A Study on Formation Pattern of DBPs by Disinfection of Drinking Raw Water II (음용 원수의 염소소독에 의한 소독부산물 생성패턴에 관한 연구 II)

  • The formation pattern of DBPs (disinfection by-products) in raw water treated with hypochlorite, chlorine disinfectant was studied. TOC (total organic carbon), residual chlorine, turbidity and 14 DBPs in raw water from Han-river and Nakdong river during 1 ~ 14 days were determined. Total DBPs in Han river was 101.3 ng/mL (789.6 nM) after 7days and THMs (trihalomethanes) are the dominant portion of 68%. HAAs (haloacetic acids) and chloral hydrate were determined 19% and 10% respectively. In Nakdong river total DBPs was 98.4 ng/mL (678.6 nM) and dominant class was HAAs. (55.8 ng/mL, 57%) THMs(34%) and N-compounds like HANs (haloacetonitriles, 5%) and chloropicrin were increased. It may be explained that high concentration of NH4-N in Nakdong river react with chlorine produced chloramine and this formed different pattern of DBPs. As a result, total DBPs formation pattern depends on raw water and disinfectant and in generally the initial concentration of acidic HAAs was high and THMs was increased gradually.

Effects of Chlorine Dioxide Treatment on Quality and Microbial Change of Agaricus bisporus Sing during Storage (이산화염소 처리가 저장 중 양송이버섯(Agaricus bisporus Sing)의 품질 및 미생물학적 변화에 미치는 영향)

  • Effects of chlorine dioxide ($ClO_2$) on weight loss, polyphenol oxidase activity, and microbial change of fresh mushrooms (Agricus bisporus Sing) were investigated. Mushrooms were treated with 5, 10, and 50 ppm of $ClO_2$ solution. Mushrooms were stored at room temperature and $4^{\circ}C$. Weight loss for $ClO_2$ treated mushrooms was smaller than that of the control group. Polyphenol oxidase activity of the $ClO_2-treated$ samples stored at room temperature increased until day 2 of storage and then decreased, and the activity of the samples stored bacteria, yeast and mold counts than the control during storage. Aerobic bacterial counts for 50 ppm of $ClO_2$ treatment at $4^{\circ}C$ increased from $1.62{\times}10^2\;CFU/g\;to\;1.66{\times}10^7\;CFU/g$ at day 10 of storage, while control groups were from $1.00{\times}10^3\;CFU/g\;to\;3.72{\times}10^8\;CFU/g$. These results indicate that $ClO_2$ treatment could be useful in improving the quality and microbial safety of mushrooms.

Effects of Combined Chlorine Dioxide Gas Treatment Using Low-Concentration Generating Sticks on the Microbiological Safety and Quality of Paprika during Storage (저농도 서방형 이산화염소 가스 병합처리가 파프리카의 저장 중 미생물 성장과 품질에 미치는 영향)

  • Chlorine dioxide ($ClO_2$) gas treatment (75 ppmv, 30 min) has been suggested to improve the microbial safety of postharvest paprika in a previous study. Based on these results, in this study, an additional combined treatment using low-concentration $ClO_2$ gas-generating sticks (3 ppmv) in paprika samples during storage was carried out at $8^{\circ}C$ and 90% relative humidity to further enhance the quality and reduce the decay rate of paprika for the purpose of lengthy storage. After the combined treatment, the initial populations of total aerobic bacteria as well as yeast and molds in the paprika samples decreased by 3.04 and 2.70 log CFU/g, respectively, compared with those of the control samples, and this microbial inactivation was maintained by the low-concentration $ClO_2$ gas-generating sticks during storage. In particular, the decay rate of samples with combined treatment was significantly lower than that of the control. Vitamin C content, hardness, and color quality parameters of paprika samples were not altered by treatment, while weight loss of the samples treated with the combined $ClO_2$ gas was lower than that of the control during storage. These results indicate that the combination of two different $ClO_2$ gas treatments is effective for retaining the quality of paprika during prolonged storage.

