  • 잉어, 가물치 및 메기의 체표면 점액내에 있는 기흡충에 데한 살충성물질의 계절적 변동에 대하여 in vitro에서 검토하였다. 이들 어류 체표점도의 에테르추출물량은 겨울철에는 그 밖에 계절에 비하여 현저하게 감소함에도 불구하고 살충성물질과 에테르추출물을 박롱 크로마토그라휘하여 얻은 각반점물질의 Rf값은 계절적 변동을 가져오지 않았다.

Histochemical Study on the Mucous Secreting Cells in the Gastrointestinal Mucosae of Rana rugosa according to Hibernation (동면에 따른 옴개구리(Rana rugosa) 위장관 점액분비세포에 관한 조직화학적 연구)

  • The mucosubstances in the gastrointestinal mucous secreting cells of Rana rugosa were detected histochmeically during pre-hibernating, hibernating, post-hibernating and active phases. The results of the observation were as follows: 1. The mucosubstances in the gastrointestinal mucous secreting cells of active frog were strongly PAS-active in stomach, PAS-active and alcianophilic at pH 2.5 in small intestine and alcianophilic at pH 2.5 in large intestine. 2. The PAS-active mucosubstances in the gastric surface epithelial cells were increased remarkably during hibernation. 3. The alcianophilic mucosubstances at both pH 2.5 and pH 1.0 were decreased remarkably in the goblet cells of small intestine during hibernation, but a little PAS-active ones were increased. 4. The alcianophilic mucosubstances at pH 2.5 were decreased remarkably and a lttle PAS-active ones also were done in the goblet cells of large intestine during hibernation. 5. The increases of the contained quantity of mucosubstances in the gastric surface epithelial cells during hibernationi may have theeffects of preventing cohesions of gastric mucosae and suppressing activities of gastric acid and enzymes. The mucosubstances neutral acidity in the intestine during hibernation may be secreted, because of acidity being done near neutrality in its lumen, due to remarkable decrease of intestinal juices and gastric acids.

Biocontrol Activity of Myxococcus sp. KYC 1126 against Phytophthora Blight on Hot Pepper (점액세균 Myxococcus sp. KYC 1126을 이용한 고추 역병 생물학적 방제 효능)

  • Bacteriolytic myxobacteria have been known to secrete various antifungal metabolites against several soilborne phytopathogens including Phytophthora. Among the three isolates of Myxococcus spp., KYC 1126 and KYC 1136 perfectly inhibited the mycelial growth of Phytophtora capsici in vitro. In order to show the biocontrol activity on Phytophthora blight of hot pepper, we tried to find the best way of application of a myxobacterial isolate. Although KYC 1126 fruiting body was easily grown on the colony of Escherichia coli as a nutrient source, it did not control the disease when it was pre-applied in soil. Before the bioassay of a liquid culture filtrate of KYC 1126 was conducted, its antifungal activity was confirmed on the seedlings applying with the mixture of the pathogen's zoospore suspension and KYC 1126 filtrate. On greenhouse experiments with five and four replications, the control value of KYC 1126 on phyllosphere and rhizosphere was 88% and 36%, respectively. Whereas, the control value of dimetnomorph+propineb on phyllosphere was 100% and that of propamorcarb on rhizosphere was 44%. There was a phytotoxicity of the myxobacterial filtrate when seedlings were washed and soaked for 24 hours. Gummy materials were covered with roots. And stem and petiole were constricted, then a whole seedling was eventually blighted.

  • 쇠오징어(Sepiella maindroni)흠반의 외측 상피조직에서 3종류의 감각세포(A, B 및 D형 감각세포)와, 1종류의 감각상피세포(C형 감각 상피세포) 그리고 2종류의 점액세포(a 및 b형 점액세포)가 각각 관찰되었다. A 및 B형 감각세포는 분포상태가 다를 뿐 형태는 거의 유사한 세포이고 많은 분비액포를 지닌 해면질 형태의 세포로서 전자 밀도가 낮은 물질을 분비하는 분비성 감각세포로 확인되었다. 0형 세포는 매우 작은 감각세포로서 세포의 자유면에 긴 융모가 밀생되어있었고, 측면원형질막은 불규칙한 수지상 연접을 이루고 있었다. 이 세포는 전자밀도가 매우 높아서 검게 보였다. C형 감각 상피세포도 세포의 자유면에 긴 미세융모가 밀생되어있는 단층 원주상 세포로서. 이 세포 역시 측면원형질막이 불규칙한 수지상 연접을 이루고 있고. 엽접내에는 많은 소포와 약간의 사립제가 관찰되었다. 3형 점액세포는 세포질속에 내강이 팽창된 활면소포체를 소지하고 있으며 전자밀도가 높은 구형의 점액과립을 형성하였고, b형 점액세포도 전자밀도가 중등도인 점액질 과립을 소지하고 있었다.

