Effect of Light Emitting Diodes Treatment on Growth and Quality of Lettuce (Lactuca sativa L. 'Oak Leaf') (LED 처리가 상추의 생육 및 품질에 미치는 영향)

  • The objective of this study was to elucidate the effect of light-emitting diode treatment on early growth and inorganic elements in leaf lettuce (Lactuca sativa L. 'Oak Leaf'). In changes to leaf morphology, shoot elongation and hypocotyl length showed poor growth under red light irradiation, while red+blue light irradiation induced shorter plant height and more leaves, resulting in increased fresh weight. With respect to Hunter's color and SPAD values, lettuce seedlings grown under red+ blue and fluorescent light irradiation had a higher $a^*$ value but showed no other changes to SPAD values. Interestingly, redness in relative chlorophyll content was 1.4 times higher under red+blue light irradiation. Inorganic element (N, Ca, Mg, and Fe) and ascorbic acid concentrations increased in lettuce plants grown under LED light irradiation compared to those of lettuce grown under fluorescent light, which showed a higher P content. In conclusion, red+blue light irradiation, which stimulates growth and higher nutrient uptake in leaf lettuce, could be employed in containers equipped with LEDs.

  • Produce, including leafy vegetables, has been implicated in several outbreaks of food illness. To evaluate microbiological safety of lettuce and it's cultivation area, a total of 147 samples were collected from lettuce farms and post harvest facility at Icheon, Gyeonggi province. The collected samples were assessed for presence of sanitary indicator microorganisms (Aerobic plate count, coliform count, Escherichia coli) and foodborne pathogens (Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus). The population of APC was over 4.0 log CFU from most of the samples. While the numbers of APC, and coliform of lettuce at 62 days after transplanting were 4.18 log CFU/g, and 1.00 log CFU/g, respectively, those of 10 days after transplanting were 5.37 log CFU/g, and 2.87 log CFU/g, respectively. B. cereus was highly detected from soil and balance which were contaminated with 3.5 log CFU/g, and 2.6 log CFU/100 $cm^2$, respectively. The number of E. coli recovered from gloves was 3.5 log CFU/hand. However, E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected. These data suggested that risk management system should be introduced to lettuce farms to enhance safety of lettuce.

Growth characteristic of lettuce (Lactuca sativa L.) based on Pulse Width modulation of artificial light sources with UV-A. (UV광원을 포함한 인공광원의 펄스폭 변조에 따른 적치마 상추의 생육특성)

  • 식물공장 내 작물의 생육은 제공되는 인공광원의 광원 및 광질에 영향을 받으며, 광흡수 파장에 따라 다르다. 또한 광합성에 효과적인 형태의 빛의 계속적인 빛의 조사보다는 광 펄스를 조절하여 공급하여 준다면 더욱더 효과적인 생육 환경을 제공해 줄 수 있다. 식물의 최적 생장을 위해 특정 파장대의 빛을 선택적으로 조사할 수 있는 LED특성을 사용하여 UV광원을 포함한 인공광원의 펄스폭 변조에 따른 적치마 상추의 생육특성을 알아보고자 하였다. 광환경은 Red(660 nm), Blue(450 nm), UV(395 nm) LED를 8:1:1 비율로 광량 $160{\mu}mol{\cdot}m^2{\cdot}s^{-1}$, 주파수 1.25, 2.5, 3.75, 5.0 kHz로 조사하여 주었으며, 온도 $20{\sim}23^{\circ}C$, 습도 50~60%, $CO_2$농도 1,000 ppm으로 조성하여 주었다. 아시아종묘 적치마상추를 파종 후 18일 째 되는날 정식, 정식 후 14일 28일 째 되는날 SPAD, 지상부 지하부의 생체중 및 건물중, 엽폭, 엽장을 측정하였으며, 측정한 엽폭과 엽장을 이용하여 엽형지수 산출, 지상부 지하부 생체중 값을 이용하여 S/R율을 산출하였다. SPAD 측정결과 생육시기가 증가할수록 SPAD함량은 감소하였으며, 1.25와 2.5 kHz에서 생육 시기 증가에 대한 SPAD함량 감소가 컸고, 3.75와 5.0 kHz의 경우 SPAD함량의 감소량은 작았다. 지상부 생체중은 3.75 kHz에서 121.51 g으로 가장 높은 값을 나타냈으며, 2.5, 1.75, 5.0 kHz 순으로 높은 값을 나타났다. 지하부 생체중의 경우 3.75 kHz에서 31.31 g으로 가장 높은 값을 나타냈으며 2.5, 5.0, 1.25 kHz 순으로 높게 나타났다. 엽형지수는 생육 시기가 증가에 따라 감소하는 것으로 나타났으며, 1.75, 2.5, 5.0, 3.75 kHz 순으로 크게 나타났다. 지상부 건물중 측정결과는 지상부 생체중 결과와, 지하부 건물중의 측정결과는 지하부 생체중 결과와 동일하였다. S/R율은 1.75 kHz를 제외하고 생육시기가 증가할수록 S/R율은 감소하는 것으로 나타났으며, 주파수가 높아질수록 S/R율은 감소하는 것으로 나타났다. 엽형지수는 엽폭/엽장으로 엽형지수를 산출한 결과로 값이 작을수록 엽폭이 넓은 형태를 의미하며, 생체중값이 가장 크게 나타났던 3.75 kHz에서 엽형지수의 값이 가장 낮게 나타났으며 3.75 kHz를 제외하고 주파수가 높을수록 엽형지수의 값이 낮게 나타났다. 이에 주파수에 따른 적상추의 생육은 3.75 kHz에서 가장 좋은 것으로 판단할 수 있었다.

