Plant regeneration system via somatic embryogenesis in leaf explant cultures of Gentiana scabra var. buergeri has been established. Leaf segments formed calli when cultured on MS medium supplemented with 0.5 mg/L 2,4-D and 2 mg/L BAP After transferred to SH medium supplemented with 0.5 mg/L 2,4-D, 2 mg/L CPA and 0.5 mg/L kinetin, the callus became embryogenic. The embryogenic callus was subcultured every 3 to 4 weeks. Upon transfer onto SH basal medium the embryogenic callus gave rise to numerous somatic embryos, which subsequently developed into plantlets. The regenerated plants were potted in an artificial soil with mixture (peatmoss : pearlite : vermiculite : 2 : 1 : 1) and transplanted to the soil after kept under a high humidity for two weeks. A total of 78 plants out of 105 regenerated plants survived in the soil. Phenotypic variations in height, number of stems and the flowering time were observed in tile regenerated plants. Cytogenetical analyses showed no chromosomal variation.
Efficient plant regeneration via shoot organogenesis from in vitro cultured leaf segments of chrysanthemum (Dendranthema grandiflora Tzvelev cv. Namjeon) was achieved. Adventitious shoot formation from leaf explants was greatly influenced by plant growth regulator, leaf age, light condition, explant number per culture vessel, and explant orientation. Leaf segments, obtained from fully expanded young 1-2nd leaves and inoculated 8 explants per petri-dish with adaxial surface contact with MS medium supplemented with 0.5 mg/L BA and 2.0 mg/L NAA, produced 100% regeneration frequency and 13.7 shoots per explant. Regenerated adventitious shoots were successfully rooted in MS medium with 0.1 mg/L NAA. The plantlets were acclimatized in artificial soil mixtures (Vermiculite:Perlite=1:1), and transferred to greenhouse for flowering. The regenerated plants showed normal phenotypes.
Mature nodal segments of 2-year-old greenhouse stock plant were cultured on Murashige and Skoog basal medium supplemented with the different kinds and various concentrations of cytokinins to produce multiple shoots in in vitro condition. The most adventitious shoots were produced from excised ends of nodal segments. The highest average number $(24.6\;{\pm}\;4.6)$ of shoots was produced with the combination of BA 1.0mg/L and TDZ 0.1mg/L in MS medium. In addition, several shoots were formed from lenticels of bark cambium with the same treatment. These concentrations promoted high shooting capability upto $94.6\%$ and NAA was the best cytokinin among five different PGR sources.

느타리 버섯의 다양한 품종과 재배형태가 급변히 요구되는 시기에 충남농업기술원 버섯연구팀의 5년 (1996~2,000)에 걸쳐 국,내외 수집 20 균주를 공시하여 개발 육성한 청풍과 명월 신품종 느타리버섯을 현재 재배되고 있는 기존 느타리품종과 동일여부를 위해 PCR-RAPD분석과 배양적, 재배적 특성을 인공재배 시험한 결과를 요약하면 다음과 같다. 1. 청풍, 명월느타리버섯 균사배 양 온도는 청 풍은 20~$25^{\circ}C$,명 월은 25~3$0^{\circ}C$, 이고 배지는 PDA배지가 양호하고 균사생육은 청풍, 명월 모두 비슷하였으며 배지 pH는 6.5~7.5 였다. 2. 버섯재배가능은 청풍은, 5~26$^{\circ}C$ 명 월은, 7~28$^{\circ}C$로 온도범위가 넓어 재배가 용이하나 적정 생육온도는 청풍, 명월 공히 15~19$^{\circ}C$가 양호 하였다 3. 청풍과 명월의 배양온도 23$^{\circ}C$에서 균사배양기간 23~24일 (850cc/pp),폐면 균상 재배에서 균배양 기간은 청풍, 명월 공히 11~12일, 초발이 소요일수는 2~3일 소요 되었다. 4.초발이시 청풍과 명월 갓 색택은 진청색이고, 생육후기 청풍은 회갈색, 명월은 진회색으로 조직이 치밀하고 저장성이 양호하여 경제성이 기대된다. 5. 다발수는 춘추2호 (13개/다발) 대 비 청 풍11개, 명월12개로 약간 적었으나 개체중은 춘추2호(14g/개) 대비 청풍 16g/개, 명월 14g/개로 비슷한 경향 이었다. 6. 수량성은 균상 폐면재배시 춘추2호 (40kg/3.3$m^2$)대비 청풍 느타리버섯은 (46kg/3.3$m^2$) 15%증수되고, 명월느타리버섯은(41kg/3.3$m^2$)로 3% 증수되었으며 병재배 겸용도 가능하였다. 7. PDA대치배양과 PCR-RAPD시험에서 청풍, 명월 신품종느타리 버섯은 기존 재배 되고 있는 느타리품종과 다른 품종으로 확인 되었다.
Armillaria isolates (KNU-A110, KNU-A234, KNU-A1022 and KNU-A1030) were excellent isolates for producing Gastrodia tuber in farm cultivation. Depth of soil between $10\;cm{\sim}19\;cm$ was favorable for producing Gastrodia tuber and rhizomorph at cultivated area. Eighty nine isolates were collected from 5 countries; 16 from Japan, 22 from USA, 26 from France, 4 from Africa and 21 from Korea. Mycelial fan of most isolates were better formed on basal medium with lemon extract than without lemon.A. mellea (KNU-A997) were strongly pathogenic to Gastrodia but A. gallica (KNU-A110) were excellent symbiotic to Gastrodia tuber. Mycelial growth were good on basal medium containing $0.25%{\sim}0.5%$ ethanol and sawdust spawn added wheat bran and corncob. Liquid culture inoculation were not only fast growth of mycelium but also reduction of contamination.
