Anti-neuroinflammatory Effect of Teleogryllus emma Derived Teleogryllusine in LPS-stimulated BV-2 Microglia (BV-2 미세아교세포에서 왕귀뚜라미 유래 Teleogryllusine의 신경염증 억제 효과)

  • The suppression of neuroinflammatory responses in microglial cells, well known as the main immune cells in the central nervous system (CNS), are considered a key target for improving the progression of neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and Huntington's disease. Teleogryllus emma is widely consumed around the world for its broad-spectrum therapeutic effect. In a previous work, we performed transcriptome analysis on T. emma in order to obtain the diversity and activity of its antimicrobial peptides (AMPs). AMPs are found in a variety of species, from microorganisms to mammals. They have received much attention as candidates oftherapeutic drugs for the treatment of inflammation-associated diseases. In this study, we investigated the anti-neuroinflammatory effect of Teleogryllusine (VKWKRLNNNKVLQKIYFVKI-NH2) derived from T. emma on lipopolysaccharide (LPS) induced BV-2 microglia cells. Teleogryllusine significantly inhibited nitric oxide (NO) production without cytotoxicity, and reducing pro-inflammatory enzymes expression such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). In addition, Telegryllusine also inhibited the expression of pro-inflammatory cytokines such as interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-α) through down-regulation of the mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B (NF-κB) signaling pathway. These results suggest that T. emma-derived Teleogryllusine could be a good source of functional substances that prevent neuroinflammation and neurodegenerative diseases.

Radiotherapy for Locoregional Recurrent Cervix Cancer after Surgery (수술후 국소 재발된 자궁경부암의 방사선 치료)

  • Purpose: The role of radiotherapy in the management of patients with locoregional recurrent cervix cancer after radical surgery were retrospectively analyzed. Methods and materials: Twenty-eight patients treated with radiotherapy for locoregional recurrence after primary surgery for carcinoma of the cervix between 1989 and 1993 were analyzed. The median follow-up of survivors was 15 months (ranged 7-43 months). Eight patients had their disease confined to the vagina and 19 patients($68\%$) had pelvic mass as part of their locoregional recurrent disease. Within 24 months after the initial surgery, $82\%$ of recurrences manifested themselves. All patients had whole pelvic irradiation with or without intracavitary radiotherapy(ICR). Results: Complete response(CR) was achieved in 18 patients($54\%$). Five of eighteen patients($28\%$) with initial CR developed second locoregional recurrence. Response to radiotherapy correlated strongly with tumor volume, site of recurrence and total radiation dose. The overall 2 year survival rate was $43\%$ and the disease free survival was $31\%$. Survival rate was significantly influenced by the factors of interval from operation to recurrence, size and site of recurrent tumor, radiation dose, response of radiotherapy, lymph node status as initial presentation, The principal cause of death was lung metastasis($36\%$). Conclusion: Radiotherapy is an excellent modality for control of locoregional recurrent cervix cancer. To improve local control and survival rate, whole pelvic external radiotherapy in addition to ICR with more than 75.0Gy at the depth of 1.0cm from vaginal mucosa is needed and frequent follow up and early detection of recurrence is suggested as well.

  • Mutations in the cystathionine ${\beta}$-synthase (CBS) gene cause homocystinuria, the most frequent inherited disorder in sulfur metabolism. CBS is the unique enzyme using both heme and pyridoxal 5-phosphate (PLP) for activity. Among the reported 140 mutations, one of the most common disease-causing alterations in human CBS is G307S mutation. To investigate the pathogenic mechanism of G307S by spectroscopic methods, we engineered the full length and the truncated G247S mutation of yeast CBS that is corresponding mutation to human G307S. Yeast CBS does not contain heme and thus gives a merit to study the spectroscopic properties. The UV-visible spectra of the purified full length and the truncated G247S yeast CBSs showed the total absence of PLP in the protein. The absence of PLP in G247S mutation was also confirmed by the PLP-cyanide adduct formation experiment, which was conducted by the incubation of the purified enzyme with KCN. The adducts were detected using a circular dichroism (CD) and a spectrofluorimeter. Radio isotope activity assay of full length and truncated G247S proteins also gave no activity. Our yeast G247S mutation data suggested that G307S might make the distortion of the active site so that cofactor PLP and substrate can not fit inside the active site. Our yeast CBS study addressed the reason why the G307S mutation in human CBS makes the enzyme inactive that consequently leads to severe clinical phenotype.

