Effect of Seed Parents on Varietal Performance in Strawberry (딸기 교잡육종 시 품종적 성능에 미치는 자방친의 효과)

  • In order to select excellent cross parents in strawberry breeding, 11 varieties were self-pollinated and tested their progenies. Among the 11 varieties, 'Akihime', 'Maehyang' and 'Seolhyang' showed superior in plant vigor, disease resistance and yield. Combining ability was tested using 15 progenies obtained from crosses between 3 selected varieties as seed parents and 5 varieties as pollen parents. Plant vigor was the strongest when 'Akihime' was used as a seed parent while ripening time tended to the fastest when 'Seolhyang' as a seed parent. Average fruit weight of the crosses 'Seolhyang' ${\times}$ 'Tochiotome', 'Seolhyang' ${\times}$ 'Geumhyang', 'Seolhyang' ${\times}$ 'Wongyo 3111' and 'Akihime' ${\times}$ 'Wongyo 3111' were above 18 g for marketable fruits. Fruit hardness was superior at crosses of 'Maehyang' as a seed parents to other crosses combinations, especially 'Maehyang' ${\times}$ 'Tochiotome'. Soluble solid contents (SSC) was not significantly different among cross combinations. The best cross combination for marketable yield was 'Akihime' ${\times}$ 'Wongyo 3111', followed by 'Seolhyang' ${\times}$ 'Geumhyang', 'Maehyang' ${\times}$ 'Geumhyang', 'Akihime' ${\times}$ 'Tochiotome', in order.

  • Purpose: Lymph node metastasis is a very important prognostic factor for all skin cancers and some sarcomas. A sentinel lymph node (SLN) biopsy is the most useful technique for identifying SLNs. Recently, a new generation of diagnostic tools, such as single photon emission computed tomography/computed tomography (SPECT/CT) and positron emission tomography/CT (PET/CT) enabled the detection of SLNs. This study compared the efficacy of PET/CT for detecting lymph node metastases with a SLN biopsy in a single medical center. Materials and Methods: From 2008 to 2018, 72 skin cancers of sarcoma patients diagnosed with some lymph node involvement in a whole body PET/CT reading were assessed. Patients suspected of lymph node metastasis were sent to biopsy and those suspected to be reactive lesions were observed. The analysis was performed retrospectively using the medical records, clinical information, PET/CT readings, and pathology results. Results: The age of patients ranged from 14 to 88 years and the mean follow-up period was 2.4 years. Twenty-two patients were suspected of a lymph node metastasis and confirmed. The sensitivity, specificity, positive predictive value and negative predictive value of PET/CT images in sarcoma and non-sarcoma tumors were increased significantly when the expert's findings were considered together. Conclusion: PET/CT is effective in detecting lymph node metastases.

  • Breeding for pear resistance to pear psylla (Cacopsylla pyricola) is one of important objective of the National Institute of Horticultural and Herbal Science breeding program. One hundred thirty three accessions from 15 Asian, Chinese and European pear species were investigated for their resistance against pear psylla. The pear psylla resistance was determined based on the following four characteristics: overwintering adult population, the number of eggs and nymphs, and the degree of soot. The different pear species showed varied resistance to pear psylla. Pyrus calleryana and P. betulaefolia indicated the highest antixenosis as ovipositional preference and antibiosis as nymphal feeding and were the most resistant genetic resources. Likewise the European pears (P. communis), 'Conference' and 'Cascade', exhibited little occurrence and damage by pear psylla. These were proved to be promising genetic materials for breeding resistant cultivars because they had good fruit quality and showed resistance to pear paylla. The observed population of overwintering adult, the number of eggs and nymphs of psylla had significant correlation each other.

