Expression of Genes Affecting Skin Coloration and Sugar Accumulation in 'Hongro' Apple Fruits at Ripening Stages in High Temperatures (고온에 의한 변색단계별 '홍로' 사과의 착색 및 당 축적 관련 유전자 발현 분석)

  • High temperature is one of the important environmental factors limiting cultivation of apple (Malus domestica Borkh). The expression of genes related with anthocyanin synthesis and sugar accumulation in response to high temperature was studied in the 'Hongro' apple fruits at different developmental stages in different temperature conditions through real-time PCR. Expression of ${\hat{a}}$-amylase (BMY) and polygalacturonase (PG) genes related with sugar synthesis was higher in late ripening stages than in initial ripening stages. Expression of four genes such as phenylalanine ammonia-lyase (PAL), chalcone synthase (CHS), flavanone 3-hydroxylase (F3H), and malate dehydrogenase (MDH), which were related with fruit skin coloration, increased gradually in apple fruits of the middle and late ripening stages. Interestingly, the expressions of all genes were highly inhibited expressed at $30-35^{\circ}C$ compared to $25^{\circ}C$ in all ripening stages. In the further work, investigation of expression levels of various genes could be conducted in the level of transcriptomics in fruits at the middle ripening stages to get meaningful information of ripening metabolism in apple in high temperatures.

Relationship between the Deposition of Tear Constituents and the Adherence of Candida albicans according to Soft Contact Lens Materials and Pigmentation (소프트콘택트렌즈 재질과 착색에 따른 눈물성분 침착과 칸디다균 흡착의 상관관계)

  • Purpose: The aim of this study was to figure out how the characteristics of soft contact lens materials and pigmentation affect the adherence of C. albicans on soft contact lenses pre-deposited with tear constituents. Methods: The adherent number of C. albicans on clear soft contact lenses (hereinafter clear lenses) and circle soft contact lenses (hereinafter circle lenses) made of etafilcon A, hilaiflcon B and nelfilcon A, respectively, was measured before and after the deposition of artificial tear. Also, bacteria adherence on lenses were observed by a scanning electron microscope. Results: Adherence of C. albicans was significantly different according to lens materials. The amount of adsorption was not different between clear lenses and circle lenses made of etafilcon A however, the number of bacteria absorption was bigger in hilafilcon B and nelfilcon A lenses. More absorption of C. albicans was found in the non-pigmented central area compared the pigmented area, and non-pigmented peripheral area has more bacterial absorption than non-pigmented central area. The number of C. albicans decreased in the case that tear protein was pre-deposited. The maintenance of antibacterial activity against C. albicans was different according to lens materials thus, etafilcon A has the longest period of its maintenance. Conclusions: It was revealed that the number of C. albicans was different according to the characteristics of lens materials, pigmentation or non-pigmentation, the pigmented area of soft contact lenses. Thus, it is suggested that the management method should be different according to the adsorption characteristics of C. albicans.

Effects of Abscisic Acid (ABA) and Fluridone on Red Coloration of 'Hongro' Apple Fruit Skins (Abscisic acid(ABA) 및 fluridone의 처리가 'Hongro' 사과의 과피 착색에 미치는 영향)

  • The objective of this study was to determine the effect of exogenous abscisic acid (ABA) on the red coloration and endogenous ABA contents of apple fruit skins. ABA and fluridone (an ABA synthetic inhibitor, FD) was sprayed on 'Hongro' apple fruit skins at 107 days after full bloom (DAFB). Visual coloration and hunter's color values were not affected by the ABA and FD treatments. Anthocyanin contents in fruit skins increased similarly to hunter $a^*$ values of fruit skins, but ABA and FD did not affect its accumulations. Liquid chromatography analysis revealed that endogenous ABA contents in control fruit increased at first and then decreased from 12 hours after the treatment. ABA treatment increased ABA contents in fruit skins from 2 hour after the treatment and it lasted until the end of the treatments. FD decreased ABA contents in fruit skins from 6 hours after the treatment. ABA treatment increased MdNCED2 (an ABA biosynthetic gene), MdACO1 (an ethylene biosynthetic gene), and MdCHS and MdDFR expressions. However, MdUFGT expressions were not affected by ABA treatment.

Developmental Characteristics and Feeding Behavior of Green Peach Aphid (Myzus persicae) on Sweet Pepper (Capsicum annuum) Cultivars (착색단고추 품종에 따른 복숭아혹진딧물의 발육특성 및 섭식행동)

  • To investigate host preferencse of Myzus persicae on 15 sweet pepper cultivars grown in Jeonnam Province, EPG (electrical penetration graph) and life table experiments were carried out in the laboratory. Phloem phase times were significantly longer on Ferrari, Jinju, Debla, and Rapido than Orobell and Thialf. Non-penetration times were conversely observed. Life span, reproduction period, total fecundity, and intrinsic rate of increase ($r_m$) of M. persicae, were higher on Ferrari, Debla, Orange glory, and Jinju than on Purple, GreenAce, Orobell, and Thialf. On the bases of these results, we conclude that M. persicae preferred Ferrari, Jinju, Orange glory, and Debla among the 15 tested sweet pepper varieties. However, we could not show the preference of the aphid for Purple, Orobell, and Thialf.

  • Ant Colony System(ACS) Algorithm is new meta-heuristic for hard combinational optimization problem. It is a population-based approach that uses exploitation of positive feedback as well as greedy search. Recently, various methods and solutions are proposed to solve optimal solution of graph coloring problem that assign to color for adjacency node($v_i, v_j$) that they has not same color. In this paper introducing ANTCOL Algorithm that is method to solve solution by Ant Colony System algorithm that is not method that it is known well as solution of existent graph coloring problem. After introducing ACS algorithm and Assignment Type Problem, show the wav how to apply ACS to solve ATP And compare graph coloring result and execution time when use existent generating functions(ANT_Random, ANT_LF, ANT_SL, ANT_DSATUR, ANT_RLF method) with ANT_XRLF method that use XRLF that apply Randomize to RLF to solve ANTCOL. Also compare graph coloring result and execution time when use method to add re-search to ANT_XRLF(ANT_XRLF_R) with existent generating functions.

