Cells first emerged 4 billion years ago and have evolved over a long period into an excellent system. Bottom-up synthetic biology is a research field that aims to develop "artificial cells" by returning to 4 billion years ago and redesigning cells from scratch. Although these artificial cells are not perfect, they are artificial cell mimicry systems that possess important characteristics of living cells. By designing the artificial cells, researchers in this field aim to explore the organization and the origins of cells from a different approach than traditional cell biology and ultimately seek to replace the use of living cells. This review aims to introduce the concepts and recent research in capsule and biocatalyst-based artificial cells, which have been actively studied recently.
Efficient production of antigen specific cytotoxic T cells is critical for appropriate adoptive immune response. In vitro culture and expansion of human T lymphocyte clones are very sophisticated and subtle procedure in immune cell therapy and hard to control. Therefore, many groups devoted their efforts to manipulate artificial antigen presenting cells (aAPCs) that can induce T cell activation and clonal expansion. To mimicking of natural antigen-presenting cells, aAPCs encompass basic signal molecules required for T cell activation: MHC:antigen complexes, co-stimulatory molecules and soluble immune modulating molecules. Orchestrated organization of these molecules is important for efficient T cell activation. Here, we discuss how those molecules have been incorporated in several aAPC models, but also how physical properties od aAPC are important for interaction with T cells.
Bakcground: To treat anastomosis site stenosis and occlusion of the artificial vessels used in vascular surgery, tissue-engineered artificial vessels using autologous cells have been constructed. We developed artificial vessels using a polymer scaffold and autologous bone marrow cells and performed an in vivo evaluation. Material and Method: We manufactured a vascular scaffold using biodegradable PLCL (poly lactide-co-${\varepsilon}$-caprolactone) and PGA (poly glycolic acid) fibers. Then we seeded autologous bone marrow cells onto the scaffold. After implantation of the artificial vessel into the abdominal aorta, we performed an angiography 3 weeks after surgery. After the dogs were euthanized we retrieved the artificial vessels and performed histological analysis. Result: Among the six dogs, 2 dogs died of massive bleeding due to a crack in the vascular scaffold 10 days after the operation. The remaining four dogs lived for 3 weeks after the operation. In these dogs. the angiography revealed no stenosis or occlusion at 3 weeks after the operation. Gross examination revealed small thrombi on the inner surface of the vessels and the histological analysis showed three layers of vessel structure similar to the native vessel. Immunohistochemical analysis demonstrated regeneration of the endothelial and smooth muscle cell layers. Conclusion: A tissue engineered vascular graft was manufactured using a polymer scaffold and autologous bone marrow cells that had a structure similar to that of the native artery. Further research is needed to determine how to accommodate the aortic pressure.
당쇄구조는 유전자에 따라 엄밀하게 규정되어 있지 않기 때문에 다양성과 유연성을 갖는 당쇄 구조는 생체기능의 안정성과 항상성을 실현하기 위해 불가피한 요소라고 생각된다 .앞으로 당쇄기능을 해명하기 위해서는 세포간, 세포내외간, 세포내의 생체정보전달기구 등에 관여하고 있는 당쇄를 떼어내어 해석하는 연구를 추진할 필요가 있다. 또 생체에 있어서 당쇄에 관한 연구로부터 얻어진 지식과 기술을 근거로 하여 인공적으로 당쇄를 설계하고 구축하여, 그것들을 단백질이나 지질에 삽입시켜 당쇄의 기능을 밝히고, 아울러 인공의 당쇄를 이용한 새로운 생체기능의 제어기술에 관한 연구도 추진해야 할 것이다.
인공장기 등 다양한 세포관련 연구를 수행할 때 요구되는 세포공배양에서 세포를 2D/3D 패터닝하는 기술은 장기를 모사하기 위한 필수적인 공정 중의 하나로 간주된다. 그러므로 이 글에서는 가장 간단하게 세포 및 표면을 이용하여 세포를 패턴화하고, 공배양할 수 있는 방법들에 대하여 설명하고자 한다.

Recently, three-dimensional scaffolds and nanofibers are being developed for bone tissue regeneration. In this study, we fabricated a hybrid scaffold using a nano-micro precision deposition system. The fabrication process involved the application of the solid freeform fabrication (SFF) technology and electrospinning. The hybrid scaffolds were combined using micro scaffolds and nanofibers. The nanofibers were deposited on each layer of the micro scaffolding using the electrospinning process. The micro scaffolds were fabricated using the SFF technology at a temperature of $100^{\circ}C$, pressure of 650 kPa, and scan velocity of 250 mm/s. Nanofiber fabrication was conducted by means of electrospinning using the flow rate, solution concentration, distance from the tip to the collector (TCD), and voltage. The nanofibers were fabricated using a flow rate of 0.1 ml/min, voltage of 5 kV, TCD of 1 mm, and 10 wt% of solution concentration. MG-63 cells were seeded into the hybrid scaffold for the purpose of its evaluation.

