Three Phases and Water Characteristics of Horticultural Substrates (원예(園藝) 상토재료(床土材料)의 삼상(三相)과 수분특성(水分特性))

  • A study was carried out to find out the basic information in physical properties for selection and manufacturing the good seedling media through the analysis of the physical properties, such as particle size, water retention and three phases of the major horticultural substrates. Easily available water(EAW), the water contents between 1kPa and 5kPa water potental, was highest in peatmoss with 39%, and perlite 34.0%, vermiculite 16.9%, but the values of osmunda and bark were lower than 4.8%. Water buffering capacity(WBC), the water content between 5kPa and 10kPa, was 6.1% in peatmoss and 2.3% in vermiculite but it was lower than 1.0% in other substrates. To adjust the suitable range of water potential at crossing point of water and air curves to 1.5~2.5kPa, more finer materials were needed in osmunda and bark, and more coarser materials must be added to peatmoss, perlite and vermiculite. Water potentials of substrates in saturated pot condition were equivalent to 2.2kPa in peatmoss and others were ranged in 1.0kPa to 4.3kPa of water potential in pressure chamber.

Mineralogical and Physico-chemical Properties of Sludge Produced During Artificial Sand Processing (국내 화강암류를 이용한 일부 인공쇄석사 제조과정에서 발생되는 슬러지의 광물.물리화학적 특성)

  • The consumption of artificially crushed sands exceeds more than 30 percent of the domestic sand supply in South Korea, and its rate is still increasing. For the manufacture of crushed sand granites and granitic gneisses are preferred, fine fractions (i.e. sludge, particles finer than 63 microns) are removed by use of flocculation agents, and its amount occupy about 15 wt%. The sludges consist of quartz, feldspars, micas, chlorite/vermiculite, kaolinites, smectites and occasionally calcite. Among the clay minerals micas are usually predominant, and $14{\AA}$ minerals, kaolinites and smectites are rather scarce. Jurassic granites usually contain more kaolinites and smectites than those of Cretaceous to Tertiary granites, probably due to longer geologic ages. On the other hand, sludge from Precambrian gneiss does not contain kaolinites and smectites. Chemical analyses for the granites and their sludges show rather clear differences in most of major chemical components. Except for $SiO_2,\;Na_2O\;and\;K_2O$, all other components represent rather clear increase. Decrease of $SiO_2$ content is attributed to the relative decrease of quartz in the sludges. And the $Na_2O decrease is caused by a relatively stronger weathering property of albite compared to Ca plagioclase. The $K_2O$ content shows rather small differences throughout the whole samples. The increases of $Al_2O_3$ and other major components resulted from weathering processes and most of colored components are also concentrated in the sludges. Particle size analyses reveal that the sludges are categorized as sandy loams in a sand-silt-clay triangular diagram. The sludge is now classified as industrial waste because of its impermeability, and this result was also confirmed by rather higher hydraulic conductivities. For the environmental problems, and accomplishing effective sand manufacture, more fresh rocks with little weathering products must be chosen.

Cold shock sensitive growth of Bacillus subtilis mutants deleted for genes involved in fatty acid synthesis (지방산 생합성 관련 유전자 결손 Bacillus subtilis 균주들의 저온충격 민감성 생장)

  • To investigate the role of bkdR, sigL, yplP, and des genes which were known to be involved in fatty acid synthesis and sensitive at low temperature, deletion mutants of Bacillus subtilis CU1065 and JH642 were constructed. To determine the low temperature sensitivity of these genes, we compared the growth curves of cells at $37^{\circ}C$ and $15^{\circ}C$. At $37^{\circ}C$, wild type and deletion mutants showed almost similar growth but only bkdR deletion strain at $15^{\circ}C$ showed very slow growing compared with wild type. At $15^{\circ}C$ sigL and yplP deletions were somewhat slower or similar to those of wild type strain. Double and triple mutants for bkdR, sigL, yplP deletions were constructed and grown at $20^{\circ}C$ in LB agar to investigate cold sensitive growth. Double or triple deletions including bkdR deletion showed cold sensitive growing. In order to identify more clearly cold sensitive growth, the experiments were carried out under cold shock conditions in which the temperature was lowered from $37^{\circ}C$ to $15^{\circ}C$ at the point of 0.4 optical densities at 600 nm. In these cold shock experiments, only bkdR deletion showed significantly lower growing and additional des deletion increases cold sensitivity. The bkdR activates the bkd operon, which catabolized isoleucine, valine and leucine, amino acids and produce precursors for the synthesis of branched fatty acids. At cold shock growing of bkdR deletion strain, isoleucine recovered cold sensitivity of bkdR deletion but valine did not restore cold sensitivity. Isoleucine is used as a precursor for the synthesis of anteiso-branched fatty acids. On the other hand, valine is used as a precursor for the synthesis of iso-branched fatty acids. This indicates that anteiso-branched fatty acid plays an important role at the cold shock condition.

