Screening for Antioxidative and Antimutagenic Capacities in 7 Common Vegetables Taken by Korean (한국인 상용채소 7종의 항산화능 및 항돌연변이능 검색)

  • This study was performed to investigate the antioxidative effect as the inhibition of malondialdehyde (MDA) and bovine serum albumin (BSA) conjugation reaction, inhibition of lipid peroxidation and the scavenging effect on 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical, and antimutagenic capacities as the Ames test in 7 common vegetables taken by Korean for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food. The water fractions of perilla leaves and sedum were most effective in the inhibition of MDA and BSA conjugation reaction showing 62.5% of inhibition rate among 7 vegetables. The inhibition rates of ethanol fractions of sedum and wild water dropwort on the lipid peroxidation were 67.1% and 61.5%, respectively. The ethanol fractions of crown daisy and wild water dropwort showed the most effective results among 7 vegetables in the DPPH radical scavenging capacities showing inhibition rate of 78.8% and 73.6%, respectively. The indirect and direct antimutagenic effects of ethanol extract of 7 vegetables were examined by Ames test using Salmonella typimurium TA98 and TA100. Inhibitory effects of wild water dropwort was superior to the other vegetables on the Ames test. These results suggest that common 7 vegetables taken by Korean are believed to be a possible antioxidative and antimutagenic capacities, although the results were different, more or less, according to the assay method and vegetables used.

  • Biological activities and anticarcinogenicity of Korean Pear peel were investigated, Electron donating activity and superoxide dismutase (SOD)-like activity of fraction II, III were up to 80% and 50-60% at 50 ppm, respectively, Inhibitory effects on xanthine oxidase were about 80% at 50 ppm, breast adenocarcinoma was about 60% at 2,000 ppm, higher III than II Inhibitory effect on prostate adenocarcinoma was about 23% at 500 ppm. In conclusion, Korean pear peel was expected to use as a functional material.

Anti-allergic Effects of Petasites japonicum (머위(Petasites japonicum) 추출물의 항알레르기 효과)

  • It is well known that the Petasites japonicum have been used for a long time in traditional medicine for the treatment of allergic diseases such as lacquer poisoning and asthma. Anti-allergic actions of Petasites japonicum extracts were asessed by testing their effects on the degranulation of mast cells. For this, hexosaminidase released (degranulation marker) from RBL-2H3 cells(mast cell line) was used. At the concentration of 300 $\mu\textrm{g}$/mL of the methanol, ethylacetate and hot water extract, the degranulation of RBL-2H3 cells were inhibited 83.33, 69.75 and 35.4%, respectively. These results suggest that the Petasites japonicum could be provide a effective resource for the control of allergic diseases.

  • The objective of this study was to evaluate the bifidogenic effect and a-glucosidase inhibitory effect of artificially cultured Phellinus linteus. The water extract of P. linteus promoted the growth of Bifidobacterium breve as well as the decrease of final pH in the media culturing intestinal bacteria. The growth of lactic acid bacteria inhibited effectively the bacterial enzymes, ${\beta}-glucosidase,\;{\beta}-glucuronidase$ and tryptophanase, of intestinal bactetria. The water extract of P. linteus inhibited maltase, sucrase and ${\alpha}-amylase$ of rat intestine.

Insecticidal activity of thiodicarb on lepidopterous pests (나방류에 대한 thiodicarb의 살충활성)

  • A series of experiments was conducted to determine the toxicities of thiodicarb on the six lepidopterous pests (Pseudaletia separata, Plutella xylostella, Palpita indica, Spodoptera exigua, Helicoverpa assulta, Spodoptera litura) and to elucidate factors insecticidal effects mechanism of thiodicarb. Thiodicarb was very effective against six lepidopterous young larvae, but less effective to the old larvae and it acted slowly. Thiodicarb inhibited acetylcholinesterase and glutathione S-transferase activities, but not inhibit esterase activity.

