고온 열처리가 홍삼물추출물의 이화학적 특성에 미치는 영향 (Effects of High Temperature Heating on the Some Physicochemical Properties of Korean Red Ginseng (Panax ginseng C.A. Meyer) Water Extract)

  • This study was carried out to investigate the some physicochemical properties of Korean red ginseng (Panax ginseng C.A. Meyer) water extract (RGWE) after heated with high temperatures above $100^{\circ}C$ for 2 hours. RGWEs were heated at 100, 110 and $120^{\circ}C$ for 2 hours by using autoclave. After RGWEs were heated at high temperature for 2 hours without not adjustment of pH, the changes of saponin, free sugars, mineral and color in the RGWEs were investigated. Total ginsenoside content in control was 1.99%, while those of RGWE were 1.65, 1.49 and 1.29% when treated at 100, 110 and $120^{\circ}C$, respectively. The contents of total ginsenoside showed decreased tendency as heating temperatures were increased. The ginsenoside-$Rh_{2}$ and $-Rg_{3}$, which have been reported as very stable red ginseng ginsenosides, showed relatively strong spots on TLC when RGWEs were heated at 110 and $120^{\circ}C$. In case of free sugars in RGWEs, fructose, glucose and maltose showed high contents when compared with control, while Fe, Ca and Mg ions showed very low contents. Value of L in RGWE treated with high temperature was almost the same with control, while values of a and b were increased. Values of a were increased from -0.86 of control to +0.04, +0.05 and +1.14 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of b also were increased from 27.68 of control to 33.61, 33.61 and 37.42 when treated with 100, 110 and $120^{\circ}C$, respectively. Values of total color in RGWEs treated with high temperatures, E, were finally increased by values of a and b.

분쇄방법에 따른 고려홍삼분말의 품질특성 (Quality Characteristics of Korean Red Ginsneg Powder on Pulverizing Methods)

  • 홍삼 분쇄의 신 가공 기술로서 비충격(非衝擊)분쇄방식인 cell cracker의 공장 적용 가능성을 제시하고 홍삼분말의 품질고급화에 기초자료로 활용하고자 기존의 hammer mill(충격분쇄방식)과 cell cracker에 의한 분쇄방식으로 홍삼분말을 제조한 후 색도, 밀도 및 관능적 특성을 비교 조사한 결과를 요약하면 홍삼분말의 색도는 cell cracker로 분쇄한 홍삼분말의 경우 색의 밝기를 나타내는 L값은 68.15로 hammer mill로 분쇄한 홍삼분말 72.08 보다 낮았으나 a(적색도) 와 b(황색도)값은 각각 4.21과 5.73, 26.56과 28.36으로 높게 나타났다. 물 추출물의 색도는 시간이 경과함에 따라 두 가지 시료 모두 흡광도가 증가하였으며 전반적으로 hammer mill로 분쇄한 홍삼분말이 cell cracker로 분쇄한 홍삼분말 보다 흡광도가 높게 나타났다. 밀도는 분말의 유동성 특성을 나타내는 이완겉보기밀도, 충격겉보기밀도, 체적감소율은 hammer mill로 분쇄한 홍삼분말보다 cell cracker로 분쇄한 홍삼분말이 모두 높게 나타났다. 미Rm럼각은 쌓임각 보다 낮았으며 이 두 가지 모두 hammer mill로 분쇄한 홍삼분말 보다 cell cracker로 분쇄한 홍삼분말이 낮게 나타났다. 관능검사결과 촉감에 있어서는 두 시료간에 유의 차가 없는 것으로 나타났지만, 홍삼분말 제품의 품질 특성에 큰 영향을 미치는 색도는 cell cracker가 좋게 평가되었으며, 전체적인 선호도에 있어서도 좋게 나타났다. 이상의 연구결과를 종합해볼 때 홍삼분말의 품질 특성에 미치는 요소로서 분쇄방법이 중요한 인자로 작용하고 있음을 알 수 있었으며 홍삼분말의 제조에는 분쇄기 선정이 무엇보다 중요하며 특히 색깔 등 관능적 품질이 강조되는 홍삼분말 제품을 제조할 경우는 기존 충격식 분쇄보다 비충격식 분쇄시스템을 적용하는 것이 품질향상 효과가 클 것으로 생각된다.