  • Twenty and 24 lines selected from Korean local cultivar, Kalmi and Punggak, respectively, were grown in the University experiment farm to select promising lines for recurrent parent in a backcross breeding program for incorporation of resistance to Phytophthora capsici and Xanthomonas campestris pv vesicatoria into the Korean local cultivars. All the selections of 'Kalmi' and 'Punggak' were relatively late in maturity and the range of the variation was wider in the selections of 'Kalmi' than in those of 'Punggak'. Many of the selections from 'Kalmi' were vigorous, high in sugar content, and tolerant to virus in the field. Many of the selections from 'Punggak' were bearing large fruits with thick pericarp and high milling percentage. Line 6, 9, 14, 18 of 'Kalmi' and line 14, 16, 18 of 'Punggak' were selected as promising lines for recurrent parent in the backcross program.

  • Korean wheat varieties were tested to evaluate quality characters and the degree of deterioration due to simulated rainfall treatment. The result indicated that old varieties were generally superior in milling, flour color and dough properties to new varieties. And the quality properties of Korean new varieties were not exceeded those of Japanese varieties.

The Test of Combining Ability and Heterosis on the Silkworm(Bombyx mori) Breeding (누에 견.사질에 관한 잡종강세 및 조합능력검정)

  • The study was conducted to obtain the genetic information on heterosis and combining ability of the quantitative characters for F1 hybrid breeding in silkworms. Six parental varieties and each set of 30 diallel crosses in F1's were used as materials, and bred on the randomized complete block design with three replications. Fourteen characters were observed with the twenty samples in each tray. The data were analyzed for (1) heterosis and combining ability in F1 hybrid. The heterosis in the weight and the length of cocoon showed positively high at 24.51%, and 23.4%, respectively and the weight of the whole cocoon as well as the weight of the whole cocoon layer showed a siginificant heterosis ranging from 15.56% to 15.71% and from 17.14% to 19.01%, but the fifth and the total instar period showed negative heterosis. It was found that the combination between, C70XRomogua and N9 X Romogua showed highly a negative heterosis on the fifth instar period and for the cocoon weight. The female of N9+Sansuian and the male of Romogua X Sansurian have a high heterosis effect, on the cocoon shell weight, and Sansurian X Romogua(reciprocal) on the length and the weight of cocoon filament with no regard to sexuality. The significant maternal and cytoplasmic effect on heterosis of the cocoon weight and the cocoon shell weight were observed with the combinations, N9 X C5, N63 X C70 and on the length of the cocoon filament with the combinations, Sansurian X N63, Sansurian X C5, Sansurian X C70 and N9 X C70, N63 X C70 on the weight of cocoon filament. As mean squared of GCA, SCA and RCA were significant with these combining ability for all characters resulted from additive and non-additive altogether and there is a significant difference between reciprocals. Sansurian showed a negative GCA effect on the fifth and total larval duration, but the higher positive GCA effects took places with varieties N9 and C5 on the length, width, weight of cocoon, cocoon shell weight, percentage of cocoon shell weight, length and weight of cocoon filament, percentage of raw-silk with no regard to both generations and silkworm sexuality. The values of SCA between the cross combinations varied generation-wise and sex-wise. It was shown that SCA value for the fifth instar period was highly negative for Sansurian X C70, Romogua X C70, Sansurian X C5, Romogua X C5, but it was positive effect on the cocoon weight, cocoon shell weight with N9 X C5, and C70 X Sansurian, on the length of cocoon filament with N9 X C5, Romogua X Sansurian on the weight of cocoon filament between Romogua and N63 and on the percentage of raw-silk between the combination of Sansurian X Romoga.

  • Seedlings and grafts of Betula pendula and Betula platyphylla var. japonica were grown under partially controlled environments in a greenhouse and in a plastic-greenhouse installed inside a laboratory. Plant growth conditions such as photoperiod, temperature, nutrient supply were partially controlled to enhance the vegetative and reproductive growth of the birch seedlings and grafts. By the treatments twenty and seventy one percents of the seedlings, respectively, for the Betula pendula and Betula platyphylla var. japonica developed visible floral organs between 250 to 508 days after seeding. By the same treatments eighty and fifty three percents of the grafts, respectively, for Betula pendula and Betula platyphylla var. japonica developed visible male catkins between 51 to 497 days and female catkins between 365 to 396 days after grafting. Breeding cycle of birch species can be reduced to a great extent by the induction of precocious flowering at early stages of seedling and graft development.

