Advances in DNA sequencing technologies have contributed to revolutionary understanding of many fundamental biological processes. With unprecedented cost-effective and high-throughput sequencing, a single laboratory can afford to de novo sequence the whole genome for species of interest. In addition, population genetic studies have been remarkably accelerated by numerous molecular markers identified from unbiased genome-wide sequences of population samples. As sequencing technologies have evolved very rapidly, acquiring appropriate individual plants or populations is a major bottleneck in plant research considering the complex nature of plant genome, such as heterozygosity, repetitiveness, and polyploidy. This challenge could be overcome by the old but effective method known as haploid induction. Haploid plants containing half of their sporophytic chromosomes can be rapidly generated mainly by culturing gametophytic cells such as ovules or pollens. Subsequent chromosome doubling in haploid plants can generate stable doubled haploid (DH) with perfect homozygosity. Here, classical methodology to generate and identify haploid plants or DH are summarized. In addition, haploid induction by epigenetic regulation of centromeric histone is explained. Furthermore, the utilization of haploid plant in the genomics era is discussed in the aspect of genome sequencing project and population genetic studies.
Pinus pumila, which occurs in the northeast Asia, is found limitedly in Daecheongbong area of Mt. Seorak in the South Korea. This population was chosen to study spatial pattern, genetic diversity and spatial genetic structure. There were 48 polymorphic and 30 monomorphic I-SSR markers. A total of 65 individuals which distributed in the study site (40 m ${\times}$ 70 m) showed weakly aggregate distribution (Aggregate Index = 0.871). A total of 40 genets were observed from 65 individuals through I-SSR genotype comparison. Proportion of distinguishable genotype (G/N), genotype diversity (D) and genotype evenness (E) were 61.5%, 0.977 and 0.909, respectively. In spite of the small number and the limited distribution, Shannon's diversity index (I = 0.567) was relatively high as compared with those of other plant species. Spatial autocorrelation using Tanimoto's distance showed that the genetic patch was established within 12 m. Based on Mantel tests, there was relatively low correlation between genetic distance and geographic distance. Therefore, it seems the P. pumila population was formed by many parent trees in early stage. For ex situ genetic conservation of P. pumila, the sampling strategy is efficient at least above 12 m between individual trees.

The diamondback moth, Plutella xylostella, overwinters in some protected areas in Korea. Using a sex pheromone trap, the adults were monitored since the occurrence of the overwintering populations. In Andong, P. xylostella exhibited four adult peaks in a year. Biological characters, such as cold tolerance, insecticide susceptibility, and developmental rate, were analyzed and showed a significant variation among different local overwintering populations. Population genetic variation was assessed with molecular markers, in which the initial high genetic variation among the overwintering populations decreased with the progress of seasons. These results suggests that there may be a significant migration of P. xylostella to decrease the genetic variation among the different local populations that are different in biological characters.
The genetic relationship was examined with PCR-RAPD markers among three scallop species, Chlamys farreri farreri, Patinopecten yessoensis, and Agropecten irradians. Six primers were selected from 60 primers used to compare PCR-RAPD profiles among species. All primers showed distinct RAPD band patterns between the three species. In Chiamys farreri farreri, the morphological characteristics such as shell size and color were considerably different between the two geographical populations. RAPD profile, however, showed that no significant genetic differences were found between the two geographical populations. Polymorphic alleles were observed within a population of each species. Thus, PCR-RAPD markers are useful in identifying scallop species and in understanding scallop population genetic structure.

