Recovery of Aglycone of Daidzein and Genistein in Soybeans (콩에 포함된 비배당체 다이드제인과 제니스테인의 회수)

  • Using aqueous solution of 60% ethanol, the isoflavones contained in the Soybean (Chungsun, Korea), Tawon (Suwon, Korea), Jackbean (Hongsung, Korea), Soybean (Jilin, China), Black Soybean (Jilin, China), Kidneybean (Jilin, China) and Soybean (California, U.S.A.) were extracted. In this work, the pretreatment step was composed of agitation extraction, filtration, concentration, ultracentrifuge, and membrane filtration. The analysis by $C_{18}$ column was performed, and the mobile phase applied was linearly changed with A/B of 85/15-65/35 vol% for 50 min (A water/acetic acid, 99.9/0.1 vol%, B acetonitrile/acetic acid, 99.9/0.1 vol%). Among the soybeans tested, the total amounts of the four isoflavones (daidzin, genistin, daidzein, and genistein) extracted by the soybeans from Korea, China and U.S.A. were 1.37, 1.60, and 2.25 mg/g, respectively. It was also found that the total amount of aglycone of daidzein and genistein from soybean (American California) was $144{\mu}g/g$, which was the largest for the soybeans experimented.

Development of a functional yogurt fortified with ubiquinone, isoflavone, and γ-aminobutyric acid (유비퀴논, 이소플라본, γ-aminobutyric acid가 강화된 기능성 요구르트 개발)

  • A potentially functional yogurt co-fermented with Monascus-fermented soybean powder (MFSP) was prepared, and its quality and antioxidant properties were investigated. Skim milk powder with (SMP+MFSP, 1:1, w/w) or without MFSP (SMP; control) was fermented by probiotic cultures consisting of L. delbrueckii subsp. bulgaricus KCTC 3635 and S. thermophilus KCTC 5092. The functional yogurt fermented with MFSP contained significantly (p<0.05) higher levels of ${\gamma}-aminobutryric$ acid (GABA; $107.22{\pm}3.06{\mu}g/g$), isoflavone aglycone (daidzein+genistein; $201.21{\pm}6.29{\mu}g/g$), and ubiquinone ($39.05{\pm}0.08{\mu}g/g$) than the control yogurt. During fermentation at $36^{\circ}C$ for 48 h, the functional yogurt displayed higher titratable acidity, viable cell numbers, and radical scavenging activity and a lower pH than the control yogurt (p<0.05). These results indicate that MFSP has great potential for enriching the free isoflavones, GABA, and ubiquinone contents in yogurt.

Analysis of Secondary Metabolites in Various Cultivars of Soybean (Glycine max (L.) Merr.) (다양한 콩 자원들의 이차대사물질 함량 분석)

  • Soybean (Glycine max (L.) Merr.) is a high-protein oilseed crop, cultivated worldwide. Soybean seeds are abundant in various secondary metabolites with physiologically active. Mature seeds of 25 soybean cultivars with various morphological and physiological characteristics analyzed for secondary metabolites, such as carotenoid, isoflavone, and soyasaponin. These secondary metabolites showed various content by genotype in 25 cultivars. Total carotenoid content ranged from 1.23 to 33.78 mg/g and three cultivars, such as IT177645, PI90763 and IT234975 with black seed coat showed high levels of total carotenoid. Total isoflavones content ranged from 20.28 to 276.35 mg/100g and were detected high levels in Savoy, PI90763 and KLG16001. In addition, total soyasaponins content ranged from 33.12 to 246 mg/100g and were detected high levels in PI90763, PI86490 and IT234975. The PI90763 was showed abundant content in all of the carotenoid, isoflavones and soyasaponins. These results could be valuable information for the development of new soybean cultivars and regulation of secondary metabolites biosynthesis in soybean.

이소플라본의 첨가가 난소절제쥐에서 골밀도 및 골함량에 미치는 영향

  • 콩단백질에 풍부한 isoflavones는 약한 estrogenic한 효과를 가진다고 보고 되면서 폐경 여성이나 난소절제쥐를 대상으로 골 대사에 미치는 효과에 대한 연구가 많이 이루어졌다. 그러나 isoflavones가 골대사에 미치는 효과는 isoflavones의 량, 섭취기간, 생리상태, 칼슘의 섭취량 등 여러 가지의 변수에 따라 다르게 나타났다. 일반적으로 isoflavones의 섭취가 너무 작거나 너무 많아도 그 효과 미미하여 isoflavones의 적당량에 대한 연구가 계속 요망되고 있다.(중략)

  • 대두에 함유되어 있는 생리활성 성분에 대한 과학적 규명에 뒤이어 몇 년 전 부터 대두와 인체생리와 관련한 여러 임상 데이터들과 대사시험에 관한 연구가 최근 집중적으로 수행되고 있다. 대두의 이소플라본은 특히 여성의 유방암, 남성의 전립선 암 등 성호르몬 관련 질환에 탁월한 효과가 있고 폐경과 관련된 호르몬 치료요법의 대체 수단으로 활발한 연구가 진행되고 있다. 그중 본문에서는 식이요인이 질병예방에 매우 중요한 인자로 작용하는 분야인 체내 지질대사 개선 및 유방암 예방과 관련된 최근 연구내용을소개한다.

Inhibitory Effects of Transglycoslyation Products of Soy Isoflavones on Cholesterol Biosynthesis (대두 이소플라본 당전이 반응 산물의 콜레스테롤 생합성 저해 효과)

  • Hydroxy-3-methylglutaryl-CoA reductase (HMG-CoA reductase) is the rate-limiting enzyme in biosynthesis of cholesterol in animals. In this study, inhibitory effects of isoflavone glycosides on HMG-CoA reductase were investigated. At sample concentration of $100{\mu}M$, genistein-7-O-triglucoside (G2-genistin) inhibited HMG-CoA reductase activity by approximately 18%, whereas daidzein-7-O-triglucoside had no inhibitory effect. In the kinetic experiments with Syrian hamster HMG-CoA reductase, G2-genistin showed inhibitory efficacy with an invariable $V_{max}$ value, suggesting that G2-genistin works as a competitive inhibitor of HMG-CoA reductase and has potential for hypocholesterolemic action through direct regulation of HMG-CoA reductase.