항산화활성을 나타내는 약용식물 소재 탐색 (Screening of the Antioxidant Activity of Some Medicinal Plants)

  • 20종 약용식물 물 추출물의 항산화활성 및 이의 폴리페놀화합물과 플라보노이드 함량을 측정 비교하였다. Linoleic acid emulsion을 이용한 total antioxidant 활성에서는 1 mg/mL 농도에서 음양곽, 오미자, 파고지, 당귀, 해동피, 토사자, 감초, 갈근, 오가피, 속단 및 산수유가 대조구에 비해 70%이상의 항산화활성을 나타내었다. 수소이온 라디칼 소거능은 오가피, 음양곽 및 산수유가 60%이상의 활성을 나타내었으며 황기는 활성이 나타나지 않았고 경포부자, 인삼, 삼칠근 및 황정은 5%이하의 매우 낮은 활성을 나타내었다. Superoxide 이온 라디칼 소거능은 수소이온 소거능 보다 활성이 낮아 1 mg/mL 농도에서는 거의 모든 시료에서 활성이 관찰되지 않았으며 5mg/mL 농도에서 음양곽이 약 42%로 가장 높았고 해동피, 오가피, 파고지, 토사자 및 속단이 20%이상의 비교적 높은 활성을 나타내었다. 총 폴리페놀은 옥죽이 약 2.6 mg/g으로 가장 작고 음양곽이 약 81.2 mg/g으로 가장 많았으며, 플라보노이드 함량은 황정이 약 0.4 mg/g으로 가장 적고, 감초가 약 55.4 mg/g으로 가장 많았다 실험에 사용한 식물 대부분에서 폴리페놀의 함량이 플라보노이드보다 많았으며, 시료 추출물의 항산화활성과 폴리페놀 함량을 비교한 결과 각 활성에 대하여 2-3종의 시료를 제외한 나머지에서 모두 폴리페놀 함량과 활성간에 양의 상관관계를 나타내어($r{\fallingdotseq}0.8$) 식물 중에 존재하는 폴리페놀화합물들이 그들이 나타내는 항산화활성과 밀접한 관련이 있음을 확인할 수 있었다. 측정한 세 가지 활성 모두에서 환성이 높고 폴리페놀화합물의 양도 많은 산수유, 오가피, 음양곽 및 해동피의 각 활성에 대한 $IC_{50}$을 측정한 결과 오가피가 세 가지 활성 모두에서 가장 낮은 값을 나타내어 활성이 가장 높았으며 음양곽 또한 superoxide 라디칼 소거능을 제외하고는 오가피와 거의 유사한 활성을 나타내었다. Positive control로 사용한 비타민 C, E, BHA 및 BHT의 경우 BHA의 superoxide 라디칼 소거능을 제외한 나머지는 모두 실험에 사용한 식물들에 비하여 $IC_{50}$ 이유의적으로 낮았다 그러나 total antioxidant 활성에서 오가피, 음양곽, 해동피는 비타민 C보다 유의적으로 높았고 산수유는 유사하였으며, superoxide 라디칼 소거능도 오가피가 BHA에 준하는 활성을 나타내어 이들 식물이 천연 항산화제로 사용이 가능할 것으로 기대되었다.

수삼, 연질 홍삼, 수액 홍삼의 항산화 효과 (Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap)

