Model Systems in Radiation Biology: Implication for Preclinical Study of Radiotherapy (방사선 생물학을 위한 모델 시스템: 방사선치료의 전임상 연구)

  • In radiation biology, analysis of various mechanisms in response to radiation has been accomplished with the use of model organisms. These model organisms are powerful tools for providing a biologically intact in vivo environment to assess physiological and pathophysiological processes affected by radiation. Accumulated data using these models have been applied to human clinical studies (including the evaluation of radiotherapeutic efficacy) and discovery of radiotherapy reagents. However, there are few studies to provide overall integrated information about these useful model organisms. Thus, this review summarizes the results of radiation biology studies using four well-known model organisms: yeast, Caenorhabditis elegans, Drosophila melanogaster, and mice.

  • Stem cell research arose from the need to explore new therapeutic possibilities for intractable and lethal diseases. Although musculoskeletal disorders are basically nonlethal, their high prevalence and the relative ease of performing clinical trials have facilitated the clinical application of stem cells in this field. On the other hand, despite the plethora of in vitro and preclinical studies in stem cell research for regenerative medicine in the musculoskeletal system, few reliable clinical studies have been published. Stem cell therapy can be applied locally for bone, cartilage, and tendon regeneration. The candidate disease modalities in bone regeneration include large bone defects, nonunion of fractures, and osteonecrosis. Focal osteochondral defect and osteoarthritis are the current targets for cartilage regeneration. For tendon regeneration, bone-tendon junction problems, such as rotator cuff tears are hot topics in clinical research. To date, the literature supporting stem cell-based therapies comprises mostly case reports or case series.

  • The advances of semiconductor and circuitry technology dovetailed with nano processing techniques have further enhanced micro-miniaturization, sensitivity, longevity and reliability in MID(Medical Implant Device). Nevertheless, one of the remaining challenges is whether power can sufficiently and continuously be supplied for the operation of the MID. Self-powered MID that harvest biomechanical energy from human motion, respiratory and muscle movement are part of a paradigm shift. In this paper, we developed a rechargeable pacemaker through self-power generation with the triboelectric nanogenerator. We demonstrate a fully implanted pacemaker based on an implantable triboelectric nanogenerator, which act as a storage as well as active movement on a large-animal(dog) scale. The self-power pacemaker harvested from animal motion is 2.47V, which is higher than the required pacemaker device sensing voltage(1.35V).

Anti-Obesity Effect of Pinus densiflora Leaf Extracts (솔잎(Pinus densiflora leaf) 추출물의 항비만효과)

  • In this study, we measured the inhibitory activity of Pinus densiflora leaf (PDL) against excessive lipid accumulation in mouse preadipocyte, 3T3-L1 cells to investigate whether PDL exerts anti-obesity activity. Lipid accumulation and the protein level were measured using Oil red O staining assay and Western blot analysis, respectively. We observed that PDL inhibited excessive lipid accumulation and decreased the expression of CEBPα, PPARγ and perilipin-1 related to lipid accumulation in 3T3-L1 cells. Therefore, considering these results, PDL can be used as a potential agent for anti-obesity.

Adaptive Image Rescaling for Weakly Contrast-Enhanced Lesions in Dedicated Breast CT: A Phantom Study (약하게 조영증강된 병변의 유방 전용 CT 영상의 대조도 개선을 위한 적응적 영상 재조정 방법: 팬텀 연구)

  • Purpose Dedicated breast CT is an emerging volumetric X-ray imaging modality for diagnosis that does not require any painful breast compression. To improve the detection rate of weakly enhanced lesions, an adaptive image rescaling (AIR) technique was proposed. Materials and Methods Two disks containing five identical holes and five holes of different diameters were scanned using 60/100 kVp to obtain single-energy CT (SECT), dual-energy CT (DECT), and AIR images. A piece of pork was also scanned as a subclinical trial. The image quality was evaluated using image contrast and contrast-to-noise ratio (CNR). The difference of imaging performances was confirmed using student's t test. Results Total mean image contrast of AIR (0.70) reached 74.5% of that of DECT (0.94) and was higher than that of SECT (0.22) by 318.2%. Total mean CNR of AIR (5.08) was 35.5% of that of SECT (14.30) and was higher than that of DECT (2.28) by 222.8%. A similar trend was observed in the subclinical study. Conclusion The results demonstrated superior image contrast of AIR over SECT, and its higher overall image quality compared to DECT with half the exposure. Therefore, AIR seems to have the potential to improve the detectability of lesions with dedicated breast CT.

Comparative Tissue Ablation Study between 532 and 980 nm (가시광선과 근적외선 파장을 이용한 조직 제거 연구)

  • In this paper, two wavelengths mainly used for laser prostatectomy have been quantitatively compared: 532 and 980 nm. Two lasers at 40 W were employed to ablate bovine liver in vitro. Ablation performance was evaluated in light of number of sweeps, ablation volume, and coagulative necrosis. 532 nm yielded up to four times higher ablation efficiency than 980 nm. Regardless of wavelength, ablation rate per sweep decreased with the number of sweeps. 532 nm generated relatively deeper ablation craters along with thinner coagulation whereas 980 nm created superficial tissue ablation with up to 2 mm thick coagulative necrosis. Due to higher light absorption and effective thermal confinement, 532 nm induced more efficient tissue ablation with a smaller coagulative necrotic zone. The current study demonstrated that 532 nm could be a more ideal wavelength for laser prostatectomy, and the future in vivo investigations will confirm these findings.

