Rediscovery of haploid breeding in the genomics era (유전체 시대에 반수체 육종의 재발견)

  • Advances in DNA sequencing technologies have contributed to revolutionary understanding of many fundamental biological processes. With unprecedented cost-effective and high-throughput sequencing, a single laboratory can afford to de novo sequence the whole genome for species of interest. In addition, population genetic studies have been remarkably accelerated by numerous molecular markers identified from unbiased genome-wide sequences of population samples. As sequencing technologies have evolved very rapidly, acquiring appropriate individual plants or populations is a major bottleneck in plant research considering the complex nature of plant genome, such as heterozygosity, repetitiveness, and polyploidy. This challenge could be overcome by the old but effective method known as haploid induction. Haploid plants containing half of their sporophytic chromosomes can be rapidly generated mainly by culturing gametophytic cells such as ovules or pollens. Subsequent chromosome doubling in haploid plants can generate stable doubled haploid (DH) with perfect homozygosity. Here, classical methodology to generate and identify haploid plants or DH are summarized. In addition, haploid induction by epigenetic regulation of centromeric histone is explained. Furthermore, the utilization of haploid plant in the genomics era is discussed in the aspect of genome sequencing project and population genetic studies.

An Experimental Study on Tip Velocity and Downstream Recirculation Zone of Single Groyne Conditions (단일 횡수제 조건에 따른 선단부 유속과 재순환 영역에 대한 실험 연구)

  • Recently, the concern of groynes that provide us with various ecological environment and improvement of scenary near river as well as hydraulic improvement has greatly increased for naturalized river and ecological river restoration. However there is no adequate design guidelines in Korea. Furthermore there is lack of research for installation of groynes in Korea. Thus, in this study, we have conducted hydraulic modeling test for kinds of groyne and changes of permeability and made a study of the analysis for installation factor of groyne. Experimental conditions were the fixed bed model for flow pattern of groyne tip and separation length about changes of groyne length and permeability. The Experiments were conducted to measure velocity and drift angle at groyne tip. The main study kas measured the groyne tip velocity for a factor of scour and drift angle for velocity increase at center of channel. We have suggested the equation about groyne tip velocity rate(tip velocity/ approach velocity) and area rate(groyne area/ flow area). And In recirculation zone of groyne downstream, To study the basic data of groyne about installation interval has analyzed the change of separation length and incidence angle. We have suggested the equation about separation length rate(separation length/ groyne length), changes of permeability and installation angle.

  • Korea's tidelands policy IS examined and criticized in the paper. Korea's major tidelands in the west coast might disappear within a decade. The anachronistic development IS driven by the political interest group, forming coalition for reclamation and busily fulfilling its own profits under the facade of public good and regional development. But all regions of the world have been gradually scrapping the massive reclamation projects since the 1970's and active movements to reverse the reclamation and restore the nature are on the rise. In 1978, the U.S. Supreme Court decided to suspend the construction of the dam and to protect the snail darter despite that over 100 million dollars had been injected. This court ruling became famous and caused the American public to change their perception about the environment. In the Netherlands, following a prolonged discussion on pros and cons of reclamation, open type seawall was adopted to strike a balance among the available alternatives. Japan's Ministry of Environment was praised for forming the National Wetland Committee in order to designate 20 new Ramsar sites by 2008, with an objective to reach 33 designated Ramsar sites. Away from the large-scale reclamation projects, Korea has to move towards smaller-scale projects focusing on the knowledege intensive and circular economy society that are more in harmony with the environment.

Comparison of Safety Margin of Shallow Foundation on Weathered Soil Layer According to Design Methods (설계법에 따른 풍화토 지반 얕은기초의 안전여유 비교)

  • In this paper bearing capacity and safety margin of shallow foundation on weathered soil ground against shear failure by using current design method of allowable stress design (ASD), load resistance factor design (LRFD) based on reliability analysis and partial safety factor design (PSFD) in Eurocode were estimated and compared to each other. Results of the plate loading test used in construction and design were collected and analysis of probability statistics on soil parameters affecting the bearing capacity of shallow foundation was performed to quantify the uncertainty of them and to investigate the resistance bias factor and covalence of ultimate bearing capacity. For the typical sections of shallow foundation in domestic field as examples, reliability index was obtained by reliability analysis (FORM) and the sensitivity analysis on soil parameters of probability variables was performed to investigate the effect of probability variable on shear failure. From stability analysis for these sections by ASD, LRFD with the target reiability index corresponding to the safety factor used in ASD and PSDF, safety margins were estimated respectively and compared.