  • 고려 인삼(Panax ginseng)은 일반적으로 안전성이 우수하고 ginsenosides or panaxosides 같은 많은 생활성적인 물질을 함유하고 있다. 특히 고려 홍삼은 교감 신경계를 안정시키고 인지력을 향상시키고 개인의 의학적 효과를 높이는 데 도움을 줄 수 있다. 새로운 가공 기술로 부드러운 홍삼을 생산하였다. 본 연구는 새로운 가공 기술에 따라 생산된 연질 홍삼이 항산화 효과를 감소시키거나 개선했는지 여부를 조사하는 데 초점을 맞췄다. 연질 홍삼과 시판 홍삼액은 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) 소거활성능에 있어 유의한 차이를 나타내지 않았다(p<0.05). 연질 홍삼은 수삼과 시판 홍삼액에 비해 DPPH (22-Diphenyl-1-pichrylic)와 OH(hydroxyl radical) 소거활성능이 높게 나타났다. OH 소거활성능은 세 그룹(수삼, 연질 홍삼과 시판 홍삼액) 간 유의한 차이를 나타내었다(p<0.05). NO (nitric oxide) 소거활성능 역시 수삼, 연질 홍삼, 홍삼 수액생산품 사이에 유의한 차이가 있었다(p<0.05). 연질 홍삼에서 많은 칼슘 결정체가 전자현미경상에서 관찰되었다. 마그네슘과 칼륨은 연질 홍상과 경질 홍삼 사이에 유의한 차이가 없었다. 연한 홍삼의 추출물은 DPPH와 OH의 존재로 인해 서로 다른 활성산소를 효율적으로 소거할 수 있어 자유 라디칼로 유도된 질환에 도움이 될 수 있다.

약용식물의 Angiotensin Converting Enzyme 저해활성 탐색 (Screening of Inhibitory activities on Angiotensin Converting Enzyme from Medicinal plants)

  • 50가지 식물자원을 대상으로 혈압상승을 주도하는 효소인 angiotensin converting enzyme의 저해활성을 검색하였다. 그결과 추출물농도 1mg/ml에서 50%이상의 높은 저해활성을 보인 식물은 산뽕나무, 머위, 산부추, 돌나물, 꿀풀, 쇠비름, 마가목, 복분자, 속새, 감초, 창포 등 11종으로 나타났다. 또한 1mg/ml농도에서 목단, 곰취, 도인, 길경, 만형자, 참취, 익모초, 산조인, 가래나무, 석곡, 오갈피, 목향 등 12종은 $40{\sim}49%$의 비교적 높은 ACE 억제활성을 보였다. 1mg/ml에서 $30{\sim}39%$의 억제율을 보인 식물은 백부자, 백출, 세신, 당귀, 인삼, 천궁, 치자 등이었으며 현호색, 행인, 단삼, 백작약, 두충 강활 박하, 진피, 방기, 고본, 구기자, 홍화, 포황, 음양곽, 원지등은 $10{\sim}29%$의 낮은 저해활성을 보였다. 앞으로 ACE 억제활성이 높은 식물자원의 유효성분에 대한 물질확인과 동물모델을 이용한 효능검증에 대하여 더 많은 연구가 있어야 할 것으로 사료된다.

보기약물(補氣藥物)인 인삼(人參), 황기(黃?), 백출(白朮), 감초(甘草)의 물 추출액이 생쥐 면역세포의 Cytokine분비에 미치는 영향 (Effect of Qi Tonifying herbs, Ginseng, Astragali, Atractylodis, Glycyrrhizae on Mouse Cytokine Secretion)

  • In Oriental medicine the primordial Qi and the defensive Qi are considered as important for immunity. Therefore it is anticipated that the improvement of the primordial Qi and the defensive Qi can enhance the ability of immune cells. This experiment was conducted to investigate how Ginseng Radix, Astragali Radix, Atractylodis Rhizoma Alba, Glycyrrhizae Radix, representative of Qi tonifying herbs, affect the immune system in terms of controlling and balancing immune cells. Using the MTS assay, increased proliferations were observed from herbal treated cells, among which Gins-eng showed the highest proliferation. When splenocytes were activated with anti-CD3 plus herbal extracts, levels of IFN-g and IL-4 were increased but those of IL-2 showed little change compared with the control cells. Levels of IL-2, IFN-g and IL-4 were increased in purified CD4 T cells when activated with anti-CD3/anti-CD28 but at $100\;{\mu}g/ml$ of Astragali and Atractylodis, levels of IL-2 were decreased by 11% and 42%, respectively and those of IFN-g were decreased by 55% and 12%, respectively. Under Th1/Th2 polarizing conditions, levels of IFN-g in Th1 cells treated with herbal extracts were all decreased but when it comes to IL-4, its levels were increased in Ginseng and Glycyrrhizae treated cells but decreased in Astragali and Atractylodis treated cells. Taken together, the data show that compared with other qi tonifying herbs, Ginseng and Glycyrrhizae have a tendency to favor Th2 cell differentiation in vitro.

