Ultrasonographic Findings of a Chondrolipoma Arising from the Left Supraclavicular Region: A Case Report (좌측 쇄골 상부에서 발생한 연골 지방종의 초음파 소견: 증례 보고)

  • Chondrolipomas, which are lipomas with chondroid metaplasia, are rare benign soft tissue tumors with no relevant epidemiological reports or radiological information. A limited number of lipomas with osteo/chondroid differentiation have been reported in the literature between 1960 and 2008. Moreover, only few studies have described the radiologic findings of chondrolipomas. Herein, we present a case of chrondrolipoma arising from the left supraclavicular region in a 77-year-old female.

Genetic variation in five species of Korean Orostachys (Crassulaceae) (한국산 바위솔속(돌나물과) 5종에 대한 유전적 변이)

  • Starch gel electrophoretic studies using 24 populations of five Korean Orostachys species were conducted to investigate allozyme variation and to test hypotheses of systematic relationships among species. The resulting phenogram showed that the populations of five Korean Orostachys species were divided into two major groups. And they were concordant with molecular and morphological data in suggesting that Orostachys was divided into two groups corresponding to the subsect. Appendiculatae and subsect. Orostachys. The low genetic identities among Korean Orostachys species indicated that the species of Orostachys have diverged gradually through the model of geographical species. Comparing the previous genetic data from the species with similar life history and mode of reproduction, most of Korean Orostachys species revealed a significant low genetic variation, while the widespread O. japonicus showed a relatively high genetic variation among the Korean species. This kind of genetic variation pattern might be the results of the isolated habitats, limited numbers of individuals within the populations, destruction of habitats, inbreeding and asexual reproduction in Korean Orostachys populations. The Jungdongjin population (POP 21) of O. malocophyllus was genetically unrelated with remaining populations of the same species, and this interpretation was consistent to the results from the previous palynological and morphological studies. Our allozyme data supported the taxonomic treatment of recently proposed taxa, O. iwarenge (Makino) Hara for. magnus and O. margaritifolius.

Efficient Callus Culture and Plant Regeneration from Mature Seed of Tall Fescue (Festuca arundinacea Schreb.) (톨 페스큐의 성숙종자로부터 효율적인 캘러스 배양 및 식물체 재분화)

  • In an effort to optimize tissue culture conditions for genetic transformation of tall fescue (Festuca arundinacea Schreb.), an efficient plant regeneration system from seed-derived calli was established. MS medium containing 6 mg/L 2,4-dichlorophenoxy acetic acid (2,4-D) and 0.1 mg/L benzyladenine (BA) were optimal for embryogenic callus formation from mature seed and had a strong effect on successive plant regeneration. The plant regeneration frequency above 50% was observed when embryogenic calli induced in this medium were transferred to N6 medium supplemented with 1 mg/L 2,4-D and 3 mg/L BA. Among several basic media, MS and N6 medium were optimal for callus induction and plant regeneration, respectively. 'Kentucky-31' showed to have high frequencies of embryogenic callus induction and plant regeneration up to 58.3 and 50%, respectively. Addition of sucrose to the regeneration medium as a carbon source increased regeneration frequency up to 55%. A short tissue culture period and high-frequency regeneration system established in this study will be useful for molecular breeding of tall fescue through genetic transformation.

Phylogenetic Study of Two Problematic Subgenera of Tomoceridae (Insecta : Collembola) from Korea (한국산 가시톡토기 과 (곤충 강: 톡토기 목)의 문제 2아속의 계통분화)

  • Two problematic subgenera of Tomoceridae (Collembola, Insecta) were investigated for their phylogenetic relationships based on morphological characters and allozyme study from three relevant species. Different dendrograms were obtained obtained between morphological and allozyme studies. The morphological data did not give rise to any result distinctive enough to separate the two subgenera whereas the allozyme analysis produced a clear separation by the high genetic distance value. They were consistent, however, whether given rise to by using distance or cladistic methods and also whether character weighting or unweighting approaches employed in the morphological character analysis. As a consequence, it is strongly suggested that any prominent morphological trait might work as a good taxonomical character when supported by a strong genetic divergence as evidenced by allozyme analysis for instance.

Effect of Orchardgrass Varieties on Callus Culture and Plant Regeneration (오차드그래스의 품종이 캘러스배양과 식물체 재분화에 미치는 영향)

  • In order to investigate the effects of genetic variations of orchardgrass in tissue culture response, calli were induced from mature seeds of eight varieties, 'Hapsung 2', '93E', 'Amba', 'Ambassdor', 'Frode', 'Frontier', 'Potomac' and 'Roughrider', and plant regeneration frequency was compared. Significant differences were observed among the varieties in both callus induction and plant regeneration. Callus induction rate of viable seeds varied from 24.3% to 71.7%. Plant regeneration frequency ranged from 76.6% to 29.7%. 'Roughrider' varieties showed higher regenerability with the frequency of 76.6%. These results can be used not only to provide additional improvements in the plant regeneration frequency from transgenic callus, but also useful for molecular breeding of orchardgrass through genetic transformation.

  • Optimum tissue culture conditions for an efficient induction of embryogenic callus from mature seeds of perennial ryegrass (Lolium perenne L.) and regeneration of plants from callus tissues were investigated. MS medium containing 3 mg/L 2,4-D and 0.1 mg/L BA was optimal for embryogenic callus induction from mature seeds. The highest plant regeneration frequency (58.3%) was observed when the embryogenic callus tissues were cultured on N6 medium supplemented with 1 mg/L 2,4-D and 3 mg/L BA. Regenerated plants were grown normally when shoots transplanted to the soil. A short tissue culture period and high-frequency regeneration system would be helpful for molecular breeding of perennial ryegrass through Agrobacterium-mediated genetic transformation.

  • In order to develop an efficient, reliable and reproducible tissue culture system for reed (Phragmites communis Trinius), an efficient plant regeneration system via callus induction was established using mature seeds as explants. MS medium containing 1 mg/L 2,4-D and 0.5 mg/L BA was optimal for callus induction from mature seeds. The highest frequency (88.7%) of callus formation was obtained in 1.0 mg/L 2,4-D. The highest plant regeneration frequency (59.6%) was found when the embryogenic calli were subcultured on MS medium supplemented with 100 mg/L myo-inositol, whereas, adding of plant growth regulators was not so promising in this case. Our result would be useful for development of transgenic reed plants.

Effects of Medicinal Herb Extracts on Osteoblast Differentiation and Osteoclast Formation (한약재 추출물의 조골세포 분화 및 파골세포 형성에 미치는 영향)

  • Bone is continuously remodeled by osteoblasts and osteoclasts. We investigated the effects of medicinal herbs, which act on bone metabolism. Fifteen kinds of medicinal herb extracts were screened for bone formation activity with osteoblastic cells, and MC3T3-E1 and bone resorption were screened with osteoclasts derived from mouse bone marrow macrophages. Among these samples, Actinidia polygama, Eucommia ulmoides Oliv., Schizonepeta tenuifolia, Sorbus commixta, and Zingiber officinale Rosc. extracts showed strong bone-forming activity accompanied with osteoblast proliferation and alkaline phosphatase activity. In addition, these extracts decreased tartrate-resistant acid phosphatase activity against osteoclast differentiation. The results indicate that these medicinal herb extracts can potentially prevent bone-related diseases such as osteoporosis by increasing osteoblast differentiation and reducing osteoclast activity.