Effective micropropagation system via somatic embryognesis was established for a Phytophthora resistant Aralia elata cultivar. Different kinds of growth regulators were needed to induce embryogenic callus with different explant sources. When leaf explants were used, a combination of 2,4-D, TDZ and L-glutamine was needed, whereas when petiole and root explants needed only 1.0 mg/L 2,4-D. Embryogenic callus induction rate under the optimum culture condition was 75.0%, 67.0% and 83.0% from leaf, petiole and root segment, respectively. Somatic embryo germination and plantlet conversion rate appeared to be influenced greatly by various osmoticums. More than 90% of embryos germinated when treated with sucrose, glucose and maltose. However, the highest conversion rate (72%) was recorded on medium with 2% sucrose only. The converted plantlets grew normally on 1/2MS basal medium, were acclimatized on artificial soil mixture and survived more than 95% in the greenhouse condition. The results suggest that the species can be clonally propagated through in vitro culture system via somatic embryogenesis.
Some factors effecting on the mycelial growth and primordia formation of Agrocybe cylindracea were investigated. Among supplements added into sawdust, wheat bran was the best for the mycelial growth, and its appropriate concentration was 30%. A variety of sawdust preparations was tested singly and in combinations. Oak sawdust and the combination of oak and larch sawdust were the most effective for the mycelial growth. The optimal moisture content was 65%, and the range of optimal temperature was $25\:{\sim}\:28^{\circ}C$. The favorable conditions for the primordia formation appeared to be similar to those of the mycelial growth. Among supplements added into sawdust, wheat bran 30% and the combination of wheat bran and rice bran 30% were the best for the primordia formation. Oak sawdust, and the oak plus larch sawdust and oak plus poplar sawdust were the best for the primordia formation. The optimal temperature for the primordia formation was $17{\pm}2^{\circ}C$.
This study was conducted to evaluate the growth characteristics of lettuce (Lactuca sativa L.) as affected by artificial light sources and different growing media in a closed-type plant production system (CPPS). The lettuce seeds were sown in the 128-cell plug tray filled with 5 different growing media such as urethane sponge (US), rock-wool (RW), Q-plug (QP), TP-S2 (TP) and PU-7B (PU). The germination rate of lettuce seeds was examined during 12 days after sowing. On the 13 days after sowing, the lettuce seedlings were transplanted in a CPPS with temperature $25{\pm}1^{\circ}C$ and nutrient solution (EC $2.0dS{\cdot}m^{-1}$, pH 6.5) using recirculating deep floating technique system. The light sources were set with FL (fluorescent lamps) and combined RB LEDs (red : blue = 7 : 3) with $150{\pm}10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD and a photoperiod of 14/10 hours (light/dark). The initial germination rate of lettuce was the highest in TP. The final germination and mean daily germination were the significantly highest in RW, QP and TP. The plant height, leaf length, leaf width, leaf area, and fresh and dry weights of shoot were the greatest in QP irradiated with RB LED. The number of leaves, fresh and dry weights of root and SPAD were the greatest in QP and TP irradiated with RB LED. The root length was the longest in TP irradiated with RB LED. Therefore, these results indicate that RB LED was effective for the growth of lettuce and it was also found that the QP and TP were effective for the germination and growth of lettuce in a CPPS. In addition, we confirmed the applicability of the newly developed growing medium TP for the lettuce production in a CPPS.

Bulbophyllum auricomum Lindl. is a rare orchid and has flowers with an attractive fragrance. The present study investigated the tissue culture method for micropropagation. Capsules derived from artificial self-pollination were obtained for the best seed germination in MS basal medium. Plant growth regulators (1.0 mg·L-1 of BAP and 2.0 mg·L-1 of NAA) were affected by callus induction from subcultured pseudobulb explants. For the callus subculture, different natural plant extracts were tested in 11 treatment media. Among them, MS medium with 150 mL·L-1 of coconut water was generally effective in fresh weight (1.75 ± 0.08) and (3.01 ± 0.20) of callus proliferation and PLBs induction at 1 and 2 months, respectively, followed by an MS combination of 30 g·L-1 of banana and 20 g·L-1 of potato extract. The results of a comparative study of different MS mediums containing plant growth regulators with a natural extract combination and MS medium supplemented with natural extract only showed that MS medium supplemented with a combination of natural extracts (150 mL·L-1 of coconut water) and plant growth regulators (2.0 mg·L-1 of BAP and 1.0 mg·L-1 of NAA) obtained the highest shoot regeneration (3.37 ± 0.17) and (6.41 ± 0.68) after 1 month and 2 months of culturing, respectively.
To establish the optimum conditions of in vitro plant regeneration, the leaf, rhizome, and root explants of Belamcanda chinensis were cultured on the MS medium supplemented with different concentration of 2,4-D and BA. The callus induction was more effective in the root explants than the leaf and rhizome explants, and was the best on MS medium containing 3.0 mg/L 2,4-D or 1.0 mg/L 2,4-D and 3.0 mg/L BA. The highest numbers of shoots were regenerated when callus were cultured on MS medium containing 3.0 mg/L 2,4-D for 4 weeks. However, the multiple shoots were induced on MS medium supplemented with the combination of 2,4-D and BA. The normal root formation from shoot was effective on the MS medium lacking any plant growth regulators. For acclimatization, the regenerated plantlets were cultured on MS medium without sucrose and plant growth regulators for 2 weeks, and then transferred to the pot.