Lipid Compositions Changes of Seasoned Pork Prepared with Medicinal Plant Extracts During Storage (한약재 추출물을 첨가한 양념 돈육의 저장 중 지질성분의 변화)

  • The seasoned porks loins manufactured using with seasoning sauce that were prepared with difference composites of medicinal plant extracts (T-0; medicinal plants extract not added, $T-1{\sim}3$ ; pork treated with mixture of 6 kinds medicinal plant, respectively). And the changes of lipids were investigated during storage at $4^{\circ}C$ for 15 days. Content of TBARS in raw pork was $0.15{\pm}0.03$ MA mg/kg at beginning storage stage, in seasoned pork group (T-1), TBARS content was $0.35{\pm}0.01$ MA mg/kg after storage for 15 days. But TBARS contents were ranged from 0.27 to 0.3MA mg/kg in seasoned pork groups (T-1, T-2 and T-3) and it significantly lower than T-0 group. Unsaturated fatty acid content tended to increase during storage, but decreased after storage 9 days. These tendency was equally appeared in the cholesterol content of $T-1{\sim}3$ groups. In conclusion, the addition of medicinal plant composites enhanced antioxidant activity of the seasoned pork.

Isolation and Morphological Identification of Fresh Water Green Algae from Organic Farming Habitats in Korea (유기농업 생태계로부터 담수 녹조류 분리 및 형태적 동정)

  • This study aimed to isolate and identify freshwater algae from the organic agricultural ecosystems and investigate its biological characteristics to study the possibility of utilizing a biomass freshwater algae in organic farming. In the survey area, average water temperature was $12.4{\sim}28.2^{\circ}C$ and the pH ranges were from 6.1 to 8.5. The solid culture method is more suitable than liquid culture method for isolation of freshwater algae with lower contamination level and higher isolation frequency. A total of 115 strains were isolated from six freshwater algae habitats in nine regions in Korea. BGMM (BG11 Modified Medium) amended with NaNO3 and $KNO_3$ as a nitrogen, and $Na_2CO_3$ as carbon source was designed to isolate and culture freshwater algae. Absorbance of freshwater algae culture has increased dramatically to four days and decreased after eight days after inoculation. CHK008 of the seven isolates showed the highest absorbance in seven days after culturing in BGMM. The optimal pH of BGMM for culturing freshwater algae was pH 6-7. As light intensity increased, growth of freshwater algae increased. Among the five kinds of carbon sources, glucose and galactose promoted good growth of freshwater algae in BGMM. The colony color of purified 16 green algae isolates showed a separation of green, dark and light green, and of them, eleven algae strains showed a strong fluorescent light under fluorescence microscopy. Cell size of the green algae showed a wide range of variation depending on the species. General morphology of the green algae strains was spherical. Chlamydomonas sp. was elliptical, and Chlorella sorokiniana was ellipsoidal and cylindrical. All strains of the green algae except for Chlamydomonas sp. did not have flagella. One isolate of Chlamydomonas sp. and five isolates of C. sorokiniana secreted mucus. Sixteen isolates of 16 green algae were identified as two family and six species, Chlorella vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp. based on their morphological characteristics.

Research Trend of Biomass-Derived Engineering Plastics (바이오매스 기반 엔지니어링 플라스틱 연구 동향)

  • Sustainable plastics can be mainly categorized into (1) biodegradable plastics decomposed into water and carbon dioxide after use, and (2) biomass-derived plastics possessing the carbon neutrality by utilizing raw materials converted from atmospheric carbon dioxide to biomass. Recently, biomass-derived engineering plastics (EP) and natural nanofiber-reinforced nanocomposites are emerging as a new direction of the industry. In addition to the eco-friendliness of natural resources, these materials are competitive over petroleum-based plastics in the high value-added plastics market. Polyesters and polycarbonates synthesized from isosorbide and 2,5-furandicarboxylic acid, which are representative biomass-derived monomers, are at the forefront of industrialization due to their higher transparency, mechanical properties, thermal stability, and gas barrier properties. Moreover, isosorbide has potential to be applied to super EP material with continuous service temperature over 150 ℃. In situ polymerization utilizing surface hydrophilicity and multi-functionality of natural nanofibers such as nanocellulose and nanochitin achieves remarkable improvements of mechanical properties with the minimal dose of nanofillers. Biomass-derived tough-plastics covered in this review are expected to replace petroleum-based plastics by satisfying the carbon neutrality required by the environment, the high functionality by the consumer, and the accessibility by the industry.

