Animal Models for the IGF-1 Signal System in Longevity (장수와 관련된 IGF-1 신호 시스템을 연구하기 위한 동물 모델)

  • Longevity is an exciting but difficult subject to study because it is determined by complex processes that require the coordinated action of several genetic factors as well as physiological and environmental influences. Genetic approaches have been applied to animal models to identify the molecular mechanism responsible for longevity. Several experimental model organisms obtained over the last decades suggest that the complete deletion of a single gene by gene targeting has proven to be an invaluable tool for the discovery of the mechanisms underlying longevity. The first discovery of long-lived mutants came from Caenorhabditis elegans research, which identified the insulin/IGF-1 pathway as responsible for longevity in this worm. IGF-1 is a multifunctional polypeptide that has sequence similarity to insulin and is involved in normal growth and development of cells. Several factors in the IGF-1 system have since been studied by gene targeting in the control of longevity in lower species, including nematode and fruit fly. In addition, significant progress has been made using mice models to extend the lifespan by targeted mutations that interfere with growth hormone/IGF-1 and IGF-1 signaling cascades. A recent finding that IGF-1 is involved in aging in mice was achieved by using liver-specific knockout mutant mice, and this clearly demonstrated that the IGF-1 signal pathway can extend the lifespan in both invertebrates and vertebrate models. Although the underlying molecular mechanisms for the control of longevity are not fully understood, it is widely accepted that reduced IGF-1 signaling plays an important role in the control of aging and longevity. Several genes involved in the IGF-1 signaling system are reviewed in relation to longevity in genetically modified mice models.

Physiological Function of Insulin-like Peptides in Insects (곤충 insulin-like peptide의 생리 조절 작용)

  • Insulin and insulin-like growth factor-1 (IGF-1) are hormones that play an important role in the physiological regulation of metabolism, growth, and longevity in vertebrates. Likewise, insulin-like peptides (ILPs), which are structurally similar to insulin and IGF-1, are crucial in insect physiology. In this review, we present an integrated summary of insect ILPs and their receptor signaling, which has been shown to be comparable to insulin and IGF-1 receptor signaling in vertebrates based on genetic studies of Drosophila melanogaster. Additionally, we review the control of ILP synthesis and secretion in the brain in response to nutrition, as well as the ILPs' physiological role in insect metabolism. Moreover, we discuss the contribution of ILPs to growth, development, reproduction, and diapause. Finally, we consider the possibility of targeting ILP receptor signaling in pest management.

Methanol extract of Lespedeza maximowiczii var. tricolor Nakai improves glucose metabolism through PPARγ agonist and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice (삼색싸리 메탄올 추출물의 3T3-L1지방세포와 db/db 마우스에서의 PPARγ 작용제와 인슐린 유사효과를 통한 혈당조절 개선효과)

  • The aim of this study is to investigate the effect of Lespedeza maximowiczii var. tricolor Nakai (LMTN) on glucose metabolism. LMTN extract significantly enhanced the glucose uptake and lipid accumulation in 3T3-L1 adipocytes compared with control. Also, LMTN extract in 3T3-L1 adipocytes significantly increased the protein expression of peroxisome proliferator-activated receptor (PPAR)γ, insulin receptor substrate-1, and glucose transporter (GLUT)4. The regulatory effect on glucose uptake or insulin signal transduction of LMTM extract was lower than troglitazone or pinitol such as the positive control, but increased PPARγ activation. Additionally, LMTM extract has an insulin-mimetic effect. In db/db mice, LMTN extract (250 mg/kg BW) significantly reduced water and food intake, blood glucose, and level of plasma triglyceride and total cholesterol. Furthermore, the expression of PPARã and GLUT4 mRNA in adipose or muscle tissue effectively was increased by oral treatment of LMTN extract. Thus, our results suggest that LMTN extract improves the glucose metabolism through PPARγ and insulin-mimetic effect in 3T3-L1 adipocytes and db/db mice.

