한국 재래닭의 산란형질에 대한 능력변이

  • The observed means and standard divisions of the major laying traits per generation in korean native chickens were as follow. The age at first egg, the egg weight at the first egg, the weight at 270days and the egg production to 270days were 147.1$\pm$1.9 days. 32.3$\pm$0.9 g, 49.4$\pm$0.7 g and 76.2$\pm$1.7 eggs. The regression coefficients were -0.75$\pm$0.01 days, 0.33$\pm$0.08 g, 0.19$\pm$0.05 g and 0.47$\pm$0.3 eggs respectively.

  • The observed means and standard divisions of the egg shape per generation in korean native chickens were as follow. The egg major axis, the egg miner axis and the egg shape index were 53.0$\pm$0.6~55.6$\pm$0.8 mm, 39.1$\pm$0.8~40.9$\pm$0.2 mm and 72.7$\pm$1.5~75.9$\pm$0.5 mm. The regression coefficients were -0.02~0.20 mm, 0.10~0.52 mm and 0.19~1.00 mm respectively.

  • The observed means and standard divisions of the egg texture per generation in korean native chickens were as follow. The egg shell strength, the egg shell thickness and the egg shell colour were 3.8$\pm$0.2~4.2$\pm$0.1 kg/$\textrm{cm}^2$, 356.4$\pm$4.9~368.8$\pm$6.5${\mu}{\textrm}{m}$ and 47.0$\pm$1.7~50.0$\pm$3.3%. The regression coefficients were -0.03~0.03 kg/$\textrm{cm}^2$, -0.84~4.40${\mu}{\textrm}{m}$ and -0.88~-0.36% respectively.

  • The significant strain effects were investigated In all the major economic traits in Korean native chicken. The highest Least Square(LS) means of the body weights at 150 and 270days of age were observed in Black strain as 1, 594.38 and 1, 911.57 g. The earliest LS mean of the ages at first egg was 146.88 days in White strain. The heaviest LS means of egg weights at the first egg and 270 days of age were observed in Red brown strain as 33.20 and 50.74 9. The LS mean of the largest number of egg production was 79.50 eggs in Yellow strain.

  • This study was conducted to obtain for genetic information with 11, 538 progenies of 5 strains in Korean native chicken. The heritability estimates of the body weights at 150 and 270days of age were 0.25~0.61 and 0.32~0.59 in 5 strains, the heritabilities of the body weight at 270 days were relatively higher than that of the body weight at 150 days. The medium or low heritability estimates were observed in the ages at first egg as 0.15 ~0.31, and heritability estimates of the egg weights at the first egg and 270 days of age were 0.06~0.09 and 0.24~0.41. Medium or low heritability estimates were observed for the number of egg production as 0.09 ~0.26, respectively.

  • The observed genetic gains per generation in Korean native chicken on the basis of the 7 generation data collected from 11,583 birds from 1995 to 2001 at National Livestock Research Institute, Korea. Results obtained were summarized as follows : The body weight at 150 days, -5.292 ∼ 3.682 g ; for the body weight at 270 days, -6.184 ∼ 4.591 g ; for the age at first egg, -0.399 ∼ -0.102 ; for the egg weight at the first egg, -0.026 ∼ 0.016 ; for the e99 weight at 270 days, -0.039 ∼ 0.068 g ; for the egg production at 270 days, 0.222 ∼ 0.587, respectively.

Identification of Differentially Expressed Genes in Four Different Growing Stages in Korea Native Chicken Liver (황갈색 재래닭의 간에서 성장 단계별 차등 발현 유전자 분석)

  • The chicken liver has been involved in various biological functions including detoxification, glycogen storage and plasma protein synthesis. The aim of this study was to investigate differentially expressed genes in chicken liver in four different growing stages. Using 10 arbitrary Annealing Control Primers (ACPs), five differentially expressed genes have been identified. Based on the Basic Local Alignment Search Tool (BLAST) search results, three of them were matched with previously known genes, and the other two were matched with unknown EST sequence and a hypothetical protein, respectively. In order to confirm the expression results, quantitative real-time PCR was also performed. The high similarities between the expression data using arbitrary ACPs and quantitative real-time PCR indicate that the identified genes are the real differentially expressed genes in different growing stages. The genes identified in this study can be used as valuable biomarkers in chicken with further investigation of the functions.

Genetic Relationship between Populations and Analysis of Genetic Structure in the Korean Native Chicken and the Endemic Chicken Breeds (한국재래닭 및 토착화 품종간의 유연 관계 및 유전 특성 분석)

  • The purpose of this study was to assess the genetic variation and establish the relationship amongst breeds and strains using 7 chicken specific microsatellite markers. A total of 317 DNA samples from four Korean native chicken (KNC) strains (KR: Korean Native Red chicken strain, KY: Korean Native Yellow chicken strain, KL: Korean Native Black chicken strain, KO: Ogol chicken strain) and three introduced endemic chicken breeds (LE: Leghorn chicken breed, RI: Rhode Island Red chicken breed, CO: Cornish chicken breed). The size of microsatellite markers was decided using GeneMapper Software (v.4.0) after being analyzed using an ABI 3130 Genetic Analyzer. Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. The lowest distance (0.074) was observed between the KY and KL breeds and the highest distance (0.779) between the KL and LE breeds. The KNC strains (KR, KY, KL) have comparatively near genetic distance each other. On the other side, each individual was not ramified to different groups and were spread evenly in phylogenetic dendrogram about all the KNC of each strain populations. But the endemic breed populations (LE, RI, CO) were ramified to different groups. The microsatellite polymorphism data were shown to be useful for assessing the genetic relationship between Korean native strains and other foreign breeds.

한국 재래닭의 발생.발육단계별 telomere와 telomerase activity 분석

  • This study was carried out to analyze the amount of telomeres and telomerase activity of several chicken cells. Telomere quantity and telomerase activity were analyzed during organ development, growth and aging in embryonic and adults chicken. Analyzed cells were whole embryos and the cells from brain, heart, liver, kidney, lymphocytes and germinal tissues in Korean Native Chicken. The amount of telomeric DNA was analyzed by quantitative fluorescence in situ hybridization (Q-FISH) techniques using a chicken telomere repeat probe. Telomerase activity was performed by Telomeric Repeat Amplification Protocol (TRAP) assay. In results, telomerase activity was highly detectable in early embryonic cells, germinal cells and kidney cells. Whereas the cells from brain, heart, and liver had gradually down-regulated pattern of telomerase activity. Analyzing the telomere quantities on chicken cells, the amount of telomeric DNA of most chicken cells gradually decreased as growth. From these results, the amount of telomeric DNA was directly affected by telomerase activity. Consequently the telomere quantity and telomerase activity are closely relate to cell differentiation and tissue specificity during developmental and growing stages.
