The segregation mode of plant height in the crosses of rice varieties 1. Indica X Indica crosses (수도 품종간 교잡에 있어서의 초장의 유전 분리 1. Indica x Indica 조합)

  • A genetrc study was made on plant height of indica rices with a few segregating F2 populations involving three semi-dwarf varieties such as T(N)I, CP231-SLO17, and B569A12. These populations were grown in breeding nursery at the International Rice Research Institute (IRRI) during several seasons. 20 to 25 day old seedlings grown at upland seedbed were transplanted to the paddy in a single plant hill spacing 30 cm ${\times}$ 25cm. Measurements of plant height were made from the juncture between culm and roots to the tip of the longest panicle of a plant pulled out from the paddy when they are matured. The results are summarized as follows: 1. Tall indica varieties originated from the south-east Asian countries could be classified into two groups depending upon their allelism whether they showed monogenic segregating ratio of 3:1 or not when they were crossed to T(N)1. 2. Most of U.S. varieties, especially semi-dwarf breedirg materials such as CP231 ${\times}$ SIO17 and B569A12, did not show monogenic segregating mode of 3:1 ratio when they were crossed to T(N)1 or to other varieties bearing the same genetic allele of T(N)1 such as Peta and Sigadis.

A Comparative Study of Inductive and Deductive Instructional Effects on the Learning of Population Genetic Concepts (집단유전 개념 학습에서 귀납적 - 연역적인 수업효과 비교)

  • The purposes of this study was to investigate the effect of inductive Instructional method and deductive one concerning the learners' population genetic concepts and achievement according to learners' cognitive characteristics. For the study, 180 students were sampled from a boys' high school: 90 students for inductive teaching method and 90 students for deductive teaching method. Group Assessment of Logical Thinking(GALT) and Group Embedded Figure Test (GEFT) were used as the measure of cognitive characteristics. The results of this study were as follows. 1) The inductive instructional method was more effective in the understanding of population genetic concepts and their achievement. 2) Inductive instructional method was more effective than deductive one for the learners in formal operational level and in field independent cognitive style. 3) For the learners in a transitional level and field dependent cognitive style, deductive instructional way was more effective than inductive way on the average, but it was not statistically significant. It was turned out that learners' cognitive level was one of important factors when teachers instruct the concept of population genetics.

  • For development of all male seed production at hatchery scale, a comparative study was made on the seed production traits of supermale (IT - t) and superfemale (IT - L1 ~) Nile tilapia, Oreochromis niloticus. Supermales were crossed with normal females (XX- f) and the superfemales with normal males (XY - t ) or sex reversed males (XX- L1 t); progeny survival of these crossings and sex ratio were evaluated. Hatching success of the eggs, fertilized by the supermale was significantly lower than that by the normal male. Over 95-99 % progenies sired by crossing supermales with normal females were males, while 52-55 % progenies alone were males with the cross of normal males and normal females. Hatching success and survival of alevins were significantly higher for the progenies of the crosses between superfemales and sex reversed males than those resulting from the crosses between superfemales and normal males. However, there was no significant difference in the sex ratio among the progenies of these crosses. Therefore, crossings of superfemales with sex reversed males provide the highest percentage of survival and male progenies.

Assessment of genetic diversity and population structure of commercial button mushroom (Agaricus bisporus) strains in Korea (한국의 상업적 양송이 균주의 유전적 다양성 및 집단 구조)

  • Agaricus bisporus is a functional food and among the most widely cultivated mushrooms in the world. In this study, we analyzed the genetic diversity and population structure of 23 Korean and 42 foreign A. bisporus cultivars using SSR (Simple sequence repeat) markers. Genetic diversity of A. bisporus cultivars was as follows: number of alleles was approximately 13; observed and expected heterozygosity were approximately 0.59 and 0.74, respectively; and polymorphic information content value was about 0.71. A. bisporus cultivars were divided into three groups using distance-based analysis. Genetic diversity of Group 2, which consisted of cultivars from various countries, was high. In addition, model-based subpopulations were divided into two, and the genetic diversity of Pop2 (Population 2), which had many cultivars, was high. The results of this study could be used in a breeding program for A. bisporus, such as the development of new genetic resources and securing diversity.