Fagopyritol, a Derivative of D-chiro-inositol, Induces GLUT4 Translocation via Actin Filament Remodeling in L6-GLUT4myc Skeletal Muscle Cells (랫드 근육세포에서 fagopyritol이 액틴 필라멘트 구조와 포도당 수송체 4에 미치는 영향)

  • Insulin induces glucose transporter 4 (GLUT4) translocation to the muscle cell surface. As fagopyritol has insulin-like effects, the effects of fagopyritol on GLUT4 translocation and filamentous (F) actin remodeling in L6-GLUT4myc skeletal muscle cells were investigated. Fagopyritol significantly increased plasma membrane GLUT4 levels compared with the basal control in L6-GLUT4myc myoblast cells. Phosphatidylinositol (PI) 3-kinase inhibitor (LY294002) treatment prevented GLUT4 translocation to the plasma membrane in the myoblasts. Fagopyritol treatment apparently stimulates F-actin remodeling in myoblasts. In addition, fagopyritol treatment induced GLUT4 translocation and F-actin remodeling in myotubes. Taken together, these results suggest that fagopyritol promotes GLUT4 translocation and F-actin remodeling by activating the PI 3-kinase-dependent signaling pathway.

브라디키닌 수용체 발현 및 길항제개발에 관한 연구

  • 신장에서 브라디키닌(Bradikinin, BK)의 생리적 역할을 규명하기 위한 첫 단계로 토끼신장 근위세뇨관의 일차배양세포를 이용, BK 수용체를 ($^3$H) BK를 이용하여 수용체결합실험을 함과 아울러 세포배양과정에서 여러 성장인자들의 BK 수용체발현에 미치는 영향을 관찰하였다. 첫째, 토끼 신장의 피질, 수질 및 근 위세뇨관 둥 각 부위에 대한 BK 수용체결합 실험결과 신수질 부분에 가장 많은 BK 수용체가 발견되었으며 이때 해리항수는 0.52 nM, 그리고 최대결합부위는 mg 단배질당 112.5 fmol이었다. 둘째, serum free 배지에서 insulin, transferrin 그리고 hydrocortisone이 성장인자로 사용될 때 BK 수용체발현이 가장 높았으며 이 중 인슐린에 가장 큰 영향을 받았다. 한편 인슐린의 최적농도를 결정하는 실험의 결과 5 $\mu\textrm{g}$/ml매서 가장 높은 수용체발현을 나타내었으며 그 이상에서는 별다른 차이를 보이지 못했다. 세째, 위의 세 성장인자에 prostaglandin E$_1$이나 triiodothyronine을 첨가시 BK 수용체발현은 오히려 저하되었다. 네째, fetal bovine serum (FBS)과 위의 세 성장인자간의 수용체발현능을 비교한 실험에서 세포배양 후 첫 일주일에는 FBS가 세 성장인자보다 약간 나은 수용체발현능을 나타내었으나 그후 이주째에는 세 성장인자가 BK 수용체발현에 더 적절한 요소임을 보여주었다. 이상의 결과로 보아 토끼 근위세뇨관 상피세포에서의 BK 수용체발현은 세포성장인자로 insulin, transferrin, hydrocortisone 중 insulin에 가장 큰 영향을 받는것으로 보이며, 이들 세가지 성장인자는 serum free 배지에서 세포성장 및 기능에 많은 영향을 주는 것으로 생각된다.prolidine이 $K_{M}$ /K$_{H}$ 비가 가장 높았고 diphenidol이 가장 낮았다. 이상의 결과로 보아 항 histamine제의 muscarinic receptor 차단작용은 이들 약물의 항 alleragy 효과에 필요한 작용이 아니며 본 실험에서 추정된 항 histamine제의 H$_1$-receptor와 muscarinic receptor에 대한 상대적 역가는 이들 약물의 선택과 평가에 중요한 지표가 될수 있을 것으로 생각된다.ing ischemic insults. Expression of IGF-1 and Its Receptor Genes in the Oocytes and Preimplantation Embryos in Mouse (생쥐 난자와 착상전 초기배아에서 IGF-1과 IGF-1 수용체 유전자 발현)

