tissue, fibrosis were developed forming incomplete septal fibrosis. Whereas, in the RIP-treated rat liver tissues, the fibrosis were decreased recovering to normal morphology. The expressions of $TGF-{\beta}_1$, ${\alpha}$-smooth muscle actin($\alpha-SMA$), and vimetin were increased in the DMN-treated rat liver tissues, but decreased in the various areas of RIP-treated rat liver tissues. According to these results, RIP could be a possible therapeutic agent to reduce hepatic fibrosis, and the $TGF-{\beta}_1$, ${\alpha}$-SMA, and vimentin could be possible indicative markers of hepatic fibrosis development and recovery.

  • 나노섬유를 제조하는 방법 중에는 상분리 현상을 이용한 방법, 자가 조립성을 이용한 방법, 템플레이트를 이용한 방법, 전기방사법이 있으며 특히 전기방사법은 연속적으로 균일한 나노섬유를 제조할 수 있다. 또한 전기방사법은 장비가 간단하며 고분자 blend ratio와 무기재료의 함량에 따라 뛰어난 특성을 나타내는 나노복합섬유를 만들 수 있다. 최근 식물에서 추출한 단백질을 전기방사법을 이용하여 나노입자 및 나노섬유를 제조하고 이를 의료 분야 등에 적용하기 위한 연구가 활발히 진행되고 있으며 이런 식물성 단백질은 동물성 단백질에 비하여 인체 적용이 용이하고 매장량이 풍부한 장점이 있다. 본 연구에서는 전기방사법을 이용하여 옥수수에서 추출한 단백질인 zein의 나노입자 및 나노섬유를 제조하였다. 또한 천연 추출물이 혼입된 복합 나노입자 및 나노섬유를 제조하여 zein이 가진 고유 특성 이외에 천연 추출물의 특성을 추가로 부여해서 더욱 발전된 나노입자 및 나노섬유를 제조하였다. 고분자 농도, 전압, 방사거리 등 다양한 공정변수를 조절하여 최적의 조건을 확립하였으며 제조된 나노입자 및 나노섬유는 field-emission type scanning electron microscope (FE-SEM), transmission electron microscopy (TEM), ultraviolet-visible spectroscopy (UV/vis), fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC)를 이용하여 특성분석을 실시하였다.
