결핵성 림프절염의 진단를 위한 세침흡인 세포검사 및 중합효소연쇄 반응과 효소면역법을 이용한 Mycobacterium tuberculosis의 검출 (Polymerase Chain Reaction Detection of Mycobacterium tuberculosis and Fine Needle Aspiration Cytology for the Diagnosis of Tuberculous Lymphadenitis)

  • Tuberculous lymphadenitis is not uncommon in Korea. Therefore, an inexpensive, safe and rapid method is needed to diagnose the tuberculous lymphadenitis. Flne needle aspiration cytology Is a good method for this purpose, but has several limitations in the diagnosis of tuberculous lymphadenitis, especially when the presence of acid-fast bacilli is not proved. To evaluation the usefulness of the polymerase chain reaction with enzyme immunoassay technique in the detection of Mycobacterium tuberculosis (M. tuberculosis) In the cervical Iymph node asplrates, the authors performed fine needle aspiration cytology and M. tuberculosis PCR with enzyme immunoassay for mycobacterial DNA sequences from 15 cases of the fine needle aspirates. Cytomorphologically, the cases were categorized into three types: predominantly necrotic materials; typical epithelioid cell granulomas with or without slant cells and caseous necrosis; and non-tuberculous lesions, such as reactive lymphadenitis, abscess, metastatic carcinoma and malignant lymphoma. M. tuberculosis DNA was found in 8 of 15 cases by PCR with enzyme immunoassay. Negative findings on PCR were achieved in 7 cases, which revealed non-tuberculous tymphadenopathy. In conclusion, we suggest that M. tuberculosis PCR with enzyme immunoassay using the fine needle aspirates is a very useful tool for the diagnosis of tuberculous lymphadenitis.

삼자삼상탕(三子蔘相湯)이 알러지피부염 유발 NC/Nga 생쥐의 피부 병변 면역 관련 인자에 미치는 영향 (Effects of SJSBT on Various Immunological Factors in Skin Related to Pathogenesis of Allergic Dermatitis in DNCB Treated NC/Nga Mice)

  • This study was investigated the anti-allergic effect of SJSBT on DNCB induced atopic dermatitis in NC/Nga mice. We summerized as the follow. SJSBT significantly decreased the clinical manifestations of atopic dermatitis including itching, dryness, edema, hemorrhage, and lichenification in dose dependent manner. SJSBT markedly suppressed invasion and edema of leukocytes and mast cell in dorsal skin and ear tissue. SJSBT significantly reduced the number of CD11b+/Gr-1 cell compared with positive control, but it had no affect the number of CD3+ and CCR3+/CD3+ cell. SJSBT markedly suppressed the expression of cytokine and chemokine such as IL-6, $TNF-{\alpha}$, eotaxin and CCR3 compared with positive control group. SJSBT significantly decreased the invasion of CD4+ and CCR3+ cell in ear and dorsal skin tissue compared with positive control group by using immunohistochemical staining. Taken together, these findings suggested that SJSBT has an anti-allergic activity and this might be useful for the clinical application to treat allergic diseases such as atopic dermatitis.

무비강장주의 면역증진 효과 (Immune-enhancing Effect of Mubigangjang-Ju)

  • Mubigangjang-Ju (MGJ) is a traditional wine, fermented extract of Cynanchum wilfordii, Angelicae gigantis and Epimedium koreanum etc. In the present study, we comparatively investigated the immune-enhancing effect of fermented extract (MGJ) and water extract (WE). Forced swimming test (FST) was performed as a model of activity test in mice and measured blood urea nitrogen (BUN), aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactic dehydrogenase (LDH), glucose (Glc) and total protein (TP) in the serum. Each extracts were orally administered into mice, 10 ㎖/kg, once per day for 7 days using a feeding atraumatic needle. After 3 days, on FST, the immobility time was decreased in the MGJ-fed group (133.7±18.6 s) in comparison with the saline-fed group (155.8±16.6 s). After 7 days, the immobility time was significantly decreased in the MGJ-fed group (105.3±12.7 s) in comparison with the saline-fed group (171.3±8.1 s). In addition, the content of AST was significantly decreased and the contents of BUN, ALT and LDH in the blood serum was also decreased. Whereas, the content of Glc tend to increase and TP level was not changed. However, WE had no effect on all experiments. The present results suggest that fermented extract was more effective than water extract and it may be useful for the immune-enhancing agent.