The invasive red swamp crayfish, Procambarus clarkii, is native to south-central United States and northeastern Mexico. Recently, it has been being spreading in the wild in South Korea. However, its primary sources, introduction routes, establishment, and expansion in South Korea remain unclear. Here, we analyzed genetic diversity and population genetic structures of its domestic natural populations during early invasion, commercial stock from local aquaria (a suspected introduction source), and original United States population using mitochondrial COI gene sequences for 267 individuals and eight microsatellite markers for 158 individuals. Natural and commercial populations of P. clarkii showed reduced genetic diversity (e.g., haplotype diversity and allelic richness). The highest genetic diversity was observed in one original source population based on both genetic markers. Despite a large number of individuals in commercial aquaria, we detected remarkably low genetic diversity and only three haplotypes among 226 individuals, suggesting an inbred population likely originating from a small founder group. Additionally, the low genetic diversity in the natural population indicates a small effective population size during early establishment of P. clarkii in South Korea. Interestingly, genetic differentiation between natural populations and the United States population was lower than that between natural populations and aquarium populations. This suggests that various genetic types from the United States likely have entered different domestic aquariums, leading to distinct natural populations through separate pathways. Results of our study will provide an insight on the level of genetic divergence and population differentiation during the initial stage of invasion of non-indigenous species into new environments.
Considerably large phenotypic and genotypic variations for the important agronomic traits were found in a soybean population consisted with 825 local lines collected in Korea. Genetic coefficient of variation, heritability, and genetic gain for seed weight, plant height. and number of pods per plant were relatively high. Maturity showed a high heritability value with a low genetic gain and the result may be due to the narrow range of maturity date of the population studied.
Genetic variation in Asaum misandrum, a woodland herb in Korea, was investigated based on allozyme analysis with starch gel electrophoresis. All of previously reported populations in Korea were sampled and seven loci from six enzymes were analyzed. Overall genetic variation of A. misandrum population showed considerably high levels of genetic variation within the species (A = 2.05, P = 71.4, $H_E$ = 0.294). A positive $F_{IS}$ value of A. misandrum indicated overall deficiency of heterozygotes, and a low $F_{ST}$ value (0.112) meant very little differentiation among populations. Factors contributing to the high levels of genetic diversity found within populations of A. misandrum include population maintenance via wide distribution range from Korea to Japan and primarily outcrossing breeding system. Although it showed moderate genetic diversity level, most habitats of the species were scattered and discontinuous. Besides, low numbers of individuals were found in the most habitats and individuals are collected frequently from the wild due to the unique shape of the flowers as well as the rarity of the species. Thus, there is a need to set up a reasonable conservation strategies including the maintenance mechanism of genetic diversity of A. misandrum.

Schisandra nigra Max. has been cultivated far a medical use as well as food. It is an endemic species which has a unique habitat at the altitude of 600-1,400 m in Cheju island. In this study, three natural populations of S. nigra were investigated by using of starch-gel electrophoresis to determine the extent and distribution of genetic diversity. Except 2 monomorphic locus (Mdh-2 and Pgi-1), 4 of the 6 isozyme locus (Idh, Mdh-2, Mnr, and Pgi-2), verified from 4 isozymes, revealed polymorphism in the three populations of S. nigra. The mean number of allele per locus was 1.7 and the percentages of polymorphism loci were 38.9% at 95% level and 50% at 99% level respectively. The observed and estimated heterozygosities were 0.141 and 0.147 respectively. Although plants which were in the face of crisis and distributed in the restricted area, have been known to the very low degree of genetic variation, S. nigra showed higher genetic variation than others. Genetic variation was mostly allocated within population and individuals than that among populations. The result of Wright's F analysis estimates of $F_{IT}$ and $F_{IT}$ showed that S. nigra population revealed Hardy-Weinberg steady state.
Surveys of elecirophoretic variations in isozymes and proteins encoded by 25 persumtive loci were conducted to estimate the degree of genic variation and to assess species recognition for three species of the Family Gasterosteidae (Pisces) in Korea. The average degree of genic variation of Gasterosteus aculeatus was HD = 0.086, HG = 0.084, and P =36% and it was similar to Pungitius sinensis (HD = 0.080, HG = 0.091, P = 29%). But P.kaibarae was less variable than these species (HD = 0.034; HG = 0.034, P = 13%). The average genetic similarities among the populations in each species of Gasterosteus aculeatus pungitius sinensis and P. kaibarae were S = 0.963, S = 0.953 and S = 0.800 respectively.Yongchon and Kyongiu populations of P. kaibarae were genetically remote from the rest of the conspecific populations (S = 0.690). Chae (1988) also observed morphological differences between them. The value of genetic similariiy coefficients of S = 0.606 between P. sinensis and P. kaibarae is low enough to consider as specific level of genetic difference, but moderate rate of hybridization (44%) between them at a synpatric area of Kangnung indicates that isolating mechanism is incomplete. Therefore it is concluded that P. sinensis and P. kaffarae are semispecitic status. Intergeneric genetic similarities between the genus Gasrerosteus and Pungitius were S = 0.052 and this value is the lowest estimate up to date in freshwater fishes.