  • Insulin-like growth factors (IGF-1 and IGF-2) play an important regulatory role in premplantation embryonic development. To study the role of IGF-1 during premplantation embryonic development in mouse, the presence of mRNA transcripts for IGF-1 and IGF-lR in the oocytes and preimplantation embryos was examined. In this study, the transcripts of IGF-1 was detected in oocytes using primers for IGF-1. The PCR products were identified by Msp I restriction enzyme digest. We revealed that the transcripts of IGF-1 and IGF-1R were presented in the oocytes and preimplantation embryos. The highest mRNA levels in GV stage oocytes were decreased at 4- or 8-cell stage and then reincreased upto blastocyst. The presence of IGF-1 and IGF-lR in GV-oocytes suggests that the transcripts in the early stage embryos were derived from maternal genome. Additionally, the presence of IGF-1 and IGF-lR in the oocytes and preimplantation embryos suggests that IGF-1 plays an autocrine role during preimplantation embryonic development through IGF-lR as a signalling pathway.

LC15-0133, a DPP IV Inhibitor: Efficacy in Various Animal Models (LC15-0133, DPP IV 저해제: 여러 동물 모델에서의 효능)

  • GLP-1-based drugs (GLP-1 analogues and DPP IV inhibitors) and incretin mimetics are currently one of the most exciting classes of agents for type II diabetes. GLP-1, a gut peptide, is an incretin that potentiates glucose-dependent insulin release from the pancreas, slows GI-transit and stimulates the proliferation of beta-cells. DPP IV inhibitors act like incretins by inhibiting DPP IV which inactivates GLP-1. LC15-0133 is a competitive, reversible DPP IV inhibitor ($IC_{50}$ = 24 nM, Ki=0.247 nM) with excellent selectivity over other critical human proteases such as DPP II, DPP 8, elastase, trypsin. and urokinase. LC15-0133 showed long half-life and good bioavailability in rats and dogs. Inhibition of plasma DPP IV activity by LC15-0133 was kept more than 50% 24 hours after oral dosing in rats and dogs at 0.1 mg/kg and 0.02 mg/kg, respectively. The Minimum effective doses of LC15-0133 were 0.01 mg/kg for lowering blood glucose excursion during oral glucose tolerance test and 0.1 mg/kg for increasing glucose-induced GLP-1 response in C57BL/6 mice. Repeat oral administration of LC15-0133 for 1 month delayed the progression to diabetes and reduced HbA1c levels in a dose-dependent manner in Zucker Diabetic Fatty rats. In conclusion, LC15-0133 is a novel, potent, selective and orally active DPP IV inhibitor and showed an excellent blood glucose lowering effects in various animal models.

Effect of Fish Meal Replacement on Insulin-like Growth Factor-I Expression in the Liver and Muscle and Implications for the Growth of Olive Flounder Paralichthys olivaceus (사료의 어분함량대체가 넙치(Paralichthys olivaceus)의 간과 근육 내 인슐린유사성장인자의 발현과 체성장에 미치는 영향)

  • This study examined the effect of insulin-like growth factor (IGF)-I expression in the liver and muscle on the growth of Paralichthys olivaceus fed diets low in fish meal. A feeding experiment was conducted at Jeju National University, Jeju Island, Korea. Groups of P. olivaceus (total initial weight: 200 g) were maintained for 20 weeks on one of five experimental diets containing different proportions of fish meal. Diets containing 0%, 20%, 30%, 40%, and 50% fish meal were labeled FM0, FM20, FM30, FM40, and FM50, respectively. Fish growth was observed every 4 weeks during the feeding experiment, and plasma and liver and muscle tissues were sampled. Plasma IGF-I levels were analyzed using an ELISA kit. The mechanism of IGF-I receptor signaling was examined using immunoblotting and reverse transcription-polymerase chain reaction. The greatest total weight increase was observed in the FM30 group. In parallel, plasma levels of IGF-I and IGF-binding protein were highest in the FM30 group, and mRNA and protein expression were also significantly higher in this group. The first step in the IGF-I signaling pathway, tyrosine-phosphorylation checking, occurred smoothly until 20 weeks. These results suggest that a dietary ratio of 30% fish meal best promotes growth in this species. The IGF-I signaling pathway in the liver and muscle is associated with growth in P. olivaceus.