가토 대동맥 평활근에서 인삼 알콜 추출물에 의한 Calcium 동원에 관한 연구 (A Study on the Mobilization of Calcium by Ginseng Alcohol Extract in Rabbit Vascular Smooth Muscle)

  • There have been conflicting reports concerning the effect of Panax ginseng on the contractility of vascular smooth muscle, i.e., Panax ginseng extract has been reported to cause relaxation, contraction or to have no effect on the tension of vascular smooth muscle. A further investigation of $Ca^{++}$ stores which supply $Ca^{++}$ for contraction of vascular smooth muscle is needed to understand the underlying mechanisms of this conflicting effect of ginseng alcohol extract (GAE). The present study was intended to examine the sources of calcium mobilized for contraction of vascular smooth muscle by GAE. Aortic ring preparations were made from the rabbit thoracic aorta and endothelial cells were removed from the ring. The contractility of the aortic ring was measured under various experimental conditions and $Ca^{++}$ flux across the membrane of aortic ring and the sarcoplasmic reticulum and mitochondria were measured with a calcium selective electrode. The result were summarized as follows; 1) At low concentration of extracellular $Ca^{++}$, GAE increased the contractility of vascular smooth muscle in dose-dependent fashion except high concentration $Ca^{++}$ (1 mM). 2) In the presence of ryanodine, GAE still increased contractility of vascular smooth muscle as much as control group, but in the presence of caffeine, GAE increased it significantly. i.e. Their effects seemed to be additive. 3) In the presence of verapamil+lanthanum, and verapamil+lanthanum+ryanodine, the contractility of the vascular smooth muscle was decreased, but a dose dependent increase in vascular tension was still demonstrated by GAE although total tension was low. 4) GAE increased $Ca^{++}$ efflux from vascular smooth muscle cells, but have no effect on $Ca^{++}$ influx. 5) GAE increased $Ca^{++}$ efflux from sarcoplasmic reticulum and mitochondria vesicles. From the above results, it may be concluded that GAE increased the release of $Ca^{++}$ from sarcoplasmic reticulum, mitochondria or other intracellular $Ca^{++}$ stores of vascular smooth muscle, but it does not increase $Ca^{++}$ influx across the plasma membrane.

인삼 품종별 뿌리 추출물의 NMDA 수용체 길항 효과 및 진세노사이드 함량 (N-Methyl-D-Aspartate (NMDA) Receptor Antagonistic Effect and Ginsenoside Content of Panax ginseng C. A. Meyer Cultivar Root Extracts)

  • Background: Although a number of Panax ginseng cultivars have been developed by Korean researchers in recent years, there has been insufficient analysis of their beneficial properties. In this study, we sought to identify useful ginseng varieties as functional materials. Methods and Results: We evaluated effects of root extracts of 10 ginseng cultivars (Cheongsun; CS, Chunpoong; CP, Gopoong; GP, Gumpoong; GMP, K1, Sunhyang; SH, Sunone; SO, Sunpoong; SP, Sunun; SU and Yunpoong; YP) against the inhibitory effects of nitric oxide (NO) and reactive oxygen species (ROS) production in mouse brain microglial BV2 cells, as well as the binding of N-methyl-D-aspartate receptor (NMDAR), a marker related to memory. Ginsenosides, such as 20 (S)-protopanaxadiols (PPDs), including ginsenoside-Rb1, -Rb2, -Rb3, -Rc, -Rd, and - Rg3 and 20 (S)-protopanaxatriols (PPTs) including -Re, -Rg1, and -Rg2 were analyzed by HPLC. We observed that the cultivar GMP showed the highest inhibitory effect (60.8%) against NO production at 20 ㎍/㎖. Those cultivars showing the significantly highest inhibition effects against ROS at 20 ㎍/㎖ were K1 (57.3%), SP (54.5%), YP (53.1%), CP (51.7%), CS (50.9%) and SH (49.6%). At 50 ㎍/㎖, K1 showed the most potent inhibitory effect (51.2%) on NMDAR binding. The total phenol content of SH (1.89 mg/g) and K1 (1.73 mg/g) were higher than those of the other cultivars, whereas in terms of PD/PT ratios, the values of CP (0.98), K1 (1.05) and SO (1.05) were lower than those of the other cultivars. On the basis of correlation coefficient (0.7064) between NMDAR inhibition and ONOO- scavenging activity. Conclusions: The findings of this study indicate that the cultivars K1 and SH could be useful ginseng resources as functional materials with favorable cognition-improving and antioxidative properties.