삭시톡신 분석을 위한 항체의 제조 및 항-삭시톡신 항혈청의 민감도 분석 (Production of antibodies for saxitoxin analysis and sensitivity analysis of anti-saxitoxin antiserum)

  • 해양미세조류 유래 독성물질에 대한 이해와 활용에 있어서 가장 중요하지만 간과되고 있는 부분은 독성물질을 검출할 수 있는 빠르고, 쉽고 경제적인 검출기술을 개발하는 것이다. 이 논문에서 우리는 삭시톡신(STX)에 대한 항체를 생산하였다. 헤모시아닌(mariculture keyhole limpet hemocyanin, mcKLH)과 오브알부민(ovalbumin, OVA)을 운반단백질로 사용하였다. 면역반응을 위해서 mcKLH-STX 결합체를 BALB/c 쥐에 복강주사하였다. 채혈 후 항-STX 항혈청을 분리하였다. 항혈청의 역가분석을 위하여 유리 STX와 OVA-STX로 코팅된 microtiter plate를 이용하여 간접 ELISA 실시하였다. 발색반응을 위한 이차항체로는 goat anti-mouse IgG-phosphatase conjugate가 사용되었다. 항-STX 항혈청은 OVA-STX와 유리 STX에 특이적으로 반응하였다. 항-STX 항혈청의 민감도는 매우 높았으며, STX를 위한 검출한계는 약 64.9 ng/kg이었다.

광주기 변화에 따른 마우스의 스트레스 호르몬, 면역기능 및 혈액학적 지표에 미치는 영향 (Influence of various photoperiods on stress hormone production, immune function, and hematological parameters in ICR mice)

  • In the present study, the effects of different photoperiods on stress, immunity, and hematological parameters in ICR mice were evaluated. Fifty male ICR mice 7 weeks old (body weight, $27.3{\pm}2.5g$) were divided into five groups: DP-0 (0/24-h light/dark cycle), DP-6 (6/18-h light/dark cycle), DP-12 (12/12-h light/dark cycle), DP-18 (18/6-h light/dark cycle), and DP-24 (24/0-h light/dark cycle). During the experimental period, no significant differences in body weight or feed intake were observed between the groups. Hematological analysis revealed that white blood cell, red blood cell, and hemoglobin values for the DP-0 group were significantly different compared to those of the other groups. After 28 days, no significant difference in serum cortisol concentration was observed among the groups, but serum cortisol levels increased in a light exposure-dependent manner. Total serum immunoglobulin G (IgG) concentrations of the DP-0 and PD-6 groups were significantly increased compared to those of the other groups (p < 0.05), and serum total IgG levels decreased in a light exposure-dependent manner. Results of the present study indicated that various photoperiods affect hematological parameters and total serum IgG levels in ICR mice while having no significant effects on body weight, feed intake, or cortisol levels.

마우스 염증성 장 질환 모델에서 G-CSF (Granuocyte Colony Stimulating Factor)에 의한 염증 완화 (Granulocyte Colony Stimulating Factor (G-CSF) Attenuates 2,4,6-Trinitrobenzene Sulfonic Acid (TNBS)-induced Colitis in Mice)

  • Background: Granulocyte colony stimulating factor (G-CSF) is known as a cytokine central to the hematopoiesis of blood cells and to modulate their cellular functions. Besides granulocytes and their precursors, monocytes/macrophages and endothelial cells are direct target cells of G-CSF action. G-CSF influences immune cells in an anti inflammatory way. Methods: To evaluate whether G-CSF has a potential for preventing or ameliorating diseases characterized by mucosal inflammation, we used a mouse model with trinitrobenzene sulfonic acid (TNBS)-induced inflammatory colitis. To the mice model G-CSF was administrated daily by intraperitoneal injection. Macroscopic evaluation and immunohistochemical analysis of colonic tissues were performed. Results: Re combinant human G-CSF significantly inhibited LPS-induced TNF-${\alpha}$ mRNA expression in THP-1 cells. As for in vivo relevance, G-CSF dramatically reduced the weight loss of mice, colonic damage, and mucosal ulceration that characterize TNBS colitis. Moreover, G-CSF suppressed the expression of tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and intercellular adhesion molecule-1 in TNBS colitis. Conclusion: Current results demonstrate that G-CSF may be an effective agent for the treatment of diseases characterized by mucosal inflammation.

