  • The identification of species, hygroscopic property, and ability of ethylene gas absorption of 23 ancient charcoals excavated from wooden coffin burials and roof-tile kilns of Chudong-ri cultural site were investigated. All of the 12 charcoals excavated from wooden coffin burials were broad-leaved trees. Among the total 12 samples, 9 samples were Lepidobalanus and others were Celtis spp.. On the other hand, other 11 charcoals from roof-tile kilns were needle-leaved tree, Pinus spp.(hard pine). The broad-leaved tree charcoals from wooden coffin burials showed a higher moisture absorption capacity than needle-leaved tree charcoals from roof-tile kilns. The ethylene gas absorption was greater in the Lepidobalanus charcoal than that of Celtis spp. and Pinus spp. (hard pine) charcoal. The broad-leaved tree charcoal having high absorption ability of substances was due to a large microporous and specific surface area. Therefore, it was estimated that broad-leaved tree charcoals were filled in order to make favorable condition in tomb. The wood quality of pine is soft and easy to burn because of low specific gravity, as well as high calorific value by resin in wood. We could assume that the pine wood was used as fuel for roof-tile kilns because of easy control of heating and thermal power.

  • Taraxacum platycarpum has been used as a medicinal plant. We investigated optimal condition for efficient plant regeneration through adventitious shoot formation on medium with various kinds of growth regulators. Adventitious shoot formation was achieved when cytokinin was used alone. Shoot formation was higher on MS medium containing 2 mg/L BAP compared to that with 2 mg/L kinetin and 2 mg/L 2-ip. Among root, hypocotyl and cotyledon, roots were the best explant for the adventitious shoot induction. Adventitious shoot formation from roots declined markedly by the combination of both 0.1 mg/L NAA and 2 mg/L BAP, while shoot formation from cotyledons was stimulated by the same combination. Root formation from the regenerated shoots was achieved on 1/3MS medium containing 0.2 mg/L NAA. Regenerated plantlets was acclimatized and transplanted to the soil, showing 100% survival.

  • In our previous study, a new herbal preparation (HemoHIM) was developed as a functional food for the radioprotection and immunomodulatory agents. In order elucidate the mechanism involved, we examined the effect of HemoHIM on the compound 48/80-induced histamine release, and on the phorbol 12-myristate 13-acetate (PMA)/calcium ionophore (A23187)-induced inflammatory cytokine secretion in HMC-1. The cell culture supernatants were harvested, and the cytokines (IL-4, IL-6, IL-8, TNF-α, GM-CSF) in the supernatants were measured by enzyme-linked immunosorbent assay. The total RNA of the cells was extracted, and the cytokines or c-kit/tryptase/FcεRI's messenger RNA expressions were examined using reverse transcriptase polymerase chain reaction. Under low concentrations, HemoHIM inhibited histamine release in HMC-1 stimulated compound 48/80. Furthermore HemoHIM inhibited PMA/A23187-induced inflammatory cytokines' secreation or mRNA expression in a dose-dependent manner. But IL-8 secretion was not inhibited by low concentrayion of HemoHIM, respectively. The mRNA expression of c-kit and FcεRI were also inhibited in a dose-dependent manner. Tryptase mRNA expression was only inhibited by low concentration of HemoHIM. These results indicated that HemoHIM might be an useful agent for protection against allergy as well as immune modulation, especially since it is a relatively nontoxic natural product.