Histochemical Studies of the Goblet Cell of the Oviduct in Korean Ring-necked Pheasants(Phasianus colchicus karpowi) (한국산 꿩 난관의 술잔세포에 관한 조직화학적 연구)

  • The study was designed to investigate the aspects of the goblet cell of the oviduct in laying Korean ring-necked pheasants by histochemical methods (PAS, alcian blue-PAS, alcian blue pH 2.5, alcian blue pH 1.0, and aldehyde fuchsin-alcian blue stain). Twelve laying Korean ring-necked pheasants were used. The results were summarized as follows. The goblet cell of the oviduct of the Korean ring-necked pheasant contained sulfated acid mucosubstances and neutral mucosubstances in the infundibulum. The compounds of sulfated acid mucosubstances, nonsulfated acid mucosubstances, and neutral mucosubstances are present in goblet cells of the magnum, vagina and openings of the tubular gland of the uterus of the oviduct in the Korean ring-necked pheasant. The goblet cell of the isthmus and uterus of the oviduct of the Korean ring-necked pheasant contained neutral mucosubstances except openings of the tubular gland of the uterus. The histochemical characteristics of secretory granules of goblet cells in oviduct of the Korean ring-necked pheasant were not related to the position of eggs in oviduct.

  • Polymerase chain reactin (PCR) could be a simple way to screen new microbial strains producing useful secondary metabolites if their biosynthetic genes are known and candidate strains to be screened are available. In this study, we have screened two myxobacterial strains, KYC3047 and KYC3076, carrying genes appeared to be biosynthetic genes of soraphen A, a potent antifungal substance, out of 50 cellulose degrading myxobacteria using PCR. The two strains were identified as Sorangium cellulosum based on morphological, physiological, and molecular biological characteristics. Both of the strains produced substances having strong antifungal activities as expected against Candida albicans, a causative agent of candidiasis, and Colletotrichum acutatum, a causative agent of anthracnose on pepper.

  • A 70-year-old female who was diagnosed as myxoid chondrosarcoma by fine needle aspiration of a pleural mass is described. She presented with left chest discomfort of 4 months' duration and aggravating dyspnea and chest pain for 2 months. Chest X-ray and CT scan revealed a large lobulated low density mass invading chest wall at the left pleural cavity and massive pleural fluid. Fine needle aspiration was done under the impression of mesothelioma or metastatic cancer. The aspirates from the mass were very cellular and composed of isolated or clustered forms of large plump cells. Abundant cytoplasm was blulsh opaque and the margin was rounded in the isolated cells, whereas clustered cells show ill-defined ceil borders and aggregating tendency. The nuclei were eccentric, round to ovoid, and had fine chromatin pattern and multiple small nucleoli. Cellular pleomorphism or mitotic figure was not definite. These findings were consistent with cytologic features of chondrosarcoma. Final diagnosis was confirmed as myxoid chondrosarcoma by mediastinoscopic biopsy and the tumor showed strong positivity for S-100 protein.

Chivosazole F, An Efficient Inhibitor of Multidrug-Resistant Cancer Cells Isolated from Sorangium Cellulosum [Myxobacteria] (Chivosazole F: 점액세균 Sorangium Cellulosum이 생산하는 다제내성 암세포의 생장억제물질)

  • In the course of our search for compounds effective to multidrug-resistant cancer cells from myxobacteria with the adriamycin-resistant cancer cell line CL02, we found cytotoxic activity against the CL02 cells in culture extract of Sorangium cellulosum JW1045. Activity-guided fractionation of the culture extract led to the isolation of an active principle, chivosazole F, This compound showed high cytotoxic activity against cultured human cancer cells. The $IC_{50}$ values, measured by a SRB assay with different cell lines, ranged from 0.1 to 10 ng/ml. Furthermore chivosazole F was as active against drug-resistant cancer cells CL02 and CP70 as against the corresponding sensitive cells.

  • The chemical compositions of the root bark of Ulmus species and mucilage were investigated. The methanol extractives were fractionated with n-hexane, ethyl ether, ethyl acetate and water and antibacterial activities of each fraction were investigated. Although differences of chemical compositions of the Ulmus species were not noticeable, nitrogen and lignin contents of xylem were higher than those of phloem. However, ash and extractive contents showed adverse results. The mucilage of root bark was slowly gushing out by water and similar to pectin substance with high acidic sugar content. In fractionation experiment, the yield of methanol extraction was about 7% and the portion of water-soluble materials in methanol extractives was 78%. These water- soluble materials had the most high antibacterial activity on Staphylococcus aureus and Salmonella typhimurium.

Screening of Myxobacteria Inhibiting the Growth of Collectotrichum acutatum Causing Anthracnose on Pepper (고추탄저균 성장 억제 점액세균의 탐색)

  • As an effort to search new bacterial biocontrol agents against pepper anthracnose, we screened myxobacteria, which might inhibit the growth of Colletotrichum acutatum, the agent of that plant disease. When 93 myxobacterial strains including 59 Myxococcus spp. and 34 Corallococcus spp. were tested against C. acutatum ACYSJ001 on agar plates, 10 strains identified as the genus Myxococcus significantly obstructed the growth of C. acutatum, whereas the majority of strains belonging to the genus Corallococcus did not demonstrate any counteractive effect. Such results have indicated that the strains of the genus Myxococcus have a high potential to play roles of biocontrol agents for control of pepper anthracnose. These also have revealed that the strains of the genus Myxococcus could be used as excellent microbial resources for screening novel antifungal substances.
