In vitro Cytotoxicity Evaluation of Polydimethylsiloxane as a Biosensor Coating Material (바이오센서 코팅용 Polydimethylsiloxane의 생체외 세포독성 평가)

  • PDMS was selected for a coating material of implantable biosensors and the cytotoxicity of extracts released from a polymer was evaluated using ISO 10993-5, Biological evaluation of medical devices-Part 5: Tests for in vitro cytotoxicity. Organo-tin was used as a positive control and a medium without serum was used as a negative control. Materials extract were prepared by incubating specimens in RPMI medium without serum ($125{\mu}L/cm^2$) for 24 h, 1 week and 6 weeks at $38^{\circ}C$. The evaluation of cytotoxicity was performed by two different methods : 1) seeding cells with extracts at the beginning 2) incubating extracts with cell sheets already formed on the plate. Both cell morphology and MTT numerical data were shown for the confirmation of cytotoxicity and cell spreading on the surface of PDMS.

Combined Toxicity in the Brackish Water Flea Diaphanosoma celebensis Exposed to Benzo[a]pyrene and Mercury (벤조피렌과 수은에 대한 기수산 물벼룩 Diaphanosoma celebensis의 복합 독성 영향)

  • Polycyclic aromatic hydrocarbons (PAHs) are persistent and commonly detected in marine ecosystem. They coexist with a various contaminants including heavy metals in real environment, but most studies have been still focused on single effects of these contaminants to aquatic organisms. In this study, the single and combined effects of benzy[a]pyrene (B[a]P) and HgCl2 were investigated in the brackish water flea, Diaphanosoma celebensis. For combined effect study, three mixtures A (3:7), B (5:5), and C (7:3) of benzy[a]pyrene (B[a]P) and HgCl2 were determined by 48 h - LC50 values of single exposure. Combined effects of both chemicals were estimated by concentration addition (CA) model and independent action (IA) model. As results, 48 h - LC50 values of benzy[a]pyrene (B[a]P) and HgCl2 was 25.75 ㎍ l-1 and 3.6 ㎍ l-1, respectively. TU values of mixture A, B, and C were 1.06, 0.83, and 0.96, respectively, indicating additive effects of mixtures using CA model. This study will be helpful as basic data for understanding the combined effects of contaminants in marine environment.

Residue of the Organophosphorus Pesticides in the Coastal Environment on the Cheju island (제주도 연안 해양환경 중에서 유기인계 농약의 잔류)

  • To investigate the residue of organophosphorus pesticides such as Monocrotophos[Dimethyl-1-methyl-2-methyl carbamoylvinylphosphate]and EPN[O-ethyl-O-4-nitrophenyl phenylphosphonothioate]in the coastal environment of Cheju island, samples of sea organism, water and sediment were collected at the Sackdaldong and Daepodong near the Jungmoon golf course in July and Aug. 1997. The qualified detection limit of mono-crotophos and EPN by GC -FPD were 0.024 ng/g and 0.020 ng/g in the sea organism, respectively Neither Monocrotophos nor EPN was detected in seaweed cava (Ecklonia cava), Agar (Gelidium amansii), turban sell (Batillus cornutus) and sea urchin (Anthocidaris Crassispina). They were not detected in seawater and sediment, either. These results showed that the coastal area near the Jungmoon golf course in the Cheju island was not polluted by the organophosphorus pesticides such as Monocrotophos and EPN.

Herbicidal Effect of 5-Aminolevulinic Acid, a Biodegradable Photodynamic Substance (생분해성 광활성 물질 5-aminolevulinic acid의 제초활성)

  • Laboratory and greenhouse experiments were conducted to determine the herbicidal effect of two types of ${\delta}$-aminolevulinic acid (ALA), microbiologically-produced ALA (Bio-ALA) and synthetically produced ALA (Synthetic-ALA), on plant growth and chlorophyll content of Chinese cabbage. ALA effect on early plant growth was greatly concentration dependant, showing significant inhibition at higher concentrations. Both pre- and post-emergence application of ALA exhibited significant degree of photodynamic phytotoxicity. Older plants with many leaves were more tolerant to ALA than younger plants, showing less injury. No significant difference in herbicidal activity of two types of ALA, Bio-ALA and Synthetic-ALA, on plant height and chlorophyll content of Chinese cabbage was observed. However, residual biological activity and physico-chemical properties of Synthetic-ALA were more stable than those of Bio-ALA. Our results suggest that ALA had herbicidal potential with both pre- and post-emergence application, and that the chemical may be a valuable mean of eco-friendly weed control based on natural microbial substance.

Microbiological Assay for the Detection of Chloramphenicol in Meat and Milk (식육 및 우유 중 클로람페니콜의 미생물학적 검출법에 관한 연구)

  • Chloramphenicol (CAP) is a very effective broad-spectrum antibiotic which had been widely used in animal production. However, the drug is not approved in many countries for use in food-producing animals because of its potential toxicity and the possibility of residues in food products. In this study, a modified microbiological assay was developed for the sensitive detection of CAP residues in meat and milk. The method was characterized by the extraction of CAP with ethyl acetate, addition of $0.15\;\mu\textrm{g}$ oxytetracycline/ml in the phosphate buffer diluent (pH 6.0), a luteus ATCC 9341. The lowest levels of CAP detected in muscle tissues and milk were $0.025\;\mu\textrm{g}/ml\;and\;0.05\;\mu\textrm{g}/g$, respectively. Recovery rates free CAP from milk were 68.5%, from bovine muscle 65.1%, from swine muscle 63.8%, and from chicken muscle 59.4%, respectively, and the coefficients of variation were 1.8~15.1%. The results showed that the detection limits of CAP residues in animal products could significantly be improved by the modified microbiological assay than the conventional ones.