Improvement of Bleaching Performance of Photosensitive Electrochromic Device by the Additive of TEMPOL (TEMPOL 첨가제 적용에 의한 광감응형 전기변색 소자 탈색성능 향상)

  • We have developed photosensitive electrochromic smart windows that does not require any transparent conducting oxide (TCO) substrate. In our previous study, we demonstrated that a flexible film-type device made with a low temperature curing WO3 sol and TiO2 sol could show a reversible and rapid switching between colored and bleached state via incorporation of platinum catalysts on the surface of WO3 layer. However, when these devices were exposed to sunlight over 4 hour, it was confirmed that they did not return to fully bleached state in the darkened state due to their overcoloring process. In this study, we added 4-hydroxy-(2,2,6,6-tetramethylpiperidin-1-yl)oxyl (TEMPOL) as an additive to the electrolyte of photosensitive electrochromic device to effectively prevent the undesired overcoloring process. The resulting device with TEMPOL indeed did not undergo excessive coloration and showed great reversibility even after being exposed to sunlight for over 4 hours. Various concentrations of TEMPOL were applied to compare changes in the visible transmittance and coloring/bleaching kinetics of devices. In terms of energetic point of view, we proposed a plausible mechanism of TEMPOL to prevent excessive coloration.

  • The attractiveness of the gingiva is determined by its color, shape, and the shape and location of the boundary between the teeth and the gingival tissue. The standards beauty, balance, and health of the gingiva are all different, but the general public would agree that a coral pink gingiva is more beautiful than black or brown gingiva. Hence, one would be able to smile more confidently in public if he or she receives a gingival pigmentation removal surgery that changes the color of black or brown gums to a beautiful pink color with relative simplicity. The color of one's gingiva varies from pale pink to deep bluish purple, depending on many health components. The most prominent among these include the vascular supply, epithelial thickness, the degree of keratinization, and the presence of pigment in the epithelium. Melanin, carotene, reduced hemoglobulin, and oxyhemoglobulin are the main pigments contributing to the normal color of the oral mucosa. The health of one's gingival tissue are essential for an attractive smile. Excessive melanin deposits in the basal and early basal layers of the epithelium stored in the form of melanosomes frequently cause pigmentation. Relationship between the Deposition of Tear Constituents on Soft Contact Lenses according to Material and Pigmentation and Adherence of Staphylococcus aureus (소프트콘택트렌즈 재질과 착색에 따른 눈물성분 침착과 포도상구균 흡착의 상관관계)

  • Purpose: The study was aimed to figure out the effect of materials and pigmentation of soft contact lens on the adherence of Staphylococcus aureus upon soft contact lenses deposited with tear components. Methods: The number of adherent S. aureus on clear and circle soft contact lenses made of etafilcon A, hilafilcon B, nelfilcon A was measured before and after incubation in artificial tear. Furthermore, the denaturalization level of tear protein with time after incubation in artificial tear was estimated by electrophoresis. Results: The adherence of S. aureus was significantly different according to the lens materials. The pattern of bacterial adherence on clear and circle contact lenses was different. That is, the adherent amount of S. aureus was somewhat larger on circle lens made of etafilcon A however, amount on circle lenses made of hilafilcon B and nelfilcon A was 89.3% and 71.3% of the number on clear lenses. When the tear protein was deposited on contact lenses, the number of adherent bacteria decreased and its degree was varied according to the lens material. The degree of decrease was the biggest in clear soft lens made of etafilcon A. Anti-bacterial effect of tear protein decreased with time after deposition of tear protein on soft contact lens and the amount of lysozyme also decreased. The reduction of anti-bacterial effect and quantity of lysozyme was different according to contact lens materials and pigmentation. Conclusions: It was revealed that the adherence of S. aureus depends on contact lens materials and pigmentation, and the specification of lens material affects more on adherence of S.aureus than pigmentation. It was further figured out the denaturalization level of anti-bacterial protein on soft contact lens varies according to lens materials and pigmentation, which produces an effect on the quantity of bacterial adherence.

The residue characteristics of chlorpyrifos in chilli and sweet peppers (고추와 착색단고추 재배 중 사용한 Chlorpyrifos의 잔류특성)

  • The characteristics of chlorpyrifos was studied to clarify the effect of the cultivation season, spray intervals, spray volume and concentration on residues in chilli and sweet peppers cultivated under greenhouse condition. Residue levels of chlorpyrifos detected in chilli pepper fruit cultivated were 1.5 to 2.7 times higher in winter(February-March) than those in summer(June-July). During winter season residue levels of chlorpyrifos in chilli peppers were 6.9~3.2 times higher than those in sweet peppers for 10 days after 3 times application with 7 days interval. Residue levels in chilli pepper were from 2.070 mg/kg at 1 day to 0.929 mg/kg at 10 day, while those in sweet pepper were from 0.302 mg/kg to 0.291 mg/kg. Residue levels in sweet pepper to which are 3 times applied with 7 days interval were from 0.302 mg/kg at 1 day to 0.291 mg/kg at 7 day, and the residue levels in sweet pepper which are 3 times applied with 3 days interval were from 0.498 mg/kg at 1 day to 0.470 mg/kg at 7 day. Residue levels of chlorpyrifos in the sweet peppers applied by double concentration were 2.5 times higher than those sprayed by normal standard amounts diluted in double volume.