Effect of Nigella sativa Oil on Melanogenesis (니겔라 사티바 오일의 미백 효능에 관한 연구)

  • In order to investigate the potential of Nigella sativa (N. sativa) oil as an active ingredient for whitening cosmetics, we prepared N. sativa oil. We measured its inhibitory effects on mushroom tyrosinase activity, cellular tyrosinase activity, and melanin synthesis inhibitory activity in B16 melanoma cells. N. sativa oil and its components showed inhibitory activity against mushroom tyrosinase and melanin synthesis. In a melanin synthesis inhibition assay using mouse B16-F10 melanoma cell, it reduced melanin production up to 86 % at a concentration of 10 mg/mL without cytotoxicity. In the study on the melanogenic protein expressions by using RT-PCR and Western blot, N. sativa oil and its components inhibited expression of tyrosinase protein, which is a well-known key protein on melanogenesis, and tyrosinase expression was gradually decreased in a dose-dependent manner. Therefore, this result suggests that N. sativa oil could be used as an active ingredient for whitening cosmetics.

  • The effects of sperific inhibitors of $3\beta-hydroxy-\Delta^5-steroid\;dehydrogenase$ $(3\beta-HSD;\;an\;enzyme\;catalyzing\;conversion\;of \Delta^5\;steroids\;to\;\Delta^4 steroids),$ cyanoketone and trilostane, on $^3H-pregnenolone$ metabolism in isolated ovarian thecal layers have been investigated in vitro. At all doses of cyanoketone $(10^{-5}\;and\;10^{-4}\;M)$ and trilostane $(10^{-5}\;and\;10^{-4}\;M)$ $(3\beta-HSD$ enzyme activity that transforms pregnenolone to $17\alpha-hydroxyprogesterone$ was inhibited in the thecal layers. Trilostane appeared to be more efficient than cyanoketone. Trilostane at doses of $10^{-8},\;10^{-7},\;10^{-6},\;and\;10^{-5},\;M/ml$ caused a dose-response inhibition of $\Delta^4$ steroids accumulation in the medium from pregnenolone, but not completely blocked the conversion of $\Delta^5\;to\;\Delta^4$ steroids.

Glucosyltransferase Inhibition of Streptococcus mutans by Dioscorea batatas and Prunella vulgaris extract (마와 꿀풀 추출물에 의한 Streptococcus mutans의 산 생성 및 Glucosyltransferase 저해효과)

  • Streptococcus mutans is the major causative factor in dental caries and has been known to induce dental caries by the process of initial attachment proliferation and acid production. The pH of Streptococcus mutans(S. mutans) media and glucosyltransferase(GTase) activity were determined to evaluate the anticariogenic activity of Dioscorea batatas and Prunella vulgaris hexane fraction. In the experiment of hexane fraction of Dioscorea batatas, the relative growth ratio(RGR) against Streptococcus mutans (S. mutans) was determined as 93% in concentration of 0.125 mg/ml, 100% in 0.25 mg/ml. The hexane fraction of Prunella vulgaris revealed relative growth ratio(RGR) against Streptococcus mutans (S. mutans) as 86% in concentration of 0.25 mg/ml, 100% in 0.5 mg/ml. The relative growth ratio(RGR) against of hexane fraction were determined as 92% in concentration of 0.25 mg/ml, 100% in 0.5 mg/ml. The acidities were pH 7.2 on Dioscorea batatas and 7.0 on Prunella vulgaris in 2.0 mg/ml in contrast to pH 5.6 on Dioscorea batatas and 5.2 on Prunella vulgaris in control. The inhibitory effect to activity revealed 35% on Dioscorea batatas and 25% on Prunella vulgaris in 2.0 mg/ml. The inhibitory effect of Dioscorea batatas was more potent than Prunella vulgaris.

  • Enzymatic cellulose hydrolysis depends on the several factors such as the structural features (CrI, particle size and surface area, etc.), the nature of cellulase enzyme system, the inhibitory effects of products, and enzyme deactivation. At the presence of products on the initial hydro- lysis rate of cellulose, cellobiose has more severe inhibitory effect than glucose. Othewise, the inhibition effect of products for adsorbed enzyme is related to the glucose and cellobiose conentration hyperbolically. Enzyme deactivation of FPA and ${\beta}-glucosidase$ were expressed by exponential decay profile.

  • Antioxidant activity of monascus pigment of Monascus purpureus P-57 mutant was studied. Methanol extract from monascus pigment was separated into five organic solvent fractions; hexane, chloroform, ethyl acetate, butanol and water fractions. Hexane fraction showed the highest free radical scavenging effect on 1,l-diphenyl-2-picryl hydrazyl(DPPH), and the strongest inhibitory effect against xanthine oxidase, followed by chloroform fraction. But butanol and water fraction did not show inhibitory effect against the enzyme. Lineweaver-Burk plot showed that hexane fraction inhibited xanthine oxidase by non-competitive mode.