  • Properties of enzymatic hydrolysates from ascidian tunic were assessed on supernatant ratio, solid yields and solid concentration. The concentartion of solid and yields in the extracts were increased as the enzyme concentration raised from $100\;{\mu}l\;to\;1000{\mu}l$ during the extraction period. The optima concentration and reaction time of each enzyme on digestion were $400\;{\mu}l$ 60 minutes, through treated with Duncan's multiple test. The percent of yields of solid, protein and carotenoids for 60 minutes extraction at $400\;{\mu}l$ were $32.32\%,\;1.34\%\;and\;74.60\;mg\%$, respectively, in Viscozyme systems. The extracts were composed with many kinds of carbohydrates such as arabinose, ribose, xylose, galactose, glucose, N-acetyl-D-galactosamine, and N-acetyl-D-glucosamine. Aspartic and glutamic arid were noted as predominant amino acids in all parts. Amino acid profiles of various ascidian tunic part were similiar to each other, but most of essential amino acids content of inter coat was higher than that of root and tunic (body). About sixty six fatty acids components were observed, and their distribution among neutral and polar lipids was compared. The main fatty acids were found to be 14:0, 16:0, 16:1n7, 18:0, 18:1n9, 18:1n7, 18:2n6, 20:5n3, and 22:6n3.

  • The performances of a parallel current air driven type foam separator were evaluated in the pilot-scale recirculating aquaculture system. The system was stocked with Nile tilapia (Oreochromis niloticus) at an initial rearing densities of $2\%$, $5\%$ and $7\%$ of water volume of rearing tank and reared for 15, 30, 35 days, respectively. The condensated volume of effluent foam was increasing with increased rearing density. As increasing rearing density from $2\%$ to $7\%$, the protein concentration in rearing tank was increased from 16.6 g/$m^3$ to 21,9 g/$m^3$ and the removal amount of protein through foam separator as increased from 0.99 g/day to 2.5 g/day. But protein concentration ratio in the foam was decreased from 3.2 to 1.9. Changes of the removal amount and the concentration ratios of total suspended solid (TSS) and chemical oxygen demand (COD_cr.) were similar to proteins. The highest concentration ratios of TSS and COD_cr. were 10.2 and 8.4 at 2$2\%$ of rearing density.

THE EFFECT OF C-FACTOR AND VOLUME ON MICROLEAKAGE OF COMPOSITE RESIN RESTORATIONS WITH ENAMEL MARGINS (법랑질 변연으로 이루어진 복합레진 수복물의 체적과 C-factor가 미세누출에 미치는 영향)

  • Competition will usually develop between the opposing walls as the restorative resin shrinks during polymerization. Magnitude of this phenomenon may be depended upon cavity configuration and volume. The purpose of this sturdy was to evaluate the effect of cavity configuration and volume on microleakage of composite resin restoration that has margins on the enamel site only. The labial enamel of forty bovine teeth was ground using a model trimmer to expose a flat enamel surface. Four groups with cylindrical cavities were defined, according to volume and configuration factor(Depth x Diameter / C-factor) - Group I : 1.5 mm ${\times}$ 2.0 mm / 4.0, Group II : 1.5 mm ${\times}$ 6.0 mm / 2.0, Group III : 2.Omm ${\times}$ 1.72 mm / 5.62, Group IV : 2.0 mm ${\times}$ 5.23 mm / 2.54. After treating with fifth-generation one-bottle adhesive - BC Plus$^{TM}$ (Vericom, AnYang, Korea), cavities were bulk flted with microhybrid composite resin - Denfill$^{TM}$ (Vericom). Teeth were stored in distilled water for one day at room temperature and were finished and polished with Sof-Lex system. Specimens were thermocycled 500 times between 5$^{\circ}$C and 55$^{\circ}$C for 30 second at each temperature. Teeth were isolated with two layers of nail varnish except the restoration surface and 1 mm surrounding margins. Electrical conductivity (${\mu}$A) was recorded in distilled water by electrochemical method. Microleakage scores were compared and analyzed using two-way ANOVA at 95% level. The results were as follows: 1. Small cavity volume showed lower microleakage score than large one, however, there was no statistically significant difference. 2. There was no relationship between cavity configuration and microleakage. Factors of cavity configuration and volume did not affect on microleakage of resin restorations with enamel margins only.