인삼 추출물에 의한 Cylindrocarpon destructans의 주화성 반응 연구 (Chemotactic Response Study of Cylindrocarpon destructans towards Ginseng Root Exudates)

  • Background: Cylindrocarpon destructans (Zins) Scholten is an important pathogenic fungus that causes ginseng root rot in many ginseng growing areas in China. Although C. destructans have been studied worldwide, research on its chemotaxis towards ginseng (Panax ginseng C. A. Meyer) root exudates in the rhizosphere remains limited. Methods and Results: In this study, we collected ginseng root exudates with three different polarities from three-year-old ginseng roots, and performed chemotaxis and spore germination assays to investigate the ability of these exudates to induce the response in C. destructans. The results showed that, compared with other conditions, when C. destructans cultivated at $20^{\circ}C$ and a pH of 6 exhibited a strong positive chemotactic response toward $2mg/{\ell}$ aqueous phase, $20mg/{\ell}$ butanol phase, and $0.2mg/{\ell}$ petroleum ether from ginseng root exudates, the chemotactic moving indexes were 0.1581, 0.1638 and 0.1441, respectively. In addition, the spore germination rate with optimal chemotactic parameters were 48%, 53%, and 41% in the aqueous phase, butanol phase and petroleum ether groups, respectiviely, which were significantly higher than that in the control group (23%) (p < 0.05). The mycelial growth rate with optimal chemotactic parameters increased with culture time, and the maximum growth rates in the aqueous phase, butanol phase and petroleum ether groups were 0.425, 0.406 and 0.364 respectively, on the 4th day. The optimal chemotactic parameters were $39.73mg/50mg/{\ell}$, $48.93mg/50mg/{\ell}$, and $31.43mg/50mg/{\ell}$, in aqueous phase, butanol phase and petroleum ether respectively, from ginseng root exudates, compared with $5.5mg/50mg/{\ell}$, in the control group. Conclusions: The present study revealed that certain ginseng root exudates containing chemical attractants act as nutritional sources or signals for C. destructans and support its colonization of ginseng roots.

인삼, 산양산삼 및 홍삼 추출물의 항비만 효과 (Anti-obesity effects of cultivated ginseng, -wild simulated ginseng and -red ginseng extracts)

  • This study was to investigate whether cultivated ginseng (CG), cultivated wild simulated ginseng (CWG) and cultivated red ginseng (CRG) extracts influences on the obesity. The saponin contents of 3 kinds of ginsengs were analysed by HPLC-ESI-TOF-MS. Total saponin contents were determined in CG on the most contents and since red ginseng has the highest PD (protopanaxadiol type) / PT (protopanaxatriol type) ratio, there may be differences between ginseng, wild ginseng, and red ginseng with respect to their pharmacological effects. Male C57BL/6J mice were fed a normal diet(N), HFD (60% Kcal fat, C), HFD with CG, CWG and CRG extracts (800 mg/kg) for 5 weeks. We observed change of total body weight, degree of hepatic lipid accumulation and immunohistochemical change of GLP-1 and insulin-secreting cells. Also this study attempts to use the physiological analysis method to analyze the changes of blood lipids, insulin and leptin concentration. The change of body weight and size of accumulated lipid droplets in liver lobules decreased in all of the experimental groups than the control(C) group. In the pancreas, the immunohistochemical density of insulin-secreting cells were significantly stronger in the CWG and CRG than C group. The levels of serum insulin and leptin significantly decreased 55.6%, 54.3% respectively in CWG and CRG. The changes of triglyceride, total cholesterol in serum decreased in CRG than the C group. Obesity related CG, CWG and CRG extracts might have contribute to improvement of obesity by regulating the levels of blood lipids and biochemical indicator of fat accumulation.