Effect of Ethanolic Extract of Schizandra chinensis for the Delayed Ripening Kimchi Preparation (오미자(Schizandra chinensis) 추출물이 김치의 과숙억제에 미치는 영향)

  • This study mainly focused on to investigate the effects of Schizandra chinensis on the growth of a bacterium, CS6 which was isolated from kimchi. CS6 was final]y identified to lactobacillus plantarum that caused acidification of kimchi. The ethanolic extract of Schizandra chinensis(EES) inhibited the growth of L. plantarum. Minimum inhibition concentration of crude EES on L. plantarum was 62.5mg/$m\ell$. In broth culture, 5$\mu\textrm{g}$/$m\ell$ of EES completely inhibited the growth of L. plantarum during fermentation. The addition of 0.4% of EES has no apparent effect on quality including the taste and color on kimchi. It was expected that EES-containing kimchi could extend the period of preservation. Analysis of organic acids in water fractions of EES was carried out by HPLC. It is apparent that antimicrobial active fractions contained the highest concentration of succinic acid, a little tartaric acid and malic acid. Among these organic acids, succinic acid showed the strong inhibitory effect against L. plantarum CS6 in vitro. Succinic acid-containing kimchi with a concentration of 0.4 and 0.5% had the inhibitory effect on growth of L. plantarum. Inhibitory effect of EES on amylase, cellulase and pectinase was also tested. In conclusion, the present experiment demonstrated that EES inhibited the growth of L. plantarum, and various enzyme activity. EES-containing kimchi was sustained the hardness, and initial acidity during fermentation. EES was considered as the possible additive of kimchi process and EES added in kimchi increase the quality, and storage period of kimchi.

Effects of Hemicellulase on Purple Sweet Potato Bread (헤미셀룰레이즈 첨가가 자색고구마 식빵의 품질에 미치는 영향)

  • In this study, purple sweet potato bread was prepared by the addition of 0.005%, 0.010%, 0.015% and 0.020% hemicellulase. It's effect on product quality and consumer evaluation were examined. The results showed that the dough pH and total titratable acidity were not significantly different between samples. In the fermentation power of dough expansion, a 0.015% addition sample was the highest between the samples. The bread pH decreased significantly as hemicellulase was increased, whereas. Bread total titratable acidity was significantly decreased. The addition of hemicellulase samples were significantly higher in specific volume and baking loss than the control sample. The moisture content was not significantly different between samples. In colors, the lightness of the control sample was the highest, the redness of the 0.020% addition sample was the lowest while the yellowness of the control was the lowest. The hardness and the fracturability decreased significantly as hemicellulase was increased. The resilience indicated reverse effects. In consumer evaluation, the color and softness were not significantly different between samples. And the hemicellulase addition of samples was higher in flavor than that of the control sample. The overall acceptability was the highest at 5.67 with a 0.010% addition sample. According to these results, the addition of 0.010% hemicellulase in purple sweet potato bread would be the optimum level.

Changes of Quality Characteristics on the Bread added Chitosan (키토산 첨가에 따른 식빵의 품질 변화)

  • This was studied to evaluate the quality characteristics of the bread added chitosan during storage at room temperature(Temp. $27^{\circ}C{\pm}2$, RH $75%{\pm}10$). The volume of the dough was increased depending on the larger molecular weight and the higer concentration of chitosan but was decreased at 0.50% of 120 kDa chitosan. The water activity was low depending on the larger molecular weight and the higher concentration of chitosan at the early storage, but maintained constantly during storage totally. The colors of the bread was hardly affected by 30 kDa of chitosan. Textural characteristics was improved at 30 kDa and 120 kDa of chitosan. Especially, the change of the hardness were maintained lower at 30 kDa, 120 kDa of chitosan during storage than that of standard. These results showed that the quality of the bread by added 30 kDa of chitosan was improved highly.

Influence of resin-nanoceramic CAD/CAM block shade and thickness on the microhardness of dual-cured resin cement (레진-나노세라믹 CAD/CAM블록의 색조와 두께가 이원중합 레진시멘트의 미세경도에 미치는 영향)

  • The purpose of this study was to examine the effect of shade and thickness of resin-nanoceramic CAD-CAM block (RNB) on the microhardness of dual-cured resin cement, as well as to measure the number of photons transmitted through RNBs of different thicknesses and colors. One dual-cured resin cement was used to prepare resin cement specimens. Resin cement specimens were light-cured for 40 seconds through 3 shades (A1, A2, A3 in HT (high translucency) and LT (low translucency) respectively) and four thicknesses (1, 2, 3, 4 mm) of RNB specimens. Vickers microhardness measurements of resin cement specimens were performed using a Vickers hardness tester. The light transmission of RNB specimens was measured using a spectrometer (SpectroPro-500, Acton Research, Acton, MA, U.S.A.), and the translucency parameter was calculated using the CIEL*a*b* system. Data were statistically analyzed by ANOVA and Tukey's test. There was a significant decrease of microhardness of resin cement specimen with an overlay of 4 mm of RNB thickness and A3 shade in comparison to A1 and 1 mm, respectively (p<0.05). The translucency parameter values and light transmission of RNBs tested differed significantly, according to the thicknesses of the specimen (p<0.05). Light transmission is decreased with increase in the thicknesses of RNBs. Shade A1 transmitted more light than darker blocks. A decrease in microhardness of resin cement specimens was observed with increasing thickness and shade (A1 to A3) of RNBs.