Distribution Patterns and Ecological Characters of Paulownia coreana and P. tomentosa in Busan Metropolitan City Using MaxEnt Model (MaxEnt 모형을 활용한 부산광역시 내 오동나무 및 참오동나무의 분포 경향과 생태적 특성)

  • Paulownia species has long been recognized in Korean traditional culture and the values of the species have been researched in various focuses. However, studies on distribution and ecological characteristics of the species are still needed. This study aimed to identify distribution trends and ecological characteristics of two Paulownia species in Busan metropolitan city using the MaxEnt model. The MaxEnt model was established based on the environmental factors such as positioning information of the Paulownia species, topography, climate and degree of anthropogenic disturbance potentiality (ADP), which was collected in the on-site research. The study verified that the accuracy of the model was appropriate as the AUC value of Paulownia coreana and P. tomentosa was 0.809, respectively. In terms of the distribution trends of the two Paulownia species in the research area depending on the distribution model, they were both mainly distributed in downtown where built-up area and bare ground were densely concentrated. The potential distribution area of the two species was identified as $137.4km^2$ for P. coreana and $135.0km^2$ for P. tomentosa. The distribution probability was high in Jung-gu, Dongrae-gu, Busanjin-gu and Yeonje-gu. As a result of the analysis on contribution of the environmental factors, it was turned out that the degree of anthropogenic disturbance potentiality (ADP) contributed to distribution of P. coreana and P. tomentosa by about 50%, and the contribution of the environmental factors had a positive correlation with the degree of ADP. The elevation had a negative correlation with both the two species, which was considered because the species must compete more with native species in natural habitats as the altitude above sea level rises. The research findings demonstrated numerically that the distribution of P.coreana and P. tomentosa depended on artificial activities, and indicated the relevance with the Korean traditional landscape. These findings are expected to provide meaningful information in using, preserving and restoring Paulownia species.

A Characteristics of Visual Narrative Expression in Garden Design - Focused on the Taehwagang Garden Show 2018 - (정원디자인에 나타난 시각적 서술의 표현특성 - 2018 태화강 정원박람회 작품을 대상으로 -)

  • Appreciating gardens in garden show has its meaning in appreciating concepts and ideas of artists, hidden inside of garden, as formative arts, as well as the beauty of the nature. This study is aimed to understand the intension of artists in visual expression through formative media in the gardens by assessing structure of visual narrative in the space with 20 artworks among the ones presented in 2018 Taehwagang Garden Show. The formative structure is delivered as contents and forms through formative media and formative language. Thus, for analysis on the artworks, the researcher assessed expressive characteristics of the media, through visual and space language, that forms the formative structure in the contest of narrative structure expressed in the gardens and findings of the analysis are as follows. First, for intertextuality obtained through media image, most of the artworks delivered message through 'figure image.' This means, the concept is delivered as 'affinity of actual objects' through the media and associated 'meaning and meaning action' are expected. Second, the characteristics of signs to show symbolism in the gardens were categorized into 'icon'. 'index' and 'symbol'. The results showed that most of the artworks expressed common characteristics between image and meaning, using 'icon' and 'symbol'. Third, as space formation components, based on formative principles, the components of 'dominant' and 'subordinate' roles were defined as the key components for meaning delivery. Also, it was understood that 'space configuration with overlapped image' and 'space configuration with transparency' were adopted to strengthen conceptional layers. Forth, for space occupation types, there were mostly central hall type, corridor type and passage space type and for open space type, the entire space area was conceptualized, instead of certain object. The circulation line was defined in the frequency order of circular type, pass type and return type. The study on the expressive characteristics of visual narrative in garden design is meaningful as it could build base data for the method of spatial design for visual development of concepts in the future.