Potential Allelic Association of Microsatellite Markers on Bovine Chromosome 5 with Carcass Traits in Hanwoo (Korean cattle) (Microsatellite 의 대립유전자 빈도를 이용한 한우의 경제형질과의 연관성 규명)

  • A total of 10 polymorphic microsatellite markers on bovine chromosome 5 were used for allelic association tests with phenotypic characteristics in Hanwoo. The data analyzed in this study were collected from 326 steers. Chi-square tests were performed to compare the frequencies of individual alleles between the high and the low breeding value groups. The following breeding values were analyzed for QTL effects. The frequency of allele 239 of DIK2828 showed a significant difference between the high and the low breeding value groups in the breeding value of marbling score (MSBV). The allele 279 of BMC1009 was found to show significant differences in allelic distribution for the breeding value of cold carcass weight (CWBV) and the breeding value of backfat thickness (BFBV) and allele 285 showed significant differences in allelic distribution for CWBV, BFBV, and MSBV. The allele 200 of DIK4329 showed significant differences in allelic distributions for the breeding values of longissimus muscle area (LMABV) and BFBV. In this study, we identified the QTL for carcass traits at around 20 (DIK2828), 41 (BMC1009) and 95 (DIK4329) cM in chromosome 5. The results provided a useful reference for further positional candidate gene research and marker-assisted selection for fat metabolism and carcass traits.

Floral Characteristics and Cross Compatibility of Collected Paeonia lactiflora PALL (작약(芍藥) 수집종(蒐集種)의 화기특성(花器特性)과 교잡친화성(交雜親和性))

  • In order to obtain the basic information on cross breeding of peony, the growth and floral characteristics were investigasted for local collected strains in main cultvation area in Korea. Local peony strains were classified by early emergence lines and late emergence lines according to the emergence date. Flower color of single-petaling flower was varied with red, pink and white, on the other hand, red+red, pink+white, pink+red and pink+pink in double-petaling flower. Number of pistil of the most local peony strains was 3-4/flower, number of stamen was varied from 0 to more than 200 ea/flower, classifing by 4 groups according to the diameter of flower. Most of the cross combinations showed the cross comptibility, and number of follicle was 3-5 ea/flower, average number of seed was 15.6 ea/follicle, seed size was grouped : by small grains with less than 10g, medium grains with 10-15g and large grains with more than 15g of 100 seed weight.

Effective Screening Methods for Lipoxygenase Isozymes in Soybean Seeds (콩 lipoxygenase 효소의 효율적인 검정법)

  • Normal soybean seed contains three lipoxygenase isozymes called L-1, L-2, and L-3, respectively, which are responsible for the generation of undesirable grassy-beany flavors. Simple and effective methods for the detection of lipoxygenase isozymes were developed in soybean seeds. Sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) has been tried in separating these isozymes. It was done effectively on 7.5% separating gel and 4.5% stacking gel. However, no reliable method has been developed specifically for separating L-3, L-13 and L-23. Visual judging methods were based on the bleaching activities of lipoxygenase in contact with methylene blue and ${\beta}$-carotene. Sodium linoleate bleaching method was adopted to determine L-1 and L-2. Carotene bleaching and spectrophotometric methods were used to determine L-3. These systems were very rapid within one minute, furthermore only required a small piece of cotyledon (below 10 mg) and the other part could be used for generation advance after analysis. It was demonstrated that 200 seed samples could be analyzed per day by one laboratory assistant. The combination of visual judging methods and electrophoresis is suitable for breeding programs. It took 6.5 hours for analysis of 100 seed samples by one person.
