Abstract
Glucoamylase and ethanol productivities of HSDD-170 and HSDM-119 formed by S. cerevisiae and S. diastaticus protoplast fusion were investigated. For the production of the glucoamylase, soluble starch as carbon source, yeast extract and C. S. L as nitrogen source added into the basal medium were favorable. The production of the enzyme reached at maximum after cultivation of the fusant for 4 days at 3
세포융합에 의한 새로운 알코올 발효 효모 개발을 목적으로, S. cerevisiae와 extracellular glucoamylase를 분비하는 S. diastaticus 간의 세포융합을 통해 HSDD-170과 HSDM-119 두 균주를 선별하여 이들 융합체의 특징에 대해서는 제 1보에 발표한 바 있다. 본 보에서는 parent cell인 S. diastaticus와 fusant가 분비하는 amylase의 성질을 비교 조사하였으며, 전분을 이용한 알코올 발효에 있어서의 조건을 검토하였다. Amylase생성 및 발효에 대한 탄소원으로는 dextrin과 가용성 전분에 의해 효소생성이 유도됨을 알 수 있었으며 질소원으로는 무기질소원 보다 유기 질소원인 yeast extract와 C.S.L.가 좋았다. 이들 amylase의 성질은 pH 5.5, 반응온도 55