Anti-Inflammatory Effects of Volatile Flavor Extract from Herbal Medicinal Prescriptions Including Cnidium officinale Makino and Angelica gigas Nakai (천궁 및 당귀를 함유한 한방처방제 휘발성 향기추출물의 항염증 효과)

  • This study was conducted to develop functional sources of herbal cosmetics for treatment of skin aging and inflammatory disorders using volatile flavor extracts of four different herbal medicinal prescriptions including Cnidium officinale Makino (COM), Angelica gigas Nakai (AGN), Mentha arvense L. (MAL), Artemisiae argyi Folium (AAF), Paeonia lactiflora Pall (PLP), Rehmanniae Radix Preparata (RRP), Scutellaria baicalensis Georgi (SBG), Panax ginseng C.A. Meyer (PGM), Glycyrrhiza uralensis Fisch (GUF). The volatile flavor extracts of four different herbal medicinal prescriptions (HH-1: COM, AGN, PLP, RRP, HH-2: COM, AGN, PLP, RRP, SBG, PGM, GUF, HH-3: COM, AGN, MAL, AAF, HH-4: COM, AGN, MAL, AAF, SBG, PGM, GUF) were extracted using SDE and their antioxidant and anti-inflammatory effects were measured by using DPPH radical and SLO, respectively. As a result, HH-2 showed moderate DPPH radical scavenging activity (68.24 %) and the strongest SLO inhibitory activity (83.96 %) at 100 ${\mu}g$/mL. Moreover, HH-2 of four different prescriptions significantly inhibited NO production on LPS-stimulated RAW 264.7 cells in a dose-dependent manner without considerable cell cytotoxicity at range of 2.0 ~ 50 ${\mu}g$/mL. Additionally, HH-2 also effectively suppressed the production of $PGE_2$ and IL-6, which are responsible for promoting the inflammatory process. Major volatile components of HH-2 were identified as eugenol, paeonol, butyl phthalide, ${\beta}$-eudesmol and butylidene dihydrophthalide by GC-MS analysis. Thus, these results suggest that HH-2 may be useful as a potential source of anti-inflammatory agents in herbal medicinal cosmetics.

Evaluation of Prosthetic Reconstruction in Lower Extremity (하지 골 종양에서 종양 대치물을 이용한 사지 구제술의 평가)

  • Purpose : We evaluated the radiological and functional results of prosthetic reconstruction for locally aggressive benign and malignant tumor in the lower extremity. Materials and Methods : Eighty eight patients were followed up for an average 76 months(22~174). We examined the survival rate of prosthesis, and evaluated the final result by MSTS functional score and ISOLS radiological implants evaluation system. They were statistically analyzed according to the age(<20 year vs. ${\geq}$20 year), fixation methods, amount of bony resection, chemotherapy, local recurrence, and presence of metastasis. Results : The 5 year prosthetic survival rates were 100% in the proximal femur, 83.3% in the distal femur, 81.9% in the proximal tibia. Mean total functional scores were 73.3%, 72%, 68.7%, respectively. In distal femur, the non-chemotherapeutic group was superior in the prosthetic survival rate. Recurrence or metastasis affected the functions in the distal femur and proximal tibia. In the radiological evaluation of the distal femur, older patients over 20 years of age and with cement fixation were superior in bone remodeling(p<0.05). Postoperative infection and radiological loosening were the main causes of the prosthetic failure. Conclusion : The prosthetic reconstruction in the lower extremity led to good clinical and radiological results. Amount of bony resection, chemotherapy, recurrence and metastasis seemed to influence the prosthetic survival, and long-term follow-up will be necessary to investigate more significant prognostic factors.