Effects of Light Sources, Light Quality on the Growth Response of Leafy Vegetables in Closed-type Plant Factory System (완전제어형 식물공장에서 광원, 광질에 따른 엽채류 6종의 생육반응)

  • This study was conducted to evaluate the growth response of economical six leafy vegetables that are crown daisy, pak-choi and four kinds of lettuce (Red leaf lettuce, Green leaf lettuce, Head lettuce, Romaine lettuce) by light treatment of LED in plant factory. The light treatments were composed of red, blue, red+farred, red+blue, red+blue+white LEDs, irradiation time ratio of the red and blue LED per minute (1 : 1, 2 : 1, 5 : 1, 10 : 1), and duty ratio of mixed light (100%, 99%, 97%). The following results were obtained in different LED light sources treatments: Shoot biomass and S/R ratio of romaine lettuce were the highest under mixed red+blue LEDs. S/R ratio of head lettuce was higher under mixed red+blue+white LEDs than red+blue LEDs. The others showed no difference in LED light treatment. Shoot biomass, total biomass and S/R ratio of green lettuce, head lettuce and pak-choi were highest in the higher red ratio (5 : 1) on irradiation time of red : blue LED ratios. By the different duty ratio (red+blue and red+blue+white LEDs), Under the mixed light of red+blue, shoot and root biomass of crown daisy and romaine lettuce were high in duty ratio of 100% and 99%, and S/R ratio was highest in all the 6 kinds in duty ratio of 97%. All the 6 kinds showed a fine growth state in low duty ratio (97%). Green lettuce, romaine lettuce and pak-choi showed relatively high shoot biomass and total biomass in low duty ratio of 97% under the mixed light of red+blue+white. S/R ratio of romaine lettuce and head lettuce were highest in the duty ratio of 97% with red+blue+white LEDs. Thus, we can cultivate stably without reference to external factors, if we use appropriate light sources and light quality in closed-type plant factory.

Leakage of Sugars, Amino Acids and Protein from Differently - Aged Seeds of Sesame, Welsh Onion and Lettuce (퇴화처리에 따른 참깨, 파, 상추 종자의 당, 아미노산, 단백질 누출)

  • Experiments were conducted to obtain information on separation of nonviable seeds from seed lots by the nondestructive ways. Seeds of sesame, welsh onion and lettuce were artificially aged at 90% relative humidity and 45$^{\circ}C$ to get different seed qualities. The relationships between seed quality and leakage of total sugars, amino acids, and proteins into soaking water were determined to know a possibility of grading seeds. Dead seeds of lettuce leaked significant amounts of total sugars, amino acids, and proteins, while high quality seeds leaked negligeable amounts of total sugars and some of amino acids and proteins. Dead seeds of welsh onion leaked significant amounts of amino acids and some total sugars and proteins, while high quality seeds leaked negligeable amounts of these compounds. Sesame seeds leaked little total sugars, amino acids, and proteins regardless of seed quality.

Effect of Covering Methods with Ventilating Non-Woven Fabric on the Growth and Yield in Leaf Lettuce during Low Temperature Season (통기성 간이 피복재의 피복방법이 저온기에 잎상추의 생육, 수량 및 품질에 미치는 영향)

  • The study was conducted to estimate stimulation of plant growth of blue and red leaf lettuce (Lactuca sativa L.) as affected by covering methods with vinyl materials containing concentrated PO film. Plants were grown in direct, direct and tunnel, and tunnel covering with 'Pasraido materials' from October 13 to 31. With the covering mean air temperature, soil temperature, leaf temperature and relative humidity remarkedly increased. Higher temperature was observed in the order of direct and tunnel, direct, tunnel covering and the control. Photosynthetic photon flux (PPF, $\mu$mol m$^{-2}$ . s$^{-1}$ ) was observed in reverse order of temperature. Concentration of $CO_2$ was higher during night than during day under the light. Accumulation of highest $CO_2$ at night was obtained from direct coverings followed by direct and tunnel, tunnel covering and the control. Plant height, leaf number and leaf area of blue and red lettuces were stimulated by direct covering. However, there was no significant difference.

