Isolation of Superoxide Dismutase cDNAS from an Weedy Rice Variety and Transformation of a Cultivated Rice Variety (잡초성벼의 superoxide dismutase cDNA cloning과 재배벼로의 형질전환)

  • Two different cDNA clones for superoxide dismutase (SOD) were isolated from an weedy rice variety (Oryza sativa, cv. Bhutan14Ad) and were introduced into a cultivated rice variety (Oryza sativa, cv. Nakdong) in order to develop the environmental stress-resistant rice plants. Sequence analysis of the cloned cDNAS indicated that the deduced amino acid sequence of SOD-A is 88.4% identical to that of SOD-B. Furthermore, the nucleotide sequence of SOD-A is 99.3% identical to that of a Cu/Zn SOD gene of Oryza sativa (GenBank accession No. L36320). The nueleotide sequence of SOD-B was identical to that of the previously published SOD gene (Accession No. D01000). A cultivated rice variety, Nakdong-byeo, was transformed with chimeric SOD genes containing a actin promoter of rice and pin2 terminator using a particle bombardment technique. Transformed calli were selected on an selection medium containing phosphinothricin (PPT). Transgenic rice plants were regenerated from the PPT-resistant calli. PCR analysis with genomic DNAs from transgenic plants revealed that transgenes are introduced into rice genome.

Herbicidal Activity of New Rice Herbicide Pyrimisulfan+Mefenacet SC against Sulfonlurea-Resistant Sea Club Rush (Scirpus planiculmis) (설포닐우레아계 제초제 저항성 새섬매자기에 대한 신규 제초제 Pyrimisulfan+Mefenacet SC의 약제방제 효과)

  • This study was conducted to investigate the herbicidal activity against Scirpus planiculmis (SCPPL) which has been reported as resistant biotype on sulfonylurea (SU) herbicides at the west coast reclaimed area first and proliferated continuously at Hwasung and Pyeongtaek in Gyeonggi province, and some Chungnam province. In order to investigate resistance breakability against SCPPL, new rice herbicides containing tefuryltrione [p-hydrophenyl pyruvate dioxygenase (HPPD) inhibitor], pyrimisulfan and triafamone[actolactate synthethase (ALS) inhibitor] were tested. In both lab and field experiments, tefuryltrione GR, 4-HPPD inhibitor showed growth retardation with a bleaching on the young leaves of SCPPL at early time after treatment. However, it restored and regerminated with insufficient control effect. On the other hand, pyrimisulfan+mefenacet SC, triafamone+tefuryltrione GR, and ALS inhibitor showed excellent control effect on SU-resistant SCPPL with growth retardation and necrosis. In particular, pyrimisulfan+mefenacet SC showed excellent controlling effect on SU-resistant SCPPL in regional field experiments.

Herbicidal efficacy of flucetosulfuron+pyrazosulfuron-ethyl in controlling perennial sedges and sulfonylurea resistant weeds (Flucetosulfuron+pyrazosulfuron-ethyl 합제의 다년생 사초과 및 sulfonylurea계 저항성 잡초 방제효과)

  • This study was conducted to evaluate the herbicidal efficacy of flucetosulfuron+pyrazosulfuron-ethyl against Eleocharis kuroguwai and Scirpus planiculmis and to investigate tank-mix of flucetosulfuron + pyrazosulfuron-ethyl with bentazone and 2,4-D to control sulfonylurea resistant Monochoria vaginalis and Scirpus juncoides. In controlling E. kuroguwai, flucetosulfuron + pyrazosulfuron-ethyl showed 75% control, lower than that (95%) of bentazone + MCPA, at 29 DAA (days after application), while at 60 DAA it showed 90%, greater than that of (78%) of bentazone + MCPA, with greater control than penoxsulam at all times. In case of Scirpus planiculmis control, flucetosulfuron + pyrazosulfuron-ethyl showed lower activity than bentazone + MCPA but greater than penoxsulam. Flucetosulfuron + pyrazosulfuron-ethyl showed consistently high activities against Echinochloa crus-galli and E. kuroguwai regardless of soil flooding condition, while penoxsulam and bentazone + cyhalofop showed significantly lower activity in 5 cm flooding condition than 0 cm flooding condition. Flucetosulfuron + pyrazosulfuron-ethyl did not control sulfonylurea resistant Monochoria vaginalis and Scirpus juncoides, while its tank-mix with bentazone or 2,4-D at 2/3 or 1/2-folds of their recommended rates, respectively, provided > 90% control.

Resistance to ACCase Inhibitor Cyhalofop-butyl in Echinochloa oryzicola Collected in Gyeongsangnam-do Province of Korea (ACCase 저해 제초제 cyhalofop-butyl에 대한 경남지방 수집종 피의 저항성)

  • Repeated use of ACCase inhibiting herbicides for a long time has resulted in increases of resistant Echinochloa oryzicola populations in paddy fields in middle west area of Korea. This study aims to investigate current status of herbicide resistant E. oryzicola in Gyeongsangnam-do, in which there is less information about herbicide resistance. For resistance frequency and dose-response study, seeds from 100 individual plants of E. oryzicola in Gyeongsangnam-do were collected and tested with cyhalofop-butyl. Seven percent of plants from Gyeongsangnam-do was resistant at a recommended rate of cyhalofop-butyl. $GR_{50}$ values (herbicide rates required to reduce plant growth 50%) for one representative resistant populations and five susceptible populations were $738g\;a.i.\;ha^{-1}$ and 66-234 (average 147)$g\;a.i.\;ha^{-1}$, respectively, indicating average 5 times difference in resistance. Although lower rate of frequency of herbicide resistance in Gyeongsangnam-do than in Jeollabuk-do, increases of herbicide resistance are expected in this area because of increases of direct seeded rice fields and increases of dependence on a specific herbicide. Therefore, it is necessary to monitor herbicide resistance regularly and conduct integrated herbicide resistance management in this area.

