Effects of Bisphosphonates on the Proliferation and the AlkalinePhosphatase Activity of Human Bone Marrow Derived Mesenchymal Stem Cells (Bisphosphonate가 인간 골수유래 간엽줄기세포의 증식과 알칼리성 인산분해효소 활성에 미치는 영향)

  • Purpose: The purpose of this study is to find out the effects of bisphosphonates (BPs) on the proliferation and the alkaline phosphatase (ALP) activity of human bone marrow derived mesenchymal stem cells (hMSCs), and thus state its correlation with bisphosphonate related osteonecrosis of the jaw (BRONJ). Methods: hMSCs was obtained by collecting and culturing cancellous bone fragments from a patient undergoing iliac bone graft. Alendronate (Aln) and Pamidronate (Pam), Ibandronate (Ibn) were added to the culture media in the concentration from $10^{-3}$ M to $10^{-11}$ M and cell toxicity, viability were measured. For ALP activity evaluation, Aln and Pam were added to the culture media in the concentration from $5{\times}10^{-7}$ M to $1{\times}10^{-8}$ M and were cultured for 1 week, 2 weeks and 3 weeks. ALP activity data were standardized using protein assay. Control groups were prepared for each examination. Results: Aln, Pam and Ibn all failed to increase the proliferation of hMSCs. With 1 week, 2 weeks of $5{\times}10^{-8}$M of Aln treatment, the ALP activity increased. Pam treatment increased the ALP activity with 2 weeks of $5{\times}10^{-8}$ M and$1{\times}10^{-8}$M. Also Ibn treatment increased the ALP activity with 2 weeks of $5{\times}10^{-8}$ M and $1{\times}10^{-8}$ M. Conclusion: It is considered that BPs are not capable of improving the proliferation of hMSCs. Also, after a transient increase in the ALP activity with the lower concentration of BPs, the activity decreased again. Therefore, in patients on long-term medication of BPs, the proliferation and osteoblast differentiation of hMSCs are restrained, and thus delayed wound healing and increase in BRONJ complications may occur.

  • The aims of this study were to investigate the antioxidative and antitumor effects of crude polysaccharide fraction from Pleurtus eryngii (CPPE). CPPE inhibited autoxidation of linoleic acid at 1,000 $\mu$g/mL concentration, and the inhibitory rates of lipid peroxidation in rat liver microsome were 5.28, 9.40, and 32.5% at 10, 100, and 1,000 $\mu$g/mL concentrations, respectively. After treatment with CPPE for 72 hours, the inhibitory rates against MCF-7, A549 and AGS cell lines showed 42.3, 33.4 and 26.7% at concentration of 1,000 $\mu$g/mL, respectively. Results of CPPE treatment at 100 and 300 mg/kg/day for 7 days in sarcoma-180 bearing-mice showed survival rates of 70 and 90%, respectively. Body weights of mice treated with CPPE were significantly decreased when compared with the control.

Electricity Production by Metallic and Carbon Anodes Immersed in an Estuarine Sediment (퇴적토에 담지된 금속 및 탄소전극에 의한 전기 생산 특성)

  • One-chambered sediment cells with a variety of anodic electrodes were tested for generation of electricity. Material used for anodes was iron, brass, zinc/iron, copper and graphite felt which was used for a common cathode. The estuarine sediment served as supplier of oxidants or electron-producing microbial habitat which evoked electrons via fast metal corrosion reactions or a complicated microbial electron transfer mechanism, respectively. Maximum power density and current density were found to be $6.90\;W/m^2$ (iron/zinc) and $7.76\;A/m^2$ (iron), respectively. Interestingly, copper wrapped with carbon cloth produced better electric performance than copper only, by 60%, possibly because the cloth not only prevented rapid corrosion on the copper surface by some degrees, but also helped growing some electron-emitting microbes on its surface. At anodes oxidation reduction potential(ORP) was kept to be stationary over time except at the very initial period. The pH reduction in the copper and copper/carbon electrodes could be a sign of organic acid production due to a chemical change in the sediment. The simple estimation of interfacial, electrical resistances of electrodes and electrolyte in the sediment cell that a key to the electricity generation should be in how to control corrosion rate or microbial electron transfer activity.