  • Heat shock proteins (HSPs) are induced in response to various physiological or environmental stressors. However, the transcriptional activation of HSPs is regulated by a family of heat shock factors (HSFs). Fish models provide an ideal system for examining the biochemical and molecular mechanisms of adaptation to various temperatures and water environments. In this study, we examined the pattern differentials of heat shock factor 1 (HSF1) and expression of heat shock protein 70 (HSP70) in response to thermal stress in goldfish and mouse hepatocyte cultures by immune-blot analysis. Goldfish HSF1 (gfHSF1) changed from a monomer to a trimer at 33C and showed slightly at 37C, whereas mouse HSF1 (mHSF1) did so at 42C. This experiment showed similar results to a previous study, indicating that gfHSF1 and mHSF1 play different temperature in the stress response. We also examined the activation conditions of the purified recombinant proteins in human HSF1 (hmHSF1) and gfHSF1 using CD spectroscopy and immune-blot analysis. The purified recombinant HSF1s were treated from 25C to 42C. Structural changes were observed in hmHSF1 and gfHSF1 according to the heat-treatment conditions. These results revealed that both mammal HSF1 (human and mouse HSF1) and fish HSF1 exhibited temperature-dependent changes; however, their optimal activation temperatures differed.

  • Li-Mn spinel (LiMn2O4) is prepared by a hydrothermal process with K-Birnessite (KMnO4yH2O) as a precursor. The K-Birnessite obtained via a hydrothermal process with potassium permanganate [KMnO4] and urea [CO(NH2)2] as starting materials are converted to Li-Mn spinel nanoparticles reacting with LiOH. The molar ratio of LiOH/K-Birnessite is adjusted in order to find the effect of the ratio on the structural, morphological and electrochemical performances of the Li-Mn spinel. X-ray diffraction (XRD) and field emission scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy (EDS), and thermogravimetry (TG) are used to investigate the crystal structure and morphology of the samples. Galvanostatic charge and discharge are carried out to measure the capacity and rate capability of the Li-Mn spinel. The capacity shows a maximum value of 117mAhg1 when the molar ratio of LiOH/K-Birnessite is 0.8 and decreases with the increase of the ratio. However the rate capability is improved with the increase of the ratio due to the reduction of the particle size.

  • Euryale ferox is only annual hydrophyte among endangered plants in Korea and its seed is high valuable medicinal material. This studies were carried to find out key factors that made E. ferox endanger in Korea and to apply the results for effective management of the natural wetlands inhabited by E. ferox. We investigated the distributed sites reported by previous studies and classified the distribution-affected factors into three categories, eutrophication(hypothesis 1), soil accumulation(hypothesis 2) and increasing of water level(hypothesis 3) in the habitat. These three hypotheses were tested in the field and explained the results with the morphological characteristics of seed germination stages of E. ferox. The geographical distribution ranges of E. ferox diminished at a rate of 1 latitude with about 10 natural wetlands for last 30 years. Water eutrophication in the wetland had a positive effect on the establishment of E. ferox. But higher in soil accumulation and water depth, then lower in the establishment rate of E. ferox. These results indicate that water depth in the wetland must be below 50cm during the germination season, especially in May and emergent hydrophytes filter zone is need to protect soil accumulation from soil erosion within its watershed for the effective management of wetland with E. ferox.

  • Transgenic mice were generated by microinjecting a plasmid DNA containing the SV40 (simian virus 40) large T antigen (Tag) gene fused with mouse albumin promoter/enhancer sequences into fertilized one-cell mouse embryos. Among eleven founder transgenic animals, four developed hepatocellular carcinoma, two showed kidney cancer and one developed skin and brain tumors. Three stable transgenic lines, #1-2, #1-6 and #1-11 were established. Members of the lines #1-6 and #1-11 reproducibly developed liver tumors by 8 to 10 weeks of age but did not exhibit any phenotypic changes in other tissues. Histological changes loading to liver tumor formation occurred with predictable kinetics and could be classified into three distinct stages; (a) newborn to 3 weeks of age, characterized by hyperplastic hepatocytes with reduced amounts of cytoplasm without any nuclear alterations, (b) between 4 to 8 weeks of age, characterized by diffuse liver cell dysplasia without observable tumor nodules, and (c) 9 weeks of age and thereafter, characterized by hepatocellular carcinomas in the background of extensive liver dysplasia. Metastasis to the lung from a liver carcinoma was observed in #1-11 founder animal. This transgenic mouse system displays similarities with human liver cancers in a number of aspects and provides a useful model for the study of molecular events involved in hepatocarcinogenesis.