  • This paper reviews current status of weed science in Japanese regional agricultural systems based on the 54th Annual Meeting of the Weed Science Society of Japan. About 300 researchers from 5 countries including Korea participated in the Conference and presented 100 papers in research areas. This congress has an purpose to discuss new troubles, findings and results of weed science. Weed science faces big challenges such as increase in herbicide-resistant weeds, gene-flow from transgenic crops, and invasive weeds. Major research topics were invasive weeds and their ecology, allelopathy, weed management in paddy field, weed management in field crops, and herbicide resistance. Weed control and herbicide resistance management in paddy field were a main object of research. To prevent the increase of problematic weeds and to overcome food crisis, the importance of weed-related researches has been raised. Therefore it is expected that various weed management systems and control of herbicide resistant weeds should be studied continuously in the weed science.

The Weed Flora of Korean Mulberry Fields (뽕밭에서 발생하는 잡초 양상)

  • The weed flora in the mulberry fields were investigated in Suwon, Jeonju, and Buan in May, July, and September of 2014. The objectives of this study were to use the survey data for establishing weed control methods and to bring awareness of possible problematic weeds in the Korean mulberry fields. The survey was conducted in 53 regions, covering approximately $145,925m^2$. Altogether 153 weed species of 37 families were identified, of which 68 were annual, 39 species were biennial and 46 were perennial. The dominance was the highest with Digitaria ciliaris followed by Erigeron annuus, Chenopodium album, Echinochloa crus-galli var.crus-galli, Acalypha australis, Commelina communis etc. Exotic weeds presented 44 species with 28.8% of a total presence, of which Erigeron annuus was the highest, followed by Chenopodium album, Phytolacca americana, Conyza canadensis, Oxalis corymbosa etc. Especially, we should aware Senecio vulgaris, not controlled with glufosinate ammonium SL in the Korean mulberry fields because it was known as atrazine resistance in US, Canada, Germany etc. In the PCA plot, weeds presented in the mulberry fields were divided into two groups, Eclipta prostrata community and Stellaria aquatic community and weed flora of Suwon and Buan were different due to those only presented in Suwon.

Expression Site of Protoporphyrinogen Oxidase Influences on Herbicide Resistance in Transgenic Rice (형질전환 벼에서 Protoporphyrinogen Oxidase의 발현 위치가 제초제 저항성에 미치는 영향)

  • The effect of Protox expression site on herbicidal resistance was investigated in wild-type and transgenic rice plants imposed by peroxidizing herbicide oxyfluorfen. The transgenic rice systems involved the plastidal expression of Arabidopsis protoporphyrinogen oxidase (Protox; AP line) and the dual expression of Myxococcus xanthus Protox in chloroplasts and mitochondria (TTS line). The oxyfluorfen-treated TTS4 line showed the lower levels of cellular leakage and malonyldialdehyde and the sustained capacity of 5-aminolevulinic acid synthesis, compared to the oxyfluorfen-treated AP and wild-type lines. During oxyfluorfen action, the TTS4 line had greater herbicide resistance than the AP1 line, indicating that the dual expression of M. xanthus Protox in chloroplasts and mitochondria prevented the accumulation of photodynamic protoporphyrin IX more effectively than the expression of Arabidopsis Protox only in chloroplasts. These results suggest that the ectopic expression of Protox in mitochondria greatly contributes to the herbicidal resistance in rice plants.

Control and Occurrence of Herbicide Resistance Echinochloa oryzicola in Rice Paddy Field of Honam Area (호남지역에서 제초제 저항성 강피의 발생과 방제)

  • This study was carried out to investigate the ACCase inhibiting herbicide resistant Echinochloa oryzicola collected in a Honam rice field and to investigate the control of E. oryzicola in directly seeded paddy rice field. The study result showed that about 44% of the 67 biotypes collected in the rice field in Honam region were resistant to ACCase-inhibiting herbicides. The efficacy of several chemical herbicides on these resistant E. oryzicola biotypes was evaluated according to their application timings. Among herbicides applied 4 days before seeding, pretilachlor, butachlor, oxadiazon, oxadiargyl, oxadiazon+pretilachlor, and fentrazamide+oxadiargyl, provided 95% or higher control of the resistant E. oryzicola biotypes. Among the herbicides applied 10 days after seeding, bromobutide+fentrazamide+imazosulfuron, benzobicyclon+fentrazamide+imazosulfuron, pyrazosulfuron-ethyl+thiobencarb, fentrazamide+imazosulfuron, bromobutide+imazosulfuron+mefenacet and bromobutide+imazosulfuron+pyraclonil provided ${\geq}95%$ control of the weeds. Bromobutide+imazosulfuron+mefenacet and bromobutide+pyrazosulfuron-ethyl+pyriminobac-methyl, applied 15 days after seeding, provided ${\geq}95$ and 90% control of the weeds. Foliar application of cyhalofop-butyl+propanil at 30 days after seeding provided ${\geq}90%$ control of the weeds. In addition, oxaziclomefone provided ${\geq}90%$ control for 40 days after soil application. These results show that E. oryzicola biotypes resistant to ACCase-inhibiting herbicides could be effectively controlled in directly seeded rice in submerged paddy fields using single or tank-mixed chemical herbicides currently available on the market.

  • China is one of most important countries for production and consumption of herbicide. Although the chemical weeding technology promoted the development of agricultural modernization in Chinese, it led to the negative effects to agriculture. In particular, the weeds resistant herbicide in paddy field had been serious challenge for safe production of rice. The chemical control technology for weed resistant herbicide with effective, low cost and safety characteristic will be key problem being solved in futhure.