Effect of Chlorine Dioxide, Cold Plasma Gas Sterilization and MAP Treatment on the Quality and Microbiological Changes of Paprika During Storage (이산화염소 및 저온 플라즈마 가스 살균 및 MAP 처리가 파프리카의 저장 중 품질과 미생물학적 변화에 미치는 영향)

  • This study was conducted to investigate the effect of packaging methods and sterilization treatment on storability and microbial control in paprika fruits. When treated with chlorine dioxide gas for 3, 6, and 12 hours and cold plasma gas for 1, 3, and 6 hours, and then packed in a carton box and stored in a 8 ± 1℃ chamber for 7 days, chlorine dioxide treated 12 hours and plasma treated 6 hours was prevented the development of E·coli and YM(yeast and mold). Accordingly, the control was treated with chlorine dioxide for 12 hours and plasma for 6 hours, packed using a carton box and 40,000 cc·m-2·day-1·atm-1 OTR film (MAP), and stored in a 8 ± 1℃ chamber for 20 days. Fresh weight loss rate during storage was less than 1% in the MAP treatments, and the visual quality of the MAP treatments was above the marketability limit until the end of storage. There was no difference in the contents of oxygen, carbon dioxide, and ethylene in the film. In the case of firmness, the chlorine dioxide treatments was low, and the Hunter a* value, which showed chromaticity, was highest in the Plasma 6h MAP treatment. Off-odor was investigated in the MAP treatments, but it was very low. The rate of mold growth on the fruit stalk of paprika was the fastest and highest in the chlorine dioxide treated box packaging treatments, and the lowest in the chlorine dioxide treated MAP treatments at the end of storage. The aerobic count in the pulp on the storage end date was the lowest in the plasma treated box packaging treatments, the lowest number of E·coli in the chlorine dioxide treated MAP treatments, and the lowest yeast & mold in the chlorine dioxide treated box packaging treatments. As a result, for the inhibition of microorganisms during paprika storage, it is considered appropriate to treat plasma for 6 hours before storage regardless of the packaging method.

  • Recently, there are a lot of researches related to the high-fluidity concrete (HFC) with field applications. However, most applications and studies are with concretes with high strength level so there are little studies about durability evaluations such as chloride ion penetration properties with normal strength concrete. Therefore, to evaluate the durability of HFC with normal strength level, this study performed the chloride ion penetration test and observed the micro pore distribution with normal strength HFC which contains limestone powder. Experimental results showed that most micro-pores have diameters between 0.005 to 0.05 ${\mu}m$ with HFCs using limestone powder and the average diameter becomes larger with the increase of limestone powder content. Also, it was shown that, with the increase of the limestone powder content, penetration depth and diffusion coefficient of chloride ion increased and diffusion coefficient had good relationships with compressive strength and average pore diameter with the coefficient of determination over 0.90.

  • This study was conducted to develop an enzyme-linked immunosorbent assay(ELISA) for the determination of cooked goat meat. Muscle proteins were extracted from goat meat by heating at $98^{\circ}C$ for 15 min. Major thermostable(TS) protein, whose size and pI are 36 and 38 kDa and 4.5 respectively, were purified by DEAE-Sephadex A-50 and Sephadex G-75 column chromatography. The TS protein was immunized into rabbits in order to produce goat specific antibodies. Competitive indirect ELISA(ciELISA) was established by using the anti-TS antibody. The antibody showed high reactivity toward the TS antigen and the boiled goat meat extract but it did not show any reactivities toward extracts of boiled chicken, pork, lamb, and beef. Thus, this ciELISA developed in this study could be applicable to identify goat species from cooked meat.

  • Electrogenerated chlorine leaching of used printed circuit board was Investigated in hydrochloric acid solution. The used printed circuit board contained about 45% of metal component, in which copper was about 84%. The leaching rate was greatly effected by current density and agitation. Utilization of electrogenerated chlorine was enhanced by increasing agitation and lowering current density. Leaching of copper was suppressed, while the minor metal elements, such as aluminum, lead, and tin, dominated the leaching at the initial stage.

Determination of bromine in 1000 ㎍/g Cl standard solution by ID-ICPMS (동위원소희석 질량분석법에 의한 1000 ㎍/g 염소 표준용액 중 브롬 불순물 분석)

  • The isotope dilution method was used for the determination of Br impurity in $1000{\mu}g/g$ Cl standard solution. Since relatively pure KCl salt was used for the preparation of the Cl standard solution, the Br impurity determination suffers from both spectral and non-spectral interferences due to the presence of a large amount of K and Cl matrices. AG2-X8 anion-exchange resin was employed to separate the Br analyte from the matrices, and RF power was raised to 1500 W and nebulizer gas flow rate was lowered to 0.77 L/min to reduce background from the $ArArH^+$ molecular ions. The Br impurity in the $1000{\mu}g/g$ Cl standard solution was determined to be 43.7 ng/g with the standard addition method. The analytical result was in good agreement with 41.2 ng/g (RSD 1.6%) determined by the isotope dilution method to lower uncertainty from poor reproducibility of the anion-exchange process.
