Characteristics of Interspecific Protoplast Fusant from Lactobacillus acidiophilus 88 and Lactobacillus bulgaricus IFO 13953 (Lactobacillus acidophilus 88과 Lactobacillus bulgaricus IFO 13953간의 세포융합주의 특성에 관한 연구)

  • 유산균의 균주개량방법의 일환으로 photoplast fu-sion 기법과 electrofusion법을 이용하여 protease 활성, lipase 활성, 내열성, 내산성 등이 우수한 L. bulgaricus와 bacteriocin을 생산하는 L. acidophilus 간의 융합시켜 얻은 융합주들의 생리학적 성질을 검토하였다. 산 생성능, 내열성, 내산성 protease, lipase 활성 등 L. bulgaricus가 L. acidophilus보다 우수하였다. L. bulgaricus는 lactose와 sorbos를 이용하였으나. mal-tose 와 dorbitol을 이용하지 못하는 반면 , L. acidophilus는 matose를 이용하고 lactose 와 sorbose를 이용하지 못하였다. 융합주 가운데서는 367810번이 모균주의 발효능 특성을 함께 지님으로서 재조합체임을 확인 할 수 있었으며, sorbitol의 경우 모균주에서는 발효능이 전혀 나타나지 않았음에도 불구하고 융합주 4, 7번이 발효능을 나타내어 융합과정에서 새로운 형질을 획득하기도 한다는 사실을 알 수 있었다. Lactase 활성은 모균주 모두 높은 $\beta-galactosidase$ 활성을 보였으나, $phospho-\beta-galactosidase$ 활성은 거의 없었으며 융합주들도 다소 차이는 있었지만 모균주와 유사한 효소활성을 나타내었다. 발효에 있어서 key enzyme으로 작용하는 protease. lipase 등의 효소활성도 모균주의 활성보다 우수한 융합주도 존재하였다.

  • The activity of glucoamylase and pullulanase, properties of glucoamylase and ethanol productivities of fusants were studied. Glucoamylase and pullulanase activity of fusants were higher than parents. The optimal pH and temperature of glucoamylase of fusants were very similar to the those produced by S. diastaticus. In alcohol fermentation. fermenting ability and fermentation rate of fusants were higher and faster than either of its parental strain. The maximum of alcohol yield in 15% of liquefied potato starch was 7.8% (v/v)

  • To increase the bioavailability of Opuntia humifusa, O. humifusa was fermented using L. plantarum, and the characteristics of the fermented extract were confirmed. The characteristics of fermented extracts were investigated through viscosity, total polyphenol content, amino acid, antibacterial activity, cytokine analysis and zymography. The viscosity decreased by half and vitamin C remained almost unchanged during fermentation. Total polyphenols, most amino acids, total amino acids and major minerals were increased by fermentation. There was no cytotoxicity, and the activity of cytokines and MMP-9 in inflammation was inhibited. The inhibitory effect on inflammation and tissue destruction was found to be inhibited in L. plantarum fermentation extracts compared to hot water extracts. These results are expected to be the basis for the development of materials that enhance the functionality of L. plantarum-fermented O. humifusa extract.

  • Glucoamylase and ethanol productivities of HSDD-170 and HSDM-119 formed by S. cerevisiae and S. diastaticus protoplast fusion were investigated. For the production of the glucoamylase, soluble starch as carbon source, yeast extract and C. S. L as nitrogen source added into the basal medium were favorable. The production of the enzyme reached at maximum after cultivation of the fusant for 4 days at 3$0^{\circ}C$, aerobically. The properties of glucoamylase produced by fusants were very similar to those produced by S. diastaticus as based on optimum temperature, pH stability. In alcohol fermentation from starch, strain HSDD-170 fermented starch faster than either of its parental strains. The maximum of alcohol yield in 15% of liquefied potato starch was 7.5% (v/v).

Skin lightening effect of fermented Panax ginseng extract (자연삼 발효 추출물의 미백 활성에 대한 연구)

  • Panax ginseng is known for various pharmacological activities mainly due to saponins. Since minor saponins, generated by the decomposition of major saponins, generally exert higher activities than major saponins, the fermentation may increase the minor saponin contents in ginseng products. In this study, we tested fermented ginseng extract whether or not provide a safe cosmetic ingredient for whitening purpose. In this regard, fermented Ginseng extract was prepared and evaluated the inhibitory activity toward tyrosinase and the melanin synthesis suppression. The safety was tested via cell viability and toxicity test. The skin lightening effect was also evaluated by clinical study. The fermented Ginseng extract exerted higher activities in tyrosine inhibition and in suppressing melanin synthesis compared to Kojic acid and arbutin. In the clinical test, skin lightening effecte of the sample was clearly higher than vehicle or Vitamin C. We thus concluded that the fermented Ginseng extract may provide a safe cosmetic ingredient for skin lightening purpose.

