Associated Nitrogen Fixation in the Rhizosphere of Rice in Saline and Reclaimed Saline Paddy Soil -II.   • Associative heterotrophic nitrogen fixing bacteria were identified by immunodiffusion method in the histosphere of Planta-ginaceae, Caryophllaceae, Gramineae, and two types of rice cultivars. Twenty four strains associative heterotrophic bacteria with high ARA (more than 10nmole/tube/hr) were isolated from the histosphere of grasses and rice.* Those strains were related with 8 species of Azospirillum, 11 species of Pseudomonas, 2 species of Klebsiella and 2 species of Agrobacterium. Among them Azospirillum sp. and Pseudomonas sp. were predominant in histosphere of grasses and rice cultivars. From the histosphere of Oryza sativa, and Sagina maxima, the strains of Azospirillum, Pseudomonas, Klebsiella, and Agrobacter were identified while Pseudomonas was identified from Ischaemum anthephoroides, Plantago lanceolata, Miscanthus sacchuriflorum, and only Azospirillum was identified from Zoisia sinica, respectively. Associative nitrogen fixing heterotrophic bacteria were more abundant in the histosphere of Oryza sativa and Sagina maxima than that of other grasses grown in saline and reclaimed saline paddy soil.

Characteristics of Antifungal Bacterium, Bacillus subtilis YS1 and It′s Mutant Induced by Gamma Radiation (온천수로부터 분리한 항진균세균의 특성 및 감마선$(Co^{60})$ 조사를 이용한 돌연변이체 유기)

  • Antifungal bacterium, Bacillus subtilis YS1 was isolated from Yusong hot spring showed broad antifungal spectrum against 12 kinds of plant pathogenic fungi and Candida albicans, animal pathogen. From the gamma($Co^{60}$) radiation sensitivity test, $D_10$ value was 2.08 kGy and it survived above 20 kGy of radiation dose. Several mutants were induced by gamma radiation. Among them, YS1-1009 mutant showed resistance against tebuconazole of herbicide, increased activity against Botryoshaeria dothidea and ligninase activity. YS67 mutant was antifungal deficient auxotrophic mutants(trp-pro-or arg-ura-). From this results, it suggested that gamma irradiation could be useful method for mutant induction.

  • In this study, orchid mycorrhizal fungi (OMF) were isolated from four terrestrial orchids on Mt. Hambeak, Platnathera chlorantha, Platnathera mandarinorum, Cephalanthera falcate, and Cephalanthera longibracteata. OMF were identified using morphological and sequences analysis of fungal internal transcribed spacer (ITS) regions by specific primer of basidiomycetous orchid mycorrhizas; ITS1-OF and ITS4-OF. Four species of orchid mycorrhizal fungi were identified as Ceratobasidium sp, Epulorhiza anaticula, Tulasnella calospora and Tulasnella sp.

  • A key to the species which have been isolated from the Korean forest soils was provided, and three unrecorded species found in the forest soils of Mt. Halla were described and illustrated. Twenty dictyostelid cellular slime molds had been found in Korean forest soils. Among these, nine species were PG positive, and eleven species were PG negative. Three species described in this study were all PG positive. These were Dictyastelium aurea-stipes Cavender, Raper et Norberg vaL aurea-stipes (new strain HL-5), Dictyastelium monachasiaides Hagiwara, PalysPhandylium tenuissimum Hagiwara. Morphological features and growth habits of these species were examined and compared, and their distribution patterns were illustrated.trated.

Isolation and Identification of Yeasts from Wild Flowers in Gyejoksan, Oseosan and Beakamsan of Korea (대전 계족산과 충남 오서산 및 전북 백암산 주위 야생화들로부터 효모의 분리 및 동정)

  • Yeasts isolated from wild flowers of Gyejoksan in Daejeon city, Oseosan in Chungchungnamdo, and Baekamsan in Jeollabukdo, Korea were identified by comparison of nucleotide sequences for PCR-amplified D1/D2 region of 26S rDNA or internal transcribed spacer (ITS) 1 and 2 including 5.8S rDNA using BLAST. Twelve yeast strains of ten species and seventeen yeast strains of ten species were isolated from wild flowers of Gyejoksan and Oseosan, respectively. And thirty seven yeast strains of twenty four species were isolated from wild flowers of Baekamsan. Total thirty four yeast species were isolated from three different sample collection areas, but only nine species were overlapped from the at least two different sampling areas: Cryptococcus sp., Cryptococcus aureus, Cryptococcus flavescens, Cryptococcus flavus, Metschnikowia sp., Pseudozyma aphidis, Rhodotorula glutinis, Sporobolomyces carnicolor, and Sporobolomyces ruberrimus. Among them only Cryptococcus aureus was occurred from all three different collection sites. Other twenty five species were restricted to specific collection site suggesting that each area has distinctive yeast flora.