  • Genetic variation in field grown Korean ginseng(Panax ginseng C.A.Meyer) was evaluated using random amplified polymorphic(RAPD) markers. (This experiment was carried to collect the local native from farm of Chungnam National University in Korea in order to investigate genetic variation.) Some morphological characters showed considerable variation ranging 22 to 68cm in plant hight, 10 to 38mm in root diameter, 16 to 86g in root weight, and culum color and flowering date, respectively. Ten RAPD primers out of the 32 which produced reproducible bands in 662 Korean ginseng plants were selected for the further study. The total number of bands generated by 10 primers were 108 and among them 103 were polymorphic among the 662 plants with the polymorphism ratio of 94.5%. A total of 662 plants were classified into 16 groups based on polymorphic data with an URP 05 primer.

  • Virus diseases often cause serious damage to rice production in Asia. The lack of information on virus resistance genes has been a major obstacle for the breeding of resistant varieties. In order to identify DNA marker associated with resistance against rice stripe virus (RSV), the quantitative trait locus (QTL) was carried out using advanced backcross population developed from a cross between RSV-resistant tongil type cultivar Shinkwang and susceptible japonica cultivar Ilpum. A RSV resistance QTL $qSTV11^{SG}$ explaining 44.2% of the phenotypic variation was identified on chromosome 11 of Tongil type rice cultivar 'Shingwang'. $qSTV11^{SG}$ was tightly linked to DNA marker RM6897. The RM6897 divided as resistance type allele and susceptible type alleles. Twenty seven resistant varieties showed the resistant-type allele and 23 susceptible varieties were susceptible-type allele to the marker of RM6897. This results and the molecular markers presented here may be useful in rice breeding for improving RSV resistance in japonica rice.

Screening for Genotypes Lacking Lipoxygenase from Germplasm Collection of Korean Soybean Land Races (한국 재래종 콩집단에서 비린내 없는 콩품종 육성을 위한 Lipoxygenase 결실인자 변이 연구)

  • Soybean seeds contain lipoxygenase, which is responsible for the objectionable beany flavors in soybean seeds. The isozymes of lipoxygenase (1$\times$1, 1$\times$2, 1$\times$3) were discovered in United States of America, Japan, and Korea, and the mode of inheritance of the mutant genes was determined. This investigation was conducted to screen lipoxygenase-1, 2, and 3 lacking soybean lines from the Korean soybean land race population. Two lipoxygenase-1lacking lines, KAS 610-8 and KAS 621-8 were found in this investigation. In general, lipoxygenase acking varieties were small in seed size and low in oil content. A severe pod borer damage was observed in the two selected lipoxygenase-1 lacking lines. Lipoxygenase lacking line was not found in Korean wild soybean population used in this study and the lipoxygenase lacking lines were found only in Kyung-Nam province and the results imply that lipoxygenase lacking mutants were induced recently in cultivars.

  • Stress-susceptible pigs have been known as the porcine stress syndrome (PSS), swine PSS, also known as malignant hyperthermia (MH), is characterized as sudden death and production of poor meat quality such as PSE (pale, soft and exudative) meat after slaughtering. PSS and PSE meat cause major economic losses in the pig industry. A point mutation in the gene coding for the ryanodine receptor (RYR1) in porcine skeletal muscle, also known calcium (Ca$^{2+}$) release channel, has been associated with swine PSS and halothane sensitivity. We used the PCR-RFLP(restriction fragment length polymorphism) and PCR-SSCP (single strand conformation polymorphism) methods to detect the PSS gene mutation (C1843T) in the RYR1 gene and to estimate genotype frequencies of PSS gene in Korean pig breed populations. In PCR-RFLP and SSCP analyses, three genotypes of homozygous normal (N/M), heterozygous carrier (N/n) and homozygous recessive mutant (n/n) were detected using agarose or polyacrylamide gel electrophoresis, respectively. The proportions of normal, carrier and PSS pigs were 57.1, 35.7 and 7.1% for Landrace, 82.5, 15.8 and 1.7% far L. Yorkshire, 95.2, 4.8 and 0.0% for Duroc and 72.0, 22.7 and 5.3% for Crossbreed. Consequently, DNA-based diagnosis for the identification of stress-susceptible pigs of PSS and pigs producing PSE meat is a powerful technique. Especially, PCR-SSCP method may be useful as a rapid, sensitive and inexpensive test for the large-scale screening of PSS genotypes and pigs with PSE meat in the pork industry.y.