생체 내의 모든 기능은 유전자 발현에 의해 결정된다. 유전자 발현은 않은 인자들에 의해 조절되며, 이러한 조절 과정에 따라 유전자 발현량이 결정되는 것이다. 세포 주기 역시 유전자 발현과 밀접한 연관성을 가지고 있다. 본 논문에서는 효모에서 세포 주기의 각 단계와 관련된 유전자들의 분석을 통해서 세포주기를 조절하는데 있어서 중요한 역할을 수행하는 전사 조절 모티프들이 무엇인지를 찾아보았다. 주요 모티프의 추출은 인공신경망 모델을 학습하고. 입출력 에러 분석을 통하여 이루어진다. 그 결과 MCB 등 기존의 실험 결과를 통하여 세포주기에 관련이 있다고 알려진 모티프들이 높은 점수를 보인다는 것을 알 수 있었고. 그 외에 세포주기의 각 단계에서 유전자 발현에 중요한 역할을 수행할 것으로 예상되는 다른 모티프들도 예측해볼 수 있었다.

차세대 염기서열 분석 기술은 성능과 비용 면에서 매우 향상되어 한 개체 내 여러 세포의 유전자 분석이 가능한 수준이다. 한 개체 내 여러 조직 세포의 유전자는 모두 동일하지 않기 때문에 여러 조직 세포의 Lineage 를 계층적으로 표현하고 이를 조직 세포 간 변이 정도를 파악하는 데 활용한다면 암 돌연변이 발생 등을 미리 예측할 수 있다. 본 논문은 한 개체 내 여러 조직 간 변이를 관찰하기 위해 변이 검출 데이터를 계층적 군집 방법을 이용해 분석하고 이를 시각화 하는 방법을 제안한다. 실제의 8 개 조직 세포의 유전자를 분석하고 변이를 검출하여 Dendrogram 그래프로 시각화 하였다.

In tissue engineering, a scaffold is a three-dimensional(3D) structure that serves as a template for regeneration the functions of damaged tissues or organs. Among materials for scaffolds, polycaprolactone(PCL) and ${\beta}$-tricalcium phosphate(${\beta}$-TCP) are biodegradable and biocompatible. In this study, we fabricated 3D PCL, blended PCL (60 wt %)/${\beta}$-TCP (40 wt %), and pure ${\beta}$-TCP scaffolds by a multi-head scaffold fabrication system. Scaffolds with a pore size of $600{\pm}20{\mu}m$ was observed by scanning electron microscopy. The effects of 3D PCL, blended PCL (60 wt %)/${\beta}$-TCP (40 wt %) and pure ${\beta}$-TCP scaffolds were analyzed by evaluating their mechanical characteristics. In addition, in an in-vitro study using osteoblast-like saos-2 cells, we confirmed the effects of 3D scaffolds on cellular behaviors such as cell adhesion and proliferation. In summary, the 3D blended PCL (60 wt %)/${\beta}$-TCP (40 wt %) scaffold was found to be suitable for human cancellous bone in terms of its the compressive strength, biocompatibility, and osteoconductivity. Thus, blending PCL and ${\beta}$-TCP could be a promising approach for fabricating 3D scaffolds for effective bone regeneration.

최근 선진 각국에서는 시각장애인을 위한 인공 망막 모델구현을 위한 연구가 매우 활발히 진행 중이다. 인공 망막의 여러 기법중 시각피질자극에 의한 방법은 망막손상과 시신경 손상 환자에게 적용할 수 있도록 시피질을 자극한다. 그러나 이 방법은 시각자극전달의 중간단계를 생략하고 직접 뇌세포를 자극하는 것이다. 따라서 본 논문에서는 시각피질자극방법에 기반하여 인간시각처리와 유사하게 영상의 압축방법을 부과함으로써 세포의 특성을 고려한 모델을 나타낸다. 실험 데이터로는 자동차 번호판 숫자들이며, 인식기는 SOM을 사용하였다. 실험결과, 제안된 인식모델과 일반적 인식모델과의 차이가 없음을 알 수 있었다.