Effect of Polyolefine Greenhouse Covering Film on Growth of Lettuce and Cucumber in Cool Season Cultivation (상추와 오이재배 하우스의 저온기 PO필름 피복효과)

  • This study evaluated the effect of polyolefine greenhouse covering film on growth of lettuce and cucumber. The growth of lettuce and cucumber plants under conventional film (polyethylene plus ethylenevinyl acetate, PE + EVA) and polyolefine (PO) film were evaluated in a greenhouse. The physical qualities of PO film, such as tensile strength and tear strength, were stronger than conventional PE + EVA film. PO film showed 4% higher transmission performance in photosynthetic active radiation (PAR) than PE + EVA film. The average day and night temperature in greenhouse under PO film were $1.5^{\circ}C$ and $0.8^{\circ}C$ higher but there was no significant difference in relative humidity compared with conventional film. Chlorophyll content of lettuce and cucumber plants grown under PO film were higher than conventional film. Growth characteristics; plant height, leaf area, fresh weight and dry weight of cucumber was shown better for PO film. The yields of cucumber and lettuce was increased 14.0% and 13.6% respectively, when the crops were grown under PO covering film greenhouse.

  • 상추(Lactuca sativa L.)는 대표적인 쌈 및 샐러드 채소로 우리나라 기준(2016년) 3,387 ha의 면적에서 86,128톤을 생산하여 엽채류 중 배추, 양배추 다음으로 많이 생산되는 작물이다. 안토시아닌(Anthocyanins)은 열매, 꽃, 줄기, 잎 등 식물계에 널리 분포되어 있는 페놀 화합물 중 하나로 적색, 자색 등의 색을 나타내는 수용성 flavonoid계 색소이다. BSLs (Bitter sesquiterpene lactones)는 항암, 항균, 해열과 염증완화에 효과가 있는 것으로 알려져 있다. 본 연구는 농촌진흥청 농업유전자원센터에서 보유 중인 상추 66자원의 형태학적 특성 및 액체크로마토그래피(HPLC, UPLC)를 이용한 안토시아닌과 BSLs성분을 분석하여 함량이 높은 자원을 선발하고자 한다. 상추시료 0.05 g을 $MeOH/H_2O/AcAc$로 추출 한 후, UPLC를 사용하여 안토시아닌 함량을 분석하였으며, 상추시료 0.25 g을 100% MeOH로 추출 한 후 HPLC를 사용하여 BSLs 함량을 분석하였다. 연구 결과, 상추 유전자원의 안토시아닌 함량 범위는 0 mg/100 g에서 371.94 mg/100 g이고, BSLs성분 함량 범위는 $60.28{\mu}g/g\;DW$에서 $2821.92{\mu}g/g\;DW$ 이었다. 상추 66자원 중 안토시아닌함량이 200 mg/100 g이상인 자원은 IT217012, IT218395, IT231524, IT231525, IT260852이며, BSLs 함량이 $1700{\mu}g/g\;DW$이상인 자원은 IT231524, IT231525, IT231527, IT264971, IT271118이다. 두 성분의 함량이 모두 높은 자원 IT231524와 IT231525 이었다. 이 두자원의 형태적 특성은 초형이 잎상추로 잎이 넓은 타원형에 가장자리 결각이 강한 자주색이다. 따라서 본 연구는 다양한 상추 유전자원의 형태학적 특성 및 BSLs, 안토시아닌 성분이 높은 자원을 선발하여 육종소재로 활용하고자 한다.

Development of Potassium Concentration of Nutrient and Supply Method for Low Potassium Lettuce Production in a Closed-type Plant Factory System (완전제어형 식물공장에서 저칼륨 상추 생산을 위한 적정 칼륨 농도 조성 및 처리시기 개발)

  • Potassium in vegetables is known to have an adverse impact on a patient with chronic kidney desease. However, since vegetables also contain many other nutrient, consumption of vegetables by these patients is inevitable. The objective of this study was conducted to develop a fresh lettuce which contains low level of potassium for nephropathy in a closed-type plant factory system. Lettuce of "Charles" was used for experiment. The plants were cultivated in hydroponic system with a 16-h photoperiod at $15-21^{\circ}C$, 65% RH, $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$(LED W:R, 9:1) and $600-650mg{\cdot}L^{-1}$ $CO_2$ during 28 days. Nutrient solution which contains 1%, 5% and 10% potassium compared to conventional composition were supplied at 1 week and 2 weeks before harvest. The content of potassium and macro elements in leafy vegetables were analyzed by ICP emission spectroscopy after harvest. The potassium content in leaf of the 2 weeks before harvest treatment was significantly lower at than control. There were no significant differences between control and treatments in fresh weight and number of leaves. But there were differences among treatments. Considering the vegetable amounts consumed by nephropathy patients, the supply of nutrient which contain 1% and 5% potassium at 2 weeks before harvest was suitable for low potassium lettuce production. This study indicated that low potassium lettuce could be produced by developed nutrient composition and supply method.