매실 혼합물이 DSS로 유도된 염증성 장질환 동물모델의 면역조절에 미치는 활성 (Anti-Inflammatory Effects of Prunus mume Mixture in Colitis Induced by Dextran Sodium Sulfate)

  • This study was conducted to investigate the anti-inflammatory effects of Pruns mume, Schisandra chinensis, Chaenomeles sinensis-- Prunus mume mixtrue (PM) treatment on colitis induced in mice by dextran sodium sulfate (DSS) treatment. A total of 25 male BALB/c mice (average weight $20.7\;{\pm}\; 1.6 \;g$) were divided into 5 treatment groups and fed a commercial diet (A), PM administration (B), commercial diet + induced colitis by DSS (C), PM administration + induced colitis by DSS (D) and sulfasalazine + induced colitis by DSS (E). We found that PM treatment (D) and sulfasalazine (E) decreased the expression of $TNF-{\alpha}$ and COX-2 compared to the DSS-induced colitis group (C). The expression of IL-4, STAT6, $IFN-{\gamma}$, STAT1 was decreased in group D and group E compared to the colitis group (C), COX-2 and STAT1 were more decreased in group D. The serum IgE levels decreased in the PM treatment groups (C and D) compared to the non-PM treatment groups (A and B) although there was no significant difference between the PM treatment groups. It is notable that a therapeutic application of the PM extracts ameliorated DSS-induced colitis in mice.

  • 설사로 폐사한 자돈의 장 절편을 이용하여 돼지 유행성 설사바이러스(PEDV), 돼지 전염성 위장염 바이러스(TGEV), 돼지 로타 바이러스(PRV)에 대한 병인학적 검사를 형광항체반응을 통하여 조사하였던 바 돼지 유행성 설사 바이러스의 감염에 의한 자돈의 폐사를 확인하였다. 간접형광항체검사시 돼지 유행성 설사 바이러스에 대한 양성반응을 보인 가검재료를 이용 Vero세포에 연속 계대한 후 plaque assay를 통하여 크로닝된 돼지 유행성 설사 바이러스 KPEDV-9주를 작성하였다. 돼지 유행성 설사 바이러스에 대한 면역혈청과 바이러스가 분리된 농장에서 채취된 돼지 혈청을 이용 돼지 유행성 설사 바이러스에 대한 구조단백성분을 분석하였던 바 88K(M.W.), 74K, 70K, 58~54K, 54~46K, 44~40K 및 33~32K에 상당하는 단백성분을 검출할 수 있었다.

광섬유 소산장을 이용한 식중독균 신속검출용 형광면역센서 (Fiber-optic fluoroimmunosensor for foodborn pathogens using an optical evanescent field)

  • In this study, the fiber-optic fluoro-immunosensor was designed to detect foodborne pathogens. The fabricated system is composed of the multimode optical fiber on which antibodies are immobilized. Then, a sandwich immunoassay is applied to the fabricated the fiber-optic fluoro-immunosensor. In the "sandwich" binding format, a primary or "capture" antibody is immobilized on the core surface of the multimode optical fiber and a secondary or named as "tracer" antibody is added to the bulk solution. A tracer is labeled FITC (fluorescein isothiocyanate; ${\lambda}ex$=492 nm, ${\lambda}em$= 520 nm). Different concentrations of antigens are tested in different fibers. The detection limit of the fabricated system is 5.08×103 cfu/ml for Vibrio antigen and $0.1{\mu}g/ml$, $0.05{\mu}g/ml$ in non-labeled monolayer phosphate buffered saline (NMP), non-labeled monolayer carbonate bicarbonate buffer (NMC), respectively.

  • Objectives : The purpose of this study is to report clinical effect of oriental medical for a not improved ITP patient, who developed complication from DVT, after intravenous immunoglobulin treatment. Methods : The patient was treated using Saam acupuncture, herbal medication, moxibustion and physical treatment. And we measured of systemic symptoms and platelet counts. Results : After treatment, systemic symptoms and platelet counts were improved in case. Conclusions : Oriental medical treatment showed significant effect on ITP patient that not improved platelet counts after intravenous immunoglobulin treatment.