Isolation and Identification of Filamentous Fungi from Indoor Air of a Sogokju Traditional Rice Wine Factory (소곡주 공장의 공기로부터 곰팡이의 분리 및 동정)

  • To investigate the mycoflora of indoor air in a Sogokju, traditional rice wine, factory, fungi were sampled and analyzed from the air of several rooms in the factory using an Anderson air sampler and from two kinds of Nuruk. Twelve fungal species belonging to the genera of Aspergillus, Penicillium, Gibberella, Cladosporium, and Talaromyces were isolated. Species belonging to Aspergillus and Penicillium genera were the major species. Seven different species of Penicillium were isolated from each different room of the factory. The Aspergillus species found from indoor air of the factory was also found from Nuruk. Rhizopus sp. was commonly isolated from Nuruk but not from indoor air of the factory. This is first report of fungi present in indoor air of a traditional rice wine factory in Korea.

Antibiotics Resistance for Isolated Pathogenic Bacteria from Manufacturing Establishment of Sandwich Products (샌드위치 제조사업장에서 분리된 병원성세균에 대한 항생제 내성 검사)

  • The Collected 70 samples from 5 sandwich shops were analysed for the pathogenic bacteria such as Escherichia coli O157:H7, Salmonella spp., Listeria monocytogenes, and Staphylococcus aureus. As a result of Listeria monocytogenes and Saphylococcus aureus were detected in 1 sample, 11 samples, respectively. However, Escherichia coli O157:H7 and Salmonella spp. were not detected in anywhere. The antibiotics test of isolated bacteria was pelformed by the disk diffusion method from NCCLS. The resistance rate of Listeria monocytogenes isolates was confirmed 38.5% to 10 species such as Am, B, P, and Va for antibiotics of 26 species. MRSA was determinated 4 strains in S. aureus isolates. The resistance pattern of Staphylococcus aureus isolates were confirmed 36.4% to P Am OX B K E CXM, 18.2% to P Am B K E CXM B, 9.1% to P Am B K, 27.3% to P Am B, and 9.1% to Te B Nb. Therefore, continuous surveillance and monitoring for antibiotic resistance strains are demanded for prevention of increases in multiple antibiotic resistance strains.

Two Unrecorded Enophytic Fungi Isolated from Root of Ligusticum chuanxiong in Korea: Pithomyces chartarum and Plectosphaerella niemeijerarum (토천궁의 뿌리에서 분리된 2종의 국내 미기록 내생균: Pithomyces chartarum and Plectosphaerella niemeijerarum)

  • Endophytic fungal strains were isolated from the sterilized roots of a medicinal plant, Ligusticum chuanxiong and identified based on morphological characteristics and molecular analysis of internal transcribed spacer, large subunit rDNA, and beta-tubulin DNA regions. Our results confirmed the presence of Pithomyces chartarum and Plectosphaerella niemeijerarum in the fungal strains. To the best of our knowledge, the fungal strains have not been reported in Korea. In this report, we describe morphological characteristics and phylogenetic trees of these endophytic fungal strains.

  • In the present study, we isolated endophytic fungal strains from the rhizoids of the moss Funaria hygrometrica. The isolated strains were identified based on morphological characteristics and analysis of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA sequence regions. Consequently, we confirmed the presence of two endophytic fungal species, Curvularia protuberata and Didymella anserina, which have not been reported in Korea previously. Here, we describe the morphological characteristics and molecular analysis results of these fungal species.

Identification of Mycorrhizal Fungi Identified on Citrus Orchard Soils in The Island of Cheju (제주도 감귤원 토양에서 분리한 공생균근균의 동정)

  • VA-mycorrhizal spores were collected from the 14 citurs orchards of different soil textures and locations in Cheju island. Five species and two kinds of spores were identified as based on the morphological characteristics of the spores; Acaulospora bireticulata, Glomus deserticola, G. geosporum, G. vesiculiferum, and Sclerocystis pachycaulis. Additionally, two kind spores of Acaulospora were also observed but difficult to be identified in this moment. Glomus deserticola and unidentified spores of Acaulospora (brown spores sized 90 to $125\;{\mu}m$ in diameter) were most frequently observed in the all soil specimens in Cheju, while the other kinds of spores were rarely observed in the soil of Cheju.