Enhancement of Immune Activities of Fermented Morinda citrifolia L. (Noni) and Six Marker Compounds (노니 지표성분 6종과 발효노니의 면역활성 증진 효과)

  • This study will evaluate the effect of fermented Morinda citrifolia L. extracts and its marker compounds to provide baseline data for utilizing Morinda citrifolia L. as functional health products. Morinda citrifolia L. and six marker compounds were processed on RAW 246.7 macrophage to test for XTT Cytotoxicity, measure Nitric Oxide and Cyokine formation, and analyze the expression of immune marker genes. Furthermore, LPS and fermented red ginseng extract, a common functional ingredient, are used as positive controls. Our results showed that fermented Morinda citrifolia L and six bioactive compounds did not have any cytotoxic effect in all treatment concentrations and groups. Among six bioactive compounds, SCP and ASE confirmed the formation of NO. In addition, the ASE treatment group showed increased formation of IL-6 and IL-1β and the expression of iNOS and TNF-α. Also, fermented Morinda citrifolia L extract activated the macrophage by enhancing the production of nitric oxide (NO), interleukin (IL)-6, and IL-1β, and the expression of COX2 compared to Morinda citrifolia L. extracts. The result of the study showed that Fermented Morinda citrifolia L. (Noni) and marker compound enhance the innate immunity activity and suggested that the bioactive compound could be applied as a marker compound. Thus, Fermented Morinda citrifolia L. (Noni) could be used as functional food material to develop immunity-enhancing products, and highly functional marker compounds can be utilized as the effective components.

Preparation of Korean Traditional Alcoholic Beverage (Yakju) by a Protoplast Fusion Yeast Strain Utilizing Starch and its Quality Characteristics (전분분해 효모융합체를 이용한 전통 발효주의 제조와 품질특성)

  • The objectives of this study were to evaluate the physico-chemical and sensory characteristics of a Korean traditional alcoholic beverage (yakju) prepared using different nuruk (Korean-style koji) concentrations and yeasts such as the fusant FA776 and Saccharomyces cerevisiae KOY-1, respectively. The fusant FA776, which has alcohol-fermenting and starch-utilizing properties, was formed by Saccharomyces cerevisiae KOY-1 and Saccharomyces diastaticus KCTC1804. The fermentation trial was conducted in a 5 L lab-scale jar at $25^{\circ}C$. The maximum alcohol production of the K-100 and F-50 reached levels of 135.0 mg/mL and 119.4 mg/mL, respectively. The pH values were in a range of 4.3-4.5. Total acidity was in a range of 0.47-0.60%. Organic acids and amino acids were analyzed in order to evaluate variations in its composition and content via HPLC analysis. Organic acids including lactic acid, citric acid, malic acid, and pyruvic acid, and 16 kinds of amino acids, including aspartic acid, were detected in all treatments. K-100 showed the highest amino acid contents, whereas F-50 exhibited the lowest amino acid contents. Volatile flavor components such as phenylethyl alcohol, isoamyl alcohol, 2-methylthiophane, isobutyl alcohol, and ethyl succinate were detected as a major component in all treatments, as determined via gas chromatography. The results of our sensory evaluation demonstrated that Yakju fermented by the FA776 fusant yielded more favorable results than S. cerevisiae KOY-1.

  • L-Lysine 생성균인 Corynebacterium glutamicum의 동종간 융합주 RS 99를 담체인 Alginate와 여기에 TiCl$_4$로부터 제조된 Titanium hydroxide를 혼합하여 각각 고정하고 이들의 Gel strength, 활성도 및 회분식 발효조건을 비교하였다. 그 결과 Alginate-Titanium hydroxide를 담체로 선정하여 고정화 C, glutamicum 융합주의 재사용성 및 안정성을 검토하였으며, Continuous-Flow Stirred-Tank Reactor를 구성하여 L-Lysine 의 연속발효를 시도하였다.

  • This study was to demonstrate the anti-inflammation effect using extracts derived from fusion-fermentation to add the function of Araliae Continentalis Radix (fACR). Since there are limitations to the use of ACR, available strains were selected through fermentation using lactobacillus strains, and the increases in total amount of polyphenols and amino acids was confirmed through comparison with Hot Water Extract of ACR. To determine the anti-inflammatory effect of the fACR was measure cytokine inflammation associated with arthritis, the arthritis when cartilage destruction is accompanied mainly MMP-9 activity was confirmed to evaluate the inhibition effect. These results show that fermentation using lactobacillus increases major biological activities and inflammatory response-restraining effects of ACR. This study is expected to be a basis for studying the preventive effect of fACR on arthritis.

Manufacturing and Quality Characteristics of Puffed Black Bean Fermented by Lactobacillus plantarum Strains Isolated from Kimchi (김치 유래 Lactobacillus plantarum을 이용한 팽화 검은콩 발효물의 제조 및 품질특성)

  • The purpose of this study was to optimize the fermentation condition of black bean by lactic acid bacteria (LAB) and to evaluate the quality characteristics of fermented black bean. Lactobacillus plantarum SU22 isolated from kimchi was selected as a starter for the fermentation of black bean because the strain exhibited strong antimicrobial activity against pathogenic bacteria and did not produce biogenic amines or a carcinogenic enzyme, β-glucuronidase. Fermentation was performed with broth containing puffed black bean (PBB) inoculated with 1% (v/v) of L. plantarum SU22 at 37℃ for 48h. The viable cell count of LAB was over 9 Log CFU/mL in PBB (20%) broth fermented with L. plantarum SU22. Fermentation of alcalase-treated PBB (20%) broth with L. plantarum SU22 was found to be the optimal condition, increasing viable cell count of LAB up to 10.30 Log CFU/mL. Under the optimal condition, the total polyphenol content (94.02 mg GAE/g) and DPPH radical scavenging activity (92.50%) were significantly increased, compared to non-fermented control (87.74 mg GAE/g, 83.14%).