  • In order to analyze nutritional factors affecting in situ bioremediation of diesel degradation and cell viability were studied by varying nutritional conditions. In column experiments packed with diesel-contaminated soil, nitrogen was found to be the major limiting nutrient. When nitrogen was added to soil at four different levels of C : N (100 : 5, 100 : 10, 100 : 15, and 100 : 20 mg N/kg dry soil), the greatest simulation of microbial activity occurred at the lowest, rather than the highest nitrogen addition. However, no significant effects was observed when phosphorus and air were added. No matter how the incubation mode varied, less than 50% of the diesel was remained after 7 days of treatment, presumably because the residual hydrocarbons were adsorbed on soil particles, adsorption

  • Studies on the food-safety of pheasant and mallard, which belong to wild fowl as new meat resources. were carried out. The results were summarized as follows : 1. Food poisoning bacteria including Salmonella spp, was not detected from the inspections of small intestine, cecum, and rectum. 2. Parasite inspection tests on blood, feces, digestive organ, and thoracic organs were negative. 3. Antibiotic residues from the carcass muscle by simplified disk methods were not detected. 4. Seven different pesticide residue tests, such as DDT and BHT, on the muscle and liver were negative. 5. Four different kinds of toxic heavy metals such as Cd were much lower than the permissible concentration. Studies on the food safety tests and inspections from the pheasant and mallard were revealed that from taking this new food resources, the toxicities would be very low for the human health by the direct influences.

Resistance Mechanisms of Green Peach Aphid, Myzus persicae (Homoptera: Aphididae), to Imidacloprid (복숭아혹진딧물(Myzus persicae)의 imidacloprid에 대한 저항성 기작)

  • Resistance mechanisms in the green peach aphid (Myzus persicae) resistant to imidacloprid were investigated. Imidacloprid residues on the aphid integuments decreased slowly as time passed with no significant difference between the susceptible and resistant strains. Residue in the aphid body increased in both strains with time elapse, and was slightly more in the susceptible strain. A higher metabolic rate of imidacloprid in the resistant strain can be expected by the fact that more amount of imidacloprid were excreted in the resistant strain than in the susceptible one. The activity of AChE was higher 1.4 times in the resistant strain than in the susceptible one, and imidacloprid did not inhibit AChE at all in both strains. Piperonyl butoxide (PBO) and iprobenfos (IBP) synergized imidacloprid activity. The mixtures of imidacloprid and PBO (1 : 1 and 1 : 5) caused 69.4- and 250-fold increase of imidacloprid toxicity against the aphid. Insecticide toxicity of the mixtures of IBP and imidacloprid (1 : 1 and 1 : 5) was also increased 227 and 80.6 times. Esterase activity when $\alpha$-naphtyl butyrate and $\beta$-naphtyl acetate were used as substrates was higher in the resistance strain than in the susceptible one. This means that P450 monooxygenase and esterase are responsible for the resistance to imidacloprid in this aphid strain.

Acute Toxicity of Ozone on Survival and Physiological Conditions of Olive Flounder, Paralichthys Olivaceus (넙치(Paralichthys olivaceus)의 생존과 생리상태에 미치는 오존의 급성 독성)

  • Ozonization for rearing seawater in land-based culture system has recently been utilized for disinfection of pathogenic microorganisms and improvement of water quality. This study was conducted to examine the effects of total residual oxidants (TRO) in ozone-treated seawater on survival, blood parameters, osmolality and oxygen consumption, and gill tissue of the flounder. Paralichthys olivaceus. Experiments were carried out with the starved flounder of 12~19 cm in total length at $20^{\circ}C$. The 48-hr and 96-hr $LC_{50}$ for the flounder amounts to 26.4 ppb and 22.3 ppb, respectively. With increase of TRO concentration from 24 to 39 ppb, the values of hematocrit, hemoglobin concentration, red blood cell count and osmolality of the flounder with respect to exposure time were significantly elevated, however, the oxygen consumption rates decreased. In the case of the fish exposed to 13 ppb for 96 hrs, blood glucose increased with an elapse of exposure time, while survival rate was 100%. Death apparently resulted by massive destruction of gill lamellar epithelium, severe osmotic imbalance and the lack of oxygen uptake. The results of this experiment indicated that to protect aquaculture organisms, the ozone-treated seawater should not contain any residual oxidants, and that relatively long-term exposure to TRO of low concentration can impact on survival and physiological conditions of the flounder.

Organic Removal Efficiency and Toxicity Evaluation of Persistent Microorganism from Dye Wastewater Treatment using a Photooxidation system (광산화시스템을 이용한 염색폐수의 유기물 처리효율 및 잔류미생물의 독성 평가)

  • The removal efficiency of organic compounds and the toxicity evaluation of microorganism have been studied in dye wastewater treatment using $UV/TiO_2$ and $UV/H_2O_2$ photooxidation system. Sample waters tested in this work were raw dye wastewater and dye wastewater treated in $UV/TiO_2$ and $UV/H_2O_2$ photooxidation system respectively. Total organic carbon(TOC) removal rate was 50% in $UV/TiO_2$ process and 80% in $UV/H_2O_2$ process. It has been investigated with colony counting agar method and paper disk method whether the type of treatment process has affected the microorganism growth. In the raw wastewater, more than four types of microorganisms have survived. But, little of microorganisms were alive at TOC removal rate of 50% in $UV/TiO_2$ system. In contrast to that, two types of microorganisms were found at TOC removal rate of 80% in $UV/H_2O_2$ system.