  • In this paper, We would research and analysis for actual beauty information condition in Korea on the end of July 2004. We make an investigation into 3 beauty subject that Web service were operated by 100% regular school education. 15.5% Private school in 363 teaching institutions, 8.74% of 767 material dealers, 0.38% of 77,546 beauty salons. There are 12 companies of client management program and 3 hair changing programs. It bring to be few of technical beauty information data and creature value added that lack of full-time lecturer, according1y, KBMC and Ministry of Health & Welfare would constitute beauty information commit that make connection of each 3 beauty subjects and web database for technical beauty information data. As a result of unification and communization for beauty information system. It should be take synergy effect for Korean Beauty.

The Preclinical Study of Hyeolbuchugeo-tang (Xuefuzhuyu-tang) on Bone Healing in Rats with Rib Fracture (골절 유발 Rat에 대한 혈부축어탕(血府逐瘀湯)의 전임상 연구)

  • Objectives The purpose of this study is to evaluate the healing effect of Hyeolbuchugeo-tang (HC) in rats with rib fracture. Methods Rats were randomly divided into 5 groups (naive, control, positive control, HC-L and HC-H). All groups except naive group were subjected to bone fracture of rib. Naive group received no treatment at all. Control group was fed with phosphate buffered saline. Positive control group was orally medicated with tramadol. Experimental group was orally medicated with HC extract (50 mg/kg for low concentration [HC-L], 100 mg/kg for high concentration [HC-H]). X-ray and micro-computed tomography (micro-CT) were conducted to assess the effect of HC. We analysed the level of 2) transforming growth factor-β1 (TGF-β1), Ki67, alkaline phosphatase (ALP), receptor activator of nuclear factor kappa-β, runt-related transcription factor 2 (Runx2) and tartrate resistant acid phosphatase (TRAP) on 7 and 14 days after fracture. ALP, alanine aminotransferase, aspartate aminotransferase, blood urea nitrogen, creatinine was measured for safety assessment. Results X-ray and micro-CT, showed HC enhance bone repair process. Callus formation was increased in experimental group at 7 days after fracture, but decreased at 14 days after fracture. 7 days after fracture, the level of TGF-β1 in experimental group was decreased. The level of Ki67, Runx2 in HC-H, TRAP in HC-L was increased. 14 days after fracture, the level of Ki67 in HC-L and HC-H was decreased. The level of ALP, Runx2, BUN in HC-L, TRAP in HC-L and HC-H was increased. Conclusions Taken together the results, HC promoted healing of bone fracture. In conclusion, HC has a potential to promote healing of bone fracture.

Research about Hyperspectral Imaging System for Pre-Clinical testing of Small Animal (소형동물 전임상실험을 위한 하이퍼스펙트럼 영상장비 연구)

  • In this study we have developed a hyperspectrum imaging system for highly sensitive and effective imaging analysis. An optical setup was designed using acoustic optical tunable filter (AOTF) for high sensitive hyperspectrum imaging. Light emitted by mercury lamp gets split in to diffracted and undiffracted beams while passing though AOTF. GFP transfected HEK-293 cell line was used as a model for in vitro imaging analysis. Cells were first, analyzed by fluorescence microscope followed by flow cytometric analysis. Flow cytometric analysis showed 66.31% transfection yield in GFP transfected HEK-293 cells. Various images of GFP transfected HEK-293 cell were grabbed by collecting the diffracted light using a CCD over a dynamic range of frequency of 129-171 MHz with an interval of 3 MHz. Subsequently, for in vivo image analysis of GFP transfected cells in mouse, a whole-body-imaging system was constructed. The blue light of 488 nm wavelength was obtained from a Xenon arc lamp using an appropriate filter and transmitted through an optical cable to a ring illuminator. To check the efficacy of the newly developed whole-body-imaging system, a comparative imaging analysis was performed on a normal mouse in presence and absence of Xenon arc irradiation. The developed hyperspectrum imaging analysis with AOTF showed the highest intensity of green fluorescent protein at 153 MHz of frequency and 494 nm of wavelength. However, the fluorescence intensity remained same as that of the background below 138 MHz (475 nm) and above 162 MHz (532 nm). The mouse images captured using the constructed whole-body-imaging system appeared monochromatic in absence of Xenon arc irradiation and blue when irradiated with Xenon arc lamp. Nevertheless, in either case mouse images appeared clearly.

The Preclinical and Clinical Effects of Herbal Product Containing Rosa mutiflora Roots Extracts as a Main Component on the Hair Growth Promotion (찔레뿌리 추출물을 주성분으로 하는 천연물 조성물의 발모촉진 효과에 관한 전임상 및 임상시험 연구)

  • In this study, to develop hair growth agents using natural products which has excellent ability to promote hair growth effect and fewer side effect, animal experiment and clinical trials was performed to evaluate hair growth promotion effect of herbal product containing $Rosa$ $mutiflora$ roots extracts as a main component (RMHP). 4-week-old male C57BL/6 mice were removed the dorsal hair using thioglycolate, and applied 15% EtOH solution as a negative control, 5% minoxidil as a positive control and RMHP to dorsal skin. In the results of macroscopy and photo-interpretation, RMHP group recorded 100% (+++++) of hair growth was proved to significantly stimulate hair growth against 80% (++++) negative control group. 37 patients were treated with RMHP and evaluated the therapeutic effect at 16 weeks. Hair density was significantly increased at 16 weeks after applying RMHP ($125.0{\pm}4.9\;FU/cm^2$) compared to before treatment ($104.3{\pm}4.7\;FU/cm^2$, p < 0.05), and hair thickness were also significantly increased ($0.066{\pm}0.003$ mm) than before treatment ($0.055{\pm}0.002$ mm, p < 0.05). The result of clinical photo-interpretation using 7-point rating scale assessment, after 16 weeks clinical symptoms were evaluated to significantly improve with $1.23{\pm}0.05$ (p < 0.05). Therefore, the results of this study were observed that RMHP have hair loss prevention effect and hair growth promotion effect to hair loss patients.