Teachers' Conceptions about the Genetic Determinism of Human Behaviors (교사들이 가지는 사람의 행동적 특징에 대한 유전자 결정론적 인식)

  • Genetic determinism of human behaviors is considered as a philosophical perspective that genes in humans determine biological as well as social traits. However, many biologists agree that human traits are determined by interactions between genes and genes, as well as between genes and environments. In this context, genetic determinism still affect ideas of the general public as well as research directions of biologists. According to Cl$\acute{e}$ment's KVP model (Cast$\acute{e}$ra & Cl$\acute{e}$ment, 2012), teachers' conceptions of genetic determinism influence students' concepts of genetics. This study intends to investigate teachers' conceptions on genetic determinism of human behaviors. For this end, a questionnaire adopted from a previous research (Cast$\acute{e}$ra & Cl$\acute{e}$ment, 2012) has been administered to 308 teachers including 151 pre-service and 157 in-service. Factor analysis has been conducted to extract major factors and one-way ANOVA has been employed to find out differences in extracted factors among different groups of teachers. Four factors have been extracted from 14 items of questionnaire, including factor 1, a perspective of genetic determinism of gender differences in intellectual ability, social status, and emotional traits; factor 2, a perspective of genetic determinism of individual differences in intellectual ability; factor 3, a perspective of genetic determinism of individual differences in biological immune function and behavioral trait; and factor 4, a perspective of genetic determinism of ethnic differences. From the results of One-way ANOVA among teacher groups on four factors, first, it has shown a significant difference in factor 1 (F=3.325, p=.006), factor 3 (F=3.320, p=.006) and factor 4 (F=4.325, p=.001) due to their subject matters. In post-hoc comparison there have been no significant difference between biology teachers and other teachers. It has also been found that there was a significant difference between pre-service and in-service teachers in factor 1 (t=-3.938, p=.000) and factor 4 (t=-3.121, p=.002) and in-service teachers are more genetic deterministic than pre-service teachers. Finally, different religions have no influence on teachers' conceptions of genetic determinism of human behaviors.

Single Nucleotide Polymorphisms (SNPs) Discovery in GHSR Gene and Their Association Analysis with Economic Traits in Korean Native Chickens (GHSR 유전자 내 유전변이의 탐색과 한국재래계의 성장 및 산란 특성에 미치는 연관성 분석)

  • Recently, it was reported that certain polymorphisms in the growth hormone secretagogue receptor gene (GHSR) are associated with the growth of chickens. However, the correlation between GHSR polymorphisms and economic traits has not been investigated in Korean native chickens (KNCs). Therefore, the objective of this study was to confirm the suitability of the GHSR gene as a candidate for genomic selection and identify a genetic marker for KNCs. A total of 220 KNCs from six breeds raised at the National Institute of Animal Science were genotyped for the c.739+726 SNP in the GHSR gene using polymerase chain reaction- restriction fragment length polymorphism (PCR-RFLP), and the sequence for a subset of 30 birds was analyzed using direct sequencing. The association between the SNP genotypes and the economic traits of the KNCs was analyzed using the statistical package for the social science (SPSS) software program. The association analysis between the c.739+726T>C SNP and economic traits revealed that the SNP was significantly associated with body weight at 150 and 270 days (BW150 and BW270, respectively) in all KNCs (p<0.01), BW150 in KNC (Gary) (p<0.05), and egg production number in KNC (White, p<0.05). In addition, the SNPs discovered using direct sequencing (513A>G, 517A>T) had a significant effect on the body weight and egg production traits (p<0.05). In conclusion, these results might be useful as a basis for studies on the improvement of KNC breeds. Furthermore, these results suggest that the SNPs (c.739+726T>C, 513A>G, and 517A>T) located in the GHSR gene could be useful molecular genetic markers for KNCs.