Induction of $\beta$-Lactamase on $\beta$-Lactam Antibiotics Resistant Bacterium ($\beta$-Lactam계 항생물질 내성균주의 $\beta$-Lactamase 생산의 유도)

  • Bacillus subtilis J105, high resistant bacteria against $\beta$-lactam antibiotics, become higher resistant through induction of $\beta$--lactamase in the presence of $\beta$--lactam antibiotics. When there is no antibiotics in medium, the production of resistance-inductive $\beta$--lactamase reached its plateau 15 hours later. But when there is ampicillin (500$\mu\textrm{g}$/$m\ell$) in medium, the production of enzyme reached its plateau 25 hours later since cultivating bacteria, whereas it is found that enzyme 2,900 units/$m\ell$ about 20 times as much as compared with not-presence of antibiotics was actived. In addition, as the result of MIC comparing applying ampicillin-treated and non-treated strain MIC of ampicillin-treated strain is about 2~27 times higher. It is considered that this strain induce $\beta$--lactamase production by ampicil-lin-treatment, then increasing it resistance. It is found that this resistant strain induce $\beta$--lactamase production against cephalosporin antibiotics as well as peni-cillin. As the result of examining the time of adding antibiotics for each phase of growth, it is concluded that logarith-mic phase is the most effective. As the aboves, it is suggested that this strain is a peculia strain that its resistance is induced high by various $\beta$--lactam antibiotics.

The Effect of Forsythia suspensa Extract on the Wound Healing of Rats (백서의 창상치유에 미치는 연교액기스의 효과)

  • We examined the wound healing effect and toxicity of Forsythia suspensa extract to verify the therapeutic usefulness on wound treatment. Additionally, we examined its antibacterial effect. The wound healing effect of Forsythia suspensa extract was compared with the tetrachlorodecaoxygen anion complex (TCDO), protein free haemodialysate (Haemyl), and physiological saline. The wound areas on the 4th, 11th, 14th day of treatment with Forsythia suspensa extract, TCDO, and Haemyl were significantly smaller than with the saline treatment. The wounds on the 8th day of treatment with Forsythia suspensa extract were only siginificantly smaller than the saline treatment. On the 8th day of treatment with Forsythia suspensa extract, TCDO, and Haemyl accerlerlated the wound epithelization more than saline treatment. There was not a significant difference in liver enzyme activity (GOT, GPT) and blood cell count between the Fosythia suspensa extract and the saline treatment. In addition, the Forsythia suspensa extract did not inhibit bacterial proliferation. These results suggest that Forsythia suspensa extract has great possibility of being developed into new wound healing drugs. Its pharmacological effects seem to have no relation with antibacterial effect.

Suppressive Activity of Extract of Termialia chebula Retz. on Hepatic Fibrosis (가자(Terminalia chebula Retz.) 추출물의 간섬유화 억제활성)

  • Activation of hepatic stellate cells (HSCs) is known to be responsible for hepatic fibrosis and cirrhosis. When round-shape quiescent HSCs go to activation by liver injury, production of extracellular matrix is increased, and its shape becomes myofibroblast-like shape. The activated HSCs are characterized by the high rate of proliferation and the increased production of extracellular matrix. One way of the regeneration of activated HSCs is an apoptosis induction followed by removing the activated myofibroblast-like cells. The effect of extract of Terminalia chebula Retz. (TCE) on cytotoxicity was evaluated using the rat primary hepatocyte, HepG2 and T-HSC/Cl-6 by incubating these cells with TCE up to the dose of $1,000{\mu}g/mL$. At the maximum dose of TCE, no cytotoxicity was found on primary hepatocyte and HepG2, but cytotoxic effect of TCE was found on activated HSCs, and T-HSC/Cl-6 in a U-shaped dose-response manner with the highest effect at $500{\mu}g/mL$ of TCE. Finally, we confirmed the occurrence of apoptotic cell death by annexin-V/PI double staining. The population of annexin-V positive cells was increased in a dose dependent manner.

Selection of Constitutive Promoter for Exoinulinase Production in Fed-Batch Culture of Recombinant Yeast (재조합 효모의 유가배양에서 Exoinulinase생산을 위한 Promoter의 선별)