Efficient Integer pel and Fractional pel Motion Estimation on H.264/AVC (H.264/AVC에서 효율적인 정화소.부화소 움직임 추정)

  • Motion estimation (ME) plays an important role in digital video compression. But it limits the performance of image quality and encoding speed and is computational demanding part of the encoder. To reduce computational time and maintain the image quality, integer pel and fractional pel ME methods are proposed in this paper. The proposed method for integer pel ME uses a hierarchical search strategy. This strategy method consists of symmetrical cross-X pattern, multi square grid pattern, diamond patterns. These search patterns places search points symmetrically and evenly that can cover the overall search area not to fall into the local minimum and to reduce the computational time. The proposed method for fractional pel uses full search pattern, center biased fractional pel search pattern and the proposed search pattern. According to block sizes, the proposed method for fractional pel decides the search pattern adaptively. Experiment results show that the speedup improvement of the proposed method over Unsymmetrical cross Multi Hexagon grid Search (UMHexagonS) and Full Search (FS) can be up to around $1.2{\sim}5.2$ times faster. Compared to image quality of FS, the proposed method shows an average PSNR drop of 0.01 dB while showing an average PSNR gain of 0.02 dB in comparison to that of UMHexagonS.

The Cell Resequencing Buffer for the Cell Sequence Integrity Guarantee for the Cyclic Banyan Network (사이클릭 벤얀 망의 셀 순서 무결성 보장을 위한 셀 재배열 버퍼)

  • In this paper, we present the cell resequencing buffer to solve the cell sequence integrity problem of the Cyclic banyan network that is a high-performance fault-tolerant cell switch. By offering multiple paths between input ports and output ports, using the deflection self-routing, the Cyclic banyan switch offer high reliability, and it also solves congestion problem for the internal links of the switch. By the way, these multiple paths can be different lengths for each other. Therefore, the cells departing from an identical source port and arriving at an identical destination port can reach to the output port as the order that is different from the order arriving at input port. The proposed cell resequencing buffer is a hardware sliding window mechanism. to solve such cell sequence integrity problem. To calculate the size of sliding window that cause the prime cost of the presented device, we analyzed the distribution of the cell delay through the simulation analyses under traffic load that have a nonuniform address distribution that express tile Property of traffic of the Internet. Through these analyses, we found out that we can make a cell resequencing buffer by which the cell sequence integrity is to be secured, by using a, few of ordinary memory and control logic. The cell resequencing buffer presented in this paper can be used for other multiple paths switching networks.

Low-salinity Water and Circulation in Summer around Saemangeum Area in the West Coast of Korea (하계 서해안 새만금 연안역 주변 저염수와 순환)

  • In the mid-west coast of Korea where Mankyung and Dongjin rivers discharge fresh water, Saemangeum tidal dyke of 33 km long is under construction to reclaim the very shallow estuary region of 41,000ha. Main source of freshwater in this coastal area is Keum River locating closely north of the dyke. At present, the dyke connected with Gogunsan-Gundo separates this area into three regions; northwestern, southwestern and eastern (Saemangeum) region of the dyke, and the water in Saemangeum region is exchanged through one gap in the northern dyke and two gaps in the southern dyke. We have observed distributions and structures of temperature and salinity to examine the summer circulation related with low-salinity water in this coastal area in 1998 and 1999. In the surface layer off the northern dyke a tongue-like distribution of low-salinity extends 60 km long from Keum River estuary mouth to the northwest, forming plume front bounded by offshore water. In the inner region of Saemangeum dykes salinity distributions show that two river waters are merged together and the low salinity water is deflected toward northern gap of the dyke. In the surface layer off the southern dyke we observed small tongue-like distribution of another low-salinity water extending to the north from Gomso Bay. Based on the analysis of distributions of low-salinity water and frontal structures, we can suggest an anticlockwise circulation of coastal water around the dyke, composed by the estuarine water outgoing from the inner region of the dyke through the northern dyke's gap and the inflow through two gaps of southern dyke from offshore. After completing the dyke construction, this coastal circulation around the dyke will be, however, changed because fresh water discharge of Mankyung and Dongjin rivers will be routed artificially and directly into the area offshore of the southern dyke.

An exploratory study on reviewing journal articles of qualitative studies in the field of social welfare in Korea (국내 사회복지질적연구논문 심사의 발전적 방향에 관한 탐색)

  • This study suggests future directions or approaches as to how we should appraise qualitative studies by reviewing previous studies on this topic written in English and published outside of Korea. This study explored experiences and perspectives of qualitative researchers in Korea by conducting a focus group discussion on this topic. Results of this focus group discussion indicated that qualitative researchers often face manuscript reviewers who are overly biased with quantitative paradigm which result in feeling of frustration. They also shared feeling burdened and confused as reviewers due to their own lack of training and preparedness in reviewing qualitative articles. However, participants also stressed the feeling of gratitude and being rewarded because of the experience of mutual growth and partnership with peer qualitative researchers as they strive towards improving the quality and completion of peer researchers' work. Previous studies on this topic included wide range of issues concerning qualitative research appraisal from reflections on the experiences as submitters and reviewers of qualitative studies to academic conceptual papers that discussed researchers' dilemma on research paradigms and qualitative research appraising methods. This study ends with suggestions for the future directions for qualitative researchers in Korea.