  • In order to overexpress constitutively the Kluyveromyces marxianus exoinulinase gene (INUI) in Saccharomyces cerevisiae, four episomal expression systems employing GAPDH, ADHI, PGK and ENOI promoters were constructed as p YIGP aADHI -INU, pPGK-INU, and pENOI- INU plasmids respectively, When S cereviais transformants harboring each plasmid were batchwisely cultivated in the fermentor containing 5% glucose medium no significant differences in the cell growth are observed How- ever the experession level of exoinulinase and plasmid stability showed a strong dependency on the promoter employed. The expression levels of exoinulinase were about 1.70 unit/ml for GAPDH promoter 1.67 unit/ml for PGK promoter, 1.29 unit /ml for ADH1 promoter, and 0.80 unit/ml for ENOl promoter. The plasmid stabilites were maintaines above 80% in all experession systems. except the GAPDH promoter system of 55%, Based on the plas- mid stability and expression level of exoinulinase the ADHl and PGK promoter system were selected for the fed - batch culture to overproduce exoinulinase By the intermittent feeding of yeast extract and glucose, both promoter systems gave the cell concentration of about 30 g-dry cell weight/1 byt the maximal exoinulinase activity of 3.70 unit/ml and plasmid stability of 96% in the ADH1 promoter were higher than those (2.70 unit/ml, 80%) of PGK sys- tem Taking into account the plasmid stability and extended culture time the ADH1 promoter systems would be the most feasible expression systems for the constitutive overproduction of exoinulinase through high cell-density fed- batch cultures using non-selective rich medium.

  • Streptomycetes is a group of Gram-positive soil bacteria that growas a branching vegetative mycelium leading to the formation of spores, and display a physiological differenti-ation related to the synthesis of many secondary metabolites including antibiotics. Their complex life cycle and multicellular differentiation require various levels of regulation and types of signal transduction systems including eukaryotic-type serine/threonine protein kinases and prokaryotic-type histidine/aspartic acid protein kinases. Akt kinase that was found in cells is a sorine/threonine kinase controlling signal pathway for multi-tude of important cellular events. The activation or inactivation of Akt kinase in the cell is one of the critical regulatory points to deliver cell proliferation, differentiation, survival or apoptosis signal. To find the regula-tory protein homologous to Akt in Streptomyces, the fluorescien-labeled synthetic peptide (FITC-TRRSR-TESIT) was designed from the consensus sequence of target proteins for Akt kinase. From the difference of the mobility between the nonphosphorylated and phosphorylated synthetic peptides on Agarose gel electro-phoresis, the Akt-phosphorylating activity was monitored. The cell-free extract prepared from Streptomyces griseus IFO 13350 and the Akt homologous protein was purified by ammonium sulfate fractionation and many steps of column chromatographies such as, DEAE-Sepharose, Mono Q, Resource Phenyl-Soporose and Gel permeation column chromatographies. As a result, the protein phosphorylating the fluorescien-labeled Akt substrate was identified and it's molecular weight was estimated as 39 kDa on SDS-PAGE.

  • Bacillus polyfermenticus SCD which is commonly called as Bisroot strain is being used for functional foods through the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine in humans. The cells of B. polyfermenticus SCD were treated for 4h in artificial gastric juice (pH 2.0,3.0) and bile acid. Final viability of the strain in artificial gastric Juice (pH 2.0, 3.0) is reached to 62.8% and 81.2% respectively B. polyfermenticus SCD is resistant to antibiotics such as streptomycin, rifampicin, nystatin and ampicilin. B. polyfermenticus SCD is well known supplies the nutrients by synthesizing vitamin $B_1$, $B_2$, C and K. B. polyfermenticus SCD produces various digestive enzymes and the enzymes enable to completely digest diets in our body. Above all, $\alpha$-amylase and pretense activities are very higher than B. subtilis KCTC 1020, about two fold and twenty five fold respectively. B. polyfermenticus SCD is very stable during long-term storage period in phosphate buffers of wide-range pH, solutions of various concentrations of sodium chloride, 5% glucose solution and water.

A Study on Denitrification by Sulfur-Oxidizing Bacteria for the Industrial Wastewater Contain Fluoride and Nitrogen (불소.질소 함유 폐수의 황산화탈질에 관한 연구)

  • Nitric acid and hydrofluoric acid are used for acid pickling in zirconium alloy tubing manufacturing process. Nitrate and fluoride in the wastewater were treated by chemical coagulation and SOD (Sulfur Oxidation Denitrification) process. This study is investigated the effect of fluoride concentration and the optimal condition for SOD process. The limited fluoride concentration for SOD process was below 20 mg F-/L. The adjusted pH and alkalinity by NaOH and $NaHCO_3$ was shown to be more effective for removal of nitrate compared with using NaOH. Furthermore, the microbial activator mixed trace elements and ingredient for alkalinity did not only supplement with alkalinity but also enhance the growth and proliferation for sulfur-oxidizing bacteria. As a result, the inorganic industrial wastewater was successfully treated by the microbial activator in SOD process without continuous addition of seed sludge. Finally, SOD process was shown to remove nitrate in industrial wastewater and to contribute the microbial activator for activation of sulfur-oxidizing bacteria.