A Comparative Study on Methods for Outlier Test of Rainfall in Korea (국내 강우의 이상치검정 방법의 비교 연구)

  • 이상치는 표본자료에서 크게 어긋나 다른 자료들로부터 떨어져 표시되는 자료로써, 실제로 발생할 확률이 매우 낮은 자료로 정의되고 있다. 설계홍수량을 산정하기 위하여 적용하고 있는 극치계열의 연최대치 강우자료에는 기계오작동 및 엔지니어의 표독오류가 발생하고 있으며, 기후변화에 따른 거대태풍 및 국지적인 집중호우 발생 등으로 인한 극치값 등에서 이상치가 관측되고 있다. 통상 이상치들은 통계분석시 자료 본연의 특성을 왜곡시켜 편향된 결과를 산정할 수 있으므로 빈도해석시 이상치해석 절차를 수행하여 자료의 적정성을 확인하여야 한다. 현재 실무에서는 설계홍수량 산정요령과 하천설계기준 해설 등에서 관련 내용을 기술하고 있지만, 국내 강우자료의 기록연수의 부족으로 인하여 빈도해석시 이상치 해석이 미수행되고 있어 이상치에 따른 자료편의가 발생하면 결과물인 확률강우량이 왜곡되게 산정될 수 있다. 따라서, 본 연구에서는 국내 주요 도시의 강우자료를 이용하여 이상치검정을 수행하였다. 대상지점으로는 서울, 부산, 대전, 대구, 인천, 광주, 울산 등의 비교적 긴 관측년수를 보유하고 있는 광역시를 선정하였으며, 지속기간은 10분, 1~24시간의 25개 강우자료를 적용하였다. 이상치검정 방법으로는 타 방법에 비하여 이상치 검정력이 뛰어난 것으로 알려진 2가지 방법을 채택하였으며, 표본자료의 평균과 표준편차로 표준화된 z값을 이용하여 상 하 한계선를 초과하는 값을 확인하는 z-Score 방법중 향상된 중위수 절대편차(MAD)에 의한 수정 z-Score 방법(Hoaglin, 1993)과 Box-Plot 방법(Tukey, 1969)을 적용하였다. Box-Plot 방법(Tukey, 1969)은 전체 자료를 25%씩 사분위로 구분하는 방법으로 정렬된 자료계열을 중앙값, 박스, 수염(whiskers), 이상치로 구분한다. 정렬된 25~75% 값들을 박스로 포함하여 외곽의 수염값들을 이상치로 분류하며, 특히 사분위수의 도식화로 데이터의 분포를 파악하기 좋으며, 이상치들의 위치와 자료의 비대칭 여부를 쉽게 파악할 수 있다. 본 연구의 수행으로 수정 z-Score 방법의 경우에는 서울과 대구지점에는 이상치가 없으며, 부산지점에는 13개, 대전지점 7개, 인천지점 5개, 광주지점 32개, 울산지점 26개가 나타났다. Box-Plot 방법으로는 서울지점 35개, 부산지점 39개, 대전지점 32개, 대구지점 38개, 인천지점 51개, 광주지점 61개, 울산지점 65개의 이상치가 분석되었다. 연구를 수행한 결과, 수정 z-Score 방법에 비하여 Box-Plot 방법에 의한 이상치가 더 많이 발생하였으며, 각각의 방법으로 지속기간 및 연도별 이상치 발생자료를 확인하였다. 방법별 이상치 발생현황 등을 분석하여 지점별 발생횟수를 분석하였으며, 추후 지점 및 자료의 보완이 수행되면 활용성을 증대시킬 수 있을 것으로 판단된다.

An Analysis on Occupational Content in the Technology & Home Economics Textbooks of High School (고등학교 기술·가정 교과서의 직업 내용 분석)

  • This study analyzed content of and recognition on occupations shown from Technology Home Economics textbooks of the high school elective subject having a higher connection with the career development capability for the occupational world as shown from a curriculum analysis. Hence, this study selected 3 kinds of high school Technology Home Economics textbooks with a higher share from 6 in the 2007 revised curriculum and analyzed a frequency by characteristic of occupational content and recognition on description of a occupation. Content analyzed by researchers was examined by 2 school teachers to ensure the validity and objectivity of research findings. Research findings show that there is a great deal of frequency difference in occupations suggested by publisher and unit in the Technology Home Economics subject. Also suggestion frequency of the occupational classification by level from Korean standard industrial classification did not correspond to an actual situation of employment. Occupation names suggested from textbooks occupied 3.3% of those by Korean Occupation Dictionary, showing a higher suggestion ratio at occupations preferred by adolescents. In the textbooks, occupations were suggested from reading materials, photos and illustrations more than from texts. Recognition on descriptions of occupations showed for the most part the neutral(82.8%) and the positive16.2%)', and there was no biased intention even from some negative cases(1.0%). Based on research findings, this research has significance in that it suggested things to be considered for teaching career development capability in the high school Technology Home Economics subject and need for cultivating teachers' specialty to understand occupations and instruct career development. It also suggested improvements to be required for a new curriculum textbook.
